A functionalized product (tincture) was developed using blackberries leaves (Rubus caesius L., Fam. Rosaceae, subfam. Rosoideae species) at different stages, since they provide biologically active compounds. Physicochemical analyzes were performed, such as the evaluation of the total poly-phenol content (TPC), the antioxidant potential, evaluation of the a chlorophyll, b chlorophyll and total chlorophyll for the raw material. The influence of solvents type on the extraction and the content of the total polyphenols and the antioxidant potential were also assessed. Moreover, the antibacterial potential against Staphylococcus aureus, Salmonella typhimurium, Salmonella enteritidis and Bacillus cereus was revealed. The green and red leaves registered a decrease in TPC during maceration, meaning the polyphenols were transferred to the tinctures. In the acetone extract of the green leaves, highest TPC of 538.59 mg GAE/100g was determined, with an antioxidant potential of 93.13 %RSA DPPH inhibited, which was not statistically different from the antioxidant values of the extracts obtained with other solvents. The red leaves exhibited the highest antimicrobial activity on the tested strains, except for Salmonella typhimurium, which exhibited resistance towards each tested extract. The current study demonstrates the potential usage of blackberry leaves from sustainable sources.

The widespread and uncontrolled use of anthelmintic products has contributed to the emergence of anthelmintic resistance (AR). This phenomenon globally threatens the productivity and welfare of small ruminants. A questionnaire consisting of 34 questions was handed to 234 goat farmers across Romania to gain insight into control practices against internal parasites and the farmers’ perception of the parasitic infections present in their herds and the efficacy of anthelmintic treatments. The majority of farmers (88.5%) admitted they had never submitted fecal samples for parasitological laboratory analysis, and 77.4% had treated the animals on their own. In general, the farmers dewormed their goats based on visual body weight estimation. Prophylactic anthelmintic treatment was practiced by more than 85% of the farmers. A traditional control approach based on treating the entire herd at fixed time intervals is widespread among Romanian goat and sheep farmers. The most commonly used anthelmintic drugs in the previous 3 years (2021–2023) were benzimidazoles (85.5%) and macrocyclic lactones (81.6%). Poor anthelmintic efficacy was suspected by 14.5% of farmers, and the minority (18.0%) considered internal parasites as a problem in their herds. Regarding the farmers’ perception of the presence of parasites, there was a significant level of uncertainty. This is the first survey carried out in Romanian goat herds, and it provides up-to-date information on practices aimed at controlling internal parasites.

This study compares the effects of two different doses of propofol administered intravenously (IV), in the jugular vein, to red-eared sliders (Trachemys scripta elegans). In this crossover study, 5 or 10 mg/kg propofol was administered to six Trachemys scripta elegans after cannulation of the jugular vein. Each turtle received each dose, G1 (5 mg/kg IV) and G2 (10 mg/kg IV), after a 7-day washout period. The parameters evaluated were heart rate, palpebral reflex, cloacal reflex, muscle relaxation, ease of handling, sensitivity to anterior and posterior pinch stimuli, and possibility of intubation. Additionally, respiratory rate was measured when possible, and the times from propofol administration to full recovery and from intubation to extubation were recorded. None of the turtles in G1 could be intubated, and this dose provided little relaxation and ease of handling, with a duration of effect until full recovery of 12.16 ± 8.32 (SD) min for this group. In G2, five out of the six turtles could be intubated, and the duration of effect was 32.33 ± 5.85 (SD) min. Heart rates were influenced by manipulation for catheter placement. There were statistically significant differences (p value ≤ 0.05) between the two groups in muscle relaxation degree, handling, cloacal reflex, and possibility of intubation. The 5 mg/kg propofol dose was not sufficient to induce anesthesia, even when administered in the jugular vein, in red-eared sliders. A dose of 10 mg/kg IV or higher should be used.

