To determine intraocular pressure (IOP) estimates using rebound tonometry in juvenile northern bobwhite quails.Eighteen, 28-day-old, wild caught, captive raised, northern bobwhite quails.Birds were manually restrained in sternal recumbency with two fingers placed below the head, applying minimal pressure. Following detailed ophthalmic examination by a board-certified veterinary ophthalmologist, IOP was obtained via rebound tonometry using the Tonovet "P" setting for undefined species in animals without obvious ocular pathology. IOPs were estimated three times on each eye. The mean result of the three estimates was recorded for further analysis. Only readings with low or no reported error were recorded.No difference was detected between the eyes; therefore, a reference interval was calculated using both eyes. The overall IOP (n = 36 eyes) was 5.6 ± 1.2 mmHg (mean ± SD), the median was 6 mmHg, and the range was 2-7 mmHg. The 95% reference interval was 2.2-7.6 mmHg. All quails were ultimately released with no clinically appreciable abnormalities.This study provides information on normal IOP values for juvenile northern bobwhite quails, which can be used to evaluate the ocular health of this species. Future studies with a larger number of animals are needed, as well as validation of the rebound tonometry method using direct manometry.

Northern bobwhites (Colinus virginianus) are a well known game bird that has been extensively studied and managed throughout its range for many decades. Despite this, bobwhites have continued a steady annual decline across the United States, irrespective of many well established conservation practices in place to mitigate this. Supplemental feeding is one such technique that is a widely used tool in bobwhite management, but the methods and results of the studies to investigate the effectiveness of this method are highly variable. The effectiveness of supplemental feeding practices is further hindered by the limited knowledge regarding the nutritional requirements of wild bobwhite because many of the guidelines that researchers follow are based on species of Galliformes that are primarily used for production. Here, we review supplemental feeding studies, nutritional requirements of bobwhite, and discuss future directions.

Disparate resource use originating from phenology of biotic resources, abiotic conditions, and life cycles of exploiting organisms underscores the importance of research across time and space to guide management practices. Our goal was to evaluate resource use of northern bobwhite (Colinus virginianus; bobwhite) at two spatial scales and across three age classes, from hatching through a period of the postjuvenile molt. Our study was conducted at Tall Timbers Research Station, Tallahassee, FL, USA—situated in a landscape subjected to small scale (July 15). Broods were more likely to use areas with greater proportions of fallow fields during the day than for roosting. Broods used roosts with more woody cover and visual obscurity than at available sites. Roosts consisted of less grass and bare ground. However, these effects interacted with age; broods used sparser cover at older ages. Neonate broods were more likely to use cooler roosts with greater thermal stability, but this effect was reversed for juveniles. Broods may alter resource use with changes in vulnerabilities to threats such as thermal risks and predation., We documented resource use of a ground‐dwelling precocial bird in a fire‐managed landscape was conditional on multiple spatially and temporally variable factors. Particularly, our findings demonstrated that resource use was largely dependent on scale, physiological development, behavioral activity, and land management practices—some of which interacted with each other to create complex relationships.

International audience; Understanding how predators affect prey species is a central endeavor in applied ecology. Game birds are a culturally and economically important group of birds throughout the world. Specifically, northern bobwhite (Colinus virginianus) is an imperiled game bird native to North America that has declined precipitously over the past 65 years. Concurrently, raptor populations increased substantially as a result of pesticide bans and legal protections. However, relationships between raptors and bobwhites are not well-understood because of limited long-term data. We analyzed long-term raptor survey and bobwhite survival datasets from 2008 to 2018 to determine if oscillations in raptor abundance affected bobwhite survival. We used a novel open multi-species dynamics hierarchical distance sampling model to estimate the abundance of raptors. We used a known-fate survival model to determine if variation in raptor abundance affected bobwhite survival. We had multiple working hypotheses regarding biological relationships between raptor abundance and bobwhite survival. Raptors affected bobwhites in every biological season but were more influential in the breeding season and late winter supporting the notion of bobwhite behavior and raptor migration were driving observed patterns. Our results suggest that even in areas with abundant habitat, predators exert top-down influences on vital rates suggesting similar or greater influences on populations under poor habitat conditions.

Neonicotinoid insecticides are among the latest class of insecticides that can have harmful effects on birds. Approximately 30 000 kg of neonicotinoid insecticides are applied annually to 429 100 ha of row-crop fields within the South Texas Plains Ecoregion, Texas, USA. Various studies have demonstrated that treated seeds can be highly toxic to northern bobwhites, with the consumption of only 20 corn seeds causing a fatality. Similarly, other studies have indicated that neonicotinoid insecticides can reduce arthropod populations-a substantial prey base for northern bobwhites, especially during the breeding season-by approximately 60%. Our objective was to conduct an ecological risk assessment of neonicotinoid insecticides' impact on northern bobwhite (Colinus virginianus) populations in the South Texas Plains Ecoregion. We estimated that juvenile and adult northern bobwhites could intake from 7.32 to 27.0 mg/kg/day and from 10.0 to 37.5 mg/kg/day of neonicotinoid insecticides, respectively, which can cause adverse effects on growth, reproductive output, and long-term survival. Our study determined that the application of 30 000 kg of neonicotinoid insecticides annually in the South Texas Plains Ecoregion harms the region's northern bobwhite that are exposed to neonicotinoids. Terrestrial toxicology data are limited for comprehensive ecotoxicological risk assessment (ERA) of ecosystems contaminated by per- and polyfluoroalkyl substances (PFAS) partly due to their existence as mixtures in the environment. This complicates logistical dose-response modeling and establishment of a threshold value characterizing the chronic toxicity of PFAS to ecological receptors. We examined reproduction, growth, and survival endpoints using a combination of hypothesis testing and logistical dose-response modeling of Northern Bobwhite quail (Colinus virginianus) exposed to perfluorohexanoic acid (PFHxA) alone and to PFHxA in a binary mixture with perfluorooctane sulfonic acid (PFOS) via the drinking water. The exposure concentration chronic toxicity value (CTV) representative of the lowest-observable-adverse-effect-level (LOAEL) threshold for chronic oral PFAS toxicity (based on reduced offspring weight and growth rate) was 0.10 ng/mL for PFHxA and 0.06 ng/mL for a PFOS:PFHxA (2.7:1) mixture. These estimates corresponded to an adult LOAEL average daily intake (ADI) CTV of 0.0149 and 0.0082 µg x kg bw-1 x d-1 , respectively. Neither no-observable-adverse-effect-level (NOAEL) threshold and representative CTVs nor a dose-response and predicted effective concentration (ECx ) values could be established for these two response variables. The findings indicate that reaction(s) occur among the individual PFAS components present in the mixture to alter the potential toxicity, demonstrating that mixture effects avian PFAS toxicity. Thus, chronic oral PFAS toxicity to avian receptors represented as the sum of the individual compound toxicities may not necessarily be the best method for assessing chronic mixture exposure risk at PFAS-contaminated sites. Resource selection is a dynamic process driven by habitat valuation and risk avoidance in heterogeneous landscapes. Resource selection and movement decisions of individuals may be sensitive to intrinsic factors, such as body condition, and variation in these choices may have consequences on subsequent survival. We evaluated northern bobwhite (Colinus virginianus) brood resource selection patterns to quantify utility of different cover types during the development period using integrated step-selection analysis in a Bayesian hierarchical modeling framework with three brood stages: flightless broods ≤ 14 days old, dependent broods 15–35 days old, and independent broods over 35 days old. Broods showed strongest selection for native grasslands that were burned and grazed at least once in the previous two years, and agricultural fields. Brood mobility improved with age; broods > 35 days old travelled farther on average and took daily steps > 200 m more frequently than younger broods. Young broods ≤ 14 days old did not select for idle native grasslands, while broods > 35 days old did select for that cover type. Young broods also selected areas farther from trees compared to older broods. We evaluated the survival consequences of resource selection by comparing patterns in choices of broods that succeeded to choices of broods that failed to survive to 35 days. Successful broods chose habitats with more shrub cover and areas farther from trees compared to failed broods. Our results suggest that conservation planning should consider age-specific patterns in habitat use and demographic consequences of habitat choice for greatest effectiveness.

