4. The histidine phosphocarrier protein, HPr, binds to the highly thermostable regulator of sigma D protein, Rsd, and its isolated helical fragments

The phosphotransferase system (PTS) controls the preferential use of sugars in bacteria and it is also involved in other processes, such as chemotaxis. It is formed by a protein cascade in which the first two proteins are general (namely, EI and HPr) and the others are sugar-specific permeases. The Rsd protein binds specifically to the RNA polymerase (RNAP) sigma(70) factor. We first characterized the conformational stability of Escherichia colt Rsd. And second, we delineated the binding regions of Streptomyces coelicolor, HPrsc, and E. colt Rsd, by using fragments derived from each protein. To that end, we used several biophysical probes, namely, fluorescence, CD, NMR, ITC and BLI. Rsd had a free energy of unfolding of 15 kcal mol(-1) at 25 degrees C, and a thermal denaturation midpoint of 103 degrees C at pH 6.5. The affinity between Rsd and HPrsc was 2 mu M. Interestingly enough, the isolated helical peptides, comprising the third (RsdH3) and fourth (RsdH4) Rsd helices, also interacted with HPrsc in a specific manner, and with affinities similar to that of the whole Rsd. Moreover, the isolated peptide of HPrsc, HPr9-30, comprising the active site, His15, also was bound to intact Rsd with similar affinity. Therefore, binding between Rsd and HPrsc was modulated by the two helices H3 and H4 of Rsd, and the regions around the active site of HPrsc. This implies that specific fragments of Rsd and HPrsc can be used to interfere with other protein-protein interactions (PPIs) of each other protein.

The sequential action of glutamine synthetase (GS) and glutamate synthase (GOGAT) in cyanobacteria allows the incorporation of ammonium into carbon skeletons. In the cyanobacterium Synechocystis sp. PCC 6803, the activity of GS is modulated by the interaction with proteins, which include a 65-residue-long intrinsically disordered protein (IDP), the inactivating factor IF7. This interaction is regulated by the presence of charged residues in both IF7 and GS. To understand how charged amino acids can affect the binding of an IDP with its target and to provide clues on electrostatic interactions in disordered states of proteins, we measured the pK(a) values of all IF7 acidic groups (Glu32, Glu36, Glu38, Asp40, Asp58, and Ser65, the backbone C-terminus) at 100 mM NaCl concentration, by using NMR spectroscopy. We also obtained solution structures of IF7 through molecular dynamics simulation, validated them on the basis of previous experiments, and used them to obtain theoretical estimates of the pKa values. Titration values for the two Asp and three Glu residues of IF7 were similar to those reported for random-coil models, suggesting the lack of electrostatic interactions around these residues. Furthermore, our results suggest the presence of helical structure at the N-terminus of the protein and of conformational changes at acidic pH values. The overall experimental and in silico findings suggest that local interactions and conformational equilibria do not play a role in determining the electrostatic features of the acidic residues of IF7.

Lo scopo del lavoro è quello di proporre una metodologia di disinchiostrazione applicabile a carta da macero a elevato contenuto di giornale flessografico, secondo il tradizionale processo alcalino a flottazione opportunamente adeguato. Gli elementi salienti del metodo adottato riguardano laggiunta di agenti chimici innovativi per un incremento del grado di distacco, dispersione ed asportazione degli inchiostri e laggiunta di un ridotto lavaggio a spruzzo della pasta disinchiostrata nella fase di addensamento. Una fase di chiarificazione con limpiego di additivi a base di zinco consente inoltre il completo riciclo delle acque. Il processo di disinchiostrazione proposto, realizzato in laboratorio, con una miscela di giornali offset (30%), riviste patinate (30%) e giornali flessografici (40%) in circuito chiuso, a temperatura di 40°C ha consentito di ottenere una pasta disinchiostrata con grado di bianco di circa 60 e resa dell80%.

- Abstract
The main goal of this article is to assess the impact of product innovation on the economic performance of firms operating in Medium and High-Tech (M&HT) industries. Using information from a large and unique dataset on Italian firms we estimate, by means of propensity Score (PS) matching methods, a positive and significant ‘innovation premium’ both in terms of profitability and growth (in the short-run) for those firms who introduced new innovative products. We also find that this innovation premium is particularly large for small firms and even more so when considering new established firms.

A new synthetic reagent, 5-methyl-2-mercaptobenzoxazole (MMBO), is a highly selective and powerful collector for Cu sulphides in the presence of other sulphide minerals, such as galena and sphalerite. Laboratory and pilot tests of it on several complex ores have indicated higher grades and recoveries than those obtained with xanthates. FTIR studies of its reaction with pure chalcocite have indicated the nature of the surface phase. Adsorption isotherms obtained by the rest method have revealed the quantitative evolution of surface products, permitting the determination of surface coverage at various reagent concentrations. The electrokinetic potential has been used to verify the stability of individual species in diverse concentration ranges. FTIR studies in relation to Hallimond flotation tests have led to an interpretation of the mechanism of the collecting action of oleic acid, dodecylamine acetate and di-2-ethyl-hexyl phosphoric acid (D2EHPA), on rutile at different pH. Oleic acid and D2EHPA act as collectors at acid pH; molecular forms are precipitated on the mineral surface as surfactant aggregates. - Abstract
The initial growth of Cu on polycrystalline manganese at room temperature has been investigated by Auger Spectroscopy and UV-synchrotron radiation photoemission spectroscopy, for copper coverages ranging from 1 to 18 Å. By monitoring the changes of the Cu3dlevel, a strong intermixing at the interface has been observed. The simultaneous growth of a copper phase which is lying on top of the intermixed region has also been detected. Analysis of the Auger data indicates that copper is most probably growing in a layer-by-layer mode, with a simultaneous diffusion into the substrate. Calculations for a layer-by-layer growth model are compared with the experimental data. They support our analysis.

Vacuum techniques include: photoemission spectroscopy (XPS, UPS and ESCA); auger electron spectroscopy (AES); secondary ion mass spectrometry (SIMS); extended x-ray absorption fine structure (EXAFS); near edge x-ray absorption fine structure (NEXAFS); and electron energy loss spectroscopy (EELS). Fundamental aspects and potential applications are reviewed and compared.
