6. Impact of cannabidiol and delta-9-tetrahydrocannabinol in the angiogenic role of extravillous trophoblasts

Abstract

Background: Cannabis is the most consumed illicit drug in Europe. Alongside, the medicinal use of cannabinoids is rising. Indeed, a formulation that combines the major phytocannabinoids, cannabidiol (CBD) and delta-9-tetrahydrocannabinol (THC) in a 1:1 proportion (Sativex®), is already available for the management of multiple sclerosis. Nevertheless, a lot remains unknown in relation to cannabinoid’s impact in reproductive health, namely during pregnancy. Our research group already reported the negative effects of isolated CBD and THC in placental trophoblast cells, such as the induction of apoptosis and autophagy [1,2]. Objective: Considering that extravillous trophoblasts (EVT) participate in the remodeling of maternal vessels and produce factors that play a role in angiogenesis during placentation, our goal was to evaluate the effects of CBD and THC, isolated or in combination (1:1), in EVT’s angiogenic role, using the HTR-8/SVneo cell line (ATCC, USA). Methods: HTR-8/SVneo cells were treated with CBD, THC and CBD plus THC (1:1), at a concentration of 2 µM. Their effects on the gene expression of the angiogenesis-related factors VEGFA, PGF, FLT1 and sFLT1 were assessed through RT-PCR. Tube formation assay was used to functionally evaluate HTR-8/SVneo endothelial cell-like behavior. Activation of STAT3 signaling pathway, involved in angiogenesis, was assessed by Western blot. Results: The mRNA levels of PGF were upregulated in CBD and CBD plus THC-treated cells, while VEGFA and FLT1 were not affected. On the other hand, all the treatments increased the expression of the potent anti-angiogenic factor sFLT1 and decreased tube formation. Moreover, STAT3 was activated in all treated cells. Conclusions: Our results demonstrate that CBD and THC, either alone or in combination, may affect placental angiogenesis, through dysregulation of relevant players and interference on the endothelial-behavior of EVT cells. Abstract

Although the detrimental effects of cannabis consumption during gestation are known for years, the vast majority of studies established a link between cannabis consumption and foetal development. The complex maternal-foetal interrelationships within the placental bed are essential for normal pregnancy, and decidua definitively contributes to the success of this process. Nevertheless, the molecular signalling network that coordinates strategies for successful decidualization and placentation are not well understood. The discovery of the endocannabinoid system highlighted new signalling mediators in various physiological processes, including reproduction. It is known that endocannabinoids present regulatory functions during blastocyst development, oviductal transport, and implantation. In addition, all the endocannabinoid machinery was found to be expressed in decidual and placental tissues. Additionally, endocannabinoid’s plasmatic levels were found to fluctuate during normal gestation and to induce decidual cell death and disturb normal placental development. Moreover, aberrant endocannabinoid signalling during the period of placental development has been associated with pregnancy disorders. It indicates the existence of a possible regulatory role for these molecules during decidualization and placentation processes, which are known to be particularly vulnerable. In this review, the influence of the endocannabinoid system in these critical processes is explored and discussed.

Abstract

Abstract: The main endocannabinoids (EC) identified in mammalian tissues are N-arachidonoylethanolamide (AEA, anandamide), and 2-arachidonoylglycerol (2-AG). AEA levels are critical in pregnancy, especially during implantation, decidualization, and placental development. As 2-AG functions in pregnancy are still largely undefined, we hypothesized that it may also have a role during fetoplacental development. We showed that 2-AG is not only present in the rat mesometrial decidua and plasma during fetoplacental development, but that both 2-AG synthesizing (diacylglycerol lipase) and degradation (monoacylglycerol lipase) enzymes are expressed by decidual cells. While lower concentrations of 2-AG induced apoptosis of rat primary decidual cells, via the CB1 receptor, higher concentrations induced a dramatic effect on cell morphology, cell viability and lactate dehydrogenase release, triggered through a mechanism independent of CB1. This study provides evidences that 2-AG fluctuation in maternal tissues throughout normal pregnancy is primarily regulated by its metabolizing enzymes. Together, these data supports the hypothesis that a deregulation of the endocannabinoid system through aberrant cannabinoid signalling may impact normal uterine remodelling process and consequently normal pregnancy. Abstract