Many recent studies have been focused on prevalence and impact of two helminth parasites, eyeworm Oxyspirura petrowi and caecal worm Aulonocephalus pennula, in the northern bobwhite quail (Colinus virginianus). However, few studies have attempted to examine the effect of these parasites on the bobwhite immune system. This is likely due to the lack of proper reference genes for relative gene expression studies. Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) is a glycolytic enzyme that is often utilized as a reference gene, and in this preliminary study, we evaluated the similarity of bobwhite GAPDH to GAPDH in other avian species to evaluate its potential as a reference gene in bobwhite. GAPDH was identified in the bobwhite full genome sequence and multiple sets of PCR primers were designed to generate overlapping PCR products. These products were then sequenced and then aligned to generate the sequence for the full-length open reading frame (ORF) of bobwhite GAPDH. Utilizing this sequence, phylogenetic analyses and comparative analysis of the exon–intron pattern were conducted that revealed high similarity of GAPDH encoding sequences among bobwhite and other Galliformes. Additionally, This ORF sequence was also used to predict the encoded protein and its three-dimensional structure which like the phylogenetic analyses reveal that bobwhite GAPDH is similar to GAPDH in other Galliformes. Finally, GAPDH qPCR primers were designed, standardized, and tested with bobwhite both uninfected and infected with O. petrowi, and this preliminary test showed no statistical difference in expression of GAPDH between the two groups. These analyses are the first to investigate GAPDH in bobwhite. These efforts in phylogeny, sequence analysis, and protein structure suggest that there is > 97% conservation of GADPH among Galliformes. Furthermore, the results of these in silico tests and the preliminary qPCR indicate that GAPDH is a prospective candidate for use in gene expression analyses in bobwhite.

To further characterize avian toxicity to environmental levels of select per- and poly-fluoroalkyl substances (PFAS), we established species- and tissue-specific PFAS chronic toxicity values (CTVs) associated with a lowest-observable-adverse effect level (LOAEL) threshold previously established for northern bobwhite quail (Colinus virginianus) chronically orally exposed via drinking water to either perfluorooctane sulfonate (PFOS) or a simple PFAS mixture. Aided by advances in analytical techniques, the novel avian oral PFAS CTVs reported in the present study are lower than the previously reported toxicity reference values (TRVs) estimated for birds chronically exposed via feed. Thus, current avian PFOS TRVs may not be fully protective of wild avian populations at PFAS-impacted sites. Also, likely due to differences in bioavailability, bioaccessibility, and toxicokinetics among individual PFAS between oral exposure types, we found higher bioaccumulation factors in all assessed tissues from birds exposed via water versus feed. Thus, we propose that future characterization of chemical toxicity due to ingestion exposure initially include a full examination of all probable sources of oral exposure for the most accurate derivation of TRVs and a more complete picture of ecological risk. The avian PFAS LOAEL CTVs established in the present study can be modified with the use of uncertainty factors to derive site-specific avian TRVs for ecological risk assessment at PFAS-impacted sites. From differences observed in the behavior of PFOS when administered as either a single chemical or part of a binary mixture with perfluorohexane sulfonate (PFHxS), we verified that PFOS was absorbed and distributed differently when coadministered with PFHxS and that PFOS likely interacted with PFHxS differently among tissues, helping to explain the differences observed in avian toxicity between exposures. The potential of parasites to affect host abundance has been a topic of heated contention within the scientific community for some time, with many maintaining that issues such as habitat loss are more important in regulating wildlife populations than diseases. This is in part due to the difficulty in detecting and quantifying the consequences of disease, such as parasitic infection, within wild systems. An example of this is found in the Northern bobwhite quail (Colinus virginanus), an iconic game bird that is one of the most extensively studied vertebrates on the planet. Yet, despite countless volumes dedicated to the study and management of this bird, bobwhite continue to disappear from fields, forest margins, and grasslands across the United States in what some have referred to as “our greatest wildlife tragedy”. Here, we will discuss the history of disease and wildlife conservation, some of the challenges wildlife disease studies face in the ever-changing world, and how a “weight of evidence” approach has been invaluable to evaluating the impact of parasites on bobwhite in the Rolling Plains of Texas. Through this, we highlight the potential of using “weight of the evidence” to better understand the complex effects of diseases on wildlife and urge a greater consideration of the importance of disease in wildlife conservation.

