Tese de Doutoramento em Ciências da Saúde, The reward circuit is mainly comprised by dopaminergic projections arising from the ventral tegmental area (VTA) to the nucleus accumbens (NAc), and has been shown to mediate different aspects of reward and reinforcement. Mesopontine neurons of the laterodorsal tegmentum (LDT) tightly modulate the activity of dopaminergic neurons of the VTA, controlling the release of dopamine in the NAc. Interestingly, a recent study has shown that the LDT also sends direct projections to the nucleus accumbens, suggesting both a direct and indirect role in modulating accumbal activity. In this thesis, we first show that prenatal glucocorticoid exposure (iuCG model) impacts the electrophysiological activity of the LDT-VTA circuit, culminating with a decrease in motivational drive. LDT basal activity was decreased in iuGC-exposed animals, and we observe a bidirectional effect in evoked activity: iuGC animals present a decrease in the magnitude of excitation and an increase in the magnitude of inhibition in the VTA after electrical stimulation of LDT. Importantly, optogenetic activation of LDT-VTA projections rescues this phenotype, suggesting that motivational levels are dependent on this circuit. In line with this, we further manipulated LDT-VTA circuit during multiple phases of reward using optogenetics. We showed that optical LDT-VTA stimulation is able to causally enhance choice preference for a laser-paired reward and induce intracranial selfstimulation. Next, we investigated the recently described LDT projections to the NAc. This region is comprised by two major populations of GABAergic medium spiny neurons (MSNs), canonically segregated into those expressing dopamine receptor D1 (D1-MSNs), and those expressing D2 (D2- MSNs). Using anatomical, electrophysiological and behavioural experiments, we confirmed the presence of direct cholinergic, glutamatergic and GABAergic projections from the LDT to the NAc, evoking a predominantly excitatory response in accumbal cells. Lastly, we show for the first time that selective activation of LDT-NAc projections enhances motivational drive and induces preference for a laser-associated lever, but only when paired with a food reward. We further demonstrate that specific activation of LDT-NAc cholinergic projections is sufficient for this shift in preference, suggesting that these projections enhance and narrow incentive motivation in rodents. In conclusion, with this work we showed that i) prenatal glucocorticoid exposure negatively impacts motivation due to impairments in LDT-VTA inputs, and that activation of this circuit rescues motivational deficits; ii) that LDT also controls NAc activity through direct projections of different natures; and iii) LDT-NAc (cholinergic) projections convey positive reinforcement signals., O circuito de recompensa é composto principalmente por projeções dopaminérgicas originárias da área tegmental ventral (VTA) para o nucleus accumbens (NAc), e está envolvido na percepção de recompensa e reforço positivo. Os neurónios do núcleo tegmental dorso-lateral (LDT) modulam a atividade dos neurónios dopaminérgicos do VTA, controlando a libertação de dopamina no NAc. Um estudo recente mostrou que o LDT também envia projeções diretas para o NAc, sugerindo um papel direto e indireto na modulação da atividade do accumbens. Nesta tese, mostramos pela primeira vez que a exposição pré-natal a glucocorticóides (modelo iuGC) afeta a atividade electrofisiológica do circuito LDT-VTA, originando uma diminuição da motivação. A atividade basal do LDT estava diminuída nos animais expostos a iuGC, e observamos um efeito bidirecional na atividade evocada do VTA: após a estimulação elétrica do LDT, os animais iuGC apresentavam uma diminuição na magnitude da excitação e um aumento na magnitude da inibição. De salientar que a ativação optogenética das projeções LDT-VTA normaliza esse fenótipo, sugerindo que os níveis motivacionais dependem deste circuito. Em consonância, recorrendo à técnica de optogenética, manipulamos o circuito LDT-VTA durante múltiplas fases de recompensa. A estimulação optogenética do circuito LDT-VTA aumenta a preferência por uma recompensa associada à estimulação e induz autoestimulação intracraniana. De seguida investigamos as projeções do LDT para o NAc descritas recentemente. O NAc é composto por duas populações principais de neurónios espinhosos médios GABAérgicos (MSNs), segregados canonicamente nos que expressam o receptor de dopamina D1 (D1-MSNs) e os que expressam D2 (D2-MSNs). Dados anatómicos, eletrofisiológicos e comportamentais confirmam a presença de projeções diretas do LDT para o NAc colinérgicas, glutamatérgicas e GABAérgicas, que induzem uma resposta predominantemente excitatória no NAc. Por último, demonstramos pela primeira vez que a ativação seletiva das projeções LDT-NAc aumenta o nível motivacional e induz a preferência por uma alavanca associada à estimulação, mas somente quando associada com