Abstract

Massive stars experience strong mass-loss, producing a dense, H-rich circumstellar medium (CSM). After the explosion, the collision and continued interaction of the supernova (SN) ejecta with the CSM power the light curve through the conversion of kinetic energy into radiation. When the interaction is strong, the light curve shows a broad peak and high luminosity lasting for a relatively long time. Also the spectral evolution is slower, compared to non-interacting SNe. Energetic shocks between the ejecta and the CSM create the ideal conditions for particle acceleration and production of high-energy (HE) neutrinos above 1 TeV. In this paper, we study four strongly-interacting Type IIn SNe: 2021acya, 2021adxl, 2022qml, and 2022wed to highlight their peculiar characteristics, derive the kinetic energy of the explosion and the characteristics of the CSM, infer clues on the possible progenitors and their environment and relate them to the production of HE neutrinos. The SNe analysed in this sample exploded in dwarf, star-forming galaxies and they are consistent with energetic explosions and strong interaction with the surrounding CSM. For SNe 2021acya and 2022wed, we find high CSM masses and mass-loss rates, linking them to very massive progenitors. For SN 2021adxl, the spectral analysis and less extreme CSM mass suggest a stripped-envelope massive star as possible progenitor. SN 2022qml is marginally consistent with being a Type Ia thermonuclear explosion embedded in a dense CSM. The mass-loss rates for all SNe are consistent with the expulsion of several solar masses of material during eruptive episodes in the last few decades before the explosion. Abstract

With the increasing need for effective compounds against cancer or pathogen-borne diseases, the development of new tools to investigate the enzymatic activity of biomarkers is necessary. Among these biomarkers are DNA topoisomerases, which are key enzymes that modify DNA and regulate DNA topology during cellular processes. Over the years, libraries of natural and synthetic small-molecule compounds have been extensively investigated as potential anti-cancer, anti-bacterial, or anti-parasitic drugs targeting topoisomerases. However, the current tools for measuring the potential inhibition of topoisomerase activity are time consuming and not easily adaptable outside specialized laboratories. Here, we present rolling circle amplification-based methods that provide fast and easy readouts for screening of compounds against type 1 topoisomerases. Specific assays for the investigation of the potential inhibition of eukaryotic, viral, or bacterial type 1 topoisomerase activity were developed, using human topoisomerase 1, Leishmania donovani topoisomerase 1, monkeypox virus topoisomerase 1, and Mycobacterium smegmatis topoisomerase 1 as model enzymes. The presented tools proved to be sensitive and directly quantitative, paving the way for new diagnostic and drug screening protocols in research and clinical settings.

Abstract

ObjectiveTo evaluate the evidence regarding the prevalence and risk of bundle branch block (BBB), atrioventricular block (AVB) and pacemaker implantation (PMI) in patients with spondyloarthritis compared to a control group without spondyloarthritis.MethodsA systematic review of the literature was performed using Pubmed (Medline), EMBASE (Elsevier) and Cochrane Library (Wiley) databases until December 2021. The prevalence and risk for AVB, BBB and PMI were analyzed. Cohort, case control and cross-sectional studies in patients ≥18 years meeting the classification criteria for spondyloarthritis were included. The Odds ratio (OR), risk ratio (RR), or Hazard ratio (HR) and prevalence difference were considered as outcomes. Data was synthesized in a previously defined extraction form which included a risk of bias assessment using the Newcastle-Ottawa Scale.ResultsIn total, eight out of 374 studies were included. None of the studies provided results regarding the risk of low grade AVB and BBB in SpA patients. Only indirect results comparing prevalences from low to medium quality studies were found. According to population based registries, the sex and age adjusted HR of AVB was 2.3 (95% CI 1.6–3.3) in ankylosing spondylitis, 2.9 (95% CI 1.8–4.7) in undifferentiated spondyloarthritis and 1.5 (95% CI 1.1 a 1.9) in psoriatic arthritis. The absolute risk for AVB was 0.4% (moderate to high; 95% CI 0.34%-0.69%) for AS, 0.33% (moderate to high; 95% CI 0.21%-0.53%) for uSpA and 0.34% (moderate to high; 95% CI 0.26%-0.45%) for PsA.The RR for PMI in AS patients was 1.3 (95% CI 1.16–1.46) for groups aged between 65 and 69 years, 1.33 (95% CI 1.22–1.44) for 70–75 years, 1.24 (95% CI 1.55–1.33) for 75–79 years and 1.11 (95% CI 1.06–1.17) for groups older than 80 years. The absolute risk for PMI in AS patients was 0.7% (moderate to high risk; 95% CI 0.6–0.8%) for groups aged between 65–69, 1.44% (high risk; 95% CI 1.33–1.6%) for 70–75 years, 2.09% (high risk; 95% CI 2.0–2.2%) for 75–79 years and 4.15% (high risk; 95% CI 4.0–4.3%) for groups older than 80 yearsConclusionsVery few cases of low grade AVB and BBB were observed in observational studies. No study evaluated association measures for low grade AVB and BBB but the differences of prevalence were similar in SpA and control groups even though studies lacked the power to detect statistical differences. According to population based registries there was an approximately two fold-increased risk of high grade AVB in SpA patients. RR for PMI was higher in younger age groups.

