Abstract

Lesions of the pedunculopontine tegmental nucleus (PPTg) can impair spatial learning tasks, but it is not clear whether those detrimental effects depend on the specific training conditions (for example, number of response choices available) or are secondary to enhanced anxiety. In the present work, rats with either bilateral excitotoxic (ibotenate) lesions of the PPTg (lesion group) or with vehicle infusions (control group) were tested in an elevated plus-maze, in order to measure anxiety-like behaviours and spontaneous locomotion. Subsequently, they were trained in a delayed matching-to-position (DMTP) task in a T-maze (a two-response choice task). After reaching a predefined learning criterion, or after a maximum of 30 training sessions, the animals were trained in a delayed non-matching-to-position (DNMTP) task. Lesioned animals made less grooming episodes, stretch-attend postures and closed arm entries than controls in the elevated plus-maze, suggesting slightly lower anxiety levels. None of the lesioned rats reached the learning criterion for the DMTP, and overall accuracy levels were significantly lower in those rats, compared to controls. In the DNMTP task, lesioned animals showed lower accuracy levels and higher side bias than controls in some of the sessions, but there were no significant differences between the two groups in the proportion of animals reaching the learning criterion. It is concluded that spatial learning deficits induced by damage to the PPTg are not secondary to enhanced anxiety. Instead, those deficits seem to be influenced by several conditions that modify task demands, the number of response choices being only one of such conditions.

Consultable des del TDX, Títol obtingut de la portada digitalitzada, El núcleo pedunculopóntico tegmental (PPTg) es un grupo de neuronas (predominantemente colinérgicas) situado en el tronco del encéfalo, que se ha implicado en diversas funciones cognitivas. No obstante, el papel que juega el PPTg en los procesos cognitivos no está del todo claro; por ejemplo, no se conoce si su influencia en estos aspectos depende del nivel de dificultad de la tarea, del número de opciones de respuesta disponibles de la misma o de su dependencia de los sistemas fronto-estriatales; por otra parte, no se sabe si su integridad es necesaria para la consolidación de la memoria. Para contestar a estas cuestiones, se ha realizado un trabajo compuesto por tres experimentos en el que se analizan los efectos de la lesión del PPTg sobre: 1) la adquisición del condicionamiento de evitación activa de dos sentidos (EV2), una tarea dependiente del estriado dorsal, en dos condiciones diferentes de dificultad, 2) la retención a largo plazo, al cabo de 10 días, de este mismo tipo de condicionamiento; y 3) el aprendizaje de dos tareas espaciales dependientes de los sistemas fronto-estriatales ventrales, que implican únicamente dos opciones de respuesta, y que difieren en cuanto a su grado de dificultad (la tarea de no-alternancia forzada o DMTP y la tarea de alternancia forzada o DNMTP, ambas en un laberinto en T). En el experimento I, los animales eran sometidos a lesión electrolítica bilateral del PPTg y posteriormente recibían dos sesiones de EV2 separadas por 10 días; la mitad de los sujetos eran entrenados con un estímulo condicionado (EC) de 10 segundos de duración (condición de baja dificultad) y la otra mitad con un EC de 3 segundos (condición de alta dificultad). En el experimento II, los sujetos recibían una sesión de entrenamiento en la tarea de EV2 de mayor dificultad e, inmediatamente después, eran sometidos a lesión electrolítica del PPTg; 10 días más tarde se evaluaba el nivel de retención de la misma. In order to answer these questions, we have carried out a work composed of three experiments analyzing the effects of PPTg lesions on: 1) the acquisition of two-way active avoidance conditioning, a task that depends on dorsal striatal systems, under two conditions differing in task difficulty; 2) the long-term retention (10 days) of the same conditioning task; and 3) the acquisition of two spatial tasks that depend on fronto-ventral striatal systems, that involve only two response options, and that differ in their difficulty level (delayed matching-to-position task or DMTP, and delayed non-matching-to-position task, or DNMTP). In Experiment I, the animals were subjected to bilateral electrolytic lesions of the PPTg and were thereafter trained in a two-way active avoidance task (2 sessions separated by 10 days). Half of the subjects were trained using a conditioned estimulus of 10 second duration (low difficulty condition), while the other half were trained with a conditioned stimulus lasting 3 seconds (high difficulty condition). Abstract

The acquisition and long-term retention (LTR, 14 days) of a massed two-way active avoidance conditioning using two parameters of conditioned stimulus (CS) duration and unconditioned stimulus (US) intensity was studied. Two different tests of LTR were used: with and without the presentation of the US (Additional learning and Extinction). Results were analyzed by means of analyses of variance complemented with the use of other non-traditional statistical analyses (time-series and survival analyses). Results showed a better performance using the US of lower intensity, specially when the longer CS was used. The use of an extinction procedure does not seem to be as adequate as the use of a session of additional learning to asses LTR. On the other hand, both time-series and survival analyses can afford valuable data to complement and improve those obtained from the analyses of variante Se estudió la adquisición y retención a largo plazo (RLP, 14 días) de un condicionamiento masivo de evitación activa de dos sentidos utilizando dos parámetros diferentes de duración del estímulo condicionado (EC) y de intensidad del estímulo incondicionado (El). Se realizaron dos tipos de pruebas de RLP: con y sin presentación del El (aprendizaje adicional y extinción). Los resultados fueron analizados mediante análisis de la variancia, complementados con otros análisis estadísticos no tradicionales (el análisis de la supervivencia y el de series temporales). Los resultados indican una mejor ejecución con el El de menor intensidad, especialmente cuando el EC tiene una mayor duración. El uso de un procedimiento de extinción no parece ser tan adecuado para valorar la RLP de los sujetos como el aprendizaje adicional. 29. Treating Traumatic Brain Injury with Exercise: Onset Delay and Previous Training as Key Factors Determining its Efficacy.

Abstract

Purpose: Exercise reduces cognitive deficits in traumatic brain injury (TBI), but early post-trauma exercise is often discouraged due to potential harm. The purpose was to evaluate the interaction between pre- and post-injury physical exercise on cognition, neuronal survival and inflammation., Methods: Rats were either sham-operated and kept sedentary (Sham) or subjected to controlled cortical impact injury and then distributed into sedentary (Tbi), pre-injury exercise (Pre-Tbi), post-injury exercise with early (24 hours, Tbi-early) or late (6 days, Tbi-late) onset, and a combination of pre- and post-injury exercise with early (Pre-Tbi-early) or late (Pre-Tbi-late) onset. Object recognition memory, hippocampal volume, neuronal survival (NeuN + ) in the hippocampus and perirhinal cortex, and microglial activity (Iba-1) in the hippocampus were evaluated., Results: All exercise conditions, except TBI-early, attenuated the significant memory impairment at 24-hour retention caused by TBI. Additionally, Pre-TBI-early treatment led to memory improvement at 3-hour retention. Pre-TBI reduced neuronal death and microglial activation in the hippocampus. TBI-late, but not TBI-early, mitigated hippocampal volume loss, loss of mature neurons in the hippocampus, and inflammation. Combining pre-injury and early-onset exercise reduced memory deficits but did not affect neuronal death or microglial activation.
