Background Protein-nanoparticle (NP) interactions dictate properties of nanoconjugates relevant to bionanotechnology. Non-covalent adsorption generates a protein corona (PC) formed by an inner and an outer layer, the hard and soft corona (HC, SC). Intrinsically disordered proteins (IDPs) exist in solution as conformational ensembles, whose response to the presence of NPs is not known. Methods Three IDPs (α-casein, Sic1 and α-synuclein) and lysozyme are compared, describing conformational properties inside HC on silica NPs by circular dichroism (CD) and Fourier-transform infrared (FTIR) spectroscopy. Results IDPs inside HC are largely unstructured, but display small, protein-specific conformational changes. A minor increase in helical content is observed for α-casein and α-synuclein, reminiscent of membrane effects on α-synuclein. Frozen in their largely disordered conformation, bound proteins do not undergo folding induced by dehydration, as they do in their free forms. While HC thickness approaches the hydrodynamic diameter of the protein in solution for lysozyme, it is much below the respective values for IDPs. NPs boost α-synuclein aggregation kinetics in a dose-dependent manner. Conclusions IDPs maintain structural disorder inside HC, experiencing minor, protein-specific, induced folding and stabilization against further conformational transitions, such as formation of intermolecular beta-sheets upon dehydration. The HC is formed by a single layer of protein molecules. SC likely plays a key role stabilizing amyloidogenic α-synuclein conformers. General significance Protein-NP interactions can mimic those with macromolecular partners, allowing dissection of contributing factors by rational design of NP surfaces. Application of NPs in vivo should be carefully tested for amyloidogenic potential.

Nanoparticles are normally classified as “hard”, mainly consisting of metal or metal oxide cores, or “soft”, including polymer-based, liposomes and biomimetic nanoparticles. Soft nanoparticles have been studied in depth for drug formulation and therapeutic delivery applications, albeit hard nanoparticles may offer easier synthesis, smaller size and more effective tumor penetration. Among them, silica nanoparticles maintain excellent biocompatibility and biodegradability and can be finely adjusted in size and shape, easily produced in a large scale and functionalized or loaded with active molecules. To help filling the gap of a poor clinical translation of hard nanoparticles, we have designed and developed three different nonporous silica nanocarriers loading the chemotherapeutic doxorubicin within the core matrix, on the surface or both inside and outside, respectively. A comparative study was performed on drug loading and drug release, silica matrix degradation and nanodrug cytotoxic activity, highlighting unexpected correlation between the strategy adopted for drug incorporation and nanoparticle behavior in a physiological environment. This study offers a new insight on the impact of the choice of the prodrug nanoparticles on the kinetics and efficacy of drug delivery, which may encourage the scientific community in developing a new generation of drug delivery systems based on hard nanocarriers.

In clinic, the detection of HER2 in primary tumors and in their metastases is currently based on invasive methods. Recently, nuclear molecular imaging techniques, including positron emission tomography and single photon emission computed tomography (SPECT), allowed the detection of HER2 lesions in vivo. We have developed a 99mTc-radiolabeled nanosilica system, functionalized with a trastuzumab half-chain, able to act as drug carrier and SPECT radiotracer for the identification of HER2-positive breast cancer cells. To this aim, nanoparticles functionalized or not with trastuzumab half-chain, were radiolabeled using the 99mTc-tricarbonyl approach and evaluated in HER2 positive and negative breast cancer models. Cell uptake experiments, combined with flow cytometry and fluorescence imaging, suggested that active targeting provides higher efficiency and selectivity in tumor