Abstract Background In cattle attempts to evaluate within-herd prevalence of various infectious and parasitic diseases by bulk-tank milk (BTM) testing with ELISA have been made with moderate success. The fact that BTM is composed of variable and unknown volumes of milk from individual lactating animals weakens the relationship between numerical result of the ELISA and the within-herd prevalence. We carried out a laboratory experimental study to evaluate if a pooled milk sample created by mixing an equal volume of individual milk samples from seropositive and seronegative goats, henceforth referred to as an equal-volume milk sample (EVMS), would allow for accurate estimation of within-herd seroprevalence of caprine arthritis-encephalitis (CAE) using 3 different commercial ELISAs. By mixing randomly selected milk samples from seronegative and seropositive goats, 193 EVMS were created – 93 made of seronegative samples and 100 with the proportion of seropositive individual milk samples (EVMS%POS) ranging from 1 to 100%. EVMS%POS could be considered as a proxy for the within-herd seroprevalence. Then, OD of EVMS (ODEVMS) of the 193 EVMS was measured using 3 commercial ELISAs for CAE – 2 indirect and 1 competitive. Results The cut-off values of ODEVMS indicating SRLV infection were determined. The regression functions were developed to link ODEVMS with EVMS%POS. A significant monotonic relationship between ODEVMS measured with 2 commercial indirect ELISAs and EVMS%POS was identified. Two regression models developed on this basis described approximately 90% of variability and allowed to estimate EVMS%POS, when it was below 50%. High ODEVMS indicated EVMS%POS of > 50%. Conclusion Our study introduces the concept of serological testing of EVMS as a method of detecting SRLV-infected herds and estimating the proportion of strongly seropositive goats. Further field studies are warranted to assess practical benefits of EVMS serological testing.

This review provides a synthesis of the last ten years of research on nanodelivery systems used for the delivery of essential oils (EOs), as well as their potential as a viable alternative to antibiotics in human and veterinary therapy. The use of essential oils alone in therapy is not always possible due to several limitations but nanodelivery systems seem to be able to overcome these issues. The choice of the essential oil, as well as the choice of the nanodelivery system influences the therapeutic efficacy obtained. While several studies on the characterization of EOs exist, this review assesses the characteristics of the nanomaterials used for the delivery of essential oils, as well as impact on the functionality of nanodelivered essential oils, and successful applications. Two classes of delivery systems stand out: polymeric nanoparticles (NPs) including chitosan, cellulose, zein, sodium alginate, and poly(lactic-co-glycolic) acid (PLGA), and lipidic NPs including nanostructured lipid carriers, solid lipid NPs, nanoemulsions, liposomes, and niosomes. While the advantages and disadvantages of these delivery systems and information on stability, release, and efficacy of the nanodelivered EOs are covered in the literature as presented in this review, essential information, such as the speed of emergence of a potential bacteria resistance to these new systems, or dosages for each type of infection and for each animal species or humans is still missing today. Therefore, more quantitative and in vivo studies should be conducted before the adoption of EOs loaded NPs as an alternative to antibiotics, where appropriate.

This review article focuses on the use of canine M-mode in veterinary medicine, specifically in assessing the left ventricle measurements in several breeds. It traces the historical development of echocardiography techniques, including A-mode, B-mode, and motion mode (M-mode), which provide accurate unidimensional records of cardiac structures. This article highlights the significance of M-mode measurements in diagnosing stage B2 of MMVD, where left ventricular end-diastolic internal diameter corrected with body weight (LVIDdN) is essential for identifying cardiac enlargement. It also explains the role of M-mode in diagnosing DCM, outlining criteria such as left ventricular dilatation. The authors emphasize the importance of breed-specific reference values for echocardiographic measurements due to variations in somatotype among dogs. This review provides a comprehensive table summarizing M-mode measurements of the left ventricle for 44 different dog breeds, including interventricular septum thickness, left ventricular internal diameter, and left ventricular posterior wall thickness during systole and diastole. This review’s methodology involves compiling data from various scientific literature sources, providing an extensive tabular representation of M-mode measurements for different breeds, ages, and sexes. Overall, this review highlights the critical role of M-mode echocardiography in diagnosing and managing cardiac diseases in dogs, underscores the importance of breed-specific reference values, and presents a comprehensive summary of M-mode measurements for various dog breeds, aiding both clinicians and researchers.

