6. Formylmethionyl-tRNA Binding Properties of <em>Escherichia coli</em> Ribosomal Protein S1.

Abstract

Formylmethionyl-tRNA has been found to bind to Escherichia coli ribosomal protein SI. Com- plexes of S1 and methionine-labeled fMet-tRNAfMet were detected by binding to cellulose nitrate membranes. Complex formation was stimulated by incubation at low ionic strength, at 37°C and in the absence of Mg2+. Protein S1 also bound [3H]poly(rC) under the same conditions or in buffers of higher ionic strength, 10 mM in Mg2+, and during incubation at 0°C. Unlabeled poly(rC) was an effective inhibitor of the binding of labeled fMet-tRNA when both nucleotides were added simultaneously to the reaction mixtures; however, approximately 80% of previously bound f[35S]Met-tRNA remained bound after the subsequent addition of unlabeled poly(rC). Binding of f[3sS]Met- tRNA, but not the binding of [3H]poly(rC), was inhibited by polyamines or aminoglycoside antibiotics. Protein S1 that had been reacted with N-ethylmaleimide failed to bind f[3sS]Met-tRNA but was still capable of binding [3H]poly(rC). The data support the concept of two distinct RNA-binding sites on the S1 molecule. Initiator tRNA appears to occupy a site concerned with the RNA unwinding property of S1, whereas poly(rC) binds primarily at a separate site. During initiation on the ribosome, the poly(rC) site may interact with the 3′ end of 16-S ribosomal RNA, while the other site may interact with either mRNA or fMet-tRNA. [ABSTRACT FROM AUTHOR]

11. Template Properties of Glucose-Deficient T-Even Bacteriophage DNA

Abstract

The vegetative DNA isolated from T4-infected Escherichia coliW4597 (UDPG PPase-) was about two to six times more active in stimulating protein synthesis in cell-free extracts than that isolated from T4-infected E. coliB06. This suggested that nonglucosylated vegetative DNA may be a better template than the glucosylated form. This view was supported by experiments measuring RNA synthesis on mature T-even DNAs with a range of glucose contents. The extent of 14C-GTP polymerization was inversely proportional to the glucose content of the DNA. Differences were also observed in both the kind and quantity of polypeptides produced in response to these DNAs.

Abstract

THE etiology and pathogenesis of arthrogryposis multiplex congenita (AMC) are not well understood. Most investigators accept the concept that the disease is a syndrome with different causes. These may be genetic1 or environmental2 and can be associated with neurological lesions of supranuclear structures3 or of anterior horn cells,4,5 primary muscle disorders,6,7 or failure of muscle development (amyoplasia).8 In an attempt to evolve a unified hypothesis, McKusick9 proposed that several clinical features of AMC, such as changes in the joints or the skin and the occurrence of hernia, suggest a heritable disorder of connective tissue. Consistent with this view is the histopathological study of Hariga et al,10 suggesting that an increase of fibrous tissue precedes muscle fiber degeneration. Bacteriol. 85:141–149. 1963.—Phenylalanine accumulation and its relation to phenylalanine incorporation into trichloroacetic acid-insoluble material in Escherichia coli9723 were studied with a variety of structural analogues of phenylalanine. Inhibition of phenylalanine-C14incorporation was exerted by analogues only when their concentration was sufficient to prevent the formation of 85 to 90% of the accumulated intracellular phenylalanine “pool,” indicating that less than about 15% of the “free amino acid pool” of phenylalanine is essential for maintaining a normal rate of incorporation of phenylalanine into protein. Although certain analogues having high activity in inhibiting accumulation of phenylalanine showed low activity in preventing incorporation, no analogue was found that solely prevented accumulation.

14. Muscle ribosomal protein synthesis in normal pregnancy: implication for carrier detection in Duchenne muscular dystrophy.

Abstract

Muscle samples of rectus abdominis were obtained from 11 normal pregnant women 21-30 years old during elective Caesarean section and from 10 normal, non-pregnant controls of comparable age. The amino-acid incorporation of polyribosomes extracted from the muscle homogenates of the pregnant women showed high activity for the synthesis of non-collagen protein in six cases, while control values were found in five cases. These results indicate that carrier detection in Duchenne muscular dystrophy based on increased muscle ribosomal protein synthesis cannot be done accurately during pregnancy, unless adequate values for carriers at a comparable stage of gestation are available.