Abstract

Black hole complementarity posits that the interior of a black hole is not independent from its Hawking radiation. This leads to an apparent violation of causality: the interior can be acausally affected by operators acting solely on the radiation. We argue that this perspective is misleading and that the black hole interior must be viewed as existing in the causal past of the Hawking radiation, despite the fact that they are spacelike separated in the semiclassical description. Consequently, no operation on the Hawking radiation - no matter how complex - can affect the experience of an infalling observer. The black hole interior and the radiation only appear spacelike separated in the semiclassical description because an infalling observer's ability to access complex information is limited; the chaotic dynamics on the horizon, as viewed from the exterior, then converts any effect caused by such an observer to information in the Hawking radiation which cannot be accessed at the semiclassical level. We arrive at the picture described above by considering a unitary exterior description in which the flow of information is strictly causal, which we extend to apply throughout the entire history of black hole evolution, including its formation. This description uses the stretched event horizon as an inner edge of spacetime, on which the information inside is holographically encoded. We argue that the global spacetime picture arises from coarse graining over black hole microstates, and discuss its relationship with the exterior description.

Abstract

We present optical and near-infrared observations of two Type Ibn supernovae (SNe), SN 2018jmt and SN 2019cj. Their light curves have rise times of about 10 days, reaching an absolute peak magnitude of $M_g$(SN 2018jmt) = $-$19.07 $\pm$ 0.37 and $M_V$(SN 2019cj) = $-$18.94 $\pm$ 0.19 mag, respectively. The early-time spectra of SN 2018jmt are dominated by a blue continuum, accompanied by narrow (600$-$1000 km~s$^{-1}$) He I lines with P-Cygni profile. At later epochs, the spectra become more similar to those of the prototypical SN Ibn 2006jc. At early phases, the spectra of SN 2019cj show flash ionisation emission lines of C III, N III and He II superposed on a blue continuum. These features disappear after a few days, and then the spectra of SN 2019cj evolve similarly to those of SN 2018jmt. The spectra indicate that the two SNe exploded within a He-rich circumstellar medium (CSM) lost by the progenitors a short time before the explosion. We model the light curves of the two SNe Ibn to constrain the progenitor and the explosion parameters. The ejecta masses are consistent with either that expected for a canonical SN Ib ($\sim$ 2 M$_{\odot}$) or those from a massive WR star ($>$ $\sim$ 4 M$_{\odot}$), with the kinetic energy on the order of $10^{51}$ erg. The lower limit on the ejecta mass ($>$ $\sim$ 2 M$_{\odot}$) argues against a scenario involving a relatively low-mass progenitor (e.g., $M_{ZAMS}$ $\sim$ 10 M$_{\odot}$). We set a conservative upper limit of $\sim$0.1 M$_{\odot}$ for the $^{56}$Ni masses in both SNe. From the light curve modelling, we determine a two-zone CSM distribution, with an inner, flat CSM component, and an outer CSM with a steeper density profile. 6. Laboratory confirmation and improved Accuracy of 4f and 5d energy levels of Fe II previously identified from stellar spectra

Abstract

Many energy levels of singly ionised iron (Fe II, $Z=26$) remain uncertain or experimentally unknown. Their identification and spectral line data are required in reliable astrophysical spectral analyses. In motivation for improving the atomic data of Fe II, we analysed emission spectra of a Fe-Ne plasma produced by a Penning discharge lamp recorded by high-resolution Fourier transform spectroscopy in the region 9000-27,000 cm$^{-1}$ (11,111-3704 {\AA}). Semi-empirical transition probability calculations and stellar spectra of Fe II were used to guide the analysis. In total, 24 energy levels of the 3d$^6$4f and 3d$^6$5d configurations of Fe II lying between 122,351-127,881 cm$^{-1}$ were confirmed in the laboratory for the first time, in agreement with their identities proposed by previous investigations involving only stellar spectra. Level energy and wavelength uncertainties of the 24 levels are improved by up to an order-of-magnitude compared to previously published values. These results will enable more reliable application of Fe II in astrophysical spectroscopic analyses and support further investigations of the spectrum and energy levels of Fe II.