detection compared to passive diffusion, indicating that our radiolabeling strategy did not affect the nanoconjugate binding efficiency. Ex vivo biodistribution of 99mTc-nanosilica in a SK-BR-3 (HER2+) tumor xenograft at 4 h postinjection was higher in targeted compared to nontargeted nanosilica, confirming the in vitro data. In addition, viability and toxicity tests provided evidence on nanoparticle safety in cell cultures. Our results encourage further assessment of silica 99mTc-nanoconjugates to validate a safe and versatile nanoreporter system for both diagnosis and treatment of aggressive breast cancer. One of the goals of the pharmaceutical sciences is the amelioration of targeted drug delivery. In this context, nanocarrier-dependent transportation represents an ideal method for confronting a broad range of human disorders. In this study, we investigated the possibility of improving the selective release of the anti-cancer drug paclitaxel (PTX) in the gastro-intestinal tract by encapsulating it into the biodegradable nanoparticles made by FDA-approved poly(lactic-co-glycolic acid) (PLGA) and coated with polyethylene glycol to improve their stability (PLGA-PEG-NPs). Our study was performed by combining the synthesis and characterization of the nanodrug with in vivo studies of pharmacokinetics after oral administration in mice. Moreover, fluorescent PLGA-nanoparticles (NPs), were tested both in vitro and in vivo to observe their fate and biodistribution. Our study demonstrated that PLGA-NPs: (1) are stable in the gastric tract, (2) can easily penetrate inside carcinoma colon 2 (CaCo2) cells, (3) reduce the PTX absorption from the gastrointestinal tract, further limiting systemic exposure, (4) enable PTX local targeting. At present, the oral administration of biodegradable nanocarriers is limited because of stomach degradation and the sink effect played by the duodenum. Our findings, however, exhibit promising evidence towards our overcoming these limitations for a more specific and safer strategy against gastrointestinal disorders.

Active drug targeting and controlled release of hydrophilic macromolecular drugs represent crucial points in designing efficient polymeric drug delivery nanoplatforms. In the present work EGFR-targeted polylactide-co-glycolide (PLGA) nanoparticles were made by a blend of two different PLGA-based polymers. The first, GE11-PLGA, in which PLGA was functionalized with GE11, a small peptide and EGFR allosteric ligand, able to give nanoparticles selective targeting features. The second polymer was a PEGylated PLGA (PEG-PLGA) aimed at improving nanoparticles hydrophilicity and stealth features. GE11 and GE11-PLGA were custom synthetized through a simple and inexpensive method. The nanoprecipitation technique was exploited for the preparation of polymeric nanoparticles composed by a 1:1 weight ratio between GE11-PLGA and PEG-PLGA, obtaining smart nanoplatforms with proper size for parenteral administration (143.9 ± 5.0 nm). In vitro cellular uptake in EGFR-overexpressing cell line (A549) demonstrated an active internalization of GE11-functionalized nanoparticles. GE11-PLGA/PEG-PLGA blend nanoparticles were loaded with Myoglobin, a model hydrophilic macromolecule, reaching a good loading (2.42% respect to the theoretical 4.00% w/w) and a prolonged release over 60 days. GE11-PLGA/PEG-PLGA blend nanoparticles showed good in vitro stability for 30 days in physiological saline solution at 4 °C and for 24 h in pH 7.4 or pH 5.0 buffer at 37 °C respectively, giving indications about potential storage and administration conditions. Furthermore ex vivo stability study in human plasma using fluorescence Single Particle Tracking (fSPT) assessed good GE11-PLGA/PEG-PLGA nanoparticles dimensional stability after 1 and 4 h. Thanks to the versatility in polymeric composition and relative tunable nanoparticles features in terms of drug incorporation and release, GE11-PLGA/PEG-PLGA blend NPs can be considered highly promising as smart nanoparticulate platforms for the treatment of diseases characterized by EGFR overexpression by parenteral administration.

