An electroosmotic flow (EOF)-based pump, integrated with a sol--gel stationary phase located in the electric field-free region of a microchip, enabled the separation of six nitroaromatic and nitramine explosives and their degradation products via liquid chromatography (LC). The integrated pump and LC system were fabricated within a single quartz substrate. The pump region consisted of a straight channel (3.0 cm x 230 [micro]m x 100 [micro]m) packed with 5-[micro]m porous silica beads. The sol--gel stationary phase was derived from a precursor mixture of methyltrimethoxy-and phenethyltrimethoxysilanes and was synthesized in the downstream, field-free region of the microchip, resulting in a stationary-phase monolith with dimensions of 2.6 cm x 230 [micro]m x 100 [micro]m. Fluid dynamic design considerations are discussed, especially as they relate to integrating the EOF pump with the LC system. Pump and separation performance, as characterized by flow rate measurements, injection, elution, separation, and detection, point to a viable analytical chemistry platform that encompasses all of the benefits expected of portable, laboratory-on-chip systems, including reduced sample requirements and small packaging.

In order to understand the role of stacked micelles in sample preconcentration, it is necessary to understand the factors that contribute to the micelle stacking phenomenon. Various MEKC background electrolyte (BGE) solutions were prepared in the presence of Sudan III in order to monitor the micelle stacking phenomenon in the anionic sodium dodecyl sulfate and sodium cholate micelle systems. The data show that micelle stacking is a dynamic process that is strongly dependent upon the relative conductivities of the sample matrix and BGE, the relative column length of the sample plug, and the mobilities of the ions involved in the stacking process regardless of electric field conditions (i.e., field-amplified stacking, sweeping, or high-salt stacking). Conditions under which micelle stacking can be expected to occur are presented, and the extent of micelle stacking is quantified. The micelle stacking phenomenon is correlated to the separation performance of a series of neutral alkaloids. It is shown that neutral analytes migrate rapidly through the evolving stacked micelle region in the initial moments of the separation. As a consequence of this transient interaction, analytes with small retention factors spend less time in the stacked micelle region and experience lower stacked micelle concentrations than analytes with large retention factors that spend more time in the growing stacked micelle region. It is also demonstrated that the extent of analyte enrichment generally increases with injection length, by facilitating greater interaction time with stacked micelles; however, enrichment will eventually plateau with increasing injection length as a function of an analyte's affinity for the micelle. Finally, it is shown that, in contrast to conventional wisdom, a range of long injection plugs exist where separation efficiency can be dramatically improved due to analyte interaction with an actively growing stacked micelle region.

A microchip-based, displacement immunoassay for the sensitive laser-induced fluorescence detection of staphylococcal enterotoxin B is presented. The glass microchip device consists of a microchannel that contains a double weir structure for supporting antibody-functionalized microbeads. After a 30-min sample preparation step, the displacement assay was performed without user intervention and produced quantitative results in an additional 20 min. Linear detection responses were observed over 6 orders of magnitude and provided detection limits down to 1 fM (28.5 fg/mL). The surprisingly low detection limits are hypothesized to arise from field-based enrichment analogous to field-amplified stacking, chromatographic effects, and limited diffusion lengths in the microbead bed. The assay was challenged with bovine serum albumin, casein, and milk sample matrixes. This system has the potential to provide highly sensitive detection capabilities for target biomolecules.

The ability to detect sub-nanomolar concentrations of ricin using fluorescently tagged RNA aptamers is demonstrated. Aptamers rival the specificity of antibodies and have the power to simplify immunoassays using capillary electrophoresis. Under nonequilibrium conditions, a dissociation constant, [K.sub.d], of 134 nM has been monitored between the RNA aptamer and ricin A-chain. With use of this free-solution assay, the detection of 500 pM (~14 ng/mL) or 7.1 amol of ricin is demonstrated. The presence of interfering proteins such as bovine serum albumin and casein do not inhibit this interaction at sub-nanomolar concentrations. When spiked with RNAse A, ricin can still be detected down to 1 nM concentrations despite severe aptamer degradation. This approach offers a promising method for the rapid, selective, and sensitive detection of biowarfare agents.

