Within toxicology there is a pressure to find new test systems to replace, reduce and refine animal testing. In nanoecotoxicology this raises a number of questions.

Across many environments, nitrate ( $\mbox{NO}_{3}^-$ ) is an important form of N available for microorganisms and photosynthetic organisms. Accurate $\mbox{NO}_{3}^-$ measurements are important for examining N cycling and retention in terrestrial and aquatic ecosystems, but a common method of $\mbox{NO}_{3}^-$ analysis can underestimate $\mbox{NO}_{3}^-$ concentrations when soluble iron is present (iron > 10 mg L − 1). The basic method is robust, using copperized cadmium to reduce $\mbox{NO}_{3}^-$ and then diazotizing the resulting $\mbox{NO}_{2}^-$ in a two-step process to form an easily measured colored product. We show that iron interference is unique to using an NH4Cl and ethylenediaminetetraacetic acid (EDTA) buffer. We hypothesize that interference is through iron-catalyzed reduction of the intermediate color product, a diazonium ion. We examine three historical buffers as alternatives to NH4Cl/EDTA and recommend replacement of EDTA with diethylenetriaminepentaacetic acid, which chelates metals much like EDTA, but unlike EDTA, it does not cause interference in the presence of iron.

Nitrate has long been thought to be chemically unreactive in soil. This view was challenged by the report of an apparently abiotic process whereby nitrate (NO3 −) is incorporated into organic compounds (Dail et al. 2001). In Colman et al. (2007), we examined how common this process might be by testing for it in 45 soils collected from across a range of ecosystem types. We found no evidence of this process occurring in any of the soils, but found evidence of an analytical artifact that creates the appearance of incorporation. We suggested that prior evidence of this process might be due in part or in total to this analytical artifact. Davidson et al. (2008), however, challenged our results and conclusions, suggesting that we failed to observe the abiotic incorporation because we eliminated the anaerobic microsites they argue are necessary for the process. We address the criticisms, and show that they actually raise questions about the robustness of the only study to have reported abiotic NO3 − incorporation in sterile soils. We argue that this area of research needs new artifact-free experiments if the controversy is going to be resolved.

Harmful and nuisance algal blooms are becoming a greater concern to public health, riverine ecosystems, and recreational uses of inland waterways. Algal bloom proliferation has increased in the Upper Clark Fork River due to a combination of warming water temperatures, naturally high phosphorus levels, and an influx of nitrogen from various sources. To improve understanding of bloom dynamics and how they affect water quality, often measured as algal biomass measured through pigment standing crops, a UAV-based hyperspectral imaging system was deployed to monitor several locations along the Upper Clark Fork River in western Montana. Image data were collected across the spectral range of 400–1000 nm with 2.1 nm spectral resolution during two field sampling campaigns in 2021. Included are methods to estimate chl a and phycocyanin standing crops using regression analysis of salient wavelength bands, before and after separating the pigments according to their growth form. Estimates of chl a and phycocyanin standing crops generated through a linear regression analysis are compared to in situ data, resulting in a maximum R 2 of 0.96 for estimating fila/epip chl-a and 0.94 when estimating epiphytic phycocyanin. Estimates of pigment standing crops from total abundance, epiphytic, and the sum of filamentous and epiphytic sources are also included, resulting in a promising method for remotely estimating algal standing crops. This method addresses the shortcomings of current monitoring techniques, which are limited in spatial and temporal scale, by proposing a method for rapid collection of high-spatial-resolution pigment abundance estimates. The environmental fate and potential impacts of nanopesticides on agroecosystems under realistic agricultural conditions are poorly understood. As a result, the benefits and risks of these novel formulations compared to the conventional products are currently unclear. Here, we examined the effects of repeated realistic exposures of the Cu(OH)2 nanopesticide, Kocide 3000, on simulated agricultural pastureland in an outdoor mesocosm experiment over 1 year. The Kocide applications were performed alongside three different mineral fertilization levels (Ambient, Low, and High) to assess the environmental impacts of this nanopesticide under low-input or conventional farming scenarios. The effects of Kocide over time were monitored on forage biomass, plant mineral nutrient content, plant-associated non-target microorganisms (i.e., N-fixing bacteria or mycorrhizal fungi) and six soil microbial enzyme activities. We observed that three sequential Kocide applications had no negative effects on forage biomass, root mycorrhizal colonization or soil nitrogen fixation rates. In the Low and High fertilization treatments, we observed a significant increase in aboveground plant biomass after the second Kocide exposure (+14% and +27%, respectively). Soil microbial enzyme activities were significantly reduced in the short-term after the first exposure (day 15) in the Ambient (-28% to -82%) and Low fertilization (-25% to -47%) but not in the High fertilization treatment. However, 2 months later, enzyme activities were similar across treatments and were either unresponsive or responded positively to subsequent Kocide additions. There appeared to be some long-term effects of Kocide exposure, as 6 months after the last Kocide exposure (day 365), both beta-glucosidase (-57% in Ambient and -40% in High fertilization) and phosphatase activities (-47% in Ambient fertilization) were significantly reduced in the mesocosms exposed to the nanopesticide. These results suggest that when used in conventional farming with high fertilization rates, Kocide applications did not lead to marked adverse effects on forage biomass production and key plant–microorganism interactions over a growing season. However, in the context of low-input organic farming for which this nanopesticide is approved, Kocide applications may have some unintended detrimental effects on microbially mediated soil processes involved in carbon and phosphorus cycling.