Il polilattide (PLA) e il suo co-polimero con l’acido glicolico (PLGA) appartengono alla famiglia dei poli-esteri alifatici, polimeri di sintesi frequentemente utilizzati in ambito farmaceutico per la loro biocompatibilità e scarsa citotossicità, approvati FDA per la somministrazione parenterale. Esistono però alcune limitazioni nell’uso del PLA e derivati nella realizzazione di sistemi di rilascio controllato dei farmaci, come nanoparticelle (NP). In particolare, il PLA è caratterizzato da un’elevata idrofobicità e da un elevato grado di cristallinità che rendono il profilo di degradazione della matrice polimerica piuttosto lento e, di conseguenza, anche la cessione dell’attivo in essi incapsulato. Per queste ragioni è necessario realizzare derivati del PLA per renderlo più adatto all’applicazione farmaceutica. Inoltre, in relazione alla patologia di interesse, è auspicabile che le NP possano essere indirizzate in maniera selettiva alla cellula bersaglio, esponendo sulla superficie delle NP di un agente di targeting attivo. Il progetto di ricerca della dottoranda è stato finalizzato alla realizzazione di NP per il trattamento di diverse patologie ossee ed è stato organizzato in due filoni: nel primo, la dottoranda ha studiato l’impiego di derivati del PLA ottenuti dalla funzionalizzazione con residui gommosi, scelti come probabile alternativa ai derivati PEGilati di uso comune, per la realizzazione di sistemi nanoparticellari, mentre nella seconda parte della ricerca la dottoranda si è focalizzata sulla realizzazione di smart nanoparticles a partire da PLGA derivatizzato con agenti di targeting attivo. Nel primo caso il polimero selezionato è il PCNL, polimero sintetizzato nel laboratorio del prof. Hillmyer dell’Università del Minnesota: la presenza di una porzione gommosa dovrebbe rendere più veloce il profilo di degradazione delle NP. Innanzitutto, la dottoranda ha valutato la citocompatibilità del nuovo polimero che ha manifestato una scarsa tossicità e quindi il polimero si presenta come buon candidato per la realizzazione di sistemi nanoparticellari. Successivamente, il polimero è stato utilizzato per la realizzazione di NP. Il metodo utilizzato e le condizioni di processo sono state ottimizzate utilizzando un PLA di peso molecolare paragonabile al PLA utilizzato per la sintesi del PCNL. Nonostante gli sforzi per individuare una metodica opportuna, le NP ottenute con PCNL mediante la tecnica della nanoprecipitazione non hanno dimostrato le caratteristiche dimensionali desiderate, probabilmente a causa del peso molecolare e della Tg del residuo gommoso. Per cui il polimero è stato momentaneamente accantonato. Per quanto riguarda la seconda parte della ricerca sono state realizzate NP intelligenti a base di PLGA funzionalizzato con un piccolo peptide che riconosce in maniera selettiva il recettore EGFR. L’obiettivo della dottoranda è stato quello di realizzare sistemi terapeutici destinati al trattamento di patologie dell’osso caratterizzate da una sovraespressione del EGFR, cioè tumori e artrite reumatoide. Il ligando selezionato come oggetto della ricerca è il GE11, dodecapeptide che funge da ligando allosterico del recettore EGFR. Nello specifico, questo filone del lavoro di ricerca è stato diviso in due parti, una parte chimica di sintesi e una parte di realizzazione dei sistemi nanoparticellari. La prima parte del lavoro è stata condotta presso il dipartimento di Chimica dell’Università di Milano, nel laboratorio della prof.ssa Speranza, dove la dottoranda si è concentrata sulla sintesi del peptide GE11 e del coniugato GE11-PLGA. Il peptide è stato sintetizzato con la tecnica della sintesi in fase solida, con una resa di processo del 45% e una purezza del peptide >95%. Successivamente, il GE11 è stato legato covalentemente al PLGA mediante la chimica della carbodiimmide. Il processo ha avuto una resa del 50-60%; dall’analisi NMR si può ricavare un’efficienza di funzionalizzazione del polimero pari al 70-80%. Nella seconda parte del lavoro, la dottoranda si è occupata della realizzazione di sistemi nanoparticellari: la metodica di preparazione è stata ottimizzata inizialmente con i polimeri puri, PLGA ed un suo derivato coniugato al polietilenglicole (PEG-PLGA), al fine di settare le condizioni di processo migliori ad ottenere NP con le dimensioni adatte ad una somministrazione sistemica e locale. Prima di passare alla realizzazione delle NP con il coniugato GE11-PLGA, sono state realizzate NP modello: infatti in questa fase è stato utilizzato un tetrapeptide modello (FQPV) per indagare le criticità della sintesi di smart nanoparticles, per poi utilizzare il coniugato di interesse GE11-PLGA in una fase successiva. Il coniugato FQPV-PLGA è stato ottenuto con la stessa metodica del coniugato GE11-PLGA. Le NP con il coniugato FQPV-PLGA sono state realizzate secondo due metodiche differenti: la nanoprecipitazione, utilizzando DMSO come solvente, e una metodica “ibrida”, utilizzando una miscela 80:20 acetone-CH2Cl2, come selezionato mediante disegno sperimentale (DoE). Le NP hanno mostrato dimensioni intorno ai 200 nm con entrambe le metodiche. Inoltre, sono state sviluppate altre due strategie complementari a quelle descritte, al fine di aumentare l’entità di caricamento del peptide FQPV nelle NP di PLGA. A questo scopo, sono stati realizzati lotti in cui il peptide FQPV è stato adsorbito sulle NP di PLGA: con questa metodica è stato ottenuto un buon caricamento (1:29.9 FQPV:PLGA p:p) che però si è dimostrato instabile nel tempo. La seconda metodica ha previsto l’incapsulazione del peptide FQPV ma i risultati non sono stati ottimali: il metodo ha permesso una scarsa incapsulazione del peptide (1:150 FQPV:PLGA p:p). Per la realizzazione delle NP di GE11-PLGA solo la metodica della nanoprecipitazione utilizzando il DMSO come solvente è stata utilizzata, a causa della insolubilità del coniugato negli altri solventi comunemente utilizzati (acetone, diclorometano). Le formulazioni considerate e caratterizzate sono due: una in cui la matrice delle nanoparticelle è formata solo dal coniugato GE11-PLGA e la seconda in cui il coniugato è in miscela 1:1 p:p con un PEG-PLGA di uguale peso molecolare. Entrambe le formulazioni realizzate presentano una dimensione intorno a 140 nm e un potenziale zeta negativo (-20 mV). Di entrambe le formulazioni è stata valutata anche la citotossicità mediante MTS test su cellule caratterizzate da una sovraespressione del EGFR (A549, carcinoma polmonare, e Caki1, carcinoma renale): entrambe hanno dimostrato un’ottima citocompatibilità sulle linee cellulari selezionate in un ampio range di concentrazioni (0,1µg/ml to 1mg/ml). Le NP sono state caratterizzate mediante DSC per valutarne il comportamento termico rispetto al polimero puro. Le NP di GE11-PLGA hanno manifestato una Tg intorno a 50°C, del tutto paragonabile al comportamento del coniugato (49,33°C). Invece, le NP di GE11-PLGA/PEG-PLGA hanno una Tg a 21°C, dovuta al contributo del coniugato e del polimero PEGilato che invece ha una Tg sub-ambiente. Questo comportamento potrebbe influenzare la cinetica di rilascio del farmaco veicolato. Tutti i sistemi nanoparticellari realizzati hanno un’ottima stabilità a 4°C in acqua per 30 giorni, a simulare una possibile forma di stoccaggio prima della somministrazione al paziente. Il test di stabilità a 37°C a pH 5 e pH 7,4, per simulare un’infiammazione e il pH fisiologico, invece ha evidenziato una tendenza all’aggregazione delle NP dopo 15-20 giorni in entrambe le condizioni di pH, come se la presenza del peptide creasse instabilità alle NP. Successivamente, la dottoranda ha valutato la stabilità delle NP nei fluidi biologici, in particolare in plasma umano e in liquido sinoviale umano. Questa parte del lavoro è stata condotta presso il gruppo del prof. Braeckmans dell’Università di Gent (BE). Gli esperimenti sono stati condotti con fSPT (fluorescence Single Particle Tracking) su NP di GE11-PLGA/ PEG-PLGA marcate con RhB. Da questi test si osserva che le NP sono stabili in entrambi i mezzi considerati: solamente in plasma umano si osserva un aumento della dimensione dovuto alla formazione di una corona proteica attorno alle NP. Le dimensioni restano comunque entro valori accettabili per la loro applicazione in vivo. Sempre durante il periodo di ricerca all’estero, la dottoranda si è occupata degli studi di uptake al fine di valutare il profilo di captazione delle NP in cellule