The continuous prospection for molecules that may be useful in the development of new therapeutic agents is a highly relevant issue, mainly because the launch of new drugs on the market does not accompany the emergence of new resistant microorganisms. In this context, this work describes the synthesis of new O-alkylamidoximes and the evaluation of its antifungal activity. The new O-alkylamidoximes were prepared using easy synthetic protocols and tested against three Candida species using the broth microdilution method. The synthesized compounds were obtained in moderate to good yields in high purity and without any observable decomposition. All tested compounds shown moderate antifungal activity against at least one strain of Candida. Despite the moderate activity of the new compounds, this was the first report involving the antifungal activity of O-alkylamidoximes. In view of the low chemotherapy arsenal and the development of fungal strains resistant to traditional antifungal agents, the present study opens new possibilities for the preparation of a new class of more active antifungal agents.

This study aimed to evaluate the effectiveness of different Passive Ultrasonic Irrigation (PUI) protocols on debris removal and exposure of dentinal tubules in simulated complex root canal cavities. Twenty single-rooted human mandibular premolars with simulated root canal cavities were filled with the debris and randomly divided into ten groups based on the final irrigation protocol: 1—positive control; 2—negative control; 3—conventional irrigation (CI) with 2.5% sodium hypochlorite (NaOCl); 4—CI with 17% ethylenediaminetetraacetic acid (EDTA) followed by NaOCl; 5—three cycles of PUI for 20 s (NaOCl-NaOCl-NaOCl); 6—three cycles of PUI for 20 s (NaOCl-EDTA-NaOCl); 7—one 60 s PUI cycle (NaOCl); 8—one PUI 180 s cycle (NaOCl); 9—two cycles of PUI for 60 s (EDTA-NaOCl); and 10—two cycles of PUI for 60 s (NaOCl-EDTA). The groups were analyzed by SEM. The Kruskal-Wallis test was used at a 5% level. PUI showed a higher reduction of debris, similar to the positive control group (p > 0.05) and higher than the CI and negative control groups (p < 0.05). Regarding the exposure tubules, the CI groups were similar to the negative control group in all cavities (p > 0.05). The PUI groups were similar to the positive control group (p > 0.05). However, only groups 6, 7, and 10 were statistically different from the CI and negative control (p < 0.05). The protocols using PUI, comprising groups with three cycles of 20 s (NaOCl-EDTA-NaOCl), two cycles of 60 s (EDTA-NaOCl), or one cycle of 60 s (NaOCl), were more effective at removing debris and increasing the exposure of dentinal tubules.

Objective: To assess the efficacy and safety of the use of midazolam as monotherapy, compared to the associated use of midazolam and hydroxyzine for minimum and moderate sedation of children in dental offices, using data obtained from clinical trials. Material and Methods: A systematic review protocol was developed and registered on PROSPERO (CR42020208633). An electronic search was carried out in Pubmed, Lilacs, Science Direct, Open Gray, Web of Science, and central Cochrane Library. No language restrictions were included. Clinical trials were carried out with children aged 0-12 years, using midazolam as monotherapy compared to the use of midazolam associated with hydroxyzine to verify the effectiveness and safety of oral sedation. The quality of the studies was individually assessed and grouped using the RoB 2 (Revised Cochrane risk-of-bias tool for randomized trials) and GRADE (Grading of Recommendations Assessment, Development and Evaluation) systems, respectively. Results: A total of 749 studies were found. After analyzing the inclusion and removal of duplicates, two studies were analyzed for the quality of evidence. Through this analysis, it was possible to verify the very low level of scientific evidence on the superiority of the efficacy and safety of the combined use of midazolam and hydroxyzine for oral sedation in children in dental offices. Conclusion: The conflicting results and limitations of the studies enabled to establish that there is insufficient evidence to support the use of these drugs combined. There is only evidence for the use of midazolam as monotherapy.

