No. of bitstreams: 0 Previous issue date: 2017-12-01 This experiment compared performance and physiological responses of the offspring from cows supplemented with Ca salts of PUFA or SFA + MUFA during late gestation. Ninety-six multiparous, nonlactating, pregnant Angus × Hereford cows were ranked by BW, BCS, and age and divided into 24 groups of 4 cows/group at the end of their second trimester of gestation (d −7). Cows conceived during the same estrus synchronization + AI protocol, with semen from a single sire; hence, gestation length was 195 d for all cows at the beginning of the experiment (d 0). Groups were randomly assigned to receive (DM basis) 405 g/cow daily of soybean meal in addition to 1) 190 g/cow daily of Ca salts of PUFA based on eicosapentaenoic, docosahexaenoic, and linoleic acids or 2) 190 g/cow daily of Ca salts of SFA + MUFA based on palmitic and oleic acids (CON). Groups were maintained in 2 pastures (6 groups of each treatment/pasture) and received daily 10.1 kg/cow (DM basis) of grass–alfalfa hay. Groups were segregated into 1 of 12 drylot pens (6 by 18 m) and individually offered treatments 3 times/wk from d 0 until calving. Cow BW and BCS were recorded, and blood samples were collected on d −7 of the experiment and also within 12 h after calving. Calf BW was also recorded within 12 h of calving. Calves were weaned on d 280 of the experiment, preconditioned for 45 d (d 280 to 325), transferred to a growing lot on d 325, and moved to a finishing lot on d 445, where they remained until slaughter. At calving, PUFA-supplemented cows had a greater (P < 0.01) proportion (as % of total plasma fatty acids) of PUFA, including linoleic, linolenic, arachidonic, docosapentaenoic, and docosahexaenoic acids. At weaning, calves from CON-supplemented cows were older (P = 0.03), although no treatment differences were detected (P = 0.82) for calf weaning BW. During both growing and finishing phases, ADG was greater (P ≤ 0.06) in calves from PUFA-supplemented cows. Upon slaughter, HCW and marbling were also greater (P ≤ 0.05) in calves from PUFA-supplemented cows. Collectively, these results indicate that supplementing eicosapentaenoic, docosahexaenoic, and linoleic acids to late-gestating beef cows stimulated programming effects on postnatal offspring growth and carcass quality. Therefore, supplementing late-gestating beef cows with Ca salts of PUFA appears to optimize offspring productivity in beef production systems. No. of bitstreams: 0 Previous issue date: 2017-11-01 One hundred eight Angus × Hereford steers, originating from 7 cow–calf were obtained from an auction yard on d −2 and transported by road (800 km; 12 h) to an experimental feedlot facility. Upon arrival on d −1, shrunk BW was recorded and steers were grouped with free-choice access to grass hay, mineral supplement, and water. On d 0, steers were ranked by source and shrunk BW and assigned to 1 of 18 pens (6 steers/pen). Pens were allocated to 1) no immunomodulatory ingredient supplementation during feedlot receiving (CON), 2) supplementation with OmniGen-AF (OMN; 22 g/steer daily, as-fed basis; Phibro Animal Health Corp., Teaneck, NJ) from d 0 to 30, or 3) 2 oral capsules of Stocker Immune Primer on d 0 + 15 g/ steer daily (as-fed basis) of Stocker Preconditioned Premix (Ramaekers Nutrition, Santa Cruz, CA) from d 7 to 30 (IPF). From d 0 to 80, steers had free-choice access to grass hay and water and received a corn-based concentrate. Feed DMI was recorded from each pen, and steers were assessed for bovine respiratory disease (BRD) signs daily. Steers were vaccinated against BRD pathogens on d 0 and 21. Final shrunk BW was recorded on d 81, and blood samples were collected on d 0, 3, 7, 10, 14, 21, 31, 42, 56, and 73. Steer ADG and final BW were greater (P ≤ 0.05) in CON steers than in OMN and IPF steers (1.23, 0.76, and 1.06 kg/d [SEM 0.06], respectively, and 320, 282, and 307 kg [SEM 4], respectively) and (P < 