Abstract

Research describes the synthesis of porphyrazinediols, their conversion into solitaire complexes, and chemical oxidation of the polydentate noninnocent macrocycles. Results demonstrate that porphyrazinediols are a novel class of redox-active ligands whose diol chelating group is directly attached at the beta-positions of the macrocycle.

Abstract

Polycomb repressive complex 2 (PRC2) has been shown to play a major role in transcriptional silencing in part by installing methylation marks on lysine 27 of histone 3. Dysregulation of PRC2 function correlates with certain malignancies and poor prognosis. EZH2 is the catalytic engine of the PRC2 complex and thus represents a key candidate oncology target for pharmacological intervention. Here we report the optimization of our indole-based EZH2 inhibitor series that led to the identification of CPI-1205, a highly potent (biochemical IC50 = 0.002 μM, cellular EC50 = 0.032 μM) and selective inhibitor of EZH2. This compound demonstrates robust antitumor effects in a Karpas-422 xenograft model when dosed at 160 mg/kg BID and is currently in Phase I clinical trials. Additionally, we disclose the co-crystal structure of our inhibitor series bound to the human PRC2 complex.

10. An Examination of Executive Coaches' Use of Creativity in Their Coaching Practice

Abstract

Executive coaching has become a key strategy for organizations seeking to develop their leaders. Executive coaches who are aware of their creative strengths are positioned to rapidly respond to emerging issues taking place in coaching conversations, and thereby serve the needs of their clients. Limited research is available examining executive coaches and their use of creativity in their coaching practice. This dissertation examined the existing literature through a conceptual framework encompassing three streams of theory, research and practice: (a) bringing awareness to one's creative strengths; (b) the coach/client relationship; and (c) creative tools and methods. This research assessed how executive coaches recognize their own creativity as measured by the Reisman Diagnostic Creativity Assessment instrument, as well as through participant self-report to identify the creative tools and strategies they used in their practice. Additionally, the research analyzed their best practices as shared by study participants. This was a mixed methods study, which surveyed adults in the United States who are members of the International Coach Federation (ICF) and who have agreed to participate in ICF-supported research that advances the coaching industry. Key findings of this study examined differences between creativity and demographic factors, as well as executive coaches' awareness of their own creativity and application of creative tools and best practices. Implications and recommendations for future creativity research are discussed.

Abstract

Lot differences in the biopotency of human menopausal gonadotropin (hMG) were evaluated and the potential biochemical basis was investigated. The in vivo biopotency of hMG was assessed by a unique bioassay that evaluates the number of ova shed in the cyclic hamster in response to hMG administration. Significant variation in hMG lots was observed using this assay. When subjected to chromatofocusing, hMG displayed five immunoreactive follicle-stimulating hormone (FSH) isohormones and nine luteinizing hormone (LH) isohormones. The relative distribution of FSH, but not LH isohormones, was slightly but significantly different between the lots tested. These data indicate that significant differences exist in the ability of commercially available hMG to stimulate follicular development and ovulation. The biochemical basis for these differences in in vivo biopotency remains to be elucidated.
