Introduction: Fungi are essential to the aquatic food web, nutrient cycling, energy flow, and ecosystem regulation. Fungal community structures in water can be influenced by adjacent terrestrial environments, which drive and control some ecosystem services they provide. However, the roles of freshwater fungal communities remain underexplored compared to bacterial communities in this context. Methods: We assessed the impact of anthropological and environmental factors on freshwater mycobiota in an agriculturally dominated water basin in eastern Ontario, Canada. We undertook bi-weekly surface water sampling from 2016 to 2021 and conducted fungal internal transcribed spacer 2 (ITS2) metabarcoding on the samples, complemented by ancillary data, including water physicochemical properties, upstream land use, hydrology, and weather conditions. Results: Our study yielded 6,571 OTUs from 503 water samples, spanning 15 fungal phyla, dominated by Ascomycota, Basidiomycota, and Chytridiomycota. Agricultural land use was associated with decreased mycobiota alpha diversity and distinct fungal communities were observed at agricultural drainage ditch and mixed-land use sites compared to a forested site that had minimal anthropogenic activities in catchment. Notably, river discharge emerged as a predominant influencer of both community diversity and composition, likely amplified by precipitation-induced erosion and drainage from adjacent terrestrial environments. Discussion: Water physicochemical properties, including stream fungicide levels, explained a small proportion of the variation in mycobiota communities, underscoring the significance of unmeasured factors, alongside stochastic community assembly processes. Nevertheless, stream mycobiota demonstrated functional resilience for critical ecological processes under different environmental conditions. Altogether, these results highlight the complex interplay of factors influencing the freshwater mycobiota, which is essential for elevated understanding of the ecosystem services these fungi provide. Introduction: Agricultural drainage ditches are essential for maintaining flow efficiency and are often managed by brushing (i.e., vegetation clearing along ditch banks) and dredging. These maintenance practices not only impact flow but also the ditches' capacity to process and assimilate agricultural chemicals. However, the influence of ditch management on microbial activities, essential in processing and transforming these contaminants, remains underexplored. Methods: We conducted a four-year surface water sampling campaign in two long-established (over 40 years) agricultural drainage ditches situated in eastern Ontario, Canada. One drainage ditch underwent intensive bank brushing and channel dredging, while the other remained unmanaged. We assessed the impact of these contrasting ditch management practices on the diversity and compositional structure of surface water microbiome communities, with a particular focus on the distribution of genes associated with nitrogen (N) cycling, using shotgun metagenomics sequencing and an assembly-based metagenome approach. Results and discussion: From 117 surface water samples, we reconstructed 157 metagenome-assembled genomes (MAGs), predominantly from the Patescibacteria superphylum. Brushing alone, conducted about 10 months prior to dredging, had an insignificant impact on the aquatic microbial community. However, dredging led to a significant reduction in community diversity and abundance of genes affiliated with denitrification, dissimilatory nitrate reduction to ammonia, and complete nitrification through comammox. Ditch management resulted in noticeable shifts in the microbial community, evidenced by the enrichment of taxa in Polaromonas, Emticicia, Flectobacillus, and Patescibacteria in the water of the managed ditch sites. The metagenomes of these taxa harbor genes involved in various nitrogen biotransformation pathways. Interestingly, following dredging, the levels of ammonia and ammonium, nitrite, and total Kjeldahl N in the managed ditch did not increase, but significantly decreased; likely due to ditch substrate removal and potentially increased flow efficiency and dilution. Being one of the few studies conducted to date, this study provided unique insights into the consequences of drainage ditch management on freshwater microbiomes involved in N cycling. Controlled tile drainage (CTD) can benefit the environment and crop production. However, CTD has the potential to increase soil greenhouse gas (GHG: CO, CH, NO) emissions by increasing soil water contents and elevating field water levels. A paired-field (CTD and uncontrolled tile drainage (UTD)) approach was used to compare soil GHG emissions for silt loam corn, soybean, and forage fields under CTD and UTD management in eastern Ontario, Canada during a drier and a wetter growing season. A total of five field pairs were examined. Soil GHG emissions directly over tile drains (OT) and between tile drains (BT) in the CTD fields were also assessed. Average soil GHG emissions did not significantly differ ( p > 0.05) for CTD and UTD field pairs, except for CO emissions (greater emissions from UTD fields) among two field pairs studied (forage in the drier growing season and soybean in the wetter growing season), and NO emissions from a soybean field pair in the wet growing season (greater emissions from CTD field). Significantly higher soil water contents in the UTD forage field may have augmented CO fluxes there. There were some significantly higher NO (in the wetter growing season) and CO emissions (in both growing seasons) BT relative to OT locations in some fields; but these differences were not translated significantly to other BT and OT site comparisons. The wetter growing season examined resulted in greater average daily soil CO fluxes overall, but similar CH and NO fluxes for soybean fields compared to soybean fields in the drier growing season. Overall, there were no spatially or temporally systematic differences in GHG emissions among CTD and UTD field pairs, or among BT and OT locations in CTD fields. [ABSTRACT FROM AUTHOR]

Acinetobacter calcoaceticus-baumannii (ACB) complex has been identified as a group of emerging opportunistic pathogens that cause nosocomial infections. The current study investigates the prevalence, distribution, and diversity of pathogenic ACB complex in various aquatic systems with different uses. Of the total 157 agricultural, raw drinking water intake, recreational beach, and wastewater treatment plant (WWTP) effluent samples, acinetobacters were isolated, quantified, and confirmed by genus- and ACB complex-specific PCR assays. Of all agricultural surface water samples, A. calcoaceticus (65%) was more frequently detected than A. pittii (14%), A. nosocomialis (9%), and A. baumannii (3%). In WWTP effluent samples, A. baumannii was more prevalent in de-chlorinated (60%) samples compared to both A. pittii and A. nosocomialis (40%). Interestingly , A. nosocomialis (43%), A. calcoaceticus (29%), and A. baumannii (14%) were detected in raw drinking water intake samples, whereas A. pittii (50%) and A. nosocomialis (25%) were detected in beach samples. Although no sampling location-specific differences were recorded, significant ( P < 0.05) seasonal differences were observed when agricultural surface water samples collected in spring were compared with the summer and fall. Whereas effluent chlorination significantly impacted the degree of prevalence of Acinetobacter in WWTP effluent samples, overall, the prevalence of ACB complex in all sampling locations and seasons indicates that these water sources, containing human-associated ACB complex, may pose potential health risks as community-acquired opportunistic infections.IMPORTANCE Acinetobacter calcoaceticus-baumannii (ACB) complex is a group of organisms known to cause problematic nosocomial opportunistic infections. A member of the species complex, A. baumannii , is becoming a global threat to infection treatment as strains are increasingly develop resistance to antibiotics. The prevalence and distribution of potentially pathogenic Acinetobacter calcoaceticus-baumannii complex species remain poorly understood, and there is a need to better understand the occurrence of A. baumannii in non-nosocomial environments. Our research details the spatial-temporal distribution of ACB complex species in a regional watershed and highlights the presence of ACB complex in wastewater effluent that is discharged into a river.
