1. High-Resolution HLA-DRB1 Allele Frequencies in a Romanian Cohort of Stem Cell Donors

Abstract

The goal of the current study was to determine the high-resolution frequencies of the HLA-DRB1 alleles among the analyzed Romanian cohort of healthy stem cell donors. Using Next Generation Sequencing (NGS), we estimated class II HLA-DRB1 allele frequencies to a 6-digit resolution through HLA typing in a Romanian cohort of healthy individuals. The study for HLA genotyping included 420 willing donors from the National Registry of Voluntary Hematopoietic Stem Cell Donors (RNDVCSH). In 2020 and 2021, peripheral blood samples were collected and transported to the Fundeni Clinical Institute. We used the Immucor Mia Fora NGS MFlex kit for HLA genotyping. Forty-one different alleles were detected in 420 analyzed samples, out of which the most frequent HLA-DRB1 alleles were DRB1*16:01:01 (12.6%), DRB1*11:04:01 (12.1%) and DRB1*03:01:01 (12%). The HLA-DRB1*11:01:02 and -DRB1*08:04:01, -DRB1*05:01:01, -DRB1*13:05:01, -DRB1*14:07:01, -DRB1*09:01:02, -DRB1*11:02:01, -DRB1*04:07:01, -DRB1*15:03:01, -DRB1*03:02:01, -DRB1*04:06:02, -DRB1*04:08:01, -DRB1*14:05:01 were identified only once. The results revealed similarities with countries belonging to the Eastern Europe, the Balkans and the Caucasus regions. Further studies on larger Romanian cohorts are needed for confirming the current results.

Abstract

The European polecat (Mustela putorius Linnaeus, 1758) is in decline in Romania, often living near human settlements, from mountains to lowlands. They feed on a wide variety of small animals, including rodents, such as mice or rats. The occurrence of this parasite in polecats from Romania was mentioned only once in 1991, but the parasite species was not confirmed by molecular biology. The study aimed to investigate the occurrence of Trichinella spp. in European polecats from Romania and to identify the parasite species by molecular tools. A total of 75 wild European polecats were examined by trichinoscopy and artificial digestion. A large number of animals were examined because of their wide distribution in Romanian territory and their presence near human settlements. For species determination, the positive muscle samples and the larvae recovered from artificial digestion were collected for DNA isolation and further processed by means of Multiplex PCR.

4. Attentional Blink Differences between Adolescent Dyslexic and Normal Readers

Abstract

The goal of this study was to evaluate the possibility that dyslexic individuals require more working memory resources than normal readers to shift attention from stimulus to stimulus. To test this hypothesis, normal and dyslexic adolescents participated in a Rapid Serial Visual Presentation experiment (Raymond, Shapiro, & Arnell, 1992). Surprisingly, the result showed that the participants with dyslexia produced a shallower attentional blink than normal controls. This result may be interpreted as showing differences in the way the two groups encode information in episodic memory. They also fit in a cascade-effect perspective of developmental dyslexia.

Abstract

Background: Autosomal dominant polycystic kidney disease (ADPKD) is the most common monogenic cause of renal failure. For several decades, ADPKD was regarded as an adult-onset disease. In the past decade, it has become more widely appreciated that the disease course begins in childhood. However, evidence-based guidelines on how to manage and approach children diagnosed with or at risk of ADPKD are lacking. Also, scoring systems to stratify patients into risk categories have been established only for adults. Overall, there are insufficient data on the clinical course during childhood. We therefore initiated the global ADPedKD project to establish a large international pediatric ADPKD cohort for deep characterization. Methods: Global ADPedKD is an international multicenter observational study focusing on childhood-diagnosed ADPKD. This collaborative project is based on interoperable Web-based databases, comprising 7 regional and independent but uniformly organized chapters, namely Africa, Asia, Australia, Europe, North America, South America, and the United Kingdom. In the database, a detailed basic data questionnaire, including genetics, is used in combination with data entry from follow-up visits, to provide both retrospective and prospective longitudinal data on clinical, radiologic, and laboratory findings, as well as therapeutic interventions. Discussion: The global ADPedKD initiative aims to characterize in detail the most extensive international pediatric ADPKD cohort reported to date, providing evidence for the development of unified diagnostic, follow-up, and treatment recommendations regarding modifiable disease factors. Moreover, this registry will serve as a platform for the development of clinical and/or biochemical markers predicting the risk of early and progressive disease. Abstract

Background: Autosomal dominant polycystic kidney disease (ADPKD) is the most common monogenic cause of renal failure. For several decades, ADPKD was regarded as an adult-onset disease. In the past decade, it has become more widely appreciated that the disease course begins in childhood. However, evidence-based guidelines on how to manage and approach children diagnosed with or at risk of ADPKD are lacking. Also, scoring systems to stratify patients into risk categories have been established only for adults. Overall, there are insufficient data on the clinical course during childhood. We therefore initiated the global ADPedKD project to establish a large international pediatric ADPKD cohort for deep characterization. Methods: Global ADPedKD is an international multicenter observational study focusing on childhood-diagnosed ADPKD. This collaborative project is based on interoperable Web-based databases, comprising 7 regional and independent but uniformly organized chapters, namely Africa, Asia, Australia, Europe, North America, South America, and the United Kingdom. In the database, a detailed basic data questionnaire, including genetics, is used in combination with data entry from follow-up visits, to provide both retrospective and prospective longitudinal data on clinical, radiologic, and laboratory findings, as well as therapeutic interventions. Discussion: The global ADPedKD initiative aims to characterize in detail the most extensive international pediatric ADPKD cohort reported to date, providing evidence for the development of unified diagnostic, follow-up, and treatment recommendations regarding modifiable disease factors. Moreover, this registry will serve as a platform for the development of clinical and/or biochemical markers predicting the risk of early and progressive disease.