Low-input farming subject’s swine to increased immune stress and subsequent infectious risk, which could be prevented by phytotherapy. Hypopphae rhamnoides is a widespread shrub in both Europe and Asia. The research aimed at investigating the influence of a whole fruit extract on antibacterial effect of the serum in extensively raised, two batches, sea buckthorn in vivo treated/untreated pigs. Scavenging effect over DPPH was used to estimate the antioxidant potential of the sea buckthorn commercial syrup (Steaua Divina©). Minimal inhibiting (MIC) and minimal bactericidal (MBC) capacity of the serum were tested and spectrophotometrically measured against Shigella spp., Kitococcus sedentarius, Staphylococcus werneri and Staphylococcus sciuri. The antioxidant activity of the syrup was intermediate (54.65 %). Sera from treated pigs had bactericidal and bacteriostatic effect on Shigella spp. and S. sciuri, respectively. Shigella spp. growth was inhibited at serum dilutions of ½-1/32 while much lower ½-1/4 dilutions were active against S. sciuri. In untreated pigs, the sera were inefficient in either stopping or totally inhibiting bacterial growth. The MBC and MIC values obtained for the sera of pigs treated with Hippophae rhamnoides syrup supported the positive effect of the in vivo treatment inducive of potential protection against bacterial diseases.

The cutaneous microbiome and also its resistance to antibiotics is exposed to change, depending on different habitat factors. This research investigates the composition of cutaneous microflora and its antibiotic resistance in pigs raised on free range farms (mix breed swine, from low input small farms). Swabs were collected from the skin surface and subjected to classical microbiological methods (simple broth and nutrient agar cultivation, colony isolation and biochemical API identification). The antibacterial resistance to gentamicin, streptomycin, oxitetracycline, tylosin, amoxacillin-clavulanic acid, marbofloxacin, tulatromycin, cefotaxime and doxycycline was estimated by Kirby Bauer method and multiple antibiotic resistance (MAR) index was calculated. Strains from Staphylococcus (sciuri and warnerii), Shigella spp., Kytococcus sedentarius, Salmonella spp. and Citrobacter freundii genera and species were identified in the collected samples. The most resistant was a S. warnerii strain, but the MAR index was high (0.33) in 50% of the strains. The most efficient antibiotic was cefotaxime and the least efficient was oxitetracycline. The results indicated the presence of antibiotic resistant ubiquitous and pathogenic strains in the investigated pigs which need caution, since they could express pathogenicity under appropriate conditions which low input farming system could provide.

Taraxacum officinale of Compositae family, well-known for its therapeutic effects, abundantly grows on Romanian pastures. The research aimed to investigate its alcoholic extract’s effects on cell-mediated immunity in anti-anthrax vaccinated, grazing sheep. Local Turcana ewes kept on pasture from spring to fall were sc vaccinated with R1190 attenuated anthrax strain, and blood was sampled before and 14 days after the vaccination. In vitro cell-mediated immunity was monitored employing phagocytosis and blast transformation tests in the presence of T. officinale alcoholic extract. Florfenicol is a broad-spectrum bacteriostatic antibiotic used exclusively in veterinary medicine in order to treat the pathology of farm and aquatic animals. It is a synthetic fluorinated analog of thiamphenicol and chloramphenicol that functions by inhibiting ribosomal activity, which disrupts bacterial protein synthesis and has shown over time a strong activity against Gram-positive and negative bacterial groups. Florfenicol was also reported to have anti-inflammatory activity through a marked reduction in immune cell proliferation and cytokine production. The need for improvement came from (1) the inappropriate use (to an important extent) of this antimicrobial, which led to serious concerns about florfenicol-related resistance genes, and (2) the fact that this antibiotic has a low water solubility making it difficult to formulate an aqueous solution in organic solvents, and applicable for different routes of administration. This review aims to synthesize the various applications of florfenicol in veterinary medicine, explore the potential use of nanotechnology to improve its effectiveness and analyze the advantages and limitations of such approaches. The review is based on data from scientific articles and systematic reviews identified in several databases.

The avocado seed is an underused waste resulting from the processing of pulp. Polyphenols, fibers, and carotenoids are present in the seed, which also exhibits prophylactic, fungicidal, and larvicidal effects. Developing food products with avocado seed as an ingredient or spice is highly desired for nutritional, environmental, and economic reasons. The present study proposed its valorization in a hot drink, similar to already existing coffee alternatives, obtained by infusing the roasted and grinded avocado seed. The proximate composition of the raw or conditioned avocado seed and that of the novel drink were determined. The total phenolic content was assessed using the Folin-Ciocâlteu method. The total carotenoids were extracted and assessed spectrophotometrically. Starch determination was performed by the Ewers Polarimetric method. The highest content of polyphenols, 772.90 mg GAE/100 g, was determined in the crude seed, while in the drink was as low as 17.55 mg GAE/100 g. However, the proposed drink demonstrated high antioxidant capacity, evaluated through the DPPH method. This might be due to the high content of the total carotenoid compounds determined in the roasted seed (6534.48 µg/100 g). The proposed drink demonstrated high antiproliferative activity on Hs27 and DLD-1 cell lines.

