1. Dynamic behaviour and stability analysis of a compensated aerostatic pad

Abstract

Thanks to their low friction, aerostatic pads have important applications in precision positioning systems and linear guides. A simple and cheap solution to increase the static stiffness of aerostatic pads is to add a proper designed pneumatic valve to regulate the air flow supplied to the bearing. However, integrating aerostatic pads with additional devices can reduce its dynamic performance. This paper presents a numerical study on the dynamic behaviour and stability a commercial aerostatic pad controlled by a custom-built diaphragm valve. The bearing performance is studied by means of a lumped parameters model. Air bearing stiffness and damping are analysed in the frequency domain. Subsequently, the lumped model is linearized to investigate the stability of the system by means of Routh-Hurwitz method. The performance of the controlled air pad is compared to that of a simple commercial air pad.

Abstract

Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its Minimal Information for Studies of Extracellular Vesicles, which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly.

3. On the Design of a Diaphragm Valve for Aerostatic Bearings

Abstract

Because of their almost zero friction, cleanness and long life, aerostatic bearings are commonly used in many applications where high precision of positioning is required, e.g. machine tools, measuring machines, semiconductor manufacturing and power board testing. However, air bearings suffer from low relative stiffness and poor damping. Active and passive compensation are two effective methods to enhance the static and dynamic performance of these kinds of bearings. Despite their higher performance, active compensation solutions are too expensive to be used in industrial applications, as a consequence of the costs related to their controllers, actuators and sensors. This paper presents the design and performance of a passive compensation method that exploits a diaphragm valve. Thanks to its ease of integration, satisfactory performance and relatively low cost, this method could be a valuable solution to increasing the stiffness of aerostatic bearings. This work provides a procedure to design diaphragm valves depending on the type of the integrated pad and the desired nominal air gap height. Results demonstrate that, once correctly designed, the diaphragm valve makes it possible to obtain bearings with quasi-static infinite stiffness at the selected air gap height.

Abstract

Because of their almost zero friction and wear, aerostatic pads are widely used in applications where very precise positioning is required. However, this kind of bearing suffers from poor damping and low specific stiffness. This paper presents a new compensation strategy to increase air pad stiffness. This method exploits a custom-built pneumatic valve which can be easily integrated with any commercial pad. The design and the working principle of the proposed system are described and studied with the aid of a lumped parameter model. The effectiveness of the proposed compensation is numerically and experimentally evaluated. The results demonstrate that the solution represents a good and cost-effective method to enhance the static stiffness of aerostatic pads.

Abstract

This paper deals with the stabilization of an inverted pendulum on cart; the latter is pneumatically actuated by a double acting cylinder controlled by low cost proportional valves. In particular, a numerical model of whole system is developed in order to find the ability of the pneumatic actuation in stabilizing the pendulum and evaluate its bandwidth. A cascade of two control loops (the inner one for the pendulum angle, the outer one for the cart displacement) are analyzed and proper compensators are defined. The possibility of introducing an additional loop to control the force exerted by the actuator on the cart is evaluated.

27. Drug coated balloon angioplasty for de novo coronary lesions in large vessels: a systematic review and meta-analysis.

Abstract

We aimed to investigate the safety of drug-coated balloon (DCB)-only percutaneous coronary intervention compared to drug-eluting stent (DES) for de novo lesions in large vessels. To pursue this goal, we conducted a systematic review and meta-analysis following the PRISMA guidelines. The analysis included studies that utilized DCB-only or hybrid angioplasty for de novo lesions in large coronary vessel (> 2.75 mm). The primary outcome was to assess the target lesion revascularization (TLR) rate, while secondary outcomes included cardiac death, myocardial infarction (MI), and the composite of these. A total of 15 studies, comprising 3975 patients (of whom 2114 treated with DCB) were included. Median age was 62 ± 1.5 years, with 77.4% being male. Overall, 26.9% had diabetes, and 67.6% were diagnosed with acute coronary syndrome. Over a pooled follow-up of 20.6 ± 1.9 months, the incidence of TLR was 4% in the pooled DBC group. Additionally, over a pooled follow-up of 25.8 ± 2.7 months, no significant differences were observed in incidence of TLR between the DCB group and the DES group (4.3% vs. 6.9%, odds ratio 0.71, 95% confidence interval 0.50–1.01, p = 0.059). Furthermore, there were no differences in incidence of cardiac death and MI. DCB angioplasty treatment of de novo lesions in large coronary vessels could be a safe and effective strategy in both acute and chronic coronary settings. Abstract

Introduction Despite recent clinical breakthroughs in adoptive T cell transfer approaches and checkpoint blockade in treating hematopoietic and solid tumors, suppression of the antitumor immune response in the tumor microenvironment [...], The molecular mechanisms responsible for the high immunosuppressive capacity of [CD4.sup.+] Tregs in tumors are not well known. High-dimensional single-cell profiling of T cells from chemotherapy-naive individuals with non-small-cell lung cancer identified the transcription factor IRF4 as specifically expressed by a subset of intratumoral [CD4.sup.+] effector Tregs with superior suppressive activity. In contrast to the [IRF4.sup.-] counterparts, [IRF4.sup.+] Tregs expressed a vast array of suppressive molecules, and their presence correlated with multiple exhausted subpopulations of T cells. Integration of transcriptomic and epigenomic data revealed that IRF4, either alone or in combination with its partner BATF, directly controlled a molecular program responsible for immunosuppression in tumors. Accordingly, deletion of Irf4 exclusively in Tregs resulted in delayed tumor growth in mice while the abundance of IRF4+ Tregs correlated with poor prognosis in patients with multiple human cancers. Thus, a common mechanism underlies immunosuppression in the tumor microenvironment irrespective of the tumor type.

Abstract

Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its 'Minimal Information for Studies of Extracellular Vesicles', which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly.
