A technique is described allowing the quantification and the characterization of specific beta-adrenergic receptors in intact living human lymphocytes. 125I-Iodohydroxyhenzylpindolol, a potent beta-adrenergic antagonist was used to label specific binding sites on unfractionated lymphoid cells and on purified subpopulations of T (F1 and F2) and B cells. F1 and F2 were obtained by filtration through nylon wool column as previously described (Delespesse et al., 1976), they differ in their response to mitogens, and in their interactions with adherent cells and B cells. 125I-HYP binding to unfractionated lymphocytes was a saturable, stereospecific and rapid process with a dissociation constant of 2.5 10-10 M and a binding capacity of 400-600 sites/cell. Bindings on unfractionated lymphocytes, purified B cells and T cells of the F2 fraction were similar. No detectable binding was noted on T cells from the F1 fraction. The quantified lymphocyte response to phytohaemagglutin (PHA) was investigated in fifty-six control subjects, classed according to age and sex, iii twenty-nine insulin- dependent (IDD) and eight non-insulin-dependent diabetics. In nine cases one-way mixed lymphocyte cultures (MLC) were performed using mixed lymphocytes from I DD and paired controls. In the controls, a reduction of the response to PHA was observed with ageing; this was more marked in women than in men. The diabetics gave a greatly impaired response to PHA but only a slightly depressed response in MLC These anomalies are not influenced by age hut by the severity of the diabetes. Increased DNA synthesis and immunoglobulin secretion was observed in human peripheral blood lymphocytes (PBL) cultured in vitro with soluble protein A from Staphylococcus aureus (Sp-A). Optimal Ig secretion was obtained in 6 day cultures containing 1 × 106 cells/mI and 10 μg/ml Sp-A. The presence of 5 × 10-5 M 2-mercaptoethanol in the culture medium as well as careful selection of foetal calf serum were needed for optimal results. In response to Sp-A stimulation, the three main classes of immunoglobulins were secreted by PBL. In some individuals, concentration of Sp-A optimal for lgG and IgM secretion inhibited IgA production. Immunoglobulin-con-taming cells were less abundant (0.5-5% of total cells) and smaller in Sp-A than in PWM-stimulated cultures (5-15%). The data suggest that Sp-A and PWM stimulated different subsets of circulating lymphocytes. [ABSTRACT FROM AUTHOR]

The lymphocyte transformation test (LTT) with thyroglobulin was correlated with humoral thyroglobulin antibodies (TGA) in different thyroid diseases. All patients with significant TGA had a positive LTT, while two LTT positive patients either had no TGA or revealed them a few weeks later. Lymphocyte transformation was found in all investigated cases of chronic thyroiditis and Hashimoto's disease, half of the patients with multinodular goitres and Graves' disease, and some cases (five out of sixteen) of simple goitres. Background and Aims: Gastric emptying scintigraphy (GES) is the criterion standard for the diagnosis of gastroparesis. However, data are lacking regarding the prognostic value of preoperative intragastric meal distribution during GES in patients undergoing gastric peroral endoscopic myotomy (G-POEM) for gastroparesis. This study investigated the association of GES morphologic parameters and the long-term clinical success of G-POEM., Methods: This retrospective study included patients who underwent G-POEM for refractory gastroparesis in a tertiary center with preoperative GES data. Intragastric meal distribution was measured using the proximal to distal count ratio (PDCR) at 0, 1, 2 and 4 hours, and the retention index was calculated. Clinical success was defined as a decrease of at least 50% in the Gastroparesis Cardinal Symptom Index total score after G-POEM., Results: In total, 77 patients were included with a mean follow-up of 40.14 months. Clinical success was observed in 54.55% of patients. The retention index was not associated with clinical success. Only PDCR at 0 hours (PDCR0) was associated with clinical success. In univariate analysis, the median PDCR0 was 6.0 (interquartile range, 5.59) in patients with clinical success and 4.29 (interquartile range, 4.51) in patients with clinical failure (P = .019). Objectives: So far, no 87 Sr/ 86 Sr mobility studies have been done for Neolithic remains from Belgium and information on the Sr isotopic variability in the region is scarce. This study aims to explore mobility in a Final Neolithic population from the funerary cave 'Grotte de La Faucille', contribute to the understanding of the isotopic composition of bioavailable Sr in Belgium, assess evidence for male mobility using proteomic analysis, and explore possible places of origin for nonlocal individuals., Materials and Methods: The 87 Sr/ 86 Sr isotope ratio of dental enamel from six adults and six juveniles was determined. Liquid chromatography mass spectrometry-based protein analysis was employed to identify individuals of male biological sex. 87 Sr/ 86 Sr of micromammal teeth, snail shells, and modern plants from three geological areas in Belgium were measured to establish isotopic signatures for bioavailable strontium. Nonlocality was assessed by comparing human 87 Sr/ 86 Sr isotope ratios to the 87 Sr/ 86 Sr range for bioavailable Sr., Results: Four individuals yielded 87 Sr/ 86 Sr isotope ratios consistent with a nonlocal origin. No statistical differences were found between adults and juveniles. Three males were detected in the sample set, of which two show nonlocal 87 Sr/ 86 Sr values., Discussion: This study provides evidence for mobility in Final Neolithic Belgium. The four nonlocal 87 Sr/ 86 Sr signatures correspond with the 87 Sr/ 86 Sr of bio-available Sr in Dutch South Limburg, the Black Forest in Southwest Germany, and regions of France, such as parts of the Paris Basin and the Vosges. Click chemistry by copper-mediated azide/alkyne cycloaddition reaction is known as a very efficient synthetic pathway for chemical coupling and for surface functionalization. The attractiveness of this reaction is due to the selectivity of the reaction of azides and alkynes. Nevertheless, the control of the kinetics of the reaction and the characterization of the reaction products requires the use of adequate analytical techniques. This is especially the case for the monitoring of the functionalization of polymeric compounds and the control of the homogeneity of the chemical composition of these products. In this work, the third generation of dendrigraft poly-L-lysines derivatized with acetylenic groups was decorated with tryptophan-PEG-azide arms via triazole ring formation. Pressure assisted capillary electrophoresis was used to monitor the progress and the kinetics of the click chemistry reaction. As expected the click product was obtained with excellent yields within 20 min reaction time at room temperature. Diffusion coefficients and hydrodynamic radii of dendrigraft poly-L-lysines and their click reaction products were determined by Taylor dispersion analysis. Finally, capillary electrophoresis was found very useful to check the purity and the homogeneity in the chemical composition of the decorated structures., (Copyright © 2012 Elsevier B.V.