0.01) in IPF steers than in OMN steers. No treatment effects were detected (P ≥ 0.76) for BRD incidence (66 ± 4%) and DMI, whereas G:F was greater (P < 0.01) in OMN steers than in CON steers. Mean plasma cortisol concentration was greater (P = 0.01) in CON steers than in OMN and IPF steers. Plasma haptoglobin concentrations tended (P = 0.10) to be greater in CON steers than in IPF steers on d 3, were greater (P = 0.04) in IPF steers than in CON steers on d 7, and tended (P = 0.10) to be less in OMN steers than in IPF and CON steers on d 21. Blood mRNA expression of interleukin 8 was greater (P ≤ 0.05) in OMN and IPF steers than in CON steers on d 3 and in OMN steers than in CON and IPF steers on d 14. Blood mRNA expression of tumor necrosis-α was greater (P ≤ 0.05) in OMN and IPF steers than in CON steers on d 10. Plasma IGF-I concentrations, serum antibody titers to BRD pathogens, and blood mRNA expression of chemokine ligand 5, cyclooxygenase 2, interleukin 8 receptor, and L-selectin did not differ (P ≥ 0.21) among treatments. Collectively, the immunomodulatory feed ingredients evaluated herein impacted adrenocortical and innate immune responses but failed to mitigate BRD incidence and improve performance of receiving cattle. No. of bitstreams: 0 Previous issue date: 2017-01-01 This experiment compared DMI, ruminal forage degradability, and liquid parameters as well as physiological responses in beef cattle receiving a lipopolysaccharide (LPS) challenge or not. Eight ruminally cannulated Angus × Hereford steers (485 ± 16 kg BW) were housed in individual pens on d −7, ranked by BW, and allocated to 1 of 2 treatments administered on d 0: 1) an intravenous (i.v.) bolus dose (0.5 μg/kg of BW, diluted in 5 mL of 0.9% sterile saline) of bacterial LPS (Escherichia coli 0111:B4) or 2) a 5-mL i.v. injection of 0.9% sterile saline (CON). Steers had free-choice access to mixed alfalfa–grass hay, water, and a commercial vitamin + mineral mix during the experiment (d −7 to 6). Hay DMI was evaluated daily from d −5 to 6. Immediately prior to treatment administration (h 0), polyester bags containing 4 g of ground dietary hay (DM basis) were immersed into the rumen of each steer and incubated for 0, 4, 8, 12, 24, 36, and 48 h for DM and NDF degradability evaluation. Steers were also intraruminally pulse-dosed with 5 g of Co-EDTA immediately prior to treatment administration, and rumen fluid samples were collected at 0, 2, 4, 6, 8, 12, 16, and 24 h for ruminal liquid volume and dilution rate calculations. Blood was collected every 2 h from −2 to 8 h, every 4 h from 8 to 16 h, every 12 h from 24 to 72 h, and every 24 h from 96 to 144 h relative to treatment administration. Values obtained before treatment administration were used as a covariate within each respective analysis. Steers receiving LPS had less (P ≤ 0.03) DMI on d 0 and 1 compared with CON steers. Steers receiving LPS had reduced (P ≤ 0.05) rumen liquid volume and dilution rate as well as ruminal disappearance rate and effective degradability of DM and NDF compared with CON steers. Steers receiving LPS had greater (P ≤ 0.05) plasma tumor necrosis factor α at 2 h, greater plasma hapto-globin from 24 to 72 h, greater plasma cortisol from 12 to 16 h, greater serum NEFA from 6 to 48 h, greater plasma insulin and glucose at 2 h, reduced plasma glucose from 4 to 12 h, greater plasma cholecystokinin at 16 h, and greater plasma leptin concentrations at 8, 12, 16, 36, 48, and 60 h after treatment administration compared with CON steers. Hence, LPS administration transiently reduced DMI in steers via physiological reactions that modulate gastrointestinal motility and satiety centers in the central nervous system, in addition to potential host–microbiome endocrine interactions that impaired ruminal hay DM and NDF degradability. A total of 953 lactating, multiparous, non-pregnant Nelore cows (age = 99 +/- 2 mo; days post-partum = 51.4 +/- 0.3 d; BCS = 5.34 +/- 0.04; BW = 430 +/- 2 kg) were allocated into 8 groups of approximately 120 cows each. Groups were assigned to an estrus synchronization + timed-AI protocol at the beginning of the breeding season. Concurrently with AI, blood samples were collected, hair samples were clipped from the tail switch, and cow temperament was evaluated via chute score and exit velocity. Individual exit score was calculated within each group by dividing exit velocity into quintiles and assigning cows with a score from 1 to 5 (1 = slowest; 5 = fastest cow). Temperament scores were calculated by averaging cow chute score and exit score, and used to define cow temperament ( 3 = excitable, n = 227; EXC). Cows not pregnant to AI were assigned to a second timed-AI protocol (n = 184 ADQ and 72 EXC) or exposed (n = 269 ADQ and 90 EXC) to bulls for 60 d. Pregnancy status was verified 30 d after each AI and 45 d after the breeding season via transrectal ultrasound. Cow age, BW, BCS, and d post-partum at the beginning of the breeding season were similar (P >= 0.27) between ADQ and EXC cows. At first timed-AI, EXC had greater (P < 0.01) serum cortisol but similar (P >= 0.87) serum haptoglobin and hair cortisol concentrations compared with ADQ cows (49.1 vs. 39.1 ng/mL of serum cortisol, SEM = 1.0). Pregnancy rate to first timed-AI tended (P = 0.09) to be less in EXC vs. ADQ cows (41.0 vs. 47.3%; SEM = 3.6), whereas no treatment differences were detected (P >= 0.23) for subsequent pregnancy outcomes. Calving rate was less (P = 0.04) in EXC vs. ADQ cows (68.3 vs. 74.8%; SEM = 2.2), which can be attributed to the greater (P = 0.05) pregnancy loss detected in EXC cows (9.9 vs. 5.9%; SEM = 1.4). Weaning rate tended (P = 0.09) to be less, whereas calf weaning BW and age were less (P = 0.05) in EXC vs. ADQ cows (63.9 vs. 69.4%, SEM = 2.4; 209 vs. 212 d, SEM = 1; 204 vs. 210 kg, SEM = 2). Hence, kg of calf weaned/cow exposed to breeding was reduced (P = 0.04) in EXC vs. ADQ cows (130 vs. 146 kg, SEM = 5). In summary, B. indicus cows with excitable temperament had reduced reproductive performance and overall productivity compared to cohorts with adequate temperament when exposed to timed-AI + natural breeding. No. of bitstreams: 0 Previous issue date: 2016-11-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) The objective of this experiment was to compare hormonal, uterine, and conceptus factors associated with pregnancy establishment in Bos indicus beef cows supplemented or not with Ca salts of soybean oil (CSSO) for 21 d beginning after timed AI. One hundred lactating multiparous Nelore cows were allocated to 20 groups of 5 cows/group and timed inseminated on d 0 of the experiment. After AI, groups were randomly assigned to receive (as-fed basis) 100 g of protein-mineral mix + 100 g of ground corn per cow per day, in addition to 1) 100 g/cow daily of CSSO (n = 10) or 2) 100 g/cow daily of kaolin (CON; rumen-inert indigestible substance; n = 10). Groups were maintained in 4 Panicum maximum pastures (5 groups from the same treatment within each pasture) with ad libitum access to forage. Groups were segregated daily and individually offered treatments from d 0 to 21. Blood samples were collected and transrectal ultrasonography was performed to verify ovulation and corpus luteum (CL) volume immediately before AI (d 0) and on d 7 and 15. After ultrasonography on d 15, 60 cows (30 cows/treatment and 3 cows/group) diagnosed without the presence of a CL on d 0 but with a CL greater than 0.38 cm3 in volume on d 7 and 15 were assigned to conceptus collection via transcervical flushing with PBS followed by endometrial biopsy in the uterine horn ipsilateral to the CL. Additional blood samples were collected for wholeblood RNA extraction (d 20), and pregnancy status was verified by transrectal ultrasonography (d 30) in cows not assigned to conceptus collection. Cows receiving CSSO had greater (P = 0.30) for endometrial mRNA