Rambutan (Nephelium lappaceum) is an important fruit tree. Recently, leaf spots were observed in a tree in Los Baños, Laguna, Philippines. This study aims to identify the causal agent of rambutan leaf spot in the Philippines. Two fungal isolates, MBRAM001B and MBRAM003B, were isolated from the diseased leaves on potato dextrose agar medium. Colony growth and conidia characteristics of both isolates showed high resemblance with Colletotrichum species in the C. gloeosporioides species complex. Molecular characterisation of the isolates using the partial internal transcribed spacer rDNA and TUB2 regions identified the pathogens as C. tropicale MBRAM001B and C. siamense MBRAM003B. Leaf spots developed on rambutan leaves three days after inoculation and the same pathogens were isolated from the diseased leaves, establishing Koch's Postulate. In conclusion, this study identified C. tropicale MBRAM001B and C. siamense MBRAM003B as causal agents of leaf spots in rambutan species in the Philippines. The survival and possible infection of these two Colletotrichum species on rambutan fruits and other fruit trees are worth further investigating. [ABSTRACT FROM AUTHOR]

Abstract Mango (Mangifera indica L.) has great socioeconomic importance to Brazil, but its production is affected by anthracnose. Chitosan films have shown potential in controlling this disease. In this study, the effect of the association of chitosan with phenolic compounds and extracts on the Colletotrichum tropicaledevelopment was evaluated. Phenolic compounds and extracts from mango peel were incorporated into 2.0% chitosan solutions and tested in vitro. In the in vivo experiment, after pathogen inoculation and application of treatments, fruits were evaluated for anthracnose incidence and severity. Controls consisted of the application of water or pure culture medium and fungicide imazalil. The experimental design was completely randomized and data were submitted to analysis of variance. Heliconia subulata is a common ornamental plant, it has been widely planted in southern China for greening parks, roads, and residential areas. H. subulata plants with spots on their leaves were observed in East Coast Wetland Park (18°16'53.37″N, 109°30'19.36″E), Sanya City, Hainan Province, China on Aug. 31, 2023. The symptoms of the leaves are irregular gray-white, spots, that develop into brown and black, with yellow halos at the disease-health junction. Following an on-the-spot investigation, it was found that the incidence of the disease was 40 to 50%. The leaves were disinfected with 70% ethanol for 1 min, rinsed with sterile water 3 times, disinfected for 1 min with 0.1% HgCl2, rinsed with sterile water 3 times, dried, put on potato dextrose agar (PDA) and incubated at 28℃ for 7 days. The red conidia pile was selected from the culture, dispersed in sterile water and diluted to 20 μL containing 1 to 2 conidia. After absorbing 20 μL spore suspension for many times and inoculating it on the new PDA plate, five pure cultures of single spore, J-1-1 to J-1-5, were obtained. After 7 days of growth, the colonies were grayish aerial mycelium on the front and light orange conidia on the reverse. The white aerial mycelia, conidia, acervulus, and appressorium were observed (Supplementary Fig. S1). The morphological characteristics showed that the isolate had the same characteristics as the previously described Colletotrichum spp. (Wang et al. 2021). The genomic DNA of isolates J-1-1 and J-1-5 were extracted by Fungal DNA Kit (OMEGA bio-tek, Guangzhou, China). The internal transcribed spacer (ITS), glyceraldehyde-3-phosphate dehydrogenase (GADPH), and β-tubulin 2 genes (TUB2) were amplified by primers ITS1/ITS4, GDF/GDR, and Bt2a/Bt2b, respectively (Weir et al. 2012). Based on sequencing and gene sequence alignment analysis, it was found that the consistency between the ITS sequences of isolates J-1-1 and J-1-5 was 99.82%. The consistency between GADPH and TUB2 sequences was 100%. The gene sequences of isolates J-1-1 and J-1-5 were submitted to GenBank with accession numbers PP455510/PP455511 (ITS), PP510210/PP510211 (GADPH) and PP510212/PP510213 (TUB2) respectively. Based on the BLAST analysis, the three sequences were more than 99% identical to those of the C. tropicale strain FC1 (ITS: MT192648, GAPDH: MT155819, TUB2: MT199874; Duan et al. 2022). A phylogenetic tree was constructed by MEGA 11 based on the ITS, GADPH, and TUB2 gene sequence by the maximum-likelihood method. The results showed that the isolates J-1-1 and J-1-5 were clustered with C. tropicale CBS:124949 (Supplementary Fig. S2). Based on morphological and molecular biological analysis, two isolates were identified as C. tropicale. To further test the pathogenicity of isolates J-1-1 and J-1-5, spore suspensions (1×106 conidia/mL) were prepared and 20 μL spore suspensions were inoculated on the leaves of healthy H. subulata potted plants stabbed with sterile toothpicks. Three leaves were inoculated in each treatment, and sterile water was inoculated as a control. The treated plants were placed in an incubator with a temperature of 28℃, relative humidity of 90%, and light/dark (12h/12h). After 15 days, the spore suspension treatment showed the same symptoms as the naturally diseased H. subulata plants in the field, but the leaves treated with sterile water were not infected (Supplementary Fig. S1). The morphology of the isolates obtained from diseased leaves was the same as that of isolates J-1-1 and J-1-5 on the PDA plate. To our knowledge, this is the first report of H. subulata, a new host of C. tropicale causing anthracnose in China.

