Abstract

Objetivo: Los implantes óseos son utilizados cada vez con mayor frecuencia en la práctica clínica y, entre los materiales, el Ti o sus aleaciones son los de mejor rendimiento por sus propiedades fisicoquímicas. Aleaciones como TiNbTa han demostrado mejorar las características biomecánicas del Ti puro comercial (c.p.), sin embargo, su capacidad osteointegradora necesita ser evaluada. El objetivo del presente estudio fue valorar la citotoxicidad y la capacidad de adhesión, proliferación y diferenciación de células osteoblásticas en cultivo, influida por discos de material TiNbTa frente a Ti c.p. Materiales y métodos: Analizamos a los 4 y 7 días del cultivo la línea celular MC3T3, la viabilidad celular (AlamarBlue Cell Viability Reagent. Invitrogen, España), así como la proliferación y diferenciación celular (actividad de fosfatasa alcalina (ALP) y microscopía electrónica de barrido (Fijación para SEM). Se realizó la prueba t de Student para determinar diferencias estadísticamente significativas entre los dos grupos de discos de estudio. Resultados: Los resultados obtenidos demuestran muy buena viabilidad celular durante el periodo de estudio, sin diferencias significativas para ambos materiales. MRSA is among the major causes of hospital-acquired and community infections.Methods: Therefore, in the present study, we evaluated the antibacterial activity of silver nanoparticles synthesized by Fusarium oxysporum (AgNPbio) in combination with simvastatin against reference and multidrug-resistant bacterial strains.Results: Simvastatin showed a minimal inhibitory concentration (MIC) ranging from 0.062 to 0.25 mg mL−1 against MRSA. AgNPbio with a size of 77.68± 33.95 nm and zeta potential −34.6 ± 12.7 mV showed an MIC of 0.212 mg mL−1 against S. aureus including MRSA strains. The checkerboard assay and time-kill curves exhibited a synergistic effect of the simvastatin-AgNPbio combination on antibacterial activity against MRSA strains. The combination of simvastatin and AgNPbio demonstrated antibacterial activity against Escherichia coli producing ESBL. Scanning electron microscopy showed the formation of cell surface protrusions after treatment with AgNPbio and the formation of a large amorphous mass after treatment with simvastatin, both in MRSA.Conclusion: Our results indicate that the combination of AgNPbio and simvastatin could be a great future alternative in the control of bacterial infections, where, when combined with simvastatin, smaller doses of AgNPbio are required, with the same antibacterial activity.Keywords: antibacterial, metallic nanoparticles, multidrug-resistant bacterial, statins, synergism

Abstract

Background Acute respiratory distress syndrome (ARDS) is associated with high in-hospital mortality. Alveolar recruitment followed by ventilation at optimal titrated PEEP may reduce ventilator-induced lung injury and improve oxygenation in patients with ARDS, but the effects on mortality and other clinical outcomes remain unknown. This article reports the rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART). Methods/Design ART is a pragmatic, multicenter, randomized (concealed), controlled trial, which aims to determine if maximum stepwise alveolar recruitment associated with PEEP titration is able to increase 28-day survival in patients with ARDS compared to conventional treatment (ARDSNet strategy). We will enroll adult patients with ARDS of less than 72 h duration. The intervention group will receive an alveolar recruitment maneuver, with stepwise increases of PEEP achieving 45 cmH2O and peak pressure of 60 cmH2O, followed by ventilation with optimal PEEP titrated according to the static compliance of the respiratory system. In the control group, mechanical ventilation will follow a conventional protocol (ARDSNet). In both groups, we will use controlled volume mode with low tidal volumes (4 to 6 mL/kg of predicted body weight) and targeting plateau pressure ≤30 cmH2O. The primary outcome is 28-day survival, and the secondary outcomes are: length of ICU stay; length of hospital stay; pneumothorax requiring chest tube during first 7 days; barotrauma during first 7 days; mechanical ventilation-free days from days 1 to 28; ICU, in-hospital, and 6-month survival. ART is an event-guided trial planned to last until 520 events (deaths within 28 days) are observed. These events allow detection of a hazard ratio of 0.75, with 90% power and two-tailed type I error of 5%. All analysis will follow the intention-to-treat principle. Discussion If the ART strategy with maximum recruitment and PEEP titration improves 28-day survival, this will represent a notable advance to the care of ARDS patients. Conversely, if the ART strategy is similar or inferior to the current evidence-based strategy (ARDSNet), this should also change current practice as many institutions routinely employ recruitment maneuvers and set PEEP levels according to some titration method. Esta comunicação tem como objetivo apresentar um conjunto de análises sobre os caminhos da empregabilidade e da profissionalização dos licenciados e pós-graduados em sociologia, em Portugal. Concretamente, procura-se compreender qual o contributo de diferentes gerações de sociólogos, homens e mulheres, nos processos de requalificação e recomposi ção dos vários sectores de atividade económica, ao longo dos últimos 40 anos de regime democrático. Abstract