Abstract Objective This study aimed to evaluate the cytotoxicity and synergistic effect of epigallocatechin gallate (EGCG) and fosfomycin (FOSFO) on biofilms of oral bacteria associated with endodontic infections. Methodology This study determined minimum inhibitory and bactericidal concentration (MIC/MBC) and fractionated inhibitory concentration (FIC) of EGCG and FOSFO against Enterococcus faecalis, Actinomyces israelii, Streptococcus mutans, and Fusobacterium nucleatum. Monospecies and multispecies biofilms with those bacteria formed in polystyrene microplates and in radicular dentin blocks of bovine teeth were treated with the compounds and control chlorhexidine (CHX) and evaluated by bacterial counts and microscopy analysis. Toxicity effect of the compounds was determined on fibroblasts culture by methyl tetrazolium assays. Results The combination of EGCG + FOSFO demonstrated synergism against all bacterial species, with an FIC index ranging from 0.35 to 0.5. At the MIC/FIC concentrations, EGCG, FOSFO, and EGCG+FOSFO were not toxic to fibroblasts. EGCG+FOSFO significantly reduced monospecies biofilms of E. faecalis and A. israelli, whereas S. mutans and F. nucleatum biofilms were eliminated by all compounds. Scanning electron microscopy of multispecies biofilms treated with EGCG, EGCG+FOSFO, and CHX at 100x MIC showed evident biofilm disorganization and substantial reduction of extracellular matrix. Confocal microscopy observed a significant reduction of multispecies biofilms formed in dentin tubules with 84.85%, 78.49%, and 50.6% of dead cells for EGCG+FOSFO, EGCG, and CHX at 100x MIC, respectively. Conclusion EGCG and fosfomycin showed a synergistic effect against biofilms of oral pathogens related to root canal infections without causing cytotoxicity.

Desde as últimas décadas pesquisas apontam alguns desafios encontrados no Ensino de Física, entre esses estão o ensino descontextualizado, tendo como foco a preparação dos estudantes para testes, resultando numa aprendizagem marcada pela memorização de fórmulas matemáticas e conceitos. Buscando minimizar esses desafios, pesquisas vêm propondo metodologias alternativas com a intenção de auxiliar o professor durante o processo de ensino e aprendizagem. Entre as metodologias propostas, destacamos as sequências de ensino no formato de Unidades de Ensino Potencialmente Significativas (UEPS), essa metodologia didática é fundamentada nos princípios da teoria da aprendizagem significativa. Desse modo, o objetivo dessa pesquisa é identificar as contribuições e limitações das UEPS investigadas em trabalhos científicos por meio de uma Revisão Sistemática da Literatura. Para alcançar esse objetivo realizou-se a busca de publicações por meio do Portal de Busca Integrado da Universidade de São Paulo (USP), utilizando as palavras-chaves "Unidade de Ensino Potencialmente Significativa" e sua terminologia em língua inglesa e espanhola, como também as siglas "UEPS" e "PMTU". A partir de uma triagem inicial e da leitura flutuante dos artigos foram criados critérios de inclusão, exclusão e de qualidade, sendo selecionados 12 artigos. A partir dos resultados da análise, os artigos foram agrupados em duas categorias: formação continuada e construção de conceitos, conforme a finalidade da aplicação da UEPS. Por meio da análise foi possível identificar as contribuições e as limitações das UEPS apresentadas nos artigos, bem como verificar a existência de lacunas presentes na literatura. Para os trabalhos futuros que pretendem adotar a abordagem quantitativa sugere-se a avaliação formativa dos participantes durante toda a aplicação da pesquisa. Além da realização de investigações buscando a formação inicial ou continuada de professores. Bem como pesquisas que investiguem a construção de conceitos nos anos finais do fundamental e na educação de jovens e adultos.

The molecule vitamin C, in the chemical form of ascorbic acid (AsA), is known to be essential for the metabolism of humans and animals. Humans do not produce AsA, so they depend on plants as a source of vitamin C for their food. The AsA synthesis pathway occurs partially in the cytosol, but the last oxidation step is physically linked to the respiratory chain of plant mitochondria. This oxidation step is catalyzed by l-galactono-1,4-lactone dehydrogenase (l-GalLDH). This enzyme is not considered a limiting step for AsA production; however, it presents a distinguishing characteristic: the l-GalLDH can introduce electrons directly into the respiratory chain through cytochrome c (Cytc) and therefore can be considered an extramitochondrial electron source that bypasses the phosphorylating Complex III. The use of Cytc as electron acceptor has been debated in terms of its need for AsA synthesis, but little has been said in relation to its impact on the functioning of the respiratory chain. This work seeks to offer a new view about the possible changes that result of the link between AsA synthesis and the mitochondrial respiration. We hypothesized that some physiological alterations related to low AsA may be not only explained by the deficiency of this molecule but also by the changes in the respiratory function. We discussed some findings showing that respiratory mutants contained changes in AsA synthesis. Besides, recent works that also indicate that the excessive electron transport vial-GalLDH enzyme may affect other respiratory pathways. We proposed that Cytc reduction by l-GalLDH may be part of an alternative respiratory pathway that is active during AsA synthesis. Also, it is proposed that possible links of this pathway with other pathways of alternative electron transport in plant mitochondria may exist. The review suggests potential implications of this relationship, particularly for situations of stress. We hypothesized that this pathway of alternative electron input would serve as a strategy for adaptation of plant respiration to changing conditions.