Long-chain per- and poly-fluoroalkyl substances (PFAS) have been the active ingredients in firefighting foams for more than 50 years. Due to their extreme persistence, regulatory agencies are concerned about their potential adverse environmental and health impacts. Recently, nonfluorinated chemical constituents have been proposed for use in fire-fighting foams in an effort to reduce the potential negative impacts of PFAS on terrestrial and aquatic flora and fauna. However, it is important to also determine the potential ecotoxicity of these nonfluorinated foam products, because we have little toxicological information for many of them. In preparation for a chronic study, we conducted an acute (24-h) oral toxicity test in northern bobwhite quail (Colinus virginianus) using six different fluorine-free foams; five were commercial foams (BioEx ECOPOL A, Fomtec Enviro USP, National Foam Avio Green KHC, National Foam NFD 20-391, and Solberg Re-Healing Foam), and one was an experimental foam (NRL 502W). A short-chain PFAS-based foam (Buckeye Platinum Plus C6) was also evaluated for comparative purposes. Groups of five birds were initially pseudogavaged with a volume of each product corresponding to a "limit" (the highest exposure concentration expected to occur environmentally). Only one bird (1 of 35) died during the limit test, indicating that all seven products have an acute median lethal dose in adult quail at or above the limit (~1500 mg/kg body wt). According to the adaptive modulation hypothesis, digestive enzyme activities are matched to their respective dietary substrate level so that ingested nutrients are not wasted in excreta due to insufficient digestive capacity, and so membrane space or expenditures building/maintaining the intestinal hydrolytic machinery are not wasted when substrate levels are low. We tested predictions in juvenile northern bobwhites (Colinus virginianus) and juvenile and adult domestic chickens (Gallus gallus domesticus) by feeding them on diets varying in starch, protein, and lipid composition for 7-9 d (bobwhites) or 15 d (chickens). Birds were euthanized, intestinal tissue harvested, and enzyme activities measured in tissue homogenates from proximal, medial and distal small intestine. We found that (1) α-glucosidase (AG; maltase and sucrase) activities were induced by dietary starch in both juvenile and adult chickens but not in northern bobwhites; (2) aminopeptidase-N (APN) activities were induced by dietary protein in both bobwhites and juvenile but not adult chickens; (3) AG activities were suppressed by an increase in dietary lipid in both bobwhites and juvenile but not adult chickens; and (4) APN activities were not suppressed by high dietary lipid in any birds. We review findings from 35 analogous trials in 16 avian species. 100% of avian omnivores modulate at least one enzyme in response to change in dietary substrate level. AG induction by dietary carbohydrate occurs in more members of Galloanserae than in Neoaves, and all omnivorous members of Neoaves tested so far increase APN activity on high dietary protein, whereas fewer of the Galloanserae do.

Background The influence of vegetative changes due to livestock grazing on grassland birds is well-recognized because these birds are heavily influenced by vegetative structure. Traditionally, species distribution models (SDMs) use direct variables, resources that the animal consumes or requires to persist in an area (e.g., water) to define and project a species’ niche and distribution. Indirect variables, which are features the animal does not consume or require for persistence but with which it may still interact, are often excluded. Our objective was to improve the traditional SDMs projecting the distribution of three summer resident South Texas grassland birds (Northern Bobwhite Colinus virginianus, Eastern Meadowlark Sturnella magna, and Cassin’s Sparrow Peucaea cassinii) by incorporating livestock grazing pressure, an indirect variable, into five SDM algorithms: BioClim, generalized linear model, MaxEnt, boosted regression tree, and random forest. We collected data from the Coloraditas Grazing Research and Demonstration Area (CGRDA), a 7684-ha area located on the San Antonio Viejo Ranch (SAV) in South Texas. We used several relevant environmental characteristics to build SDMs and compared model performance (AUC and TSS) with and without grazing pressure as an indirect variable. Results Machine learning models (MaxEnt and random forest) had the highest predictive performance for all species, with random forest being the most consistent for each analysis. BioClim and generalized linear model remained constant or only marginally improved with the addition of the grazing pressure. Conclusions Our findings suggest that model selection for SDM should include consideration of species prevalence, and machine-learning algorithms should be preferred when the target species is of low or unknown prevalence. Further, livestock grazing has measurable influence on grassland bird species’ distributions and should be included in SDMs as an indirect variable in addition to associated vegetative changes.

Northern bobwhites (Colinus virginianus) are small quails in the New World Quail family (Odontophoridae) and are one of the most phenotypically diverse avian species. Despite extensive research on bobwhite ecology, genomic studies investigating the evolution of phenotypic diversity in this species are lacking. Here, we present a new, highly contiguous assembly for bobwhites using tissue samples from a vouchered, wild, female bird collected in Louisiana. By performing a de novo assembly and scaffolding the assembly with Dovetail Chicago and HiC libraries and the HiRise pipeline, we produced an 866.8 Mb assembly including 1,512 scaffolds with a scaffold N50 of 66.8 Mb, a scaffold L90 of 17, and a BUSCO completeness score of 90.8%. This new assembly represents approximately 96% of the non-repetitive and 84% of the entire bobwhite genome size, greatly improves scaffold lengths and contiguity compared to an existing draft bobwhite genome, and provides an important tool for future studies of evolutionary and functional genomics in bobwhites.

The effects of flock size of European starlings (Sturnus vulgaris) was experimentally manipulated to assess the potential of influenza A virus (IAV; H4N6) transmission from a flocking passerine to bobwhite quail (Colinus virginianus) through shared food and water resources to mimic starling intrusions into free-range and backyard poultry operations. Of the three starling flock sizes tested (n = 30, n = 20, and n = 10), all successfully transmitted the virus to all or most of the quail in each animal room (6/6, 6/6, and 5/6) by the end of the experimental period, as determined by seroconversion and/or viral RNA shedding. Although starlings have been shown to be inconsistent shedders of IAVs and when they do replicate and subsequently shed virus they typically do so at low to moderate levels, this study has provided evidence that relatively small flocks (i.e., 10 or possibly a smaller number) of this species can collectively transmit the virus to a highly susceptible gallinaceous bird species. Future work should assess if starlings can transmit IAVs to additional poultry species commonly found in backyard or free-range settings. This article is protected by copyright. All rights reserved.