An integrated microdevice was utilized for the autonomous coupling of solid-phase extraction (SPE) to micellar electrokinetic chromatography (MEKC). Porous plugs of polymethacrylate polymer (~200 [micro]m in length) were fabricated by ultraviolet irradiation in microchannels. Microcolumns of hydrophobic beads packed against the polymethacrylate plugs were utilized for the quantitative extraction of rhodamine B, yielding preconcentration factors over 200 for a 90-s extraction. The calculated detection limit for this dye was 60 fM. A sample of coumarin dyes were concentrated by SPE, eluted in a nonaqueous solvent from a separate on-chip reservoir, and injected by a gated valve onto a separate column for MEKC analysis. Using the integrated device, a completely automated sequence of extraction, elution, injection, separation, and detection were performed in less than 5 min. Observed separation efficiencies were high, with plate heights below 2 [micro]m. The analysis was at least 3 times faster than semiautomated, conventional, solid-phase extraction, while requiring no user intervention. The design, fabrication, and autonomous operation of the device is discussed.

Signal amplification using enzyme multilayers on carbon nanotube (CNT) templates is shown to yield a remarkably sensitive electrochemical detection of proteins and nucleic acids. The electrostatic layer-by-layer (LBL) self-assembly onto CNT carders maximizes the ratio of enzyme tags per binding event to offer the greatest amplification factor reported to date. Absorption spectroscopy, TEM, and electrochemical characterization confirm the formation of LBL enzyme nanostructures on individual CNT carders. The enzymatic activity is found to increase with the number of enzyme layers. The new protocol is illustrated for monitoring sandwich hybridization and antibody-antigen interactions in connection with alkaline phosphatase tracers. Factors affecting the enzyme loading and the analytical performance have been optimized. Such amplified bioelectronic assays allow detection of DNA and proteins down to 80 copies (5.4 aM) and 2000 protein molecules (67 aM), respectively. Given the enormous amplification afforded by the new CNT-LBL biolabel, such route offers great promise for ultrasensitive detection of infectious agents and disease markers.

An electrochemical immunoassay protocol for the simultaneous measurements of proteins, based on the use of different inorganic nanocrystal tracers is described. The multiprotein electrical detection capability is coupled to the amplification feature of electrochemical stripping transduction (to yield fmol detection limits) and with an efficient magnetic separation (to minimize nonspecific adsorption effects). The multianalyte electrical sandwich immunoassay involves a dual binding event, based on antibodies linked to the nanocrystal tags and magnetic beads. Carbamate linkage is used for conjugating the hydroxyl-terminated nanocrystals with the secondary antibodies. Each biorecognition event yields a distinct voltammetric peak, whose position and size reflects the identity and level, respectively, of the corresponding antigen. The concept is demonstrated for a simultaneous immunoassay of [beta.sub.2]-microglobulin, IgG, bovine serum albumin, and C-reactive protein in connection with ZnS, CdS, PbS, and CuS colloidal crystals, respectively. These nanocrystal labels exhibit similar sensitivity. Such electrochemical coding could be readily multiplexed and scaled up in multiwell microtiter plates to allow simultaneous parallel detection of numerous proteins or samples and is expected to open new opportunities for protein diagnostics and biosecurity.

A microchip protocol for the capillary electrophoresis separation and electrochemical detection of thiol-containing degradation products of V-type nerve agents is described. The microchip assay relies on the derivatization reaction of 2-(dimethylamino)ethanethiol (DMAET), 2-(diethylamino)ethanethiol (DEAET), and 2-mercaptoethanol (ME) with o-phthaldialdehyde in the presence of the amino acid valine along with amperometric monitoring of the isoindole derivatives. Both off-chip and on-chip derivatization reactions have led to highly sensitive and rapid detection of the thiol degradation products. Various parameters influencing the derivatization, separation, and detection processes were examined and optimized. These include the amino acid co-reagent, reagent-mixing ratio, reaction time, injection time, separation voltage, and detection potential. The chip microsystem offers a rapid (< 4 min) simultaneous detection of micromolar concentrations of DMAET, DEAET, and ME. Linear calibration plots were observed for the V-type nerve agent thiol degradation products, along with good stability and reproducibility (RSD < 8.0%). Detection limits of 5 and 8 [micro]M were obtained for the off-chip reaction of DMAET and DEAET, respectively, following a 2-s injection. The suitability for assays of environmental matrixes was demonstrated for the determination of DMAET and DEAET in untreated tap and river water samples. The favorable analytical performance makes the new microfluidic device attractive for addressing the needs of various security scenarios.

