21. 11582 Examining Various Induction Methods of Endometriosis in Translational, Pre-Clinical Mouse Models.

Abstract

To evaluate five methods of endometriosis induction in immunocompetent mouse models to determine which models would be the most useful for pre-clinical trials related to endometriosis. Preclinical. Translational, pre-clinical mouse model. Intact 8 ± 8-week old female immunocompetent mice, with B6(Cg)- Tyrc-2J Tg(UBC-mCherry)1Phbs/J (stock #017614) used as donor mice and B6(Cg)- Tyrc-2J /J (stock # 000058) used as recipient mice. Five methods of endometriosis induction were used: (1) laparoscopic placement of decidualized endometrial implants, (2) placement of decidualized endometrial implants via laparotomy, (3) laparoscopic tissue slurry injection, (4) tissue slurry injection via laparotomy, and (5) discrete implantation into menstruating mice via laparoscopy (N=10/paradigm). After induction of endometriosis, all mice demonstrated normal estrus cyclicity. Endometriosis occurred in 100% of mice in group 1, 80% of group 2, 20% of group 3, 10% of group 4, and 100% of group 5. There was a statistically significant difference between group 1 and 2 of 1.6 (p =.009, 95% CI (.39, 2.80)), indicating that laparoscopic placement of endometrial implants was superior to laparotomy in induction of endometriosis. The methods used in group 1 and group 5 were equally effective, with a difference of 0.10 in the estimated average number of lesions (p =.89, 95% CI (-1.33, 1.53)). Induction of endometriosis was most successful via laparoscopic placement of endometrial implants using menstruating donor and recipient mice. Normal estrus cyclicity was achieved afterwards, and lesions were identified >30 days post-induction. This indicates a new mouse model that can be used in preclinical endometriosis research. 34. 11580 The Association of Interleukins With Endometriosis: A Systematic Review and Meta-Analysis.

Abstract

To compare the serum levels of interleukins (ILs) in individuals with untreated endometriosis to those without endometriosis through a systematic review and meta-analysis. Systematic review and meta-analysis. Endometriosis research. Articles related to IL serum levels in individuals with and without endometriosis through May 2023. Each article was evaluated for inclusion, data were extracted, and meta-analysis was performed. Thirty studies met inclusion criteria. These studies included IL-1Ra, IL-1b, IL-2, IL-4, IL-6, IL-8, IL-10, IL-12, IL-17A, IL-18, IL-23, and IL-37. Patients with endometriosis were found to have higher serum levels of IL-1Ra, IL-6, and IL-37 compared to those without endometriosis. They were found to have lower serum levels of IL-23 as compared to patients without endometriosis. The serum levels of IL-6 were 1.38 SDs higher in those with endometriosis than in those without endometriosis (study n=16 Hedges' g effect size = 1.38, 95% CI 0.58−2.17 , I <.001, I2=97.19%). Meta-regression suggested that IL-6 in endometriosis was not significantly impacted by the stage (I/II vs. III/IV) of the disease (Hedge's g effect size; Regression coefficient = −0.53, 95% CI−1.44 to 0.38, p =.253, change in R2 = 2.89%). There is evidence supporting the association of serum IL levels, particularly IL-6, with endometriosis. This suggests that treatment and diagnostic studies related to these inflammatory pathways may be effective for patients with endometriosis. [ABSTRACT FROM AUTHOR]

35. 11246 Factors Affecting the Immunocompetent Mouse Model of Endometriosis: A Systematic Review, Meta-Analysis, and Call to Action.

Abstract

To compile all available research pertaining to immunocompetent mouse models of endometriosis to evaluate which models may be the most effective in preclinical trials for endometriosis treatment. Systematic review and meta-analysis. Translational, preclinical mouse model. Articles using an immunocompetent mouse model that included abdominal endometriosis and reported at least one quantifiable lesion measurement with associated standard deviations or standard errors. Each article was evaluated for inclusion, data were extracted, and risk of bias and quality of articles were determined by 2 or more independent reviewers. The search strategy identified 124 articles (total number of mice, n =3056) for inclusion. There were approximately 26 total (SD= 21; median=20) mice on average per article in the suture sub-group and approximately 25 total (SD= 26; median=17) mice per article on average in the injection sub-group. The average mice per experiment per article had approximately 11 mice (SD= 6) in the injection sub-group and approximately 9 mice (SD= 5) in the suture sub-group (p=.022981). Meta-regression showed recipient exogenous supplemental hormones (p= 0.01856) and BALB/c strain (p= 0.036) affected lesion weight, autologous grafts (p =0.004) affected lesion volume, syngeneic grafts (p =0.008) affected lesion area, myometrium with endometrium (p =<0.001) affected lesion diameter, and the BALB/c strain (p =0.032) and recipient exogenous supplemental hormones (p =0.004) affected the number of lesions. The degree of heterogeneity, risk of bias, and low or very low quality of evidence emphasizes the need for a call to action. Model standardization, agreed upon by the research community, would improve reproducibility and allow for evidenced-based translational outcomes, especially in the setting of pre-clinical endometriosis studies. 36. The Effect of Video Games on Laparoscopic Skills: A Prospective Randomized Controlled Trial and Quality Improvement Project.

Abstract

To compare medical students' performance on LapMentorTM skills tests with and without the use of a video game warmup with different controllers and to determine the effect of a video game on the frequency of simulator use in OB/GYN residents. Prospective RCT combined with a QI project. Department of OB/GYN at Creighton University School of Medicine. Medical students and OB/GYN residents. First, students were randomized into groups: Group 1 (warm-up with UndergroundTM game using standard WiiUTM video game controller), Group 2 (warm-up with UndergroundTM game using custom-made laparoscopic controller), and Group 3 (no warm-up). Students completed a pretest and posttest on a validated laparoscopic simulator (LapMentorTM) according to their assigned group. Next, a pre-test questionnaire on the use of simulators for laparoscopic skills was given to OB/GYN residents. They completed a post-test questionnaire after being provided with a WiiUTM videogame system that included a custom controller and UndergroundTM video game. Two analyses were performed for the first part of the project: ANCOVA, which used data from the pretest and posttest, and the mixed model, which included participants who did not return for the posttest. Subgroup analysis was conducted on all participants and only those who completed every LapMentorTM task. A statistically significant (p <0.001) decrease from pretest to posttest was constant across groups, regardless of analysis, with no between-group differences at time of pretest and posttest. For the second part of the project, there was a statistically significant increase in laparoscopic simulator use after providing residents with a WiiUTM video game system as compared to prior (p =0.027). The use of a video game warm-up is associated with faster completion of laparoscopic skills tests in medical students regardless of controller type. Additionally, providing OB/GYN residents with access to the UndergroundTM video game is associated with increased use of the laparoscopic simulator. [ABSTRACT FROM AUTHOR]
