Coral reefs globally are experiencing more frequent and severe warming events due to anthropogenic driven climate change. Subtropical reefs experience more seasonal variability than lower latitude reefs making them typically more resilient to climate change. With relatively stable coral cover in comparison to other global coral reefs, Flower Garden Banks National Marine Sanctuary (FGBNMS) in the Gulf of Mexico is a series of 17 reefs and banks located on the continental shelf‐edge containing a variety of shallow (0–30 m) and mesophotic (30–150 m) coral reef habitats. Here, we use satellite data products to associate open ocean Argo float profiles with eddy features over FGBNMS to study the shelf‐edge reef environment spanning nearly two decades (2003–2022). Satellite data show that FGBNMS is frequently influenced (∼15 days/month) by mesoscale eddies. The upper water column variability (0–25 m) is most influenced by the seasonal mixed layer despite eddy interaction. Subsurface seasonal ranges of temperature and salinity are enhanced or suppressed depending on the influence of eddies in relation to the mixed layer depth. Within the mesophotic zone (0–150 m), the largest range of thermal variability between anticyclonic and cyclonic eddies is between 50 and 150 m upwards of 5°C. However, these observed dynamics will likely change as a result of eddy variability associated with projected warming and Loop Current weakening, leading to increased thermal stress in the future. Plain Language Summary: Coral reefs globally are threatened by climate change, yet some reefs are less impacted than others, such as coral reef habitats in Flower Garden Banks National Marine Sanctuary (FGBNMS) located in the northwestern Gulf of Mexico. This study demonstrates an exposure to large temperature and salinity ranges across different depths throughout the year which may aid in the resiliency and longevity of this ecosystem. The Gulf of Mexico contains some of the most energetic eddies, or spinning currents, in the world. While these eddies originate in the open ocean, they are mobile features that move onto FGBNMS over the shelf‐edge ∼15 days/month, translating open ocean physical and biogeochemical signatures. Here we find that the eddies on FGBNMS show significant alterations to temperature and salinity conditions. Importantly, the physical oceanography driving the eddy field is expected to weaken under climate change, potentially threatening this unique shelf‐edge reef system, and subjecting the coral reef habitats to warmer ocean temperatures in the future. The Mediterranean Sea has been sampled irregularly by research vessels in the past, mostly by national expeditions in regional waters. To monitor the hydrographic, biogeochemical and circulation changes in the Mediterranean Sea, a systematic repeat oceanographic survey programme called Med-SHIP was recommended by the Mediterranean Science Commission (CIESM) in 2011, as part of the Global Ocean Ship-based Hydrographic Investigations Program (GO-SHIP). Med-SHIP consists of zonal and meridional surveys with different frequencies, where comprehensive physical and biogeochemical properties are measured with the highest international standards. The first zonal survey was done in 2011 and repeated in 2018. In addition, a network of meridional (and other key) hydrographic sections were designed: the first cycle of these sections was completed in 2016, with three cruises funded by the EU project EUROFLEETS2. Biogeographical classifications of the global ocean generalize spatiotemporal trends in species or biomass distributions across discrete ocean biomes or provinces. These classifications are generally based on a combination of remote-sensed proxies of phytoplankton biomass and global climatologies of biogeochemical or physical parameters. However, these approaches are limited in their capacity to account for subsurface variability in these parameters. The deployment of autonomous profiling floats in the Biogeochemical Argo network over the last decade has greatly increased global coverage of subsurface measurements of bio-optical proxies for phytoplankton biomass and physiology. In this study, we used empirical orthogonal function analysis to identify the main components of variability in a global data set of 422 annual time series of Chlorophyll a fluorescence and optical backscatter profiles. Applying cluster analysis to these results, we identified six biomes within the global ocean: two high-latitude biomes capturing summer bloom dynamics in the North Atlantic and Southern Ocean and four mid- and low-latitude biomes characterized by variability in the depth and frequency of deep chlorophyll maximum formation. We report the distribution of these biomes along with associated trends in biogeochemical and physicochemical environmental parameters. Our results demonstrate light and nutrients to explain most variability in phytoplankton distributions for all biomes, while highlighting a global inverse relationship between particle stocks in the euphotic zone and transfer efficiency into the mesopelagic zone. Measuring the underwater light field is a key mission of the international Biogeochemical-Argo program. Since 2012, 0-250 dbar profiles of downwelling irradiance at 380, 412 and 490 nm besides photosynthetically available radiation (PAR) have been acquired across the globe every 1 to 10 days. The resulting unprecedented amount of radiometric data has been previously quality-controlled for real-time distribution and ocean optics applications, yet some issues affecting the accuracy of measurements at depth have been identified such as changes in sensor dark responsiveness to ambient temperature, with time and according to the material used to build the instrument components. Here, we propose a quality-control procedure to solve these sensor issues to make Argo radiometry data available for delayed-mode distribution, with associated error estimation. The presented protocol requires the acquisition of ancillary radiometric measurements at the 1000 dbar parking depth and night-time profiles. A test on >10,000 profiles from across the world revealed a quality-control success rate >90% for each band. The procedure shows similar performance in re-qualifying low radiometry values across diverse oceanic regions. We finally recommend, for future deployments, acquiring daily 1000 dbar measurements and one night profile per year, preferably during moonless nights and when the temperature range between the surface and 1000 dbar is the largest.

