8. Cortinarius subgenus Leprocybe in Europe: expanded Sanger and Next Generation Sequencing unveil unexpected diversity in the Mediterranean

Abstract

Molecular phylogenies in the past decade have demonstrated that the described diversity of Cortinarius is still underestimated, especially outside continental and boreal ecoregions where the genus has been historically investigated. We tackled this issue by revisiting the so far unresolved subgenus Leprocybe, and focused on the largely unexplored Mediterranean hotspot of biodiversity. The sequencing and phylogenetic analysis of 161 vouchered collections from Austria, Cyprus, France, Germany, Italy and Spain, including 16 types, allowed for the delineation of 11 species in this lineage, three of them recognised as new to science and formally introduced as C. jimenezianus, C. selinolens and C. viridans spp. nov., respectively. Interestingly, the newly described species exhibit a strict Mediterranean distribution, and one of them is putatively endemic to the island of Cyprus, highlighting the remarkable potential of this neglected ecoregion to uncover further undescribed diversity of Cortinarius in the future. The present work also unveils 23 synonymies in this subgenus, as well as previously undetected crypticism within C. venetus. Next Generation Sequencing carried out on three old and contaminated holotypes, successfully decrypts their phylogenetic identity, including that of C. leproleptopus, finally settling the long-standing controversy over the taxonomic status of this species. A brief overview of each species in the subgenus is lastly provided and a key is proposed to facilitate the identification of presently known European taxa of Leprocybe in the field.

Abstract

Nomenclatural type definitions are one of the most important concepts in biological nomenclature. Being physical objects that can be re-studied by other researchers, types permanently link taxonomy (an artificial agreement to classify biological diversity) with nomenclature (an artificial agreement to name biological diversity). Two proposals to amend the International Code of Nomenclature for algae, fungi, and plants (ICN), allowing DNA sequences alone (of any region and extent) to serve as types of taxon names for voucherless fungi (mainly putative taxa from environmental DNA sequences), have been submitted to be voted on at the 11th International Mycological Congress (Puerto Rico, July 2018). We consider various genetic processes affecting the distribution of alleles among taxa and find that alleles may not consistently and uniquely represent the species within which they are contained. Should the proposals be accepted, the meaning of nomenclatural types would change in a fundamental way from physical objects as sources of data to the data themselves. Such changes are conducive to irreproducible science, the potential typification on artefactual data, and massive creation of names with low information content, ultimately causing nomenclatural instability and unnecessary work for future researchers that would stall future explorations of fungal diversity. We conclude that the acceptance of DNA sequences alone as types of names of taxa, under the terms used in the current proposals, is unnecessary and would not solve the problem of naming putative taxa known only from DNA sequences in a scientifically defensible way. 15. New species of Hohenbuehelia, with comments on the Hohenbuehelia atrocoerulea – Nematoctonus robustus species complex

Abstract

Four new species of Hohenbuehelia (Fungi: Pleurotaceae) are described in the group of Hohenbuehelia atrocoerulea and Hohenbuehelia grisea. Hohenbuehelia algonquinensis, found on Pinus in Ontario, Canada, may be distinguished macroscopically from bluish collections of H. atrocoerulea and watery grey-brown collections of H. grisea by its coal-black pileus. Hohenbuehelia canadensis, on or associated with Pinus in both Ontario and Alberta, Canada, and Hohenbuehelia nimueae, on Salix in Ontario and Abies in Wyoming, USA, have similarly dark fruiting bodies and were previously misidentified as H. approximans (which we treat as a synonym of H. grisea), H. atrocoerulea, H. mustialensis or H. nigra. The latter species is shown to be a member of Resupinatus, despite the presence of prominent metuloid cystidia in its hymenium. Hohenbuehelia carlothornii has been found in Costa Rica; collections of the sexual fruiting bodies were previously identified as H. grisea and isolates from soil nematodes were identified by the anamorph name Nematoctonus robustus. That name has been treated as a synonym of H. atrocoerulea but, given the genetic and geographic variation within this complex, we transfer it to Hohenbuehelia as a distinct species. Sequences of the nuclear ribosomal DNA internal transcribed spacer region (ITS), the D1/D2 variable region of the large subunit gene, and a portion of the translation elongation factor (TEF1) gene provide good separation and support for these new species. A key to the dimidiate species of Hohenbuehelia of North America and Europe is provided.