Plants host a vast diversity of fungal symbionts inside their tissues that live in close proximity with each other to form rich and dynamic communities. Although endophytes can affect plant-herbivore interactions in several ways, it is still not known to what extent such effects are influenced by the properties of endophyte communities or by particular species traits. Here we compared the effects of high versus low foliar fungal endophyte diversity on the preferences of laboratory and wild colonies of leaf-cutting ants. We found that when endophyte densities were high, the ants responded similarly to leaves hosting one endophyte species, Colletotrichum tropicale, or those hosting a species-rich endophyte community. Results were also consistent when comparing the laboratory versus wild ant colonies. We discuss the significance of these results with respect to the ecological effects of plant-endophyte interactions in natural and agricultural ecosystems. [ABSTRACT FROM AUTHOR]

Dalbergia odorifera T. Chen (Family: Fabaceae) is an endangered, wild and second class key protected tree species in China. It has been cultivated widely in south China because of its valuable rosewood and pharmacological uses. During January and September 2022, anthracnose was observed on D. odorifera in Chikan, Zhanjiang, Guangdong (N21°15' 9.4" E110°22' 25.9"). All of the D. odorifera plants were infected and the leaf disease incidence was above 30%. The lesions were 0.35 to 0.65 cm in size, greyish-white, and gradually expanded into brown black circular spots surrounded by a yellowish halo. Later, the centers of spots were white with dark-brown borders. In severe cases, the leaves turned yellow, withered and fell off. To isolate the pathogen, about 5×5 mm tissues at the disease-health junction of spots were sterilized with 75% ethanol for 30s and 3% hydrogen peroxide for 3 min, rinsed with sterile water three times, and plated onto PDA for incubation at 25-28℃ in the dark. Isolates were obtained by picking the tip of hypha growing from the tissues, further purified by coating the diluted spore suspension to obtain single-spore isolates. Colonies with abundant aerial mycelium were white to light gray, 4.0 to 5.0 cm in diameter after 5 days at 25 to 28℃ in the dark. Conidiogenous cells were hyaline, cylindrical and monoblastic. Conidia were unicellular, subcylindrical, hyaline, guttulate, 3.5 to 5.6×11.1 to 16.8 (av. 4.6±0.48 × 14.0±1.25) μm (n>50). Appressoria were dark brown subglobose, clavate, fusiform, occasionally lobed, terminal, 7.8 to 16.3×4.6 to 8.9 μm. No swollen cells and setae were observed. To further confirm identity of the fungus, partial regions of six genetic loci of isolate ACCC 35246 (stored in Agricultural Culture Collection of China) were amplified and sequenced: actin (ACT), chitin synthase (CHS), glyceraldehyde-3-phosphate dehydrogenase (GPDH), histone (HIS), beta-tubulin (TUB) and internal transcribed spacer (ITS) utilizing the primer pairs (Damm et al. 2012; Weir et al. 2012; White et al 1990) (OP314900-OP314904 and OP269660, respectively). Based on a BLAST analysis, the six sequences were about 99% identical to those of Colletotrichum tropicale holotype strain CBS124949 (ACT: JX009489 270/272; CHS: JX009870 295/299; GPDH: JX010007 274/279; HIS: KY856395 638/670; ITS: JX010264 546/548 and TUB: JX010407 494/497, respectively) (Weir et al. 2012). Phylogenetic analyses, using a combined dataset of ACT, CHS, GPDH, ITS and TUB were carried out in MEGA X using the maximum likelihood method, placed the fungus within the C. tropicale clade. Based on morphogical and molecular analyses, the fungus was identified as C. tropicale (Rojas et al 2010). To test the pathogenicity, five healthy leaves of 1 year-old potted plants were stab-wounded with a sterile needle and inoculated with 10 µL of spore suspension (105 conidia/mL). Another five healthy leaves were inoculated with sterile water as the control. Symptoms expressed by the inoculated leaves, 10 days after inoculation, were the same as those of the diseased plants, while the non-inoculated leaves remained asymptomatic. The same fungus based on morphogical and molecular criteria was re-isolated from the spots. As an endophyte or plant pathogen, C. tropicale inhabits a wide host range of plant species belonging to 31 genera in 25 families (Farr and Rossman 2022), D. odorifera is a new host of C. tropicale causing anthracnose in China. This work will be helpful for the diagnosis and control of this disease.