Sciurus vulgaris (the Eurasian red squirrel) is native to Europe and Asia, but due to habitat destruction or fragmentation, interspecific competition, and infectious diseases, especially in European island areas the species finds itself at the brink of extinction. The repopulation of such bare habitats requires healthy squirrel specimens, either translocated from other wild habitats or reintroduced to the wilderness following captive breeding. Captivity, nonetheless, has shown an immense capacity to reshape the structure of wild species’ microbiota, adapting it to the less diverse diet and fewer environmental challenges. Therefore, assessing the differences between “wild” and “captive” microbiota in this species could elucidate if special living conditions are needed in order to augment the survival rate of specimens reintroduced into the wild. Furthermore, the microflora profile of the normal flora of healthy red squirrels raised in captivity could support clinicians in addressing infectious diseases episodes and also raise awareness on the zoonotic risk. Hence, this study aimed at documenting the bacterial species carried by S. vulgaris, disclosing overall similarities and variability patterns of the microbiota identified in individuals from two different living environments. We anticipated that the bacterial community would be less diverse in individuals raised in captivity, owing to their restrictive diet and to unchanging conditions in the enclosure. We also hypothesized that there would be a higher prevalence of zoonotic microorganisms in the captive animals, due to the proximity of humans and of other domestic species. To test this, samples (n = 100) were taken from five body regions of 20 red squirrels, both free-ranging and bred in captivity, processed by classical microbiology techniques, and further identified by biochemical assay (VITEK®2 Compact System). A relatively poor bacterial community, comprising 62 bacterial strains belonging to 18 species and 8 different genera, was identified. Most of these microorganisms were reported for the first time in S. vulgaris. With no discrimination between living environments, the highest prevalence (p < 0.001), was registered in Staphylococcus sciuri (60%; 12/20), followed by Escherichia coli (45%; 9/20) and Bacillus cereus (35%; 7/20). The results suggest unremarkable differences in diversity and richness of the resident aerobic microbiota of S. vulgaris, in relation to the living environment.

Currently, there are three classes of anthelmintics most commonly used in small ruminants: the benzimidazoles (BZs), macrocyclic lactones (MLs) and cholinergic agonists (especially levamisole; LEV). The widespread use of those products has led to the emergence of drug-resistant parasite strains which represents a serious threat to the livestock industry. In the present study, we describe for the first time a case of resistance to anthelmintics in goats in Romania. The study was carried out in 2021 in a dairy goat herd from the Transylvania region. Two types of diagnostic methods were used to confirm anthelmintic resistance (AR). First, the faecal egg count reduction test (FECRT), an in vivo AR diagnostic method, was used to evaluate the efficacy of eprinomectin (EPM). The results of this test were analysed applying two different calculative methods that are used only in treated animals (without the control group). Furthermore, two in vitro methods were used: the egg hatch test (EHT) for the detection of resistance to BZs, and the larval development test (LDT) for detection of resistance to all three classes of anthelmintics. The results of FECRT indicate the resistance of gastrointestinal nematodes (GINs) to EPM in both calculative methods (FECR1 = −88% and FECR2 = −202%). In addition, the results obtained for ivermectin aglycone (IVM-AG) in LDT also indicate resistance to drugs from MLs group, especially avermectins. Similarly, the results of in vitro methods (EHT and LDT) indicate resistance to BZs in this herd. LEV was the only drug that stopped the development of L3 larvae 100% (LDT). H. contortus was the only nematode species found in coproculture after EPM treatment. Furthermore, H. contotus L3 larvae was the only species found in the wells with the highest concentrations of thiabendazole (TBZ) and IVM-AG in LDT. This suggests that resistance to both BZs and MLs was present for that species.