Abstract

Atomic short-range order (SRO) in direct-bandgap GeSn for infrared photonics has recently attracted attention due to its notable impact on band structures. However, the SRO in GeSn thin films grown by different methods have hardly been compared. This paper compares SRO in GeSn thin films of similar compositions grown by molecular beam epitaxy (MBE) and chemical vapor deposition (CVD) using atom probe tomography. An $\sim$15% stronger preference for Sn-Sn 1$^{st}$ nearest neighbor (1NN) is observed in MBE GeSn than CVD GeSn for both thin film and quantum well samples with Sn composition ranging from 7 to 20%. Interestingly, samples grown by different deposition tools under the same method (either MBE or CVD) showed remarkable consistency in Sn-Sn 1NN SRO, while MBE vs. CVD showed clear differences. Supported by theoretical modeling, we consider that this difference in SRO originates from the impact of surface termination, where MBE surfaces are exposed to ultrahigh vacuum while CVD surfaces are terminated by H to a good extent. This finding not only suggests engineering surface termination or surfactants during the growth as a potential approach to control SRO in GeSn, but also provides insight into the underlying reasons for very different growth temperature between MBE and CVD that directly impact the strain relaxation behavior.

8. Iso-m Based Adaptive Fractional Order Control With Application to a Soft Robotic Neck

Abstract

This article proposes an adaptive fractional feedback control using recursive least squares algorithm for plant identification and a recent real-time method (iso-m) for fractional controller tuning. The combination of both methods allows keeping the same original performance specifications invariant, combining adaptability and robustness in a single scheme. Thanks to the robust controller, the system performance is maintained around a specified operating point, and due to the adaptive scheme, this operating point is adjusted depending on plant changes. Extensive experimentation of the proposal is carried out in a real platform with non-linear time varying properties, a soft robotic neck made of 3D printer soft materials. The experiments proposed consist in the neck inclination control using tilt sensors installed on the tip. According to expectations, an invariant performance despite plant parameter changes was observed throughout the experiments. The good results obtained in the proposed test platform suggest that the benefits of this control scheme are suitable for other nonlinear time varying applications.

Abstract

Replicating the behavior and movement of living organisms to develop robots which are better adapted to the human natural environment is a major area of interest today. Soft device development is one of the most promising and innovative technological fields to meet this challenge. However, soft technology lacks of suitable actuators, and therefore, development and integration of soft actuators is a priority. This article presents the development and control of a soft robotic neck which is actuated by a flexible Shape Memory Alloy (SMA)-based actuator. The proposed neck has two degrees of freedom that allow movements of inclination and orientation, thus approaching the actual movement of the human neck. The platform we have developed may be considered a real soft robotic device since, due to its flexible SMA-based actuator, it has much fewer rigid parts compared to similar platforms. Weight and motion noise have also been considerably reduced due to the lack of gear boxes, housing and bearings, which are commonly used in conventional actuators to reduce velocity and increase torque.

10. Potential Applications of an Exopolysaccharide Produced by Bacillus xiamenensis RT6 Isolated from an Acidic Environment

Abstract

The Bacillus xiamenensis RT6 strain was isolated and identified by morphological, biochemical and molecular tests from an extreme acidic environment, Rio Tinto (Huelva). Optimisation tests for exopolysaccharide (EPS) production in different culture media determined that the best medium was a minimal medium with glucose as the only carbon source. The exopolymer (EPSt) produced by the strain was isolated and characterised using different techniques (GC-MS, HPLC/MSMS, ATR-FTIR, TGA, DSC). The molecular weight of EPSt was estimated. The results showed that the average molecular weight of EPSt was approximately 2.71 × 104 Da and was made up of a heteropolysaccharide composed of glucose (60%), mannose (20%) and galactose (20%). The EPSt showed antioxidant capabilities that significantly improved cell viability. Metal chelation determined that EPSt could reduce the concentration of transition metals such as iron at the highest concentrations tested. Finally, the emulsification study showed that EPSt was able to emulsify different natural polysaccharide oils, reaching up to an 80% efficiency (olive and sesame oil), and was a good candidate for the substitution of the most polluting emulsifiers. The EPSt was found to be suitable for pharmaceutical and industrial applications.