caratterizzate da una sovraespressione del EGFR nel tempo. Il test è stato condotto con microscopia confocale, andando a quantificare la fluorescenza delle NP marcate con RhB all’intero delle cellule. Il test ha dimostrato una internalizzazione attiva delle NP con il coniugato GE11-PLGA che è risultata veloce e prolungata nel tempo, visto che dopo 24 ore le NP erano ancora presenti all’interno delle cellule. Anche le NP di solo PLGA, utilizzate come controllo, sono captate dalle cellule con un meccanismo attivo anche se il profilo è differente rispetto alle NP con il peptide GE11: l’uptake è più lento e dopo 24 ore le NP non sono più all’interno delle cellule. Le NP selezionate sono state quindi caricate con Desametasone, farmaco utilizzato per il trattamento di numerose patologie reumatiche. Diverse strategie sono state considerate al fine di aumentare il grado di incapsulazione; i risultati migliori sono stati ottenuti utilizzando Etilacetato come solvente insieme al DMSO (300 µg/100 mg polimero). I risultati raccolti relativi alle nanoparticelle di GE11-PLGA sono promettenti, se si pensa alla mancanza di aggregazione nei fluidi biologici e alla buona captazione all’interno delle cellule che sovraesprimono EGFR. Sono in corso studi mirati allo studio dei meccanismi di uptake cellulare e trafficking intracellulare delle NP e alla valutazione dell’uso di uno spaziatore tra PLGA e GE11, alternativo al PEG, al fine di migliorare l’esposizione superficiale del peptide.

A stability study was performed on ivermectin (IVM)-loaded biodegradable microparticles intended for injection in dogs. The rational was to evaluate the performances upon irradiation of a drug, such as IVM, with a few criticalities with respect to its stability, and toxicity. The goal was to provide valuable information for pharmaceutical scientists and manufacturers working in the veterinary area. The microspheres based on poly(D,L-lactide) and poly-(ε-caprolactone) and loaded with IVM and with the addition of alpha-tocopherol (TCP) as antioxidant were prepared by the emulsion solvent evaporation method and sterilized by gamma irradiation. Microsphere characterization in term of size, shape, polymer, and IVM stability upon irradiation was performed. The results show that the type of polymer significantly affects microsphere characteristics and performances. Moreover, suitably stable formulations can be achieved only by TCP addition.

The aim was to design sterile biodegradable microparticulate drug delivery systems based on poly(dl-lactide) (PLA) and poly(e-caprolactone) (PCL) and containing ivermectin (IVM), an antiparasitic drug, for subcutaneous administration in dogs. The drug delivery system should: (i) ensure a full 12-month protection upon single dose administration; (ii) be safe with particular attention regarding IVM dosage and its release, in order to prevent over dosage side effects. This preliminary work involves: polymer selection, evaluation of the effects of γ-irradiation on the polymers and IVM, investigation and set up of suitable microparticle preparation process and parameters, IVM-loaded microparticles in vitro release evaluation. Results of gel permeation chromatography analysis on the irradiated polymers and IVM mixtures showed that combination of IVM with the antioxidant α-tocopherol (TCP) reduces the damage extent induced by irradiation treatment, independently on the polymer type. Solvent evaporation process was successfully used for the preparation of PLA microparticles and appropriately modified; it was recognized as suitable for the preparation of PCL microparticles. Good process yields were achieved ranging from 76.08% to 94.72%; encapsulation efficiency was between 85.76% and 91.25%, independently from the polymer used. The type of polymer and the consequent preparation process parameters affected microparticle size that was bigger for PCL microparticles (480-800 µm) and solvent residual that was >500 ppm for PLA microparticles. In vitro release test showed significantly faster IVM release rates from PCL microparticles, with respect to PLA microparticles, suggesting that a combination of the polymers could be used to obtain the suitable drug release rate.

