Abstract

Background. COVID-19, whose etiologic agent is SARS-CoV-2, an RNA virus, is characterized by a high mutation rate. Therefore, while more subjects are infected, greater probability that the virus will potentially undergo changes that confer evolutionary advantages (immune response evasion, increased virulence, and reduced vaccination efficacy). Efforts to acquire herd immunity through vaccination may be compromised in low- and middle-income countries, where the vaccination process is slow and inequitable. This may lead to new variant outbreaks with greater transmission capacity. Therefore, it is important to surveillance the circulating variants in the populations. Methods. In this sense, in Tepic, Nayarit, Mexico, 100 viral genomes of positive patients were sequenced during the beginning and end of the third (August 4th to September 3rd, 2021) and fourth (January 3rd, to February 2nd, 2022) COVID-19 waves. Results. Sequence analysis revealed the presence of several variants; alpha (B.1.1.7), gamma (P.1), local variant (B.1.1.519), mu (B.1.621), delta (B.1.617.2), and its subtypes (AY.3, AY.4, AY.10, AY.11, AY.20, and AY.23.1) during the third wave. Later, during the fourth wave, delta subtypes were still detected (AY.26 and AY.113), as well as omicron (B.1.1.529) and omicron subtypes (B.A.1 and BA.1.1). Conclusion. Obtained data revealed a progressive shift of the dominant variants, delta, and subtypes in the third wave and omicron and subtypes in the fourth wave., Introducción. El COVID-19, cuyo agente etiológico es el SARSCoV-2, un virus de ARN, se caracteriza por una elevada tasa de mutación. Por lo tanto, a mayor número de sujetos infectados existe, mayor probabilidad de que el virus sufra cambios que le confieran ventajas evolutivas (evasión de la respuesta inmunitaria, aumento de la virulencia y reducción de la eficacia de la vacunación). Los esfuerzos por adquirir inmunidad de rebaño mediante la vacunación pueden verse comprometidos en los países en vías de desarrollo, donde el proceso de vacunación es lento y poco equitativo. Esto puede dar lugar a nuevos brotes de variantes con mayor capacidad de transmisión. Objetivo. Vigilar las variantes circulantes en la población Nayarita. Métodos. En este sentido, en Tepic, Nayarit, México, se secuenciaron 100 genomas virales de pacientes positivos durante el inicio y final de la tercera (4 de agosto al 3 de septiembre de 2021) y cuarta (3 de enero, al 2 de febrero de 2022) olas de COVID-19. Resultados. El análisis de secuencias reveló la presencia de diversas variantes; alfa (B.1.1.7), gamma (P.1), variante local (B.1.1.519), mu (B.1.621), delta (B.1.617.2), y sus subtipos (AY.3, AY.4, AY.10, AY.11, AY.20, AY.23.1) durante la tercera ola. Posteriormente, durante la cuarta ola, se siguieron detectando subtipos delta (AY.26 y AY.113), así como ómicron (B.1.1.529) y los subtipos de ómicron (B.A.1 y BA.1.1). Conclusiones. Los datos obtenidos revelaron un desplazamiento progresivo de las variantes dominantes, delta, y subtipos en la tercera ola y ómicron y sus subtipos en la cuarta ola.

Abstract

The mitochondrial membrane potential (ΔΨm) is a parameter often used to determine mitochondrial function; therefore, it can be used to determine the integrity and functionality of cells. A decrement of ΔΨm is implicated in several inflammatory‐related pathologies, such phenomena can be related to COVID‐19 infection. The present work aimed to compare the ΔΨm in leucocytes (human PBMCs; HPBMC) isolated from healthy control (HC) subjects, patients with COVID‐19 (C‐19), recovered subjects at 40 ± 13 (R1) and 335 ± 20 (R2) days after infection (dai). Obtained data showed that ΔΨm decreased in HPBMC of subjects with C‐19, R1, and R2 compared with HC. When analyzing the ΔΨm data by sex, in females, a significant decrease was observed in R1 and R2 groups versus HC. Regarding men, a significant decrease of ΔΨm was observed in R1, with respect to HC, contrary to R2 group, who reestablished this parameter. Abstract

