- Abstract
Traditional wastewater treatment plants (WWTPs) are electrical consumers, with a usual high demand in the range of 0.3 to 0.6 kWh/m3 of wastewater treated. Their digesters are commonly oversized, and consequently operated at low organic loading rates (OLRs). This opens a great opportunity for anaerobic co-digestion (AcoD) as an interesting technology to increase methane productivity and the electrical self-production in WWTPs. However, there is a quite limited implementation of AcoD at full-scale plants, since the transition from mono- to co-digestion and the further AcoD optimisation is a crucial and delicate step that could lead to the inhibition of the process if not thoroughly controlled. In this study, a methodology based on an optimum control strategy is explained in detail and it was applied to safely and optimally steer the transition from mono- to co-digestion and to maximize methane production during AcoD. A lab-scale anaerobic digester of 14 L mimicking the full-scale one (3,500 m3) was operated 30 days in advance to anticipate and if needed correct any operational destabilization that might occurr. As a result, the treatment of sewage sludge with two co-substrates (coming from a pig slaughterhouse and from a frying industry), which accounted for just 11% of the feeding flowrate, at a hydraulic retention time of 20 days allowed to raise the OLR and the methane production by 2-fold and 3-fold, respectively, increasing the self-produced electricity from 25% to 75% of the total demand of the WWTP. The diagnosis indicators proved to be accurate to take decisions concerning wastes blending and the strategy of increasing OLR. Besides, the proposed control system provides the steps to ensure a safe transition from anaerobic mono- to co-digestion and further optimisation at full-scale plants

Abstract : This work is the result of an investigation about the integration policy of professional education to basic education in the modality of youth and adults education presented by the Brazilian federal government having as its goals the universal system of basic education combined with training to the world of work, welcoming the young people and adults population segment who have discrete learning paths. The aim of the research was to investigate how the implementation of this policy has been configured in federal education in Santa Catarina. It analyzes the integration of professional education to basic education in the form of youth and adult education, in order to check the understanding of this policy in the educational units within the Federal Institute of Santa Catarina, Florianópolis campus and Florianópolis campus continent, delimited as units of analysis of this research. The youth and adult education in high school, is still incipient. To discuss a professional education policy which is integrated to high school education in youth and adult modality implies to draw landmarks of references about what is meant by this educational policy and particularly the PROEJA, as well as to identify the categories that enable and mark out the theoretical frameworks of this object of analysis. The present research takes as the methodological framework the critical ontology presented by György Lukács, based on Karl Marx´s background, due to our beliefs that this is the appropriate method to our objectives because it provides the powerful and necessary tools for the understanding of the origin, development, structure and the operating mechanism of the social relationship dominated by capital. On the basis of ontological methodology perspective, we seek to understand PROEJA in its complex relationship of social phenomena, in the process of seizing the mediation of social being, between the apparent phenomenon and the ownership of their existing reality that allow to extend the understanding of the social being as a historical and contradictory being, making possible a more accurate catchment of the educational complex and its articulation with the reality .The research presents from the study of primary and secondary sources, some themes / categories which allowed the analysis: resources and sources of financing and infrastructure financing: buildings and equipment; pre-service and in-service teacher education and training, educational integration and work as an educational principle; working conditions for education professionals; formative path of students, which allowed an understanding of how teachers, students, coordinators and managers ofthe courses have reflected about the world of work and its relations with the educational field, after the implementation of the integration national programme of professional education with the basic education for youth and adult education mode in the daily life of these educational institutions. Trying to understand the determinant aspects and the PROEJA courses configuration which were analyzed, the research results allow to consider that the lack of initial, permanent formation and continued formation held in poor conditions, the lack of infrastructure to carry out pedagogical work and the lack of permanent policy suitable to the students needs, impact in providing quality education also affect the permanence of students in the courses. We highlight that the integration policy of the professional education with Basic Education in the form of Youth and Adult Education, established by the Federal Government for the training of young and adult workers, reveals in its configuration, as just one of the educational policies that have the exclusive function to meet the interests of capital in productive restructuring time, in the context of capital crisis and work reorganization and new configurations of employment and unemployment. Finally the study also presents conceptual elements for teacher training process on professional education and the possibility of integration of primary education in the form of youth and adult education.