Antimicrobial and multidrug-resistant bacteria are a major problem worldwide and, consequently, the surveillance of antibiotic-resistant bacteria and assessment of the dissemination routes are essential. We hypothesized that migratory birds, coming from various environments, would carry more numerous Vibrio strains than sedentary species, with increased risk to be passed to their contacts or environment in habitats they transit or nest in. Similarly, we presumed that strains from migratory birds will show multidrug resistance. A total of 170 oral and rectal swabs were collected from wild birds captured in different locations of the Danube Delta (Malic, Sfantu-Gheorghe, Letea Forest) and processed using standardized selective media. V. cholerae strains were confirmed by serology and molecular methods and, subsequently, their susceptibility was evaluated. The prevalence of Vibrio species by host species, habitat type, and location was interpreted. The isolated Vibrio species were identified as Vibrio cholerae 14.33%, V. fluvialis 13.33%, V. alginolyticus 12%, V. mimicus 17.33%, V. vulnificus 10.88%, with V. parahaemolyticus and V. metschnikovii (16%) also being prevalent. Of the 76 Vibrio spp. isolates, 18.42% were resistant towards at least three antimicrobials, and 81.57% demonstrated a multidrug resistance phenotype, including mainly penicillins, aminoglycosides, and macrolides. The results of the present study indicate higher numbers of Vibrio strains in migratory (74.66%) than in sedentary birds (25.33%), confirming our hypothesis. Furthermore, the increased pathogenicity of Vibrio spp. strains, isolated from wild migratory and sedentary birds, was confirmed by their increased multiple antibiotic resistance (MAR) index (0.09–0.81).

The field of veterinary medicine needs new solutions to address the current challenges of antibiotic resistance and the need for increased animal production. In response, a multitude of delivery systems have been developed in the last 20 years in the form of engineered nanoparticles (ENPs), a subclass of which are polymeric, biodegradable ENPs, that are biocompatible and biodegradable (pbENPs). These platforms have been developed to deliver cargo, such as antibiotics, vaccines, and hormones, and in general, have been shown to be beneficial in many regards, particularly when comparing the efficacy of the delivered drugs to that of the conventional drug applications. However, the fate of pbENPs developed for veterinary applications is poorly understood. pbENPs undergo biotransformation as they are transferred from one ecosystem to another, and these transformations greatly affect their impact on health and the environment. This review addresses nanoparticle fate and impact on animals, the environment, and humans from a One Health perspective.

Today, zeolite appears as an interesting alternative for the symptomatic treatment of acute diarrhea. Therefore, this study aimed to investigate the properties of activated micronized (5 microns) zeolite clinoptilolite (MZC) from Transylvania, Romania, first by testing it in vitro and then in vivo on calves with diarrhea. To assess the toxic potential of the MZC, we performed a cell cytotoxicity assay using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) technique on primary bovine intestinal epithelial cells (BIECs). The antimicrobial activity of MZC was investigated by measuring the minimal inhibitory concentration (MIC) on Escherichia coli (ATCC 25922). The uncontrolled in vivo study was carried out over 8 days on a fattening farm, with some 650 calves. Selected calves were randomly assigned to four groups of 20 individuals. Several combinations between MZC, chestnut extract, and oxytetracycline were tested. At the beginning of the study, all calves had diarrhea, while at the end of the study, the prevalence of diarrhea was significantly lower in all four groups (p < 0.001), including the ones treated with MZC. Due to its low cytotoxicity on the intestinal cells and with regards to the results we obtained in vivo, MZC may be considered an alternative for the symptomatic treatment of undifferentiated diarrhea in calves.

Objective: The experiment aimed at establishing the effects of an alcoholic Urtica dioica extract on the in vitro blastogenic response in antigen stimulated, immunologically mature chickens. Materials and Methods: Three equal groups (n=17) of 47 days old, Rock x Cornish chickens were subjected to oral administration of: a) 0.5 ml/chicken/ day of an alcoholic stinging nettle extract (I), b) 0.5 ml/chicken/day alcohol (II, solvent control), and c) 0.5 ml chicken/day water (III-environment control) for seven days. All birds were injected with 0.5 ml/bird of a 5% SRBC suspension (days 0 and 7). Leukocyte numbers (Burker Turk method) and their blastogenic capacity (blast transformation test) were monitored on days 0, 7 and 14. The specific response was evaluated against a SRBC lysate. Student's t-test was used to evaluate the statistical significance of the differences. Results and Discussion: The leukocyte numbers increased to from 18,083.33 ±4,879.81/mm3 to 42,833.33±7,547.99/mm3 by day 14. In vitro responses to C. officinalis and E. angustifolia decreased while SRBC response increased in the nettle treated group. Conclusion: The results did not validate the implemented protocol for the alcoholic stinging nettle treatment. Other administration routes, schemes or dosages should be tested to improve the functional capacity of the leukocytes. [ABSTRACT FROM AUTHOR]