As phylogenomics focuses on comprehensive taxon sampling at the species and population/subspecies levels, incorporating genomic data from historical specimens has become increasingly common. While historical samples can fill critical gaps in our understanding of the evolutionary history of diverse groups, they also introduce additional sources of phylogenomic uncertainty, making it difficult to discern novel evolutionary relationships from artifacts caused by sample quality issues. These problems highlight the need for improved strategies to disentangle artifactual patterns from true biological signal as historical specimens become more prevalent in phylogenomic datasets. Here, we tested the limits of historical specimen-driven phylogenomics to resolve subspecies-level relationships within a highly polytypic family, the New World quails (Odontophoridae), using thousands of ultraconserved elements (UCEs). We found that relationships at and above the species-level were well-resolved and highly supported across all analyses, with the exception of discordant relationships within the two most polytypic genera which included many historical specimens. We examined the causes of discordance and found that inferring phylogenies from subsets of taxa resolved the disagreements, suggesting that analyzing subclades can help remove artifactual causes of discordance in datasets that include historical samples. At the subspecies-level, we found well-resolved geographic structure within the two most polytypic genera, including the most polytypic species in this family, Northern Bobwhites (Colinus virginianus), demonstrating that variable sites within UCEs are capable of resolving phylogenetic structure below the species level. Our results highlight the importance of complete taxonomic sampling for resolving relationships among polytypic species, often through the inclusion of historical specimens, and we propose an integrative strategy for understanding and addressing the uncertainty that historical samples sometimes introduce to phylogenetic analyses.

Per- and polyfluoroalkyl substances (PFAS) are globally distributed and present in nearly every environmental compartment. Characterizing the chronic toxicity of individual PFAS compounds and mixtures is necessary because many have been reported to cause adverse health effects. To derive toxicity reference values (TRVs) and conduct ecotoxicological risk assessments (ERAs) of PFAS-contaminated ecosystems for wildlife, species-specific PFAS chronic toxicity values (CTVs) are needed. The present study quantified PFAS residues from liver and eggs of birds chronically exposed to perfluorohexanoic acid (PFHxA) or a mixture of perfluorooctane sulfonate (PFOS) and PFHxA that produced a no-observable-adverse-effect level (NOAEL) and/or a lowest-observable-adverse-effectlevel (LOAEL). The CTVs we present are lower than those previously reported for birds and should be considered in future regulatory evaluations. From the estimated species- and tissue-specific PFAS CTVs, we found that PFOS and perfluorohexane sulfonate (PFHxS) were more bioaccumulative than PFHxA in avian tissues, but PFHxA was more toxic to reproducing birds than either PFOS or a PFOS:PFHxS mixture. We further determined that avian toxicity was not necessarily additive with respect to PFAS mixtures, which could have implications for PFAS ERAs. West Nile virus (WNV) has been implicated in regional declines of numerous North American bird species, although its potential impact upon many species, including some game birds, remains unknown. Specifically, information about susceptibility to infection and infection outcome are crucial to assessing health risks. Northern bobwhite quail (Colinus virginianus) are a popular and common game bird across much of the United States, as well as in captive breeding programs and as backyard birds. Two age groups of bobwhites were subcutaneously inoculated with WNV and euthanatized on 15 days postinoculation (DPI). Three of 10 inoculated 5-wk-old and 4/10 inoculated 15-wk-old birds developed detectable viremia titers during 1–5 DPI, with low peak titers (101.7–103.0 plaque-forming units [PFU]/ml). Three of 10 inoculated 5-wk-old and 1/10 inoculated 15-wk-old birds shed low viral titers (peak 100.7–101.8 PFU/swab) either orally or cloacally or both for limited periods from 2 to 6 DPI. All inoculated birds (n = 20) remained apparently healthy and seroconverted by 15 DPI. No infectious virus was detected in select tissues: heart, kidney, brain, skeletal muscle, spleen (15-wk-old group only), and feathers from any of the bobwhites. No sham-inoculated, contact control birds (n = 8) became viremic or had virus isolated from tissues or swabs. The most consistent microscopic lesion was minimal to mild, lymphoplasmacytic myocarditis (6/10 in 5-wk-olds; 5/10 in 15-wk-olds). Immunohistochemical labeling was most often in macrophages in spleen and bone marrow, likely reflective of clearance of infection. There were no statistically significant differences in the peak viremia and shedding titers between age groups and no differences in the development of WNV-associated lesions between the two age groups. These results suggest that WNV is unlikely to pose a health risk to bobwhites and that bobwhites likely are an incompetent reservoir host species in WNV transmission.

Flush-count surveys of game bird broods are a common method of deriving estimates of brood survival, but detection of chicks during surveys is < 1 due to factors such as vegetation obscurity, adult brooding behavior and variation by observer. Radio-telemetry is an alternative method for estimating survival that circumvents such factors and allow for higher detection. However, this practice is costly and labor-intensive and therefore not readily adopted. We sought to estimate detection probability of chicks during flush-counts as a function of vegetation height and adult brooding behavior. Secondly, we evaluated compromises in detection by comparing estimates of brood survival derived from flush-counts and radiotelemetry. Lastly, we compared counts between two observers to discern whether an additional observer could increase accuracy of counts.We radiomarked 247 northern bobwhite Colinus virginianus chicks at 10–12 days of age and conducted 46 flush-counts at 21 days of age. Vegetation height substantially decreased detection (β = –1.18; 95 CrI: –1.68 to –0.73); mean detection probability was 0.30 (95 CrI: 0.22–0.40). Observers failed to detect radio-marked chicks when adults exhibited running behavior (n = 16 chicks, n = 6 surveys), orphaning occurred (n = 11 chicks, n = 5 surveys) or brooding adults died or had transmitter failures (n = 4 chicks, n = 3 surveys). An additional observer did not affect counts with a mean difference of –0.6 chicks (95 CrI: –4.0 to 2.7) counted between observers. Chicks were not detected during 47% of surveys when ≥ 1 radio-marked individuals were known to be alive. Brood survival was 0.83 (95 CrI: 0.70–0.92) and 0.48 (95 CrI: 0.34–0.62) for radiotelemetry and naive flush-counts, respectively. Because of low detectability of chicks during flush-counts, alternative methods should be considered by future researchers.