Onion anthracnose, caused by Colletotrichum gloeosporioides, is one of the main diseases of onions in the State of Pernambuco. We examined the pathogenicity of 15 C. gloeosporioides strains and analyzed their genetic variability using RAPDs and internal transcribed spacers (ITS) of the rDNA region. Ten of the strains were obtained from substrates and hosts other than onion, including chayote (Sechium edule), guava (Psidium guajava), pomegranate (Punica granatum), water from the Capibaribe River, maracock (Passiflora sp), coconut (Cocus nucifera), surinam cherry (Eugenia uniflora), and marine soil; five isolates came from onions collected from four different regions of the State of Pernambuco and one region of the State of Amazonas. Pathogenicity tests were carried out using onion leaves and bulbs. All strains were capable of causing disease in leaves, causing a variable degree of lesions on the leaves; four strains caused the most severe damage. In the onion bulb tests, only three of the above strains caused lesions. Seven primers of arbitrary sequences were used in the RAPD analysis, generating polymorphic bands that allowed the separation of the strains into three distinct groups. The amplification products generated with the primers ITS1 and ITS4 also showed polymorphism when digested with three restriction enzymes, DraI, HaeIII and MspI. Only the latter two demonstrated genetic variations among the strains. These two types of molecular markers were able to differentiate the strain from the State of Amazonas from those of the State of Pernambuco. However, there was no relationship between groups of strains, based on molecular markers, and degree of pathogenicity for onion leaves and bulbs.

Abstract

Ao longo dos seus primeiros três anos de desenvolvimento, o Plano Nacional de Leitura tem sido acompanhado regularmente por estudos de avaliação externa, de acordo com o Sistema de Avaliação do PNL que foi elaborado e testado no ano de lançamento do Plano. Esse sistema encontra-se delineado e fundamentado na primeira publicação destes estudos de avaliação do PNL1. Esta nova publicação dos estudos de avaliação do PNL incide sobre o segundo e o terceiro anos do Plano Nacional de Leitura. No primeiro capítulo faz-se um balanço da execução geral do Plano neste período. No segundo capítulo apresentam-se os resultados do Inquérito às Escolas abrangidas pelas actividades do PNL, aplicado on-line pela segunda vez, no final do segundo ano do Plano, analisando a sua evolução em relação aos resultados do primeiro ano. No terceiro capítulo são abordados os programas de promoção da leitura nas escolas, dando-se destaque ao alargamento destes programas ao 3º ciclo do ensino básico, e referindo-se ainda a iniciativa Um Novo Livro para cada Novo Leitor. O quarto capítulo debruça-se sobre o projecto a LeR+, que adquiriu um relevo particular no desenvolvimento do Plano, pelo seu carácter inovador, experimental e intensivo. O quinto capítulo trata do projecto Leitura em Vai e Vem, desenvolvido nos jardinsde- infância. O sexto capítulo constitui uma primeira análise do projecto Ler+ dá Saúde, o qual representa uma nova e muito importante frente de acção do PNL, dirigida às crianças a partir dos seis meses de idade e às suas famílias, através da participação directa dos profissionais de saúde. O sétimo capítulo analisa os resultados da segunda aplicação nacional do Barómetro de Opinião Pública sobre o PNL, comparando-os com os resultados da primeira aplicação do Barómetro, em 2007. Nas notas finais, apresenta-se um breve conjunto de aspectos de síntese, relativos ao PNL e à sua avaliação.