11. First detection in space of the high-energy isomer of cyanomethanimine: H2CNCN

Abstract

We report the first detection in the interstellar medium of $N$-cyanomethanimine (H$_2$CNCN), the stable dimer of HCN of highest energy, and the most complex organic molecule identified in space containing the prebiotically relevant NCN backbone. We have identified a plethora of $a$-type rotational transitions with 3 $\leq J_\text{up} \leq$ 11 and $K_\text{a} \leq$ 2 that belong to this species towards the Galactic Center G+0.693-0.027 molecular cloud, the only interstellar source showing the three cyanomethanimine isomers (including the $Z$- and $E$- isomers of $C$-cyanomethanimine, HNCHCN). We have derived a total column density for H$_2$CNCN of (2.9$\, \pm \,$0.1)$\times$10$^{12}$ cm$^{-2}$, which translates into a total molecular abundance with respect to H$_2$ of (2.1$\, \pm \,$0.3)$\times$10$^{-11}$. We have also revisited the previous detection of $E$- and $Z$-HNCHCN, and found a total $C/N$-cyanomethanimine abundance ratio of 31.8$\, \pm \,$1.8 and a $Z/E$-HNCHCN ratio of 4.5$\, \pm \,$0.2. While the latter can be explained on the basis of thermodynamic equilibrium, chemical kinetics are more likely responsible for the observed $C/N$-cyanomethanimine abundance ratio, where the gas-phase reaction between methanimine (CH$_2$NH) and the cyanogen radical (CN) arises as the primary formation route., Comment: 22 pages, 8 figures, 3 tables, 2 appendix - Accepted for publication in The Astrophysical Journal

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). Abstract

In this work, we obtain closed expressions for the transfer matrix and the transmittance of electromagnetic waves propagating in finite 1D anisotropic periodic stratified media with an arbitrary number of cells. By invoking the Cayley-Hamilton theorem on the transfer matrix for the electromagnetic field in a periodic stratified media formed by N cells, we obtain a fourth-degree recursive relation for the matrix coefficients that defines the so-called Tetranacci Polynomials. In the symmetric case, corresponding to a unit-cell transfer matrix with a characteristic polynomial where the coefficients of the linear and cubic terms are equal, closed expressions for the solutions to the recursive relation, known as symmetric Tetranacci Polynomials, have recently been derived, allowing us to write the transfer matrix and transmittance in a closed form. We show as sufficient conditions that the $4\times4$ differential propagation matrix of each layer in the binary unit cell, $\Delta$, a) has eigenvalues of the form $\pm p_1$, $\pm p_2$, with $p_1\ne p_2$, and b) its off-diagonal $2\times2$ block matrices possess the same symmetric structure in both layers. Otherwise, the recursive relations are still solvable for any $4\times4$-matrix and provide an algorithm to compute the N-th power of the transfer matrix without carrying out explicitly the matrix multiplication of N matrices. We obtain analytical expressions for the dispersion relation and transmittance, in closed form, for two finite periodic systems: the first one consists of two birefringent uniaxial media with their optical axis perpendicular to the z-axis, and the second consists of two isotropic media subject to an external magnetic field oriented along the z-axis and exhibiting the Faraday effect. In this world of the digital era, in which we are living, one of the fundamental competences that students must acquire is the competence in Computational Thinking (CT). Although there is no general consensus on a formal definition, there is a general understanding of it as a set of skills and attitudes necessary for the resolution, with or without a computer, of problems that may arise in any area of life. Measuring and evaluating which of the CT skills students have acquired is fundamental, and for this purpose, previously validated measuring instruments must be used. In this study, a previously validated instrument is applied to know if the new students in the Engineering Degrees of the University of the Basque Country have the following skills in CT: Critical Thinking, Algorithmic Thinking, Problem Solving, Cooperativity and Creativity.

Abstract

Temperature dependence of vibrational modes in semiconductors depends on lattice thermal expansion and anharmonic phonon-phonon scattering. Evaluating the two contributions from experimental data is not straightforward, especially for epitaxial layers that present mechanical deformation and anisotropic lattice expansion. In this work, a temperature-dependent Raman study in epitaxial Ge and Ge$_{1-x}$Sn$_{x}$ layers is presented. A model is introduced for the Raman mode energy shift as a function of temperature, comprising thermal expansion of the strained lattice and anharmonic corrections. With support of x-ray diffraction, the model is calibrated on experimental data of epitaxial Ge grown on Si and Ge$_{1-x}$Sn$_{x}$ grown on Ge/Si, finding that the main difference between bulk and epitaxial layers is related to the anisotropic lattice expansion. The phonon anharmonicity and other parameters do not depend on dislocation defect density (in the range $7\cdot 10^6$ - $4\cdot 10^8$ cm$^{-2}$) nor on alloy composition in the range 5-14 at.%. The strain-shift coefficient for the main model of Ge and for the Ge-Ge vibrational mode of Ge$_{1-x}$Sn$_{x}$ is weakly dependent on temperature and is around -500 cm$^{-1}$. In Ge$_{1-x}$Sn$_{x}$, the composition-shift coefficient amounts to -100 cm$^{-1}$, independent of temperature and strain., Comment: 8 pages, 5 figures

