This article explores scaling deep through transformative learning in Public Sector Innovation Labs (PSI labs) as a pathway to increase the impacts of their work. Using literature review and participatory action research with two PSI labs in Vancouver and Auckland, we provide descriptions of how they enact transformative learning and scaling deep. A shared ambition for transformative innovation towards social and ecological wellbeing sparked independent moves towards scaling deep and transformative learning which, when compared, offer fruitful insights to researchers and practitioners. This paper seeks to bring together two seemingly disparate conversations, design and environmental education, with the intent to offer an interesting, new, useful approach to developing educational responses to the climate and ecological crises engendered by the Capitalocene. Beginning with observations on the relevance of design to the creation of alternative futures, we outline results from a six-person year-long research project that led us to identify six principles for guiding eco-social-cultural change: all my relations, abundant time, mystery/unknowability, embeddedness/integration, ancient futures, and (re)creative dissonance. We situate this work within transformative orientations to design, which are shown to parallel critical threads in the environmental education literature. We then extend, rework, and reimagine the six principles by suggesting how they can serve as prompts to assist environmental educators to reexamine and move beyond problematic norms of the Capitalocene in their thinking and practice. Idiopathic hypogonadotropic hypogonadism (IHH) with anosmia (Kallmann syndrome; KS) or with a normal sense of smell (normosmic IHH; nIHH) are heterogeneous genetic disorders associated with deficiency of gonadotropin-releasing hormone (GnRH). While loss-of-function mutations in FGF receptor 1 (FGFR1) cause human GnRH deficiency, to date no specific ligand for FGFR1 has been identified in GnRH neuron ontogeny. Using a candidate gene approach, we identified 6 missense mutations in FGF8 in IHH probands with variable olfactory phenotypes. These patients exhibited varied degrees of GnRH deficiency, including the rare adult-onset form of hypogonadotropic hypogonadism. Four mutations affected all 4 FGF8 splice isoforms (FGF8a, FGF8b, FGF8e, and FGF8f), while 2 mutations affected FGF8e and FGF8f isoforms only. The mutant FGF8b and FGF8f ligands exhibited decreased biological activity in vitro. Furthermore, mice homozygous for a hypomorphic Fgf8 allele lacked GnRH neurons in the hypothalamus, while heterozygous mice showed substantial decreases in the number of GnRH neurons and hypothalamic GnRH peptide concentration. Transient flavin intermediates in the flavin-dependent halogenase RebH reaction catalyzing chlorination are characterized and the kinetics of 7-chlorotryptophan formation is determined. The kinetic analysis establishes that substrate chlorination occurs after completion of flavin redox reactions, consistent with a mechanism whereby hypochlorite is generated in the RebH active site from the reaction of FaD[H.sub.2], chloride ion and [O.sub.2].

The properties of p-Hydroxybenzoate hydroxylase was studied as a model for single-component flavoprotein monoxygenases. It showed that transient formation of enzymes that stabilized in the open conformation was the mechanism for sequestering p-hydroxybenzoate to initiate catalysis.

Women's involvement in decision-making around antenatal care is an issue of ongoing debate and discussion. Most research on the topic has used interview and focus group methods to examine women's perspectives. The present study uses a different kind of evidence. By analyzing recordings of actual antenatal consultations, this paper presents a preliminary exploration of model-of-care talk in a hospital setting where a policy of woman-centered care underpinned practice. Conversation Analysis was used to examine how model-of-care pathways were introduced by midwives and discussed with women in consultations. Drawing on interactional work on deontic (i.e., the rights and responsibilities of speakers to determine courses of action) and epistemic (i.e., speakers' claims to knowledge) orientations, this paper offers an account of how woman-centered care is accomplished in a hospital setting. The findings demonstrate how midwives routinely relied on their epistemic knowledge regarding women's health to invoke a "normal" categorization that worked to position midwifery-led care as an appropriate pathway. Examination of model-of-care talk also demonstrated how authority to choose a pathway was typically managed so as to reside with the woman. Talk that topicalized epidural forms of pain management were also examined, as institutional policy around where birth could occur in the hospital system under study restricted women's options (a planned epidural precluded woman access to midwifery-led care during delivery). The findings demonstrate the various ways in which midwives created opportunities for woman-centered care in an institutional setting in which there were logistical restrictions on women's choices.