Faz-se aqui a primeira apresentação de resultados do projecto de investigação «Estudantes universitários: classes sociais e representações», que sobre esses estudantes vem construindo um observatório sociográfico extensivo e permanente. A exposição está dividida em quatro partes. Na primeira, além do enunciado dos objectivos, debate-se a problemática da caracterização de classe dos estudantes e da sua relação com as famílias de origem e com a trajectória virtual futura. São também tratadas com algum pormenor questões de operacionalização das categorias analíticas das classes sociais dos indivíduos e dos grupos domésticos. Na segunda - «Painéis transversais» - compara-se os estudantes das licenciaturas de Gestão, Sociologia e Antropologia do ISCTE quanto à origem familiar e quanto a representações sobre licenciatura e profissão. Na terceira - «Painéis diacrónicos» - retoma-se as mesmas variáveis para as analisar ao longo de uma sequência de três anos, num dos cursos. Termina-se, discutindo algumas linhas de aprofundamento e extensão da pesquisa. This is the first presentation of the research project «University students: social classes and representations», that has been developing a permanent and extensive sociographic observation of these students. The exposition is divided into four parts. In the first one it is discussed the problematic of the class characterisation of students and its relationship with their families and with their virtual forthcoming trajectory. Some concerning the operationalisation for the empirical research of analytical concepts of individuals and households social classes are also mentioned. In the second part «Transversal Panels» we compare the students of Management, Sociology and Anthropology of ISCTE concerning their origin family and their representations about university credentials and profession. In the third part «Diachronic Panels» we have taken the some variables to examine them during a three-year period. We conclude discussing some ideas for further research. Les résultats du projet de recherche «Étudiants universitaires: classes sociales et représentations» sont ici présentés pour la première fois. Il s"agit d"une observation sociographique continue et permanente de ces étudiants. L"exposé est divisé en quatre parties. La première s"occupe de la problématique de la caractérisation de classe des étudiants et de leur relation avec les familles d"origine et avec leur trajectoire virtuelle future. Les questions opérationnelles relatives aux catégories analytiques des classes sociales des individus et des ménages sont également analysées d"une façon assez détaillée. La deuxième partie «Tableaux transversaux» - procède à l"étude comparative des étudiants de licence en Gestion, en Sociologie et en Anthropologie du ISCTE quant à leur origine familiale et à leurs représentations de la licence et de la profession. La troisième partie «Tableaux diachroniques» reprend les mêmes variables en vue de l"analyse sur trois années consécutives de l"un des cours. L"étude se termine par un débat de quelques lignes sur l"approfondissement et l"extension de la recherche. Thus, it is important to assess the bone health of adolescents in order to prevent osteopenia / osteoporosis, diseases considered as public health issues of great economic impact. This thesis consists of two articles. In the first, the prevalence and factors associated with low bone mineral density (BMD) of adolescents among adolescents was estimated. This study was carried out with 2515 adolescents aged 18-19 years from the São Luís birth cohort of 1997/98. Cross-sectional study where sociodemographic conditions, lifestyle, sleep quality, physical activity and depression were studied. Abstract

Background: LI-RADS version 2018 (v2018) is used for non-invasive diagnosis of hepatocellular carcinoma (HCC). A recently proposed modification (known as mLI-RADS) demonstrated improved sensitivity while maintaining specificity and positive predictive value (PPV) of LI-RADS category 5 (definite HCC) for HCC. However, mLI-RADS requires multicenter validation., Purpose: To evaluate the performance of v2018 and mLI-RADS for liver lesions in a large, heterogeneous, multi-national cohort of patients at risk for HCC., Study Type: Systematic review and meta-analysis using individual participant data (IPD) [Study Protocol: https://osf.io/duys4]., Population: 2223 observations from 1817 patients (includes all LI-RADS categories; females = 448, males = 1361, not reported = 8) at elevated risk for developing HCC (based on LI-RADS population criteria) from 12 retrospective studies., Field Strength/sequence: 1.5T and 3T; complete liver MRI with gadoxetate disodium, including axial T2w images and dynamic axial fat-suppressed T1w images precontrast and in the arterial, portal venous, transitional, and hepatobiliary phases. Diffusion-weighted imaging was used when available., Assessment: Liver observations were categorized using v2018 and mLI-RADS. The diagnostic performance of each system's category 5 (LR-5 and mLR-5) for HCC were compared., Statistical Tests: The Quality Assessment of Diagnostic Accuracy Studies version 2 (QUADAS-2 was applied to determine risk of bias and applicability. Diagnostic performances were assessed using the likelihood ratio test for sensitivity and specificity and the Wald test for PPV. The significance level was P < 0.05., Results: 17% (2/12) of the studies were considered low risk of bias (244 liver observations; 164 patients). Abstract