Abstract

In this paper, a new pseudorandom number generator (PRNG) based on the logistic map has been proposed. To prevent the system to fall into short period orbits as well as increasing the randomness of the generated sequences, the proposed algorithm dynamically changes the parameters of the chaotic system. This PRNG has been implemented in a Virtex 7 field-programmable gate array (FPGA) with a 32-bit fixed point precision, using a total of 510 lookup tables (LUTs) and 120 registers. The sequences generated by the proposed algorithm have been subjected to the National Institute of Standards and Technology (NIST) randomness tests, passing all of them. By comparing the randomness with the sequences generated by a raw 32-bit logistic map, it is shown that, by using only an additional 16% of LUTs, the proposed PRNG obtains a much better performance in terms of randomness, increasing the NIST passing rate from 0.252 to 0.989. Finally, the proposed bitwise dynamical PRNG is compared with other chaos-based realizations previously proposed, showing great improvement in terms of resources and randomness.

17. Structured factor copulas for modeling the systemic risk of European and United States banks

Abstract

In this paper, we employ Credit Default Swaps (CDS) to model the joint and conditional distress probabilities of banks in Europe and the U.S. using factor copulas. We propose multi-factor, structured factor, and factor-vine models where the banks in the sample are clustered according to their geographic location. We find that within each region, the co-dependence between banks is best described using both, systematic and idiosyncratic, financial contagion channels. However, if we consider the banking system as a whole, then the systematic contagion channel prevails, meaning that the distress probabilities are driven by a latent global factor and region-specific factors. In all cases, the co-dependence structure of bank CDS spreads is highly correlated in the tail. The out-of-sample forecasts of several measures of systematic risk allow us to identify the periods of distress in the banking sector over the recent years including the COVID-19 pandemic, the interest rate hikes in 2022, and the banking crisis in 2023.

Abstract

BACKGROUND: Dilated cardiomyopathy (DCM) is one of the most common causes of heart failure. Multiple identified mutations in nexilin (NEXN) have been suggested to be linked with severe DCM. However, the exact association between multiple mutations of Nexn and DCM remains unclear. Moreover, it is critical for the development of precise and effective therapeutics in treatments of DCM. RESULTS: In our study, Nexn global knockout mice and mice carrying human equivalent G645del mutation are studied using functional gene rescue assays. AAV-mediated gene delivery is conducted through systemic intravenous injections at the neonatal stage. Heart tissues are analyzed by immunoblots, and functions are assessed by echocardiography. Here, we identify functional components of Nexilin and demonstrate that exogenous introduction could rescue the cardiac function and extend the lifespan of Nexn knockout mouse models. Similar therapeutic effects are also obtained in G645del mice, providing a promising intervention for future clinical therapeutics. CONCLUSIONS: In summary, we demonstrated that a single injection of AAV-Nexn was capable to restore the functions of cardiomyocytes and extended the lifespan of Nexn knockout and G645del mice. Our study represented a long-term gene replacement therapy for DCM that potentially covers all forms of loss-of-function mutations in NEXN.

20. Corneal retardation time as an ocular hypertension disease indicator

Abstract

Objective. A detailed analysis of the corneal retardation time $\tau$ as a highly related parameter to the intraocular pressure (IOP), and its plausible role as an indicator of ocular hypertension disease. Approach. A simple theoretical expression for $\tau$ is derived within the corneal viscoelastic model of Kelvin-Voigt with 3 elements. This retardation time can be easily calculated from the well-known signal and pressure amplitudes of non-contact tonometers like the Ocular Response Analyzer (ORA). Then, a population-based study was performed where 100 subjects aged from 18 to 30 were analyzed (within this group, about 10% had an elevated IOP with more than 21 mmHg). Main results. A clear relationship between the corneal retardation time and the corneal-compensated intraocular pressure was found, underlying the risk for ocular hypertensive (OHT) subjects with lower $\tau$ values to develop hypertension illnesses (due to the inability of poorly viscoelastic corneas to absorb IOP fluctuations, resulting in probable optic nerve damage). Significance. Our results might provide an useful tool to systematically discern which OHT patients (and even those with normal IOP values) are more likely to suffer glaucoma progression and, consequently, ensure an early diagnosis., Comment: Accepted for publication in Biomedical Physics and Engineering Express

