Abstract

Wound healing involves both local cells and inflammatory cells. Alkali burn of ocular surface tissue is a serious clinical problem often leading to permanent visual impairment resulting from ulceration, scarring and neovascularization during healing. Behaviors of corneal cells and inflammatory cells are orchestrated by growth factor signaling networks that have not been fully uncovered. Here we showed that adenoviral gene introduction of peroxisome proliferator-activated receptor-[gamma]/(PPAR[gamma]) inhibits activation of ocular fibroblasts and macrophages in vitro and also induced anti-inflammatory and anti-fibrogenic responses in an alkali-burned mouse cornea. PPAR[gamma] overexpression suppressed upregulation of inflammation/scarring-related growth factors and matrix metalloproteinases (MMPs) in macrophages. It also suppressed expression of such growth factors and collagen I[alpha]2 and myofibroblast generation upon exposure to TGF[beta]1. Exogenous PPAR[gamma] did not alter phosphorylation of Smad2, but inhibited its nuclear translocation. PPAR[gamma] overexpression enhanced proliferation of corneal epithelial cells, but not of fibroblasts in vitro. Epithelial cell expression of MMP-2/-9 and TGF[beta]1 and its migration were suppressed by PPAR[gamma] overexpression. In vivo experiments showed that PPAR[gamma] gene introduction suppressed monocytes/macrophages invasion and suppressed the generation of myofibroblasts, as well as upregulation of cytokines/growth factors and MMPs in a healing cornea. In vivo re-epitheliazation with basement membrane reconstruction in the healing, burned, cornea was accelerated by PPAR[gamma]-Ad expression, although PPAR[gamma] overexpression was considered to be unfavorable for cell migration. Abstract

Corneal keratocytes have a remarkable ability to heal the cornea throughout life. Given their developmental origin from the cranial neural crest, we asked whether this regenerative ability was related to the stem cell-like properties of their neural crest precursors. To this end, we challenged corneal stromal keratocytes by injecting them into a new environment along cranial neural crest migratory pathways. The results show that injected stromal keratocytes change their phenotype, proliferate and migrate ventrally adjacent to host neural crest cells. They then contribute to the corneal endothelial and stromal layers, the musculature of the eye, mandibular process, blood vessels and cardiac cushion tissue of the host. However, they fail to form neurons in cranial ganglia or branchial arch cartilage, illustrating that they are at least partially restricted progenitors rather than stem cells. The data show that, even at late embryonic stages, corneal keratocytes are not terminally differentiated, but maintain plasticity and multipotentiality, contributing to non-neuronal cranial neural crest derivatives. Abstract

The phospholipid growth factors (PLGFs), including lysophosphatidic acid (LPA), have been implicated in corneal wound healing. PLGF concentrations and activities are elevated after corneal injury. Using real-time PCR, we quantified receptor mRNA levels in the healing rabbit cornea. In intact corneas, transcripts for [S1P.sub.1], [LPA.sub.1], and [LPA.sub.3] receptor subtypes were detected, as was lipid phosphate phosphatase 1 (LPP1). After wounding, the trend for endothelium and keratocytes was for significant decreases in transcript numbers for the three receptor subtypes, whereas epithelial cells showed increased transcript numbers, except for an [S1P.sub.1] decrease in healing cells. [LPP.sub.1] transcript numbers were decreased in keratocytes and endothelium, although LPP-specific activity was unchanged. LPA-elicited [Ca.sup.2+] transients were significantly reduced in the healing endothelium. Consistent with reduced [LPA.sub.3] receptor numbers, dioctylglycerol pyrophosphate, a selective antagonist, reduced LPA-induced [Ca.sup.2+] transients 2.7-fold in nonwounded epithelium but only 1.5-fold in wound-healing endothelium. These data for the first time establish physiologically relevant differential changes in the expression of PLGF receptor subtypes and provide evidence for the changing role of [LPA.sub.3] receptors in endothelial cells. calcium; lysophosphatidic acid; phospholipid; wound healing; diacylglycerol pyrophosphate