West Nile virus (WNV) has been implicated in regional declines of numerous North American bird species, although its potential impact upon many species, including some game birds, remains unknown. Specifically, information about susceptibility to infection and infection outcome are crucial to assessing health risks. The Northern bobwhite quail (Colinus virginianus) is an important gamebird among hunters that has been experiencing a nationwide decline for > 50 yr. In West Texas, one of the last regions to experience this downward trend, research on bobwhite populations has focused on habitat variables and, increasingly, on parasitic infection. In bobwhite, two of the most common parasites are the caecal worm (Aulonocephalus pennula) and eyeworm (Oxyspirura petrowi). To better document the state of bobwhite populations in the Rolling Plains Ecoregion, trapping, summer rooster counts, fall covey counts, and parasitic infection assessments were conducted in three counties during 2018. These efforts were compared with previous years for a longitudinal perspective. In 2018, bobwhite populations experienced a widespread decline, although some counties surveyed fared slightly better than others. More effort was required to trap fewer total bobwhite, and fewer roosters and coveys were counted than in previous years. In addition, in 2018, parasitic infection levels of caecal and eyeworms were higher than or similar to levels in previous years. Additional research is necessary to understand which factors influence bobwhite populations in allopatric locations and over time.

Human hunting activity and disturbance can significantly impact prey species through both consumptive and nonconsumptive effects. The nonconsumptive effects of rabbit hunting on Northern Bobwhite (Colinus virginianus; hereafter, bobwhite) are currently unknown. Increased perceived risk of predation by bobwhite during rabbit hunting events may elicit antipredator responses among bobwhite that impact fitness via changes in behavior that ultimately impact population growth. We estimated the nonconsumptive effects of rabbit hunting on bobwhite behavior using telemetry across varying rabbit hunting intensities. Movements were analyzed using Bayesian hierarchical modeling with a before‐after‐control‐impact (BACI) design to determine the effect of rabbit hunting on bobwhite. We observed an overall reduction in bobwhite movement in the presence of rabbit hunting, with a 38% (Posterior Overlap = 0.01) increase in bobwhite step length in the absence of rabbit hunting. We also observed bobwhite maintaining closer proximity to hardwood and escape cover under high rabbit hunting intensity, with a 59% (Posterior Overlap = 0.03) increase in distance from hardwood and a 28% (Posterior Overlap = 0.14) increase in distance from escape cover when rabbit hunting was removed. Synthesis and applications. Heightened antipredator behavior through decreased movement may assist with bobwhite predator avoidance. However, decreased movement and increased use of poor habitats may also have negative effects as a result of reduced foraging time or increased susceptibility to other predators. Future research should attempt to quantify the effect of decreased movement on bobwhite fitness through the evaluation of foraging time and survival in order to continue to improve management efforts for the species.

Despite the fact that the northern bobwhite (Colinus virginianus Linnaeus; hereafter, bobwhite) is one of the most well-studied and widely distributed wildlife species in North America, we know little about how bobwhite respond to oil and gas infrastructure. We investigated the impacts of oil and gas development on space use of bobwhite using a multiseason approach. We captured and monitored bobwhite in the breeding season (1 April–30 September, n = 135 individuals) and nonbreeding season (1 October–31 March, n = 30 coveys) and modeled their habitat selection in a resource-utilization function (third order, within home range selection) and resource-selection function (second order, home range selection) format. Generally, energy infrastructure effects on bobwhite were neutral, but breeding season bobwhite did select for areas near low-traffic roads (β = − 0.31 ± 0.15 SE). In the nonbreeding season, coveys selected for areas within their home range with a limited viewshed (i.e., areas with limited visibility of anthropogenic structures; β = − 0.03 ± 0.02 SE). Selection differed between sexes for well pads (t = − 2.12, P = 0.04) but was otherwise similar. At the level of home range selection, bobwhite exhibited a preference for areas with a low density of oil and gas wells and a high density of low-traffic roads during both the breeding and nonbreeding seasons (breeding: βwell = − 0.14 ± 0.02, βroad = 0.26 ± 0.27; nonbreeding: βwell = − 0.08 ± 0.03, βroad = 0.16 ± 0.03). As a generalist species, bobwhite appear to be largely tolerant of energy infrastructure and associated disturbances at moderate levels of development but may be sensitive to high densities of oil and gas pads.

The northern bobwhite quail (Colinus virginianus) is a popular gamebird in the Rolling Plains Ecoregion of West Texas. However, there has been a population decline in this area over recent decades. Consistent reports indicate a high prevalence of the eyeworm (Oxyspirura petrowi) and caecal worm (Aulonocephalus pennula), which may be of major influence on the bobwhite population. While research has suggested pathological consequences and genetic relatedness to other pathologically significant parasites, little is known about the influence of climate on these parasites. In this study, we examined whether seasonal temperature and precipitation influences the intensity of these parasites in bobwhite. We also analyzed quantitative PCR results for bobwhite feces and cloacal swabs against temperature and precipitation to identify climatic impacts on parasite reproduction in this region. Multiple linear regression analyses were used for parasite intensity investigation while binary logistic regression analyses were used for parasite reproduction studies. Our analyses suggest that caecal worm intensity, caecal worm reproduction, and eyeworm reproduction are influenced by temperature and precipitation. Temperature data was collected 15, 30, and 60 days prior to the date of collection of individual bobwhite and compared to qPCR results to generate a temperature range that may influence future eyeworm reproduction. This is the first preliminary study investigating climatic influences with predictive statistics on eyeworm and caecal worm infection of northern bobwhite in the Rolling Plains. One challenge in avian embryology is establishing a standard developmental timetable, primarily because eggs incubated for identical durations can vary in developmental progress, even within the same species. For remedy, avian development is classified into distinct stages based on the formation of key morphological structures. Developmental stages exist for a few galliform species, but the literature is lacking a description of normal stages for California valley quail (Callipepla californica). Thus, the objective of this study was to stage and document the morphological and structural development of California valley quail. Over two laying seasons, 390 eggs were incubated at 37.8֯ C in 60% RH for ≤23 days. Eggs were opened every ≤6 hours to document embryonic development, including, blastoderm diameter, anterior angle of nostril to beak tip, and lengths of wing, tarsus, third toe, total beak, total foot, and embryo. California valley quail embryos were staged and compared to domestic chicken (Gallus gallus domesticus), the staging standard for galliformes, as well as Japanese quail (Coturnix japonica), blue-breasted quail (Synoicus chinensis) and northern bobwhite quail (Colinus virginianus). This study produced the first description of the 43 normal stages of development for California valley quail. Compared with other galliformes, the California valley quail has a different number of stages and displays developmental heterochrony in stages 1–24, and morphological and developmental differences in stages 25–hatch. The observed differences emphasize the importance of staging individual avian species instead of relying on poultry animal models or close relatives for developmental reference. This is extremely important in species-specific embryological studies that evaluate critical windows of development or evaluate the impacts of environmental change on avian development. This study also suggests that staging frequencies of ≤6 hours and egg transport protocols should be standardized for future staging studies.

