We previously demonstrated expression of full-length transcripts for sublingual mucin apoprotein, Muc19, of approximately 24 kb (Fallon MA, Latchney LR, Hand AR, Johar A, Denny PA, Georgel PT, Denny PC, and Culp DJ. Physiol Genomics 14: 95-106, 2003). We now describe the complete sequence and genomic organization of the apomucin encoded by 43 exons. Southern analyses indicate a central exon of approximately 18 kb containing 36 tandem repeats, each encoding 163 residues rich in serine and threonine. Full-length transcripts are an estimated 22,795 bp in length that span 106 kb of genomic DNA. The transcriptional start site is 24 bp downstream of a TATA box and 42 bp upstream of the conceptual translational start codon. The putative apoprotein has an estimated mass of 693.4 kDa and contains 7,524 amino acids (80% serine, threonine, glycine, alanine, and proline). We present a model for rat Muc19 transcripts and compare the conceptually translated Muc19 proteins for mouse, rat, pig, and the 3' end of human Muc19. Conserved among these apoproteins are a signal peptide, a large tandem repeat region, von Willebrand factor type C and D domains, a trypsin inhibitor-like Cys-rich domain, and a COOH-terminal cystine knot-like domain. Southern blot analyses indicate transcripts for Muc19 and Smgc (submandibular gland protein C) are splice variants of a larger gene, Muc19/Smgc. Comparative Northern analyses between the major salivary glands demonstrate highly selective Muc19 expression in neonatal and adult sublingual glands, whereas Smgc is expressed in neonatal submandibular and sublingual glands. Regulation of Muc19/Smgc gene expression is discussed with respect to alternative splicing and mucous cell cytodifferentiation.

Female nonobese diabetic (NOD) mice develop spontaneous autoimmune sialadenitis and loss of salivary flow, and are a widely used model of Sjögren's syndrome. We examined the feasibility of local salivary gland immunomodulatory gene delivery to alter these sequelae in NOD mice. We constructed recombinant adeno-associated virus (rAAV) vectors encoding either human interleukin 10 (rAAVhIL-10) or beta-galactosidase (rAAVLacZ, control vector). Mice received rAAVhIL-10 or rAAVLacZ by retrograde submandibular ductal instillation either at age 8 weeks (early, before onset of sialadenitis), or at 16 weeks (late, after onset of sialadenitis). As a systemic treatment control, separate mice received intramuscular delivery of rAAVhIL-10 at each time point. Both submandibular and intramuscular delivery of vector led to low circulating levels of hIL-10. After submandibular administration of rAAVhIL-10, salivary flow rates at 20 weeks for both the early and late treatment groups were significantly higher than for both rAAVLacZ-administered and untreated mice. Systemic delivery of rAAVhIL-10 led to improved salivary flow in the late treatment group. Inflammatory infiltrates in submandibular glands, however, were significantly reduced only in mice receiving rAAVhIL-10 locally in the salivary gland compared with mice receiving this vector intramuscularly, or rAAVLacZ or no treatment. In addition, after submandibular rAAVhIL-10 delivery, NOD mice exhibited significantly lower blood glucose, and higher serum insulin, levels than all other groups, indicating some systemic benefit of this treatment. These studies show that expression of hIL-10 by rAAV vectors can have disease-modifying effects in the salivary glands of NOD mice, and suggest that local immunomodulatory gene transfer may be useful for managing the salivary gland pathology in Sjögren's syndrome.

The expression of tissue-specific enamel matrix genes is believed to require both instructive and permissive interactions of enamel organ epithelium with dental papilla mesenchyme and/or extracellular matrix during a restricted period of development. Biosynthesis of amelogenin gene products has been found to be associated with the terminal differentiation of inner enamel organ epithelium. The developing mouse first mandibular molar was used for a detailed examination of the temporal initiation and developmental pattern of amelogenin transcription. These studies define temporally instructive versus permissive influences on amelogenin transcription. During in vivo development, amelogenin transcripts were detected in late cap (15 days in utero; E15) through bell stage (E16 through E19) mouse molar tooth formation utilizing reverse transcription coupled to polymerase chain reaction amplification. Alternatively spliced amelogenin transcripts were detected in late bell stage (E18) molars. Amelogenin transcripts were also detected in isolated late cap stage (E15) enamel organ epithelium dissected free of dental papilla mesenchyme and cultured within a substitute basement membrane gel, but not in identical cap stage enamel organ epithelium cultured on plastic or a laminin-coated filter. Amelogenin transcripts were also found in early cap stage (E14) isolated enamel organ epithelium cultured within a basement membrane gel, but were not detected in enamel organ epithelium isolated from earlier stages of odontogenesis and cultured within a basement membrane gel. The results of these experiments indicate that a basement membrane gel is a useful extracellular substrate which provides permissive interactions required for the expression of amelogenin transcripts by enamel organ epithelium and that instructive interactions which determine enamel organ epithelium to become committed to amelogenin transcription occur prior to the early cap stage (E14) of odontogenesis. The results also suggest that continued interactions of enamel organ epithelium with dental papilla mesenchyme serve to regulate amelogenin transcription and post-transcriptional amelogenin RNA splicing in a complex manner during odontogenesis.