Abstract

The concept of composite tissue allotransplantation (CTA) for restoration of congenital or acquired deformities is not new and the recent success of clinical composite tissue allotransplantation (CTA) attests to the fact that composite tissue allografts have tremendous potential in these life-enhancing reconstructions. A hand transplant, unlike a solid organ transplant, involves multiple tissues (skin, muscle, tendon, bone, cartilage, fat, nerves and blood vessels) and can be considered the 'gold standard' in CTA. In this regard, no other organ or tissue transplant matches the hand transplant in its immunogenicity as well as complexity. Development of assays that allow us to monitor the current state of an immune response (rejection/tolerance) is of great interest and requires an in-depth understanding of the complex and rare phenomenon of tolerance.

Abstract

The immune response consists of two main components: humoral immunity represented by B lymphocytes and cellular immunity maintained by the T lymphocytes. Immunoglobulins, produced by B– lymphocytes, are the main mediators of humoral immunity, and deficiencies at this level affect the body's response to infection. Plasmocytes produce nine antibody izotypes: immunoglobulins G (IgG 1, lgG2, lgG3, lgG4), immunoglobulins M (IgM), immunoglobulins A (lgA1, lgA2), immunoglobulins D (IGD) and immunoglobulins E (IgE). Primary hypogammaglobulinemias are characterized by the occurrence of recurrent infections and, paradoxically, by the occurrence of autoimmune diseases. Characteristic for these diseases is that symptoms occur at 7–9 months after birth, when transplacental antibody titers transmitted from the mother decrease, and the infant's body is unable to synthesize them to normal levels. Primary hypogammaglobulinemias are transmitted genetically, but mutations at the molecular level are still not fully understood. The most common are: Bruton agammaglobulinemia , transient newborn hypogammaglobulinemia, selective immunoglobulin deficiency and variable common immunodeficiency. Treatment consists of monthly antibiotics and immunoglobulins, depending on antibody titers (except for IgA deficiency).

Abstract

Introduction and Objectives. estimating the quality of life in relation to the health status is a helpful tool in assessing the results and determining the efficacy of any form of treatment. The main purpose of this study is to rate the patients' quality of life after renal transplantation. The objectives of this paper are: to describe the demographic characteristics of the included patients, to compare their auto perceived quality of life before and after kidney transplant surgery, and to find and analyze correlations between the demographic characteristics and the quality of life after renal transplantation. Materials and Methods. This observational descriptive study was performed in a longitudinal and prospective manner. 106 patients that had been treated for chronic kidney disease through renal transplantation were included, and they were assessed before surgery, and 1, 3, 6 and 12 months after surgery. In order to evaluate them we used the Quality of Life Index - Kidney Transplant. Results. Most of the patients are males. The quality of life scores for the four individual sub-domains follow the overall score model, slightly dropping one month after transplantation compared to the pre surgery level, then constantly rising going through all the other three assessment moments (3, 6 and 12 months after surgery). Conclusions. This study analyzes the impact of renal transplantation on the quality of life of the chronic kidney disease patients. Abstract

Introduction. Renal transplantation represents the best treatment choice for patients with chronic kidney disease, as it provides a better quality of life in comparison with chronic dialysis. Materials and Methods. This longitudinal prospective study comprises of a number of 106 patients with chronic kidney disease, assessed before and after the renal transplant surgery. The main tool used to accomplish this task was the SF-36 Quality of Life Questionnaire, that rated the autoperceived physical and mental health. Results. Renal transplanted patients have significantly higher scores at the six months postoperative follow-up concerning Physical Functioning (PF) (p=0.003), General Health (GH) (p<0.001), Vitality (VT) (p<0.001), Social Functioning (SF) (p=0.005) and Mental Health (MH) (p=0.014) as opposed to before surgery. Other aspects of the autoperceived health have only improved somewhat, like Bodily Pain (BP) (p=0.102), Physical Role (RP) (p=0.102) and Emotional Role (RE) (p=0.666). The average physical health score at the six months postoperative follow-up is significantly higher as opposed to the average physical health score before transplantation (p<0.001). Also, the average mental health score at the six months postoperative follow-up is significantly higher as opposed to the average mental health score before transplantation (p<0.001), and it has a negative correlation with the age of the patients (p=0.032), meaning that this score improves greater for younger patients, and less for the older ones. Conclusions. After the renal transplant surgery we observed a significant increase of the autoperceived health status, both for physical and mental functions. Abstract

Vibration sources are described for pneumatic hammers used in the mining industry (pick hammers), in boiler shops (riveting hammers), etc., bringing to light the fact that the principal vibration source is the variation in air pressure inside the cylinder. The present state of the art of vibration control of pneumatic hammers as it is practiced abroad, and the solutions adopted for this purpose, are discussed. A new type of pneumatic hammer with a low noise and vibration level is presented.