Background: Emerging evidence suggests that DNA methylation plays an expansive role in the central nervous system (CNS). Large-scale whole genome DNA methylation profiling of the normal human brain offers tremendous potential in understanding the role of DNA methylation in brain development and function. Methodology/Significant Findings: Using methylation-sensitive SNP chip analysis (MSNP), we performed whole genome DNA methylation profiling of the prefrontal, occipital, and temporal regions of cerebral cortex, as well as cerebellum. These data provide an unbiased representation of CpG sites comprising 377,509 CpG dinucleotides within both the genic and intergenic euchromatic region of the genome. Our large-scale genome DNA methylation profiling reveals that the prefrontal, occipital, and temporal regions of the cerebral cortex compared to cerebellum have markedly different DNA methylation signatures, with the cerebral cortex being hypermethylated and cerebellum being hypomethylated. Such differences were observed in distinct genomic regions, including genes involved in CNS function. The MSNP data were validated for a subset of these genes, by performing bisulfite cloning and sequencing and confirming that prefrontal, occipital, and temporal cortices are significantly more methylated as compared to the cerebellum. Conclusions: These findings are consistent with known developmental differences in nucleosome repeat lengths in cerebral and cerebellar cortices, with cerebrum exhibiting shorter repeat lengths than cerebellum. Our observed differences in DNA methylation profiles in these regions underscores the potential role of DNA methylation in chromatin structure and organization in CNS, reflecting functional specialization within cortical regions. - Abstract
Panic disorder PD is a debilitating anxiety disorder, characterized by recurrent episodes of intense fear that are accompanied by autonomic and psychological symptoms leading to behavioral impairment. Basic research implicates neuropeptidesignaling genes in the modulation of anxiety and stress. The genes encoding corticotropin releasing hormone receptor 1 CRHR1, tachykinin receptor 1 TACR1, gastrin releasing peptide GRP, and gastrin releasing peptide receptor GRPR were selected as candidates for PD based on their biology. Linkage and association analysis in 120 multiplex U.S. PD pedigrees was performed using 21 single nucleotide polymorphisms SNPs. Parametric and nonparametric linkage tests in pedigrees, for single point and multipoint analysis, revealed limited support for genetic linkage to TACR1 parametric and nonparametric lod scores ∼1. The familybased association test FBAT generated nominal support for allelic association in TACR1 P 0.02, and GRP P 0.02, findings which must be considered in the light of multiple comparisons. Further exploration of the GRP and TACR1 findings in large casecontrol PD samples may provide more definitive evidence implicating these loci in the genetic etiology of PD. © 2008 WileyLiss, Inc.

- Abstract

Background: Panic disorder (PD) is a common illness with a definite but “complex” genetic contribution and estimated heritability of 30–46%. Methods: We report a genome scan in 120 multiplex PD pedigrees consisting of 1591 individuals of whom 992 were genotyped with 371 markers at an average spacing of 9cM. Parametric two-point, multipoint, and nonparametric analyses were performed using three PD models (Broad, Intermediate, Narrow) and allowing for homogeneity or heterogeneity. The two-point analyses were also performed allowing for independent male and female recombination fractions (θ). Genome-wide significance was empirically evaluated using simulations of this dataset. Results: Evidence for linkage reached genome-wide significance in one region on chromosome 15q (near GABA-A receptor subunit genes) and was suggestive at loci on 2p, 2q and 9p using an averaged θ. Analyses allowing for sex-specific θ’s were consistent except that support at one locus on 2q increased to genome-wide significance and an additional region of suggestive linkage on 12q was identified. However, differences in male and female recombination fractions predicted by the sex-specific approach were not consistent with current physical maps. Conclusions: These data provide evidence for chromosomal regions on 15q and 2q that may be important in genetic susceptibility to panic disorder. Although we are encouraged by the findings of analyses using sex-specific recombination fractions, we also note that further understanding of this analytic strategy will be important.
