Corydalis bungeana Turcz. is a perennial herb belonging to the family Papaveraceae. Its chloroplast genome was sequenced and characterized. The cp genome of C. bungeana is 167,629 bp long with a GC content of 36.52%. A total of 144 genes were identified in this cp genome, including 79 protein-coding genes, 31 tRNAs and four rRNAs. A phylogenetic tree based on the complete nucleic acid sequence indicated that C. bungeana was classified into Corydaleae and had a close relationship with Lamprocapnos spectabilis.

pH-Zone-refining counter-current chromatography (pH-zone-refining CCC) was successfully applied for the large-scale separation of alkaloids from Corydalis bungeana. The crude extract was separated by a two-phase solvent system composed of petroleum ether–ethyl acetate–methanol–water (5:5:2:8, v/v) where triethylamine (10 mM) was added to the upper organic stationary phase as a retainer and hydrochloric acid (5 mM) to the aqueous mobile phase as a displacer. As a result, 285 mg of protopine, 86 mg of corynoloxine, 430 mg of coryno1ine, and 115 mg of acetylcorynoline were obtained from 3.0 g of crude extract in a one-step separation. The purities of these compounds were 99.1%, 98.3%, 99.0% and 98.5%, respectively, as determined by HPLC. The chemical structures of these isolated compounds were confirmed by ESI-MS, 1H-NMR and 13C-NMR.

Purpose: To investigate the protective effect of corynoline isolated from Corydalis bungeana Turcz on lipopolysaccharides (LPS)-induced sepsis, and determine the possible mechanism of anti-sepsis effect of the isolated corynoline. Methods: Corynoline was extracted by column chromatography. LPS (100 ng/mL) was used to induce the release of TNF-a and IL-6 in RAW 264.7 cells, and the isolated corynoline was added. ELISA method was used to determine the levels of TNF-a and IL- 6. Furthermore, sepsis in mice was established by injection of LPS (2 mg/kg, i.v.), and the levels of TNF-a and IL-6 in plasma were determined by ELISA method. For survival rate test, LPS (15 mg/kg, i.v.) and heat-killed E. coli (1.0 ×1011H CFU/kg, i.v.) were used to establish sepsis in mice model, and the mice were observed in 7 days. Results: The results indicate that corynoline significantly elevated the survival rate of septic mice induced by LPS and heat-killed E. coli, in a dose-dependent manner (p < 0.05). Corynoline decreased the release of TNF-a and IL-6 induced by LPS, in a dose-dependent manner (p < 0.05). Conclusion: Treatment with corynoline significantly inhibits the mortality of LPS-induced septic mice, and the mechanism of action is probably related to the decrease of TNF-a and IL-6 release. Four new isoquinoline alkaloids including a benzophenanthridine alkaloid (1), a morphine derivative (2), a narceine-type alkaloid (3) and a simple isoquinoline alkaloid (4), a new amide alkaloid (5) and a new phthalic acid derivative (6), together with eleven known alkaloids (7 – 17) were obtained from the whole herbs extract of Corydalis bungeana Turcz. Their structures and absolute configurations were elucidated by extensive spectroscopic data analysis including HRESIMS, NMR and electronic circular dichroism (ECD) and ECD calculation. Compounds 1 – 17 were evaluated for dopamine D2 receptor activity in CHO-D2 cells. Among them, 16 showed the highest antagonistic activity on the D2 receptor with an IC 50 value of 2.04 ± 0.01 μM. Compounds 14 and 15 exhibited moderate antagonism with IC 50 values of 13.66 ± 2.28 and 31.72 ± 2.52 μM, respectively.