Organophosphorus pesticides (OPs) are broad-spectrum insecticides that exert their neurotoxic effect by inhibiting the enzyme acetylcholinesterase (AChE). However, this enzyme is not only present in the nervous system; cholinergic activity has also been demonstrated in leukocytes, so OPs can modify neuroimmunological communication. The objective of this study was to determine the cholinergic alteration and parameters of innate immunity, caused by diazinon (DZN) in cells of the immune system, using the tilapia fish (Oreochromis niloticus) as a model. For this purpose, the fish were exposed in vivo for 6 or 24 h at 0.97, 1.95 and 3.91 mg/L at DZN. Immediately, the expression of acetylcholine muscarinic receptors (mAChR) (M2-M5 subtypes) was evaluated through leukocytes, through RT-qPCR, as well as parameters of the innate response, such as phagocytic capacity, production of reactive oxygen species (ROS) and formation of extracellular neutrophil traps (NETs), using fluorescent techniques, such as flow cytometry and microscopy. The results obtained indicate that DZN induces a decrease in phagocytic capacity and increased ROS production and NETs formation; alterations that could be related to a cholinergic disruption, since the decrease in the expression of all subtypes of mAChR evaluated (M2-M5) were observed. The results obtained can help the understanding of important biological phenomena, because the leukocyte cholinergic system is essential in basic physiological processes, as immune response induction, as well as behavioral-cognitive aspects, so that its alteration puts survival at risk of organism. Los plaguicidas organofosforados (OPs) son insecticidas de amplio espectro que ejercen su efecto neurotóxico inhibiendo la enzima acetilcolinesterasa (AChE). Sin embargo, esta enzima no sólo está presente en sistema nervioso, también se ha demostrado actividad colinérgica en leucocitos, por lo que los OPs pueden modificar la comunicación neuroinmunológica. El objetivo de este estudio fue determinar la alteración colinérgica y parámetros de inmunidad innata, provocada por diazinón (DZN) en células del sistema inmune, utilizando como modelo al pez tilapia (Oreochromis niloticus). Para este propósito, los peces fueron expuestos in vivo durante 6 o 24 h a 0.97, 1.95 y 3.91 mg/L de DZN. Inmediatamente, se evaluó en leucocitos, la expresión de los receptores muscarínicos de acetilcolina (mAChR) (subtipos M2-M5), a través de RT-qPCR, así como parámetros de la respuesta innata, como capacidad fagocítica, producción de especies reactivas de oxígeno (ROS) y formación de trampas extracelulares de neutrófilos (NETs), mediante técnicas fluorescentes, como citometría de flujo y microscopía. Los resultados obtenidos indican que DZN induce disminución de la capacidad fagocítica e incrementó la producción de ROS y formación de NETs; alteraciones que podrían estar relacionadas a una disrupción colinérgica, ya que se observó la disminución en la expresión de todos los subtipos de mAChR evaluados (M2-M5). Los resultados obtenidos, pueden ayudar a la comprensión de fenómenos biológicos importantes, debido a que el sistema colinérgico leucocitario es esencial en procesos fisiológicos básicos, como la inducción de la respuesta inmune, así como en aspectos conductuales-cognitivos, por lo que su alteración pone en riesgo la supervivencia del organismo.

Abstract

The expression of elements of the cholinergic system has been demonstrated in non-neuronal cells, such as immune cells, where acetylcholine modulates innate and adaptive responses. However, the study of the non-neuronal cholinergic system has focused on lymphocyte cholinergic mechanisms, with less attention to its role of innate cells. Considering this background, the aims of this review are 1) to review information regarding the cholinergic components of innate immune system cells; 2) to discuss the effect of cholinergic stimuli on cell functions; 3) and to describe the importance of cholinergic stimuli on host immunocompetence, in order to set the base for the design of intervention strategies in the biomedical field.

Abstract

The present review aimed to evaluate the current situation of pesticide residues detected in shrimp (commercial species) on the Mexican coasts. The organochlorine pesticides (OC), α-endosulfan (210.01 ng g−1), endosulfan sulfate (127.5 ng g−1), heptachlor (126.04 ng g−1 and γ-HCH (121.04 ng g−1) are identified as the most common pesticides in shrimp tissues, with the Northwest area reporting the highest concentrations of these OC. Given that there is an under-evaluation of pesticide residue levels, there was a greater contribution of studies directed at the Northwest of the country considering that there are states that are among the main shrimp-producing and consumers entities. The concentrations and types of pesticides banned nationally and globally, due to their toxic effects on the population, were reported. Abstract

Various exogenous factors, such as microbiological and chemical contamination condition food security. Salmonella Typhimurium (S. Typhimurium) is the cause of salmonellosis. This bacterium utilizes phagocytosis to create bacterial reservoirs. On the other hand, exposure to chemical contaminants, such as pesticides, increases susceptibility to numerous infections. Therefore, this research aims to evaluate the effect of co-exposure to diazoxon and S. Typhimurium on the in vitro infection dynamics. For this purpose, human mononuclear cells were pre-exposed in vitro to diazoxon and then challenged with S. Typhimurium at 1, 8, and 24 h. Bacterial internalization, actin polymerization, and reactive oxygen species (ROS) were analyzed. Obtained data show that mononuclear cells previously exposed to diazoxon exhibit greater internalization of S. Typhimurium. Likewise, greater ROS production and an increase in actin polymerization were observed. Therefore, in the proposed scenario, obtained data suggest that co-exposure to diazoxon and S. Abstract

The mitochondrial membrane potential (ΔΨ m ) is a parameter often used to determine mitochondrial function; therefore, it can be used to determine the integrity and functionality of cells. A decrement of ΔΨ m is implicated in several inflammatory-related pathologies, such phenomena can be related to COVID-19 infection. The present work aimed to compare the ΔΨ m in leucocytes (human PBMCs; HPBMC) isolated from healthy control (HC) subjects, patients with COVID-19 (C-19), recovered subjects at 40 ± 13 (R1) and 335 ± 20 (R2) days after infection (dai). Obtained data showed that ΔΨ m decreased in HPBMC of subjects with C-19, R1, and R2 compared with HC. When analyzing the ΔΨ m data by sex, in females, a significant decrease was observed in R1 and R2 groups versus HC. Regarding men, a significant decrease of ΔΨ m was observed in R1, with respect to HC, contrary to R2 group, who reestablished this parameter.