Microbial colonization and development of infections in wounds is a sign of chronicity. The prevailing approach to manage and treat these wounds involves dressings. However, these often fail in effectively addressing infections, as they struggle to both absorb exudates and maintain optimal local moisture. The system here presented was conceptualized with a three-layer design: the outer layer made of a fibrous polycaprolactone (PCL) film, to act as a barrier for preventing microorganisms and impurities from reaching the wound; the intermediate layer formed of a sodium alginate (SA) hydrogel loaded with ampicillin (Amp) for fighting infections; and the inner layer comprised of a fibrous film of PCL and polyethylene glycol (PEG) for facilitating cell recognition and preventing wound adhesion. Thermal evaluations, degradation, wettability and release behavior testing confirmed the system resistance overtime. The sandwich demonstrated the capability for absorbing exudates (≈70 %) and exhibited a controlled release of Amp for up to 24 h. Antimicrobial testing was performed against Staphylococcus aureus and Escherichia coli, as representatives of Gram-positive and Gram-negative bacteria: >99 % elimination of bacteria. Cell cytotoxicity assessments showed high cytocompatibility levels, confirming the safety of the proposed sandwich system. Adhesion assays confirmed the system ease of detaching without mechanical effort (0.37 N). Abstract

The genus Vigna (Leguminosae) comprises about 150 species grouped into five subgenera. The present study aimed to improve the understanding of karyotype diversity and evolution in Vigna, using new and previously published data through different cytogenetic and DNA content approaches. In the Vigna subgenera, we observed a random distribution of rDNA patterns. The 35S rDNA varied in position, from terminal to proximal, and in number, ranging from one (V. aconitifolia, V. subg. Ceratotropis) to seven pairs (V. unguiculata subsp. unguiculata, V. subg. Vigna). On the other hand, the number of 5S rDNA was conserved (one or two pairs), except for V. radiata (V. subg. Ceratotropis), which had three pairs. Genome size was relatively conserved within the genus, ranging from 1C = 0.43 to 0.70 pg in V. oblongifolia and V. unguiculata subsp. unguiculata, respectively, both belonging to V. subg. Vigna. However, we observed a positive correlation between DNA content and the number of 35S rDNA sites. In addition, data from chromosome-specific BAC-FISH suggest that the ancestral 35S rDNA locus is conserved on chromosome 6 within Vigna. Considering the rapid diversification in the number and position of rDNA sites, such conservation is surprising and suggests that additional sites may have spread out from this ancestral locus., (© 2024. Purpose: To assess the proportion of upper tract urothelial carcinomas (UTUC) that are evident without the excretory phase at CT urography (CTU), and the proportion of potentially avoidable radiation., Methods: UTUCs diagnosed between January 2008-December 2017 were retrospectively identified from a population-based cancer registry. For each patient, US, non-urographic CT, and MRI exams were assessed for a primary mass and secondary imaging findings (hydronephrosis, urinary tract thickening, luminal distention, fat stranding, and lymphadenopathy/metastatic disease). CTUs were assessed for primary and secondary findings, and whether the tumor was evident as a filling defect on excretory phase. The dose-length product (DLP) of potentially avoidable excretory phases was calculated as a fraction of total DLP., Results: 288 patients (mean age, 72±11 years, 165 males) and 545 imaging examinations were included. Of 192 patients imaged with 370 non-urographic CTs, a primary mass was evident in 154 (80.2%), secondary findings were evident in 172 (89.6%), and primary or secondary findings were evident in 179 (93.2%). Of 175 CTUs, primary and secondary findings were evident in 157 (89.7%) and 166 (94.9%) examinations, respectively, and primary or secondary findings were evident in 170/175 (97.1%). 131/175 (74.9%) UTUCs were evident as a filling defect, including the 5/175 (2.9%) UTUCs without primary or secondary findings. Of 144 CTUs with available DLP data, the proportion of potentially avoidable radiation was 103.7/235.8 (44.0%) Gy⋅cm., Conclusion: In our population, almost all UTUCs were evident via primary or secondary imaging findings without requiring the excretory phase.