Abstract

This work describes, for the first time, the synthesis of dialkyl (2-arylquinolin-8-yl)phosphonate derivatives. The preparation was carried out through a direct and simple process as a multicomponent Povarov reaction of aminophenylphosphonates, aldehydes, and styrenes and subsequent oxidation with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) or, alternatively, by a cycloaddition reaction between phosphonate aldimines and acetylenes. Based on phosphonate group structural characteristics, considered as phosphorous isosteres of carboxylic heterocycles, they may present interesting biological properties related to cell proliferation. In the current report, a new series of dialkyl (2-arylquinolin-8-yl)phosphonates have been synthesized and their antiproliferative effect evaluated on different human cancer and embryonic cells, as well as on Leishmania infantum parasites, a eukaryotic protist responsible for visceral leishmaniasis. Thereby, the antitumor effect was assessed in human lung adenocarcinoma cells (A549), human ovarian carcinoma cells (SKOV3), and human embryonic kidney cells (HEK293) versus the non-cancerous lung fibroblasts cell line (MRC5). On the other hand, the antileishmanial activity was tested against both stages of L. infantum cell cycle, namely free-living promastigotes and intramacrophage amastigotes, using a primary culture of Balb/c splenocytes to calculate the selectivity index. Besides the antiproliferative and antileishmanial capacities, their behavior as topoisomerase 1B inhibitors has been evaluated as a possible mechanism of action.

Abstract

Background: Eukaryotic topoisomerase 1 is a potential target of anti-parasitic and anti-cancer drugs. Parasites require topoisomerase 1 activity for survival and, consequently, compounds that inhibit topoisomerase 1 activity may be of interest. All effective topoisomerase 1 drugs with anti-cancer activity act by inhibiting the ligation reaction of the enzyme. Screening for topoisomerase 1 targeting drugs, therefore, should involve the possibility of dissecting which step of topoisomerase 1 activity is affected. Methods: Here we present a novel DNA-based assay that allows for screening of the effect of small-molecule compounds targeting the binding/cleavage or the ligation steps of topoisomerase 1 catalysis. This novel assay is based on the detection of a rolling circle amplification product generated from a DNA circle resulting from topoisomerase 1 activity. Results: We show that the binding/cleavage and ligation reactions of topoisomerase 1 can be investigated separately in the presented assay termed REEAD (C|L) and demonstrate that the assay can be used to investigate, which of the individual steps of topoisomerase 1 catalysis are affected by small-molecule compounds. The assay is gel-free and the results can be detected by a simple colorimetric readout method using silver-on-gold precipitation rendering large equipment unnecessary. Conclusion: REEAD (C|L) allows for easy and quantitative investigations of topoisomerase 1 targeting compounds and can be performed in non-specialized laboratories.

As enzymes work in complex biological pathways, it is important to screen for both desired and undesired inhibitory effects. We here report a screening system utilizing specific sensors for tyrosyl-DNA phosphodiesterase 1 (TDP1) and topoisomerase 1 (TOP1) activity to screen in vitro for drugs inhibiting TDP1 without affecting TOP1. As the main function of TDP1 is repair of TOP1 cleavage-induced DNA damage, inhibition of TOP1 cleavage could thus reduce the biological effect of the TDP1 drugs. We identified three new drug candidates of the 1,5-naphthyridine and 1,2,3,4-tetrahydroquinolinylphosphine sulfide families. All three TDP1 inhibitors had no effect on TOP1 activity and acted synergistically with the TOP1 poison SN-38 to increase the amount of TOP1 cleavage-induced DNA damage. Further, they promoted cell death even with low dose SN-38, thereby establishing two new classes of TDP1 inhibitors with clinical potential. Thus, we here report a dual-sensor screening approach for in vitro selection of TDP1 drugs and three new TDP1 drug candidates that act synergistically with TOP1 poisons.

27. Assessing the Viability, Functionality, and Effectiveness of the Concurrent Classroom Modality: A Quantitative Study

Abstract

The study was conducted to evaluate the implementation of the Concurrent Classroom (CC) modality, a type of hybrid learning modality wherein an in-person teacher teaches in-person and online students simultaneously. The study utilized a quantitative research design to assess the implementation of the CC based on its viability, functionality, and effectiveness. Grades 4 to 12 students, teachers, and subject coordinators participated in the study. Quantitative data were obtained from the self-rating survey, classroom observation rating and summative assessment results. Remarks about the implementation of the CC were also solicited. Obtained results show that the CC modality has high viability. It is practical, useful, and suitable for instructional delivery. CC also shows high functionality. It has enabled teachers and students to perform tasks that are expected from them. Lastly, it is an effective modality because it has produced favorable positive learning outcomes. Recommendations are focused on providing a targeted professional development to empower teachers in effectively handling CC classes, focusing on equitable cognitive engagement, active participation, and balanced support and attention for both in-person and online student groups. Also, provision and functionality of required devices, technical equipment, and connectivity in the CC learning environment must be ensured and emphasized. [For the full proceedings, see ED654100.]