expression of prostaglandin E synthase and cyclooxygenase-2. In summary, post-AI CSSO supplementation to B. indicus beef cows increased plasma concentration of linoleic acid and enhanced pregnancy establishment factors, which included CL development and plasma P4 concentration, conceptus growth, and mRNA expression of interferon-tau as well as blood mRNA expression of interferon-stimulated genes. From d −15 to 7, calves were maintained as a single group on pasture. On d 8, calves were placed into 1 of 18 drylot pens (6 pens/treatment; 5 calves/pen) and fed alfalfa–triticale hay. On d 29, calves were transported 1,440 km in a livestock trailer and unloaded on d 30 at the same feed yard with the same pen arrangement used prior to transport. From d 30 to 75, calves were fed a receiving diet based on alfalfa–triticale hay + corn-based concentrate. Calf BW was recorded on 2 consecutive days (d −15, −14, 0, 1, 28, 29, 75, and 76). Blood samples were collected on d −15, 0, 15, 30, 45, 60, and 75. The EARLY calves had less (P ≤ 0.09) ADG before weaning (d −15 to −1); however, they had greater (P ≤ 0.01) ADG during feedlot receiving (d 30 to 75) compared with calves with the other treatments. During preconditioning (d 0 to 29), CON calves had greater (P ≤ 0.04) DMI compared with EARLY and DELAYED calves. During feedlot receiving, no treatment differences were detected (P ≥ 0.17) for hay or concentrate DMI, G:F, and morbidity and mortality rates. There were no treatment effects on calf BW at weaning and at the end of the preconditioning or receiving periods (P ≥ 0.65). Plasma concentrations of antibodies against Mannheimia haemolytica were greater (P ≤ 0.05) in EARLY calves than in CON and DELAYED calves on d 0, greater (P ≤ 0.04) for CON calves than for EARLY and DELAYED calves on d 15, greater (P ≤ 0.02) in DELAYED and EARLY calves than in CON calves on d 30, and greater (P = 0.03) in EARLY calves than in CON calves on d 75. Pens were assigned to receive forage-based diets containing 1) sulfate sources of Cu, Co, Mn, and Zn (INR); 2) an organic complexed source of Cu, Mn, Co, and Zn (AAC; Availa 4; Zinpro Corporation, Eden Prairie, MN); or 3) no supplemental Cu, Co, Mn, and Zn (CON). Diets were offered from d 0 until calving and formulated to meet requirements for energy, protein, macrominerals, Se, I, and vitamins. Groups were maintained in individual pastures and assigned to the following estrus synchronization + timed AI protocol: a 2-mg injection of estradiol benzoate and an intravaginal progesterone-releasing device (CIDR) on d 0, a 12.5-mg injection of PGF(2 alpha) on d 7, CIDR removal in addition to a 0.6-mg injection of estradiol cypionate on d 9, and timed AI on d 11. No. of bitstreams: 0 Previous issue date: 2016-01-01 This study compared reproductive performance of Bos indicus cows vaccinated against the foot-and-mouth disease (FMD) virus before timed AI or during early pregnancy (Exp. 1), as well as rectal temperature (RT) and plasma concentrations of the acute-phase protein haptoglobin in cattle vaccinated or not against the FMD virus (Exp. 2). Cattle utilized in Exp. 1 and 2 originated from herds with no historical occurrences of FMD and that received vaccination against the FMD virus biannually. In Exp. 1, 604 lactating, multiparous, nonpregnant Nelore cows were randomly assigned on d -31 of the experiment to receive 1) vaccination against the FMD virus on d >= 31 (VACPRE; n = 291) and 2) vaccination against FMD virus on d 30 (VACGEST; n = 313). From d -11 to 0, all cows were assigned to an estrus synchronization + timed AI (d 0) protocol. Pregnancy status to AI was verified on d 30 and 90 via transrectal ultrasonography. A treatment x day interaction was detected (P < 0.01) for pregnancy rates to AI, which were similar (P = 0.17) between VACPRE and VACGEST on d 30 (61.8% vs. 56.2%, respectively; SEM = 2.8) but greater (P < 0.01) for VACPRE on d 90 (59.4% vs. 46.9%, respectively; SEM = 2.8). Pregnancy loss from d 30 to 90 was greater (P < 0.01) in VACGEST compared with VACPRE (16.5% vs. 3.9%, respectively; SEM = 2.2). In Exp. 2, 40 pregnant Nelore females (20 nulliparous and 20 multiparous cows; BCS = 4.73 +/- 0.12) were ranked by parity and assigned to receive (VAC; n = 20) or not receive (NOVAC; n = 20) vaccination against the FMD virus. Blood samples were collected and RT was recorded before (h 0) and 24, 72, 120, and 168 h after treatment administration. Treatment x day interactions were detected (P < 0.01) for RT and plasma haptoglobin. The RT was greater (P < 0.01) in VAC compared with NOVAC at 24 h after treatment administration and was similar (P = 0.31) between treatments at all other sampling hours. Plasma haptoglobin concentration was similar (P = 0.98) between VAC and NOVAC before treatment administration (P = 0.48) and greater (P < 0.01) in VAC at 24, 72, 120, and 168 h after treatment administration. In summary, vaccinating B. indicus beef cows against the FMD virus resulted in a 4-fold increase in pregnancy loss when the vaccine was administered 30 d after timed AI compared with 31 d before timed AI. These outcomes can be associated with inflammatory and acute-phase reactions elicited by the FMD vaccine, which are known to impair pregnancy maintenance in cattle. No. of bitstreams: 0 Previous issue date: 2015-10-01 USDA-NIFA Oregon Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) This study evaluated the effects of oral meloxicam administration on metabolic, inflammatory, and acute-phase responses of beef cattle receiving a lipopolysaccharide (LPS) challenge (Exp. 1; d - 1 to 6) or vaccinated against respiratory pathogens (Exp. 2; d 7 to 21). Twenty-one Angus steers (n = 11) and heifers (n = 10) were housed in individual pens on d - 15 and were offered free-choice water, mineral-vitamin mix, and hay until d 21. In Exp. 1, cattle were ranked on d - 1 by sex and BW and assigned to 1) oral meloxicam administration (1 mg/kg BW daily) from day - 1 to 6 (MEL8), 2) oral meloxicam administration (1 mg/kg BW) on d 0 and oral lactose monohydrate administration (1 mg/kg BW) on d - 1 and from d 1 to 6 (MEL1), or 3) oral lactose monohydrate administration (1 mg/kg BW daily) from d - 1 to 6 (CON). On d 0, cattle received an intravenous LPS bolus (0.5 mu g/kg BW) concurrently with treatment administration. Rectal temperature (RTEMP) was assessed, and blood samples were collected at -2, 0, 2, 4, 6, 8, 12, 16, 24, 36, 48, 60, 72, 96, 120, and 144 h relative to LPS administration. No treatment effects were detected (P >= 0.36) for RTEMP, concentrations of serum tumor necrosis factor a (TNF alpha), plasma haptoglobin, cortisol, insulin, and leptin, as well as blood mRNA expression of TNF alpha and cyclooxygenase-2, although all variables increased (P < 0.01) across treatments after LPS administration. In Exp. 2, cattle received the same treatments that they were assigned to in Exp. 1 from d 7 to d 13 and were vaccinated against respiratory pathogens concurrently with treatment administration on d 8. Blood samples were collected, and RTEMP was assessed as in Exp. 1 in addition to 168, 240, and 336 h relative to vaccination. No treatment effects were detected (P >= 0.26) for RTEMP, the same plasma and serum variables evaluated in Exp. 1, and serum concentrations of antibodies against Mannheimia haemolytica or serum titers against bovine respiratory syncytial virus, bovine herpesvirus-1, bovine viral diarrhea virus-1, and parainfluenza-3 virus. All variables increased (P < 0.01) across treatments after vaccination, except for serum TNF alpha and titers against bovine viral diarrhea virus-1 (P >= 0.40). Collectively, this study found no evidence that oral meloxicam administration, at the doses and intervals utilized herein, mitigated the metabolic, inflammatory, and acute-phase reactions elicited by LPS administration or vaccination against respiratory pathogens.