This paper reports on a microfiuidic device for the screening of organophosphonate nerve agent degradation products. The miniaturized system relies on an efficient chip-based separation of alkyl methylphosphonic acids (breakdown products of Satin, Soman, and VX nerve agents) followed by their sensitive contactless conductivity detection. Experimental parameters relevant to the separation and detection processes have been optimized to yield high sensitivity (with 48-86 [micro]g [L.sup.-1] detection limits), fast response (50 s for a three alkyl methylphosphonic acid mixture), high precision (RSD = 3.8-5.0%), and good linearity (over the 0.3-100 mg [L.sup.-1] range). Applicability to natural (river) water samples is demonstrated. The new microsystem offers promise for monitoring degradation products of chemical warfare agents, with advantages of speed/warning, efficiency, portability, sample size, and cost compared to conventional ion chromatography or capillary electrophoresis systems.

A single-channel chip-based analytical microsystem that allows rapid flow injection measurements of the total content of organic explosive or nerve agent compounds, as well as detailed micellar chromatographic identification of the individual ones, is described. The protocol involves repetitive rapid flow injection (screening) assays-to provide a timely warning and alarm-and switching to the separation (fingerprint identification) mode only when harmful compounds are detected. While micellar electrokinetic chromatography, in the presence of sodium dodecyl sulfate (SDS), is used for separating the neutral nitroaromatic explosive and nerve agent compounds, an operation without SDS leads to high-speed measurements of the 'total' explosives or nerve agent content. Switching between the 'flow injection' and 'separation' modes is accomplished by rapidly exchanging the SDS-free and SDS-containing buffers in the separation channel. Amperometric detection was used for monitoring the separation. Key factors influencing the sample throughput, resolution, and sensitivity have been assessed and optimized. Assays rates of about 360 and 30/h can thus be realized for the 'total' screening and 'individual' measurements, respectively. Ultimately, such development will lead to the creation of a field-deployable microanalyzer and will enable transporting the forensic laboratory to the sample source.

The photoreversible metal-ion complexation behavior of nitroquinolinospiropyranindoline (NQSP) was studied in combination with the selective identification of six different transition-metal ions, Zn(super 2+), Co(super 2+), Hg(super 2+), Cu(super 2+), Cd(super 2+), and Ni(super 2+), in single- and binary-component mixtures via partial least squares discriminant (PLSD) analysis. The plasticizer, dicapryl phthalate, was chosen as the support medium for this study on the basis of (1) its enhancement of the photoreversibility and hypsochromic shifts seen in metal complexation and (2) its potential application to supported liquid membranes for eventual sensor applications. Complexation of divalent transition-metal ions by NQSP in dicapryl phthalate produced variable hypsochromic shifts in the absorption spectra (30-60 nm), requiring chemometric techniques in order to overcome the spectral overlaps. PLSD analysis was used to build classification analysis models to differentiate between the six divalent transition-metal ions. The feasibility of performing mixture analysis was studied using the concept of net analyte signal prior to experimental verification. Single- and binary-component mixtures of metals were identified with 100 and 97.4% accuracy, respectively, which included no false positives in either the training or prediction sets.

The continual or on-demand delivery of aerosol is important for sensor validation and the development of trace particle detection methodologies. The system presented here operates by introducing aerosolized analytical standard solutions in a heated air flow, generating vapor phase analytes that are subsequently transported and cooled to provide aerosolized particles. The notable design feature was a small concentric air flow device that blends chilled dry air with vapor phase analyte to induce a phase transition without the need for volatile organic solvents. While organic solvents accelerate solvent evaporation, they present potential interferences for downstream sensors. Cyclotrimethylenetrinitramine (RDX) particles were generated and optically counted from the exhaust plume aerosol. The solid RDX material was binned into 1–2.5, 2.5–5, 5–10, and 10+ µm diameter sizes and when the system was actively cooled, the generation of particles exhibited a relative increase of 4.9%, 27.6%, 95.6%, and 78.0% in production, respectively. When observed under a microscope, collected RDX particles ranged from about 1 to 7 µm in diameter. It was determined that the production of solid material was proportional to the temperature and mass loading of the vapor, as dictated by the feed solution concentration; cooler temperatures and higher concentrations led to the increased production of particles ranging from 1 to 10 μm in diameter for the low-vapor pressure analytes studied: RDX (cyclotrimethylenetrinitramine), PETN (pentaerythritol tetranitrate), TNT (2-methyl-1,3,5-trinitrobenzene), and 2,6-DNT (2,6-dinitrotoluene). The complex 4-(N-1,4,8,11-tetraazacyclotetradecyl)benzonitrile (DMABN-Cyclam) was found to be an unusual molecule which has triple fluorescence due to the locally excited singlet state and two excited-state complexes, a twisted intramolecular charge-transfer state and an intramolecular exciplex. All three emission bands were strongly perturbed by solvent polarity, pH and metal complexation. DMABN-Cyclam may be used in the selective detection of metal cations.