Abstract: Fungal symbionts that colonize leaf tissue asymptomatically (endophytes) can alter the foraging behaviour of leaf-cutting ants, and decrease the productivity of this herbivore's mutualistic fungal cultivar, Leucocoprinus gongylophorus. This negative effect of endophytes on the ant's cultivar could be the result of direct fungal–fungal interaction or indirect reductions in the quality of leaves, the cultivar's growth substratum. To test for the indirect effects, we measured in vitro growth rates of cultivars in media that contained sterilized leaf extracts from plants with high (E high) and low (E low) endophyte colonization. We found that, opposite to our expectations, cultivars grew significantly faster in E high leaf extracts compared to E low extracts. Our results suggest that endophyte-driven changes in leaf chemistry are a less likely explanation for the observed in vivo reduction in the ant's symbiotic fungal growth and imply that the effect of direct endophyte–cultivar interactions inside nests are potentially more important. Severe blight of stems was found on Madagascar periwinkle ( Catharanthus roseus) in Shizuoka and Okinawa prefectures, Japan, in July 2003 and September 2004, respectively. Similar fungi were isolated from the diseased plants. The isolated fungi caused the disease after inoculation and subsequently were reisolated from diseased tissues. The fungi were identified as species closely related to C. siamense or C. tropicale according to sequences of the β-tubulin-2 gene, indicating that they belong to the Colletotrichum gloeosporioides species complex. This is the first report specifying the genetic relationships of the pathogens of Madagascar periwinkle anthracnose in the complex. [ABSTRACT FROM AUTHOR]

Fungal symbionts that live asymptomatically inside plant tissues (endophytes) can influence plant-insect interactions. Recent work has shown that damage by leaf-cutting ants, a major Neotropical defoliator, is reduced to almost half in plants with high densities of endophytes. We investigated changes in the phenotype of leaves that could influence ants' behavior to result in the reduction of foliar damage., We produced cucumber seedlings with high and low densities of one common endophyte species, Colletotrichum tropicale. We used the leaves in bioassays and to compare chemical and physical leaf characteristics important for ants' food selection., Ants cut about one-third more area of cucumber leaves with lower densities of endophytes and removed c. 20% more paper disks impregnated with the extracts of those leaves compared with leaves and disks from plants hosting the fungus. Colletotrichum tropicale colonization did not cause detectable changes in the composition of volatile compounds, cuticular waxes, nutrients or leaf toughness., Our study shows that endophytes changed leaf chemistry and suggests that compounds with relative low volatility released after leaf wounding are a major factor influencing foraging decisions by ants when choosing between plants with low or high endophyte loads.