Abstract

Despite the continued improvements in pancreas transplant outcomes in recent decades, a subset of recipients experience graft failure and can experience substantial morbidity and mortality. Here, we summarize what is known about the failed pancreas allograft and what factors are important for consideration of retransplantation. The current definition of pancreas allograft failure and its challenges for the transplant community are explored. The impacts of a failed pancreas allograft are presented, including patient survival and resultant morbidities. The signs, symptoms, and medical and surgical management of a failed pancreas allograft are described, whereas the options and consequences of immunosuppression withdrawal are reviewed. Medical and surgical factors necessary for successful retransplant candidacy are detailed with emphasis on how well-selected patients may achieve excellent retransplant outcomes. To achieve substantial medical mitigation and even pancreas retransplantation, patients with a failed pancreas allograft warrant special attention to their residual renal, cardiovascular, and pulmonary function. Future studies of the failed pancreas allograft will require improved reporting of graft failure from transplant centers and continued investigation from experienced centers.

Abstract

Background Best practices to improve diversity, equity, and inclusion (DEI) in the biomedical workforce remain poorly understood. The Accreditation Council for Graduate Medical Education launched the Barbara Ross-Lee, DO, Diversity, Equity, and Inclusion award for sponsoring institutions to celebrate efforts to improve DEI in graduate medical education (GME). Objective To identify themes in practices used by award applicants to improve DEI efforts at their institutions, using a qualitative design. Methods This qualitative study employed an exploratory, inductive approach and constant comparative method to analyze award applications from 2 submission cycles (2020, 2021). Data analysis involved the use of a preliminary codebook of 29 program applications used in a previous study, which was modified and expanded, to perform a subsequent analysis of 12 sponsoring institution applications. Seven adjudication sessions were conducted to ensure coding consistency and resolve disagreements, resulting in the identification of final themes. Results Institutions approaches to advancing DEI resulted from work within 5 themes and 10 subthemes. The themes encompassed organizational commitment (policies that reflect DEI mission), data infrastructure (tracking recruitment, retention, and inclusion efforts), community connection (service-learning opportunities), diverse team engagement (coproduction with residents), and systematic strategies for DEI support throughout the educational continuum. Consistent across themes was the importance of collaboration, avoiding silos, and the need for a comprehensive longitudinal approach to DEI to achieve a diverse GME workforce. Conclusions This qualitative study identified 5 themes that can inform and guide sponsoring institutions in promoting DEI.

Abstract

Research and innovation are highlighted as significant priorities among Higher Education Institutions (HEIs) in the Philippines. However, most HEIs in the country have a poor culture of research and innovation, which results in low academic productivity. This literature review was conducted to understand the dynamics of a teaching, research, and innovative university, discuss the current status of Philippine HEIs in terms of research (R&D)-innovation productivity, and excerpt best practices from leading universities in the country as a benchmark for other higher institutions toward building a culture of research and innovation. This study highlights different viewpoints but reveals one thing in common among the established research universities: they have exemplary administrative leadership and proper institutional framework, with strong qualifications from their faculty to absorb all the resources pouring in, thereby significantly boosting them and yielding a fruitful result in terms of research and innovation. With these, other Philippines HEIs need to build and develop a vital research and innovative culture among their stakeholders to produce tangible outcomes, which can be utilized for industry-related improvements and new market opportunities, and to address the nation's most pressing issues. Without the support and proper compensation within academia, industry, and government, the pool of talents will be put on the shelf. Nevertheless, leveling up from teaching institutions to become a research and innovative academia remains interesting as it depends primarily on the university's vision and actions, which are mostly affected by its internal and external environment. This paper hopes to be a springboard in formulating policy and practice toward enhancing research and innovation efficiency among the Philippine HEI's that could contribute to the overall nationwide progress.

34. A Community of Practice Shares Perspectives on Utilizing Telepresence in Doctoral Education

Abstract

In this paper, we (a) share perspectives from stakeholders who learn and teach in an EdD program located in the United States that utilizes telepresence for distance learning (TDL) in a synchronous, hybrid environment, (b) frame our learning as a community of practice, and (c) report affordances and challenges of this model. TDL students asserted that the telepresence option gave them choice in selecting their doctoral program and increased their social presence and ability to be perceived as real people, and all students recognized the importance of a cohesive community to offer support through technological challenges. Faculty were largely unfamiliar with utilizing telepresence in the classroom initially and acknowledged that revised pedagogies and instructional methods were essential to supporting all students in this model. TDL affords opportunities to increase equity and access in doctoral education; however, technological and logistical challenges must be remediated to ensure a successful learning environment.