In spite of the low kB/k0 ratios, the reactions are inferred to be genuinely base-catalyzed. The susceptibility constants estimated for the various steps of the reaction mechanism(s) have allowed to make clear the limits of the so-called "element effect" criterion.

Abstract

Dye-sensitized solar cells (DSC) recently attracted much attention as promising devices for the conversion of sunlight in electric power.[1] Such cells employ dyes, which act as photosensitizers, together with a suitable inorganic semiconductor, such as TiO2. Historically, the first class of photosensitizers employed in DSC was represented by transition metal complexes, such as the Ru-bipyridyl complex N719.[2] More recently, purely organic sensitizers, not containing any metal atom, have emerged as a cheap and efficient alternative.[3] Such substances are often based on a "donor-?-acceptor" architecture, featuring electron-donating and electron-accepting groups separated by a conjugated spacer. Here, we will present some of the dyes recently prepared in our laboratories, whose general structure (1) is shown in the figure. The synthesis of these compounds will be described together with the results of preliminary experiments conducted on test cells containing the novel sensitizers. Abstract

Cyclopentadienyl and indenyl complexes of the type (eta5-C5H5)Rh(L2) and (eta5-C9H7)Rh(L2) (L2 = chiral diphosphine) have been synthesized and characterized through multinuclear NMR spectroscopy. These formally planar complexes react with methyl iodide to give the pseudotetrahedral methyl derivatives [(eta5-C5H5)Rh(CH3)(L2)]I and [(eta5-C9H7)Rh(CH3)(L2)]I. The stereochemistry of these complexes, as far as the stereogenic metal center is concerned, is inferred by the difference of chemical shifts of the two phosphorus atoms in the P-31 NMR spectra of the diastereomeric pairs. The crystal structure of (R)C,(S)Rh- [ (eta5-C9H7)Rh(Ph2PCH-(CH3)CH2PPh2)(CH3)]BPh4 has been determined and confirms the above assignment. The electrochemical reduction of the same complex is interpreted in terms of a charge-transfer process followed by the homolytic fission of the Rh-CH3 sigma-bond of the electrogenerated rhodium-(II) complex.

Cation palladium complexes of the type [(dppp)Pd(solvent)2][X]2 2 [X = non or weakly coordinating anion; dppp = 1,3-bis(diphenylphosphino)propane] are active catalysts for the carbonylation to ketones, e.g., for the selective formation of E-1,5-diphenylpent-l-en-3-one from styrene; in the presence of water and depending on the acidity conditions dimerisation to the much less catalytically active [(dppp)Pd(mu-OH)2Pd(dppp)]X2 3 takes place, the structure of which was determined by X-ray crystallography.

Abstract

The objective of this study was to investigate correlates of risk for having microalbuminuria in individuals with one or more cardiovascular risk factors.The study involved 1919 individuals who attended general practice settings, were aged 55 to 75 yr, and did not have a history of cardiovascular events or diabetes but had one or more cardiovascular risk factors. A tree-based regression technique and multivariate analysis were used to identify distinct, homogeneous subgroups of patients with different likelihood of having microalbuminuria; interaction between correlates of microalbuminuria and risk for microalbuminuria was also investigated.The prevalence of microalbuminuria was 5.9%. Patients who did not have hypertension and had postload glycemia140 mg/dl showed the lowest prevalence of microalbuminuria (1.9%) and represented the reference class. The likelihood of microalbuminuria was seven times higher in men with hypertension and homeostatic model assessment levels in the upper tertile and four times higher in women with the same characteristics. Individuals with hypertension and lower homeostatic model assessment levels and normotensive individuals with postload glycemiaor = 140 mg/dl had a more than three-fold increased likelihood of having microalbuminuria. Treatment with statins was associated with a 54% reduction in the likelihood of having microalbuminuria, whereas levels of triglycerides150 mg/dl and fibrinogen levels in the upper tertile were associated with a significantly higher risk for microalbuminuria.The likelihood of having microalbuminuria in a population-based study of elderly individuals is strongly related to the interaction between the components of the metabolic syndrome, particularly hypertension, insulin resistance, and impaired glucose tolerance.