Deep Chlorophyll Maxima (DCM) are ubiquitous features in stratified oceanic systems. Their establishment and maintenance result from hydrographical stability favoring specific environmental conditions with respect to light and nutrient availability required for phytoplankton growth. This stability can potentially be challenged by mesoscale eddies impacting the water column's vertical structure and thus the environmental parameters that condition the subsistence of DCMs. Here, data from the global BGC-Argo float network are collocated with mesoscale eddies to explore their impact on DCMs. We show that cyclonic eddies, by providing optimal light and nutrient conditions, increase the occurrence of DCMs characterized by Deep Biomass Maxima for phytoplankton. In contrast, DCMs in anticyclonic eddies seem to be driven by photoacclimation as they coincide with Deep Acclimation Maxima without biomass accumulation. Despite considerable efforts in prevention and therapy, breast cancer remains a major public health concern worldwide. Numerous studies using breast cancer cell lines have shown the antiproliferative and pro-apoptotic effects of docosahexaenoic acid (DHA). Some studies have also demonstrated the inhibitory effect of DHA on the migration and invasion of breast cancer cells, making DHA a potential anti-metastatic agent. Thus, DHA has shown its potential as a chemotherapeutic adjuvant. However, the molecular mechanisms triggering DHA effects remain unclear, and the aim of this study was to provide a transcriptomic basis for further cellular and molecular investigations. Therefore, MDA-MB-231 cells were treated with 100 µM DHA for 1`2 h or 24 h before RNA-seq analysis. The results show the great impact of DHA-treatment on the transcriptome, especially after 24 h of treatment. The impact of DHA is particularly visible in genes involved in the cholesterol biosynthesis pathway that is strongly downregulated, and the endoplasmic reticulum (ER)-stress response that is, conversely, upregulated. This ER-stress and unfolded protein response could explain the pro-apoptotic effect of DHA. The expression of genes related to migration and invasion (especially SERPINE1 , PLAT , and MMP11 ) is also impacted by DHA. In conclusion, this transcriptomic analysis supports the antiproliferative, pro-apoptotic and anti-invasive effects of DHA, and provides new avenues for understanding its molecular mechanisms., Competing Interests: The authors declare no conflict of interest.

Chuvash polycythemia is an autosomal recessive form of erythrocytosis associated with a homozygous p.Arg200Trp mutation in the von Hippel-Lindau ( VHL ) gene. Since this discovery, additional VHL mutations have been identified in patients with congenital erythrocytosis, in a homozygous or compound-heterozygous state. VHL is a major tumor suppressor gene, mutations in which were first described in patients presenting with VHL disease, which is characterized by the development of highly vascularized tumors. Here, we identify a new VHL cryptic exon (termed E1') deep in intron 1 that is naturally expressed in many tissues. More importantly, we identify mutations in E1' in 7 families with erythrocytosis (1 homozygous case and 6 compound-heterozygous cases with a mutation in E1' in addition to a mutation in VHL coding sequences) and in 1 large family with typical VHL disease but without any alteration in the other VHL exons. In this study, we show that the mutations induced a dysregulation of VHL splicing with excessive retention of E1' and were associated with a downregulation of VHL protein expression. In addition, we demonstrate a pathogenic role for synonymous mutations in VHL exon 2 that altered splicing through E2-skipping in 5 families with erythrocytosis or VHL disease. In all the studied cases, the mutations differentially affected splicing, correlating with phenotype severity. This study demonstrates that cryptic exon retention and exon skipping are new VHL alterations and reveals a novel complex splicing regulation of the VHL gene. Resistance of Escherichia coli to third-generation cephalosporin (3GC) in fecal samples representative of French egg production was studied. The susceptibility to cefotaxime of E. coli isolates obtained by culture on nonselective media was determined. Twenty-two nonsusceptible isolates were obtained (7.51%; 95% confidence interval, 4.49 to 10.54%), the majority of which came from young birds. Most isolates carried a blaCTX-M-1 group gene, and a few carried a blaCMY-2-like gene. Control of 3GC resistance in laying hens is needed.