Objective: To evaluate both knowledge and attitude of public elementary school teachers in Alfenas, Minas Gerais, Brazil, towards Traumatic Dental Injuries (TDIs). Objectives: This systematic review (PROSPERO CRD42021227711) evaluated the influence of diabetes mellitus (DM) on the response of the pulp tissue and in the pulp cells behaviour. Materials and Methods: Searches in PubMed/MEDLINE, Embase, Web of Science and OpenGrey were performed until March 2022. Studies evaluating the effects of DM in the pulp tissue inflammation and in the cell behaviour were included, followed by risk of bias assessment (Methodological Index for Non‐Randomized Studies and SYRCLE's RoB tools). The meta‐analysis was unfeasible, and a narrative synthesis for each outcome was provided. Results: Of the 615 studies, 21 were eligible, mainly with in vivo analysis (16 studies). The pulp inflammation (10 studies) was analysed mainly by haematoxylin–eosin stain; DM increased pulp inflammation/degeneration in 9 studies, especially after dental procedures. The cell viability (5 studies) was analysed mostly using MTT assay; DM and glycating agents decreased cellular viability in 3 studies. DM reduced collagen in all of three studies. There were controversial results regarding mineralization; however, increased alkaline phosphatase was reported in three of four studies. Conclusions: DM seems to increase inflammation/degeneration and mineralization in the pulp tissue while reducing cell proliferation. Abstract Objectives: To evaluate the effect of calcium hydroxide (CH) associated with two different vehicles as a capping material for pulp tissue in primary molars, compared with mineral trioxide aggregate (MTA). Methodology: Forty-five primary mandibular molars with dental caries were treated by conventional pulpotomy using one of the following materials: MTA only (MTA group), CH with saline (CH+saline group) and CH with polyethylene glycol (CH+PEG group) (15 teeth/group). Clinical and periapical radiographic examinations of the pulpotomized teeth were performed 3, 6, and 12 months after treatment. Data were tested by chi-squared analysis and a multiple comparison post-test. Results: The MTA group showed both clinical and radiographic treatment success in 14/14 teeth (100%), at all follow-up appointments. By clinical evaluation, no teeth in the CH+saline and CH+PEG groups had signs of mobility, fistula, swelling or inflammation of the surrounding gingival tissue. However, in the CH+saline group, radiographic analysis detected internal resorption in up to 9/15 teeth (67%), and inter-radicular bone resorption and furcation radiolucency in up to 5/15 teeth (36%), from 3 to 12 months of follow-up. In the CH+PEG group, 2/11 teeth (18%) had internal resorption and 1/11 teeth (9%) presented bone resorption and furcation radiolucency at all follow-up appointments. Conclusion: CH with PEG performed better than CH with saline as capping material for pulpotomy of primary teeth. However, both combinations yielded clinical and radiographic results inferior to those of MTA alone.

RESUMO Com o objetivo de prolongar o período de conservação e manter a qualidade do mamão UENF/ Caliman01, foram testadas diferentes concentrações de CaCl2 aplicadas por infiltração a vácuo. Os frutos foram separados em seis lotes, o controle(sem tratamento) e os tratamentos que receberam a aplicação de CaCl2 a 0%, 2%, 4%, 6% e 8% CaCl2(p/v) por imersão sob vácuo, por 3 minutos, a 50kPa de tensão, com posterior análises da perda de massafresca, firmeza do fruto, firmeza do mesocarpo, ângulo de cor hue, teores de sólidos solúveis (SS), acideztitulável (AT), ácido ascórbico, açúcares solúveis totais e razão SS/AT. Os tratamentos com cálcio não interferiram na perda de massa que, de modo geral, incrementou ao longo do tempo, porém os frutos permaneceram verdes e firmes por mais tempo quando tratados com CaCl2 6% e 8% (p/v). O uso do CaCl2 não interferiu nas características químicas do fruto, tais como sólidos solúveis, acidez titulável e razão SS/AT, porém diminuiu asíntese de ácido ascórbico e a degradação dos açúcares solúveis.

In this work it was performed a selection of fungal strains able to grow in a specific medium containing chitosan (Chitosan Detector Agar) as the sole carbon source at 0.2%, and a kinetic study was performed to evaluate the best conditions for production of chitosanase from the best producing strain. The selection was made from five strains, previously isolated from soil. In the evaluation of kinetics was accompanied by cell concentration, the protein concentration (mg/mL), the enzyme activity (U/mL) for two volumes of fermentation broth (0.05 and 0.10 mL). The results showed a significant increase in all parameters studied in the first 12 hours of culture. Comparing the activity values for the two volumes of fermentation broth was observed that the volume of 0.05 mL broth showed enzymatic activity always greater than the volume of 0.1 mL