The Northern bobwhite quail (Colinus virginianus) is a popular game bird that has been experiencing a well-documented decline throughout Texas since the 1960s. While much of this decline has been attributed to habitat loss and fragmentation, recent studies have identified other factors that may also contribute to decreasing quail populations. Parasites, in particular, have become increasingly recognized as possible stressors of quail, and some species, particularly the eyeworm (Oxyspirura petrowi) and cecal worm (Aulonocephalus pennula) are highly prevalent in Texas quails. Eyeworm infection has also been documented in some passerines, suggesting helminth infection may be shared between bird species. However, the lack of comprehensive helminth surveys has rendered the extent of shared infection between quail and passerines in the ecoregion unclear. Thus, helminth surveys were conducted on bobwhite, scaled quail (Callipepla squamata), Northern mockingbirds (Mimus polyglottos), curve-billed thrashers (Toxistoma curvirostre), and Northern cardinals (Cardinalis cardinalis) to contribute data to existing parasitological gaps for birds in the Rolling Plains ecoregion of Texas. Birds were trapped across 3 counties in the Texas Rolling Plains from March to October 2019. Necropsies were conducted on 54 individuals (36 quail and 18 passerines), and extracted helminths were microscopically identified. Nematode, cestode, and acanthocephalan helminths representing at least 10 helminth species were found. Specifically, A. pennula and O. petrowi had the highest prevalence, and O. petrowi was documented in all of the study species. This research adds to the body of knowledge regarding parasitic infections in quail and passerines of the Rolling Plains ecoregion and highlights the potential consequences of shared infection of eyeworms among these bird species.

Populations of northern bobwhites (Colinus virginianus; hereafter bobwhite) have been declining across their geographic range in North America, prompting consideration of the role parasites may play. We conducted this study to learn about the helminth fauna in South Texas, a region that supports a sustainable bobwhite population. Helminths were examined from 356 bobwhites collected during the 2014-2015 (n = 124) and 2015-2016 (n = 232) hunting seasons, when increasing trends in precipitation were observed in comparison with the previous two years. Ten helminth species were found, consisting of 14,127 individuals. Of these, all are heteroxenous parasites and three are pathogenic (Dispharynx nasuta, Tetrameres pattersoni and Oxyspirura petrowi). Aulonocephalus pennula numerically dominated the component community (81% prevalence, 99% of the total helminths found), whereas each of the remaining species occurred rarely (≤9% prevalence) and contributed few individuals (≤0.4%) to the helminth community. Prevalence and abundance of A. pennula were not influenced by host age, sex or body mass, but abundance was higher during the 2014-2015 than the 2015-2016 hunting season. Our findings indicate that the helminth community in bobwhites from South Texas can vary during long-term, highly variable precipitation conditions and these communities are more similar to those found in the Rolling Plains of Texas than those found in the eastern part of the bobwhite's geographic range in the US.

Resource selection is a key component in understanding the ecological processes underlying population dynamics, particularly for species such as northern bobwhite (Colinus virginianus), which are declining across their range in North America. There is a growing body of literature quantifying breeding season resource selection in bobwhite; however, winter information is particularly sparse despite it being a season of substantial mortality. Information regarding winter resource selection is necessary to quantify the extent to which resource requirements are driving population change. We modeled bobwhite fall and winter resource selection as a function of vegetation structure, composition, and management from traditionally (intensively) managed sites and remnant (extensively managed) grassland sites in southwest Missouri using multinomial logit discrete choice models in a Bayesian framework. We captured 158 bobwhite from 67 unique coveys and attached transmitters to 119 individuals. We created 671 choice sets comprised of 1 used location and 3 available locations. Bobwhite selected for locations which were closer to trees during the winter; the relative probability of selection decreased from 0.45 (85% Credible Interval [CRI]: 0.17–0.74) to 0.00 (85% CRI: 0.00–0.002) as distance to trees ranged from 0–313 m. The relative probability of selection increased from near 0 (85% CRI: 0.00–0.01) to 0.33 (85% CRI: 0.09–0.56) and from near 0 (85% CRI: 0.00–0.00) to 0.51 (85% CRI: 0.36–0.71) as visual obstruction increased from 0 to 100% during fall and winter, respectively. Bobwhite also selected locations with more woody stems; the relative probability of selection increased from near 0.00 (85% CRI: 0.00–0.002) to 0.30 (85% CRI: 0.17–0.46) and near 0.00 (85% CRI: 0.00–0.001) to 0.35 (85% CRI: 0.22–0.55) as stem count ranged from 0 to 1000 stems in fall and winter, respectively. The relative probability of selection also decreased from 0.35 (85% CRI: 0.20–0.54) to nearly 0 (85% CRI: 0.00–0.001) as percent grass varied from 0 to 100% in fall. We suggest that dense shrub cover in close proximity to native grasslands is an important component of fall and winter cover given bobwhite selection of shrub cover and previously reported survival benefits in fall and winter.

Many recent studies have been focused on prevalence and impact of two helminth parasites, eyeworm Oxyspirura petrowi and caecal worm Aulonocephalus pennula, in the northern bobwhite quail (Colinus virginianus). However, few studies have attempted to examine the effect of these parasites on the bobwhite immune system. This is likely due to the lack of proper reference genes for relative gene expression studies. Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) is a glycolytic enzyme that is often utilized as a reference gene, and in this preliminary study, we evaluated the similarity of bobwhite GAPDH to GAPDH in other avian species to evaluate its potential as a reference gene in bobwhite. GAPDH was identified in the bobwhite full genome sequence and multiple sets of PCR primers were designed to generate overlapping PCR products. These products were then sequenced and then aligned to generate the sequence for the full-length open reading frame (ORF) of bobwhite GAPDH. Utilizing this sequence, phylogenetic analyses and comparative analysis of the exon-intron pattern were conducted that revealed high similarity of GAPDH encoding sequences among bobwhite and other Galliformes. Additionally, This ORF sequence was also used to predict the encoded protein and its three-dimensional structure which like the phylogenetic analyses reveal that bobwhite GAPDH is similar to GAPDH in other Galliformes. Finally, GAPDH qPCR primers were designed, standardized, and tested with bobwhite both uninfected and infected with O. petrowi, and this preliminary test showed no statistical difference in expression of GAPDH between the two groups. These analyses are the first to investigate GAPDH in bobwhite. These efforts in phylogeny, sequence analysis, and protein structure suggest that there is 97% conservation of GADPH among Galliformes. Furthermore, the results of these in silico tests and the preliminary qPCR indicate that GAPDH is a prospective candidate for use in gene expression analyses in bobwhite.

