Abstract

National audience; Stormwater runoffs are loaded with metals, PAHs, pesticides and pathogenic bacteria, conveyed for many in particulate form. One of the techniques used to manage this contaminated stormwater is to use retention basins in order to allow settling of the suspended solids. The deposits thus set up represent zones of contamination and a particular waste. The deposits thus constituted represent waste that must be managed. The objectives of this paper are: i) to characterize the sediments of a stormwater retention basin, ii) to identify the variability and heterogeneity of its microbiological, physical, chemical and ecotoxicological characteristics and to highlight the key factors that could explain this variability, iii) establish correlations between characteristics and identify trends in the change of sediment quality. The deriving results show that the median diameter of the sediments is between 50 and 150 μm. They are mostly contaminated with PAH, metals and 4-n-nonylphenol. Concerning the microbiological characterization, the bacteria “cores” observed in the sediments are composed of the bacterial genus which may contain pathogenic forms: Mycobacteria, Pseudomonas, Aeromonas, Nocardia. Nocardia counts are higher in sediments composed of fine and light particles (grain size between 1.1 and 3.3 μm). The ecotoxicity analysis of these sediments using the “ostracodes” test revealed that “fresh” sediments collected during a storm event were more ecotoxic than old ones. Sediment characteristics trended to become homogenous after 5 years of monitoring. Regarding investigations on the identification of the sources of microbiological contaminants observed in the retention basin, socio-urban observations revealed a direct relationship between the microbiological contamination of runoff and the presence of items and marks leave by socio-economic activities undertaken on the watershed. These results are useful for the managers in order to guide the choice or to stimulate the development of sectors or strategies of treatment and valorization of these sediments.

Abstract

International audience; Combined sewer systems convey organic matters and pathogens coming from human and animal faeces. They are mixed with rainfall runoff during storm event and can overflows to natural aquatic systems. Preliminary genetic analyses of pathogens were conducted in an experimental small water course and its lateral combined sewer system (OTHU site). Results indicate that natural system pathogens are an evolution of sewer system pathogens. However their presumed survival should not be confounded with their frequent delivery to the water course. Then a first step is to check for concentrations and frequencies that are delivered to receiving waters. First data on three pathogen species concentrations were sampled from 2009 to 2011 into the combined sewer system. Data exhibit variations that can be related to dry and wet weather sequences. However variations do not fully correlate between pathogen species. This would indicate growing rate and/or differing locations in the sewer pipe. The main hypothesis to explain concentration increase with storm events is the flushing of organic biofilms lining the immerged part of sewer conduit. This hypothesis is to be confirmed with complementary data more focussed on the water column during storm events and lining biofilms during dry weather.

Abstract

Aims To develop a q PCR approach for the detection of Pseudomonas aeruginosa in soil and manure and explore its efficacy and limitations compared with that of a classical culture-dependent approach. Methods and Results A Ps. aeruginosa ecfX q PCR assay was developed. This assay was optimized for soils of contrasting physico-chemical properties and evidenced a three-log dynamic range of detection [5 × 104 - 5 × 106 cells (g drywt soil)−1] in inoculated microcosms. Sensitivity was determined to be around 5 × 104 cells (g drywt soil)−1. In parallel, the minimum detection limit was estimated in the range of 10-100 CFU (g drywt soil)−1 using a culture-dependent approach based on the use of a selective medium (cetrimide agar base medium supplemented with nalidixic acid), coupled to ecfX gene amplification to confirm isolate identity. These soil samples led to the growth of abundant non- Ps. aeruginosa colonies mainly belonging to other Pseudomonas species but also some beta- Proteobacteria. These bacteria strongly impacted the detection threshold of this approach. Efficacy of these approaches was compared for Ps. aeruginosa enumeration among manure and agricultural soil samples from various sites in France, Tunisia and Burkina Faso. Conclusions The developed q PCR assay enabled a specific detection of Ps. aeruginosa in soil and manure samples. The culture-based approach was usually found more sensitive than the q PCR assay. However, abundance of non- Ps. aeruginosa species among the indigenous communities able to grow on the selective medium affected the sensitivity of this latter approach. Significance and Impact of the Study This study describes the first specific and sensitive q PCR assay for the detection and enumeration of Ps. aeruginosa in soil and manure and shows its complementarity with a culture-based approach.