An important issue in craniofacial biology is understanding the molecular mechanisms that regulate the transcription of genes during development. Low concentrations of the thymidine analogue, 5-bromodeoxyuridine (BrdU), have been used to perturb transcription of tissue-specific genes in a variety of tissue types, although the molecular mechanism for this inhibition has not been elucidated. The purpose of the present study was to examine the following: (1) if amelogenin transcription is inhibited in mouse molars cultured in the presence of BrdU, (2) if changes in methylation patterns of the amelogenin gene can be detected with terminal differentiation of ameloblasts in vivo and in vitro; and (3) if changes in methylation patterns of the amelogenin gene can be detected in mouse molars cultured in the presence of BrdU. Northern blot hybridization and RNA phenotyping analysis revealed that bromodeoxyuridine (BrdU) incorporation into the DNA of developing mouse mandibular first molars (M1) in vitro inhibited amelogenin transcription. Restriction endonuclease digestion of M1 genomic DNA followed by Southern blot hybridization analysis revealed that amelogenin transcriptional activity in vivo and in vitro did not correlate with changes in methylation of the amelogenin gene. These results suggested that, unlike several other developmentally regulated genes, transcriptional regulation of the amelogenin gene may not be associated with changes in DNA methylation patterns.

Electroporation was used to introduce foreign genes into cells derived from the mouse enamel organ epithelia (EOE). Optimal conditions for this electroporation were established. The introduction of a plasmid construct bearing the coding region for the large T-antigen from polyoma virus into EOE cells permitted the establishment of a derivative cell line that has the following characteristics: (1) the cells could be passaged many times; (2) they expressed a keratin-containing cytoskeleton; and (3) approx. 60% of the cells expressed amelogenin, a tissue-specific gene product unique to ameloblasts. Potential uses for such a cell line include analysis of: (1) the upstream regulatory regions required for temporally and spatially restricted expression of amelogenin; (2) the post-translational modification of amelogenin in synchronized cells and (3) the organization and biomineralization of enamel extracellular matrix in monolayer culture.

There is increasing evidence that autocrine- and paracrine-acting growth factors participate in cell and tissue development, maintenance, and renewal. Recent advances in histochemical techniques have facilitated the localization of growth factor messenger RNAs or polypeptides in tissue sections. However, the spatial relationships between the sites of growth factor transcription, translation, and post-translational processing to functional bioactive peptides have been difficult to correlate because each method of detection requires separate tissue sections. We undertook the simultaneous detection of epidermal growth factor (EGF) transcripts and EGF epitopes by combining immunohistochemistry methods with in situ hybridization. Adult mouse submandibular gland was chosen as a representative model because it contains sites of EGF biosynthesis which may participate in mediating the development, maintenance, and renewal of the organ through autocrine or paracrine mechanism(s). Granular duct (GD) cells demonstrated the presence of both EGF transcripts and EGF peptides. In contrast, the interstitial cells lying adjacent to glandular epithelium also contained relatively high levels of EGF transcripts, although no mature EGF peptides were detected. The experimental approach of co-localization and the resulting data indicate previously unreported sites of EGF transcription in glandular interstitial cells, which may provide molecular information required for the morphogenesis and differentiation of adjacent glandular epithelium.

The interpretation of recent experimental evidence has lead to several discussions at international meetings which have included the suggestion that enamelins are not tissue-specific gene products but rather are similar to, or are identical to, albumin. Further complicating the issue has been the proposal by several investigators that albumin participates in the organization of hard tissues. The experimental strategy described in this study was to hybridize the mouse albumin gene cDNA clone, palb-3, to a Northern blot of secretory phase mouse mandibular first molar RNAs. This approach would permit the evaluation of the potential transcription of the albumin gene by differentiated ameloblasts as well as other cells of the odontogenic organ. The results of this approach indicated that odontogenic cells did not transcribe the albumin gene. Albumin polypeptides, therefore, cannot be synthesized by odontogenic cells and cannot be identical to odontogenic tissue-specific gene products. If albumin is incorporated into developing enamel matrix wherein it participates in undefined roles during enamel matrix organization, it cannot arise as a biosynthetic product of the odontogenic organ.