A flock of captive bobwhite quail (Colinus virginianus) experienced loose droppings, depression, and increased mortality starting at 3 wk of age. Necropsy of the affected birds revealed intestines dilated with frothy and tan fluid. Irregular dark brown fissures within the koilin layer of the gizzard were found in 20%-30% of the birds. Histologically, gizzards showed multifocal koilin degeneration or fragmentation, degeneration and necrosis of the subjacent epithelial cells, and infiltration of macrophages, lymphocytes, and heterophils. Necrotic epithelial cells occasionally contained large, smudgy, basophilic intranuclear inclusion bodies with marginated nuclear chromatin. Adenoviral paracrystalline arrays composed of icosahedral virions (60-70 nm diameter) were seen on transmission electron microscopy in the nuclei of epithelial cells in the gizzard mucosa. Adenovirus was isolated from gizzard, liver, intestine, and trachea by inoculation of specific-pathogen-free embryonated chicken eggs. Homogenates of the gizzard, liver, and intestine were positive for the adenovirus hexon gene by PCR. Sequencing of PCR amplicons confirmed the virus as fowl aviadenovirus A. The study isolates showed more than 99% and 97% nucleotide identity with quail bronchitis virus and with aviadenoviruses from gizzard erosion and ulceration (GEU) in broilers, respectively. The viral isolates showed six substitutions (G1T, C174A, A229G, C513A, T579A, and G621C) of which two were nonsynonymous (G1T and A229G), resulting in a change in the translated amino acid as A1S and S77G, respectively. These results indicate that adenoviruses of the same type or species can cause different clinical presentations in quails, e.g., bronchitis or GEU.

Understanding how predators affect prey species is a central endeavor in applied ecology. Game birds are a culturally and economically important group of birds throughout the world. Specifically, northern bobwhite (Colinus virginianus) is an imperiled game bird native to North America that has declined precipitously over the past 65 years. Concurrently, raptor populations increased substantially as a result of pesticide bans and legal protections. However, relationships between raptors and bobwhites are not well-understood because of limited long-term data. We analyzed long-term raptor survey and bobwhite survival datasets from 2008 to 2018 to determine if oscillations in raptor abundance affected bobwhite survival. We used a novel open multi-species dynamics hierarchical distance sampling model to estimate the abundance of raptors. We used a known-fate survival model to determine if variation in raptor abundance affected bobwhite survival. We had multiple working hypotheses regarding biological relationships between raptor abundance and bobwhite survival. Raptors affected bobwhites in every biological season but were more influential in the breeding season and late winter supporting the notion of bobwhite behavior and raptor migration were driving observed patterns. Our results suggest that even in areas with abundant habitat, predators exert top-down influences on vital rates suggesting similar or greater influences on populations under poor habitat conditions.

Loss and degradation of grasslands in the Great Plains region have resulted in major declines in abundance of grassland bird species. To ensure future viability of grassland bird populations, it is crucial to evaluate specific effects of environmental factors among species to determine drivers of population decline and develop effective conservation strategies. We used threshold models to quantify the effects of land cover and weather changes in "lesser prairie‐chicken" and "greater prairie‐chicken" (Tympanuchus pallidicinctus and T. cupido, respectively), northern bobwhites (Colinus virginianus), and ring‐necked pheasants (Phasianus colchicus). We demonstrated a novel approach for estimating landscape conditions needed to optimize abundance across multiple species at a variety of spatial scales. Abundance of all four species was highest following wet summers and dry winters. Prairie chicken and ring‐necked pheasant abundance was highest following cool winters, while northern bobwhite abundance was highest following warm winters. Greater prairie chicken and northern bobwhite abundance was also highest following cooler summers. Optimal abundance of each species occurred in landscapes that represented a grassland and cropland mosaic, though prairie chicken abundance was optimized in landscapes with more grassland and less edge habitat than northern bobwhites and ring‐necked pheasants. Because these effects differed among species, managing for an optimal landscape for multiple species may not be the optimal scenario for any one species., We quantified the effects of land cover and weather changes in Galliformes in Kansas. We found evidence of nonlinear, threshold effects of land cover change in abundance of all four focal species, suggesting that the presence of both grassland and cropland is important in the landscape to simultaneously maintain these populations.

Harvest limits are an important component of wildlife management but evaluating whether realized harvest rates are close to established regulations is often not done in practice. Also, managers typically only partially control harvest rates via changes to metrics such as season length and hunting party size, but the relationships between these proxies and harvest rate are often unknown. We studied a population of northern bobwhite Colinus virginianus over a three-year period to estimate harvest, survival and relationships between harvest rate and proxies of hunting pressure. We captured and marked bobwhite with only bands (n = 479) or bands and radio-transmitters (n = 592). We monitored radio-marked individuals weekly until radio failure, lost signal or death. A comparison of annual survival rates indicated birds marked with only bands survived at similar rates (14.5%, 95% CI: 7.6, 21.4%) compared to those marked with both bands and radio-transmitters (10.6%, 95% CI: 7.9, 13.3%). A cumulative incidence function for cause-specific mortality produced an annual harvest rate estimate of 12.0% (95% CI: 8.6, 15.4%). However, this estimate was likely negatively biased in the presence of undocumented crippling loss. We also derived a 14.4% annual harvest rate using average observed levels of hunting pressure and a joint live–dead model that was not dependent on classification of mortality sources. We predicted annual harvest rates over a range of population sizes and hunting pressures to inform managers of possible thresholds to target. Hunting pressure was an important predictor of harvest rate. For instance, we estimated that adding approximately two more hunters per week would push seasonal harvest rates past the targeted harvest threshold, indicating managers should carefully regulate hunting pressure to avoid exceeding set harvest limits. Estimating relationships between weekly survival and hunting pressure can provide information to evaluate different management scenarios and help develop hunting regulations.