The formulation of quinic acid, a food constituent demonstrating potential anticaries and antigingivitis properties, was investigated in an adhesive microparticulate delivery system with the goal of improving its effect by prolonging its residence time at the site of action. Alginate and chitosan were selected as mucoadhesive polymers. The microspheres were prepared by coacervation. Different types of alginates, polymers blends and crosslinking agent concentrations were considered and evaluated. The best results in terms of encapsulation efficiency, in vitro active agent release profile and in vitro adhesive properties, both to oral mucosa and to teeth surface, were obtained with a blend of Alginate Protanal LF200S: Alginate Protanal LF120LS 1:1.5 w/w, 0.1 M CaCl2, and chitosan coating, prepared by a one-step complex coacervation method. This microparticulate delivery system showed prolonged release of quinic acid, and could be used as an active component in chewing gums or mouthwashes for both caries and gingivitis prevention. © 2012 Elsevier Ltd. Background: Smart nanoparticulate materials, namely tailored nanoparticles (NPs) with specific surface functionality, have recently attracted attention as useful tool for time- and site-specific drug delivery. Specifically, polymeric nanoparticles (NPs) can be chemically functionalized with different chemical entities, i.e. peptides, that selectively recognize biological substrates in vivo and target drug release. Divergent and very complex strategies can be pursued in order to obtain peptidedecorated NPs. Methods: A simple method was suggested for direct functionalization of poly-lactide-co-glicolide (PLGA) with small peptides. A solid-phase peptide synthesis was used to obtain a dodecapeptide (GE11) and a smaller tetrapeptide (FQPV). FQPV- and GE11-PLGA conjugates were obtained by optimized carbodiimide chemistry. Nanoprecipitation and solvent extraction/evaporation methods were purpose-built in order to prepare FQPV-PLGA NPs. NPs were characterized in terms of size, surface charge and adsorbed peptide amount; ex-vivo cytotoxicity studies were performed on FQPV-PLA NPs using adult fibroblasts. Results: Custom GE11, recently known as efficient Epidermal Growth Factor Receptor targeting agent, and FQPV, used as model peptide, were synthesized by solid-phase peptide synthesis achieving good purity (95%) and satisfactory process yields (70-85%). Then, FQPV- and GE11-PLGA conjugates were obtained by optimized carbodiimide chemistry achieving an high degree of functionalization (> 85%). Aware of different physico-chemical properties of peptide-PLGA conjugates with respect to plain PLGA, two different NPs preparation techniques, nanoprecipitation and solvent extraction/ evaporation methods, were purpose-built in order to prepare FQPV-PLGA NPs. Both methods revealed suitable to obtain NPs with proper dimensions for the parenteral administration (< 250nm), narrow size distribution (P.I. about 0.1), good morphological features and negative charge (about –20mV). A peptide adsorption protocol onto NPs was considered as additional strategy to increase peptide expression on NPs surface aimed at improving the targeting effectiveness. A Design of Experiment approach (DoE) has been successfully applied to the more versatile solvent extraction/ evaporation method in order to systematically highlight the influence of process parameters (organic solvent mixture, PVA concentration and polymeric solution volume) on NPs sizes. Conclusion: PLGA was successfully functionalized with two different peptides, FQPV and GE11, following the a versatile and simple carbodiimmide chemistry without further modifying polymer and/or peptide structure. Both nanoprecipitation and solvent extraction/evaporation NPs preparation methods were properly optimized in order to obtain peptide-PLGA based NPs and they can be alternatively selected according to solubility properties of both peptide-polymer conjugate and drug intended for encapsulation. Peptide adsorption on preformed PLGA NPs could be efficiently used to increase peptide expression on NPs surface thus improving cellular recognition in case of active targeting. Preliminary cytocompatibility evaluation of the selected peptide-PLGA based nanoparticulate materials shows a potential feasibility of the set-up synthetic procedures and NPs preparation methods for pharmaceutical purposes.