The changes in the secondary conformation and surface hydrophobicity of beta-lactoglobulin subjected to different thermal treatments were characterized at pH values of 7, 5.5 and 4 using circular dichroism (CD) and hydrophobic dye binding. Heating resulted in a decrease in alpha-helix content with a corresponding increase in random coil at all pH values, this change being more pronounced for small heating times. Heating also resulted in an increase in surface hydrophobicity as a result of partial denaturation, this increase being more pronounced at pH 4. Thermal treatment resulted in a shift of the spread monolayer isotherm at air-water interface to smaller area per molecule due to increased flexibility and more loop formation. Thermal treatment led to an increase in interfacial shear elasticity and viscosity of adsorbed beta-lactoglobulin layer at pH 5.5 and 7. Interfacial shear elasticity, shear viscosity, stability of beta-lactoglobulin stabilized emulsion and average coalescence time of a single droplet at a planar oil-water interface with adsorbed protein layer exhibited a maximum for protein subjected to 15 min heat treatment at pH 7. At pH 5.5, the interfacial shear rheological properties and average single drop coalescence time were maximum for 15 min heat treatment whereas emulsion stability was maximum for 5 min heat treatment. At pH 7, thermal treatment was found to enhance foam stability. Analysis of thin film drainage indicated that interfacial shear rheological properties do not influence thin film drainage.

Dynamics of adsorption of 14C radiolabeled beta-lactoglobulin and alpha-lactalbumin at the air-water interface was investigated through the measurement of surface pressure (pi) and surface concentration (Gamma) via a radiotracer technique. Adsorption was diffusion controlled at short times, the rates of increase of pi and Gamma being lower at longer times because of an energy barrier. At low concentrations, an apparent time lag was observed in the evolution of pi for beta-lactoglobulin but not for alpha-lactalbumin which was shown to be due to the nonlinear nature of the pi-Gamma relationship for the former. The area per molecule of an adsorbed beta-lactoglobulin during the dynamics of adsorption was smaller than that for spread monolayer since beta-lactoglobulin was not fully unfolded during adsorption. For alpha-lactalbumin, however, no such difference in the molecular areas for adsorbed and spread monolayer was observed indicating thereby that alpha-lactalbumin unfolded much more rapidly than beta-lactoglobulin. Evolution of Gamma for alpha-lactalbumin was found to occur in two steps possibly due to the change in the orientation of the adsorbed protein from a side-on to an end-on orientation. A previously developed mechanistic model (G. Narsimhan and F. Uraizee, Biotechnology Prog. 8, 187 (1992)) was improved to account for the presence of hydrophobic patches on the surface of the protein molecule as well as an adsorbed protein layer at the air-water interface. The model predictions agreed quite well with the experimental evolution of Gamma for beta-lactoglobulin and alpha-lactalbumin. The model calculations seem to indicate that alpha-lactalbumin changes its orientation at the air-water interface from side-on to other orientations at higher surface concentrations. This study of the Rennes anti-cancer Center reports the results of a clinical and isotope scan study of Dihydroergotamine-Sandoz in the treatment of post-radiotherapy hyposialorrhoea. Dihydroergotamine was used at an initial dose of 100 drops 3 times per day for two weeks, then 60 drops 3 times per day. Treatment must be continued for several months. The results of such treatment given to 11 patients in comparison with a "control" group of 19 patients were evaluated objectively using isotope studies of salivary function. It was found, in addition, that the interruption of treatment might be associated with a relapse of symptoms, reversible when the drug was restarted. Finally, and in the light of the results of this study, it would appear to be logical to treat patients immediately following the end of radiotherapy without awaiting the first symptoms of hyposialorrhoea.