Metal pollution commonly occurs in many terrestrial environments and may pose a threat for the animals inhabiting such areas. Here, we present concentrations of six metals (cadmium [Cd], copper [Cu], nickel [Ni], lead [Pb], selenium [Se], and zinc [Zn]) in the liver tissues of seven species of mammals obtained from a study that examined the impact of mesopredator removal on northern bobwhite (Colinus virginianus) populations. A total of 1326 samples were collected from 2003 to 2006 at four sites in southwest Georgia and north Florida from nine-banded armadillos (Dasypus novemcinctus), bobcats (Lynx rufus), feral cats (Felis catus), coyotes (Canis latrans), grey foxes (Urocyon cinereoargenteus), opossums (Didelphis virginiana), and raccoons (Procyon lotor). Data from armadillos, bobcats, opossums, and raccoons were published previously to examine age, sex, spatial (between sites), and temporal (between years) variation. In this paper, we present similar comparisons for the remaining three species as well as comparisons of metal concentrations among all seven species. Concentrations of Cu and Pb exhibited strong negative relationships with body weight in coyotes, while Ni was positively correlated with weight in feral cats. Concentrations of these metals, as well as the other two tested (Cd and Zn), were not significantly correlated with one another in any of the three species. The only sex difference in liver metal concentrations was observed in female feral cats, which had higher levels of Pb than did males. Coyotes exhibited significant differences in Cu concentrations between sites and between years (2005 versus 2006). We also found significant differences between sites in Pb concentrations for both feral cats and grey foxes. There were significant differences in metal concentrations among all seven species for all metals except Cd. With the exception of Cd and Se (tested only in bobcats and opossums), a three-way ANOVA with species, year, and site as the three factors revealed significant differences among species for every metal but only a single main effect of year for Cu, and no main effects of site. In sum, our results provide an extensive survey of metal concentrations in a diverse assemblage of mammals and suggest that metal accumulation may be heavily influenced by species identity, which in turn may reflect ecological lifestyle.

Examination of the endocrine system through non-invasive fecal sampling may improve population management more than using demographic indicators alone. By addressing the physiological mechanisms that are influencing fitness, management actions can be proactively developed to alleviate stressors. Proactive determination of vulnerable populations is critical for species of concern, such as the Northern Bobwhite (Colinus virginianus), which have suffered decades of population decline. We validated an assay to noninvasively measure the adrenocortical response of captive reared bobwhite through fecal corticosterone metabolites (FCM). All individuals received three sequential 48-hour treatments in which samples were collected every 4 hours, including a reference period, an adrenocorticotropic hormone (ACTH) challenge and a biological stressor (exposure to a hunting dog). Reference FCM values had a mean concentration of 16.75 pg/mg (95% CrI: 13.68, 19.91) with adrenocortical activity increasing by 73% for the duration of the ACTH challenge (29.00 pg/mg; CrI: 25.01, 33.78). FCM concentrations remained similar to that of the reference levels during the biological stressor (16.56 pg/mg; CrI: 13.33, 19.92). Our study validates the use of feces to detect changes in FCM levels in our subject species but also demonstrates the complexity of FCM and the importance of both physiological and biological validation prior to field implementation.

Per- and poly-fluoroalkyl substances (PFAS) are a broad class of environmentally persistent chemicals that include thousands of potentially toxic synthetic organic molecules. Some PFAS have been shown to cause adverse health effects including decreased total cholesterol, birth weight, and reproductive success in laboratory animals; however, a lack of chronic toxicity data exists for PFAS in avian ecological receptors. The present study reports on the chronic toxicity of perfluorooctane sulfonic acid (PFOS) and a mixture of PFOS and perfluorohexane sulfonic acid (PFHxS) to northern bobwhite quail (Colinus virginianus) via oral exposure from drinking water. Female weight gain was adversely affected at an average daily intake (ADI) of 3.10 × 10-3 ± 0.15 × 10-3 mg PFOS:PFHxS (1.2:1) mixture × kg-1 body weight × d-1 . Successful liberation from the shell once pipped was adversely affected at an ADI of 2.45 × 10-3 ± 0.01 × 10-3 mg PFOS x kg-1 body weight × d-1 . These values are comparatively much lower than the current dietary avian toxicity reference value (TRV) derived from birds that were exposed via feed, suggesting the need for updated avian TRVs. Relationships between test chemical (PFOS) and test substance (PFOS:PFHxS) showed that PFOS and PFHxS have possible interacting effects in avian receptors and likely differing mechanisms of toxicity depending on chemical co-occurrence and dose. A captive-bred Bobwhite Quail ( Colinus virginianus) ranch in southern Utah, US experienced high mortality rates in the late summer and fall of 2012. Nine juvenile birds were necropsied at the Utah Veterinary Diagnostic Laboratory. Gross lesions included pale skeletal muscle with multifocal hemorrhages and petechiae in the air sacs and serosal surfaces of most organs. Histologically there was moderate to severe, multifocal, degenerative myositis with intramyofiber schizonts and minimal lymphoplasmacytic infiltrates in the proventriculus, ventriculus, heart, and skeletal muscle. There was also moderate fibrinoid to heterophilic vasculitis in multiple organs with vascular intraendothelial or intravascular merozoites and scattered thrombosis. In the liver and spleen there were multiple degenerative schizonts that had ruptured. Blood smears from three of the birds were stained with Wright-Giemsa stain and examined at a referral laboratory. Although the blood cells were deteriorated (postmortem artifact), life stages (exact stages not specified) consistent with Haemoproteus spp. were identified in erythrocytes. Polymerase chain reaction done on pooled tissues from two birds produced an amplicon in both pooled samples, and direct sequencing confirmed the presence of 533 base pairs of a Haemoproteus sp. in the subgenus Parahaemoproteus. The identification of Parahaemoproteus spp. in quail in southern Utah implies that appropriate Culicoides spp. vectors are present in the state and that there is potential risk to other birds such as zoo and aviary populations, wild turkeys, and other game birds.