The uterine endometrium responds to blastocyst implantation with extensive proliferation and differentiation of stromal cells into decidual cells, forming the antimesometrial and mesometrial decidua. These undergo regression by apoptosis but as this process occurs at different time periods suggest that there is spatially dependent temporal control of apoptosis in these specific regions. To elucidate the role of the mitochondrion-dependent signalling pathway in tissue regression, we investigated the spatial and temporal pattern of expression of the Bcl-2 family members in uterine tissues of the implantation site, from the post-implantation period to parturition. Furthermore, the activities of the initiator caspases-8 and -9, and of the executioner caspase-3 were determined. Overall Bax and Bcl-2 were expressed from day 8 till day 19, whilst Bcl-xL was extinguished by day 16. In the antimesometrial and in the mesometrial decidua both Bcl-2 and Bax declined from days 10 to 12. In the latter Bcl-2 immunoreactivity decreased till the end of pregnancy, whilst for Bax a constant level remained thereafter. The pattern of variation of enzymatic activities throughout pregnancy for all the enzymes was similar, increasing from days 10 to 14 and decreasing towards the end of pregnancy. The increased levels of active caspase-9 correlated with Bax/Bcl-2 and Bcl-xL expression suggesting that the apoptotic mitochondrion-dependent pathway is involved in decidual regression during pregnancy progression. 49. Structure−Activity Relationships of New A,D-Ring Modified Steroids as Aromatase Inhibitors:  Design, Synthesis, and Biological Activity Evaluation

Abstract

Inhibition of aromatase is an efficient approach for the prevention and treatment of breast cancer. New A,D-ring modified steroid analogues of formestane and testolactone were designed and synthesized and their biochemical activity was investigated in vitro in an attempt to find new aromatase inhibitors and to gain insight into their structure−activity relationships (SAR). All compounds tested were less active than formestane. However, the 3-deoxy steroidal olefin 3a and its epoxide derivative 4a proved to be strong competitive aromatase inhibitors (Ki = 50 and 38 nM and IC50 = 225 and 145 nM, respectively). According to our findings, the C-3 carbonyl group is not essential for anti-aromatase activity, but 5α-stereochemistry and some planarity in the steroidal framework is required. Furthermore, modification of the steroidal cyclopentanone D-ring, by construction of a δ-lactone six-membered ring, decreases the inhibitory potency. From the results obtained, it may be concluded that the binding pocket of the active site of aromatase requires planarity in the region of the steroid A,B-rings and the D-ring structure is critical for the binding.

50. Expression of mRNA encoding decidualization-associated protein, a variant of acute-phase α2macroglobulin, by rat uterine tissues during pregnancy and pseudopregnancy

Abstract

The patterns of expression of mRNA encoding decidualization-associated protein (DAP) in the rat uterus from early, mid-, and late stages of pregnancy were examined by in situhybridization to provide an insight into the function of DAP during pregnancy. Parallel studies were performed on rat uteri during the oestrous cycle and oil-induced deciduomata. DAP transcripts were absent in virgin uteri and during the day of implantation (day 5), low in early gestation tissues (days 8, 9) and high during midgestation (days 10–14). Expression of mRNA encoding DAP was first detected on day 8 of pregnancy in decidual cells of the lateral decidual glycogen wing areas. On day 9, mRNA encoding DAP was found in the same area as on day 8 as well as in the outer layer of cells in the antimesometrial decidua adjacent to the myometrium termed the fibrinoid capsula. On days 11 and 12, expression appeared in the decidual cells of the mesometrial decidua, and was particularly strong around the ectoplacental cone and blood vessels that were invaded by the trophoblast. By day 13 and throughout the remainder of pregnancy, expression occurred in the mesometrial decidua and metrial gland. No signal was observed in granulated metrial gland cells. mRNA encoding DAP was also found in the smooth circular muscle coat adjacent to the mesometrial decidua. A similar pattern and cellular localization of expression of mRNA encoding DAP expression was detected during the development of the deciduomata, artificially induced in the absence of a conceptus. The spatial and temporal patterns of expression correlated with a possible role for DAP in mediating fetal–maternal interactions and the establishment of the placental structure.