Abstract

Heterocyclic nitrogen compounds, including fused 1,5-naphthyridines, have versatile applications in the fields of synthetic organic chemistry and play an important role in the field of medicinal chemistry, as many of them have a wide range of biological activities. In this review, a wide range of synthetic protocols for the construction of this scaffold are presented. For example, Friedländer, Skraup, Semmlere-Wolff, and hetero-Diels-Alder, among others, are well known classical synthetic protocols used for the construction of the main 1,5-naphthyridine scaffold. These syntheses are classified according to the nature of the cycle fused to the 1,5-naphthyridine ring: carbocycles, nitrogen heterocycles, oxygen heterocycles, and sulphur heterocycles. In addition, taking into account the aforementioned versatility of these heterocycles, their reactivity is presented as well as their use as a ligand for metal complexes formation. Finally, those fused 1,5-naphthyridines that present biological activity and optical applications, among others, are indicated.

This review covers the synthesis and reactivity of 1,5-naphthyridine derivatives published in the last 18 years. These heterocycles present a significant importance in the field of medicinal chemistry because many of them exhibit a great variety of biological activities. First, the published strategies related to the synthesis of 1,5-naphthyridines are presented followed by the reactivity of these compounds with electrophilic or nucleophilic reagents, in oxidations, reductions, cross-coupling reactions, modification of side chains or formation of metal complexes. 37. Tight Big-Ms for Optimal Transmission Switching

Abstract

This paper addresses the Optimal Transmission Switching (OTS) problem in electricity networks, which aims to find an optimal power grid topology that minimizes system operation costs while satisfying physical and operational constraints. Existing methods typically convert the OTS problem into a Mixed-Integer Linear Program (MILP) using big-M constants. However, the computational performance of these approaches relies significantly on the tightness of these big-Ms. In this paper, we propose an iterative tightening strategy to strengthen the big-Ms by efficiently solving a series of bounding problems that account for the economics of the OTS objective function through an upper-bound on the generating cost. We also discuss how the performance of the proposed tightening strategy is enhanced if reduced line capacities are considered. Using the 118-bus test system we demonstrate that the proposed methodology outperforms existing approaches, offering tighter bounds and significantly reducing the computational burden of the OTS problem.

Abstract

Following the Tutsi genocide in 1994, various types of literary work sprang up around the horror surrounding this event. Literary testimonies are one of these forms of literature. Reverien Rurangwa's and Yolande Mukagasana's texts are autobiographical accounts of the harrowing images, the pain, and the sorrow that both authors experienced in very different ways. These pieces and these authors must be seen in order to rehumanize us and them. We have to start looking into their words, into their eyes, as Emmanuel Levinas would say, to find ethics. Ethics cradles these two works that bear witness to agony and fear. It spreads through the description of the narrators as ends in themselves (Nkrumah) for the reader to see, here the act of bearing witness is an ethical act itself. But can we find the same kind of ethics in these two quite diverse ways of living the genocide? How can these two texts create an ethical dialogue with their readers? Abstract

The Maximal Covering Location Problem (MCLP) is a classical location problem where a company maximizes the demand covered by placing a given number of facilities, and each demand node is covered if the closest facility is within a predetermined radius. In the cooperative version of the problem (CMCLP), it is assumed that the facilities of the decision maker act cooperatively to increase the customersz' attraction towards the company. In this sense, a demand node is covered if the aggregated partial attractions (or partial coverings) of open facilities exceed a threshold. In this work, we generalize the CMCLP introducing an Ordered Median function (OMf), a function that assigns importance weights to the sorted partial attractions of each customer and then aggregates the weighted attractions to provide the total level of attraction. We name this problem the Ordered Cooperative Maximum Covering Location Problem (OCMCLP). The OMf serves as a means to compute the total attraction of each customer to the company as an aggregation of ordered partial attractions and constitutes a unifying framework for CMCLP models. We introduce a multiperiod stochastic non-linear formulation for the CMCLP with an embedded assignment problem characterizing the ordered cooperative covering. For this model, two exact solution approaches are presented: a MILP reformulation with valid inequalities and an effective approach based on Generalized Benders' cuts. Abstract

The local ordering of atoms in alloys directly has a strong impact on their electronic and optical properties. This is particularly relevant in nonrandom alloys, especially if they are deposited using far from the equilibrium processes, as is the case of epitaxial Ge1-xSnx layers. In this work, we investigate the arrangement of Ge and Sn atoms in optoelectronic grade Ge1-xSnx epitaxial layers featuring a Sn content in the 5-14% range by using polarization-dependent Raman spectroscopy and density-functional-theory calculations. The thorough analysis of the polarization-dependent spectra in parallel and perpendicular configuration allowed us to properly tag all the observed vibrational modes, and to shed light on that associated to disorder-assisted Raman transitions. Indeed, with the help of large-scale atomistic simulations, we were able to highlight how the presence of Sn atoms, that modify the local environments of Ge atoms, gives rise to two spectral features at different Raman shifts, corresponding to distortions of the atomic bonds.