uma recompensa. Mostramos ainda que a ativação específica das projeções colinérgicas LDTNAc é suficiente para a mudança de preferência. Em conclusão, com este trabalho demonstramos que i) a exposição pré-natal a glucocorticóides reduz a motivação devido a défices nas projeções do LDT para o VTA, e que a ativação deste circuito normaliza estes défices; ii) que o LDT também controla diretamente a atividade do NAc através de projeções diretas de tipos diferentes; e iii) as projeções LDT-NAc (colinérgicas) codificam sinais de reforço positivos., The work presented in this thesis was performed in the Life and Health Sciences Research Institute (ICVS), Minho University. Dissertação de Mestrado em Engenharia Zootécnica Este trabalho apresenta um estudo realizado numa exploração comercial com reprodutores da estirpe Ross 308, no qual se procurou determinar o efeito da relação macho:fêmea nas performances reprodutivas e nos resultados da exploração. Foram estudadas duas relações macho:fêmea (8% e 10%) analisando parâmetros tais como o peso dos machos, uniformidade, estimativa de alimentação, parâmetros de bem-estar (estado das patas, cloaca, cauda e emplumação), mortalidade das aves, percentagem de postura semanal e postura no chão, fertilidade, eclodibilidade e número de pintos viáveis nascidos por galinha. Foi ainda realizada uma breve análise económica. Este estudo foi dividido em duas fases. Na primeira fase do estudo, realizada nos pavilhões de produção, foi efetuada uma avaliação física dos galos reprodutores tendo em conta parâmetros de bem-estar como a saúde das patas, estado da cloaca, emplumação do corpo e da cauda, e ainda o peso destas aves. O estudo realizou-se ao longo de 16 semanas e em cada semana 48 galos escolhidos aleatoriamente na relação 8/100 e 56 galos escolhidos aleatoriamente na relação 10/100 foram observados e pesados. Na segunda fase do estudo, realizada no centro de incubação, avaliou-se a fertilidade do bando através da observação dos ovos eclodidos e não eclodidos (férteis), e os ovos inférteis e as mortalidades embrionárias nas diversas fases de incubação foram registados. Os dados foram recolhidos e analisados numa base semanal tendo-se procedido à sua análise estatística. Em ambos os tratamentos os galos apresentaram pesos corporais superiores aos recomendados pelos guias de maneio, sendo que esta diferença foi mais evidente no tratamento 8/100, o que parece ter influenciado a fertilidade deste tratamento. Os resultados não mostraram diferenças significativas entre os dois tratamentos nos parâmetros indicadores de bem-estar. No entanto verificou-se que a taxa de fertilidade dos ovos e o número de pintos nascidos viáveis foram superiores no tratamento 10/100 (73,6% vs 83,9% e 3,51 pintos nascido/galinha vs 3,70 pintos nascido/galinha). A análise económica mostrou que a relação 10 machos para 100 fêmeas permitiu um aumento de rendimento de 48 cêntimos por galinha presente no período de 8 semanas de produção quando comparada com a relação 8 machos 100 fêmeas. This work is presents a study carried out on a commercial farm with Ross 308 broiler breeders, looking to study the male:female ratio effect on the reproductive performances and its advantages for the producer. The relationships of 8% and 10% of males were studied considering parameters such as the male’s weight, uniformity, feeding estimate, welfare parameters (heath of feet and legs, cloaca (vent) condition and body and tail feathering), mortality rate, weekly percentage of egg production and posture on the floor, fertility, hatchability and number of viable chicks hatched. Additional feeding costs and the profit made on the sale of day-old chicks in the two cases were also taken into account in this study. This study was divided in two different phases. In the first phase of the study, carried out on de production pavilions, a physical evaluation of broiler breeders was conducted over 16 weeks (48 random males in the 8/100 ration and 56 random males in the 10/100 ration weekly), taking into account parameters such as the health of feet and legs, cloaca (vent) condition, body and tail feathering, and the weight of the animals. This study was carried out over 16 weeks and each week 48 random males in the 8/100 ration and 56 random males in the 10/100 ration were observed and weighed. In the second phase of this work, the fertility of flocks was studied with the observation of unhatched eggs and the number of infertile eggs and embryonic mortality at various stages of incubation was recorded. Data were collected and analyzed on a weekly basis having proceeded to their statistical analysis. In both treatments the roosters had body weights higher than those recommended by management guides, and this difference was more evident in the treatment 8/100, which seems to have influenced the fertility of this treatment. No significant differences between the two treatments were found as regards to indicators welfare parameters. However it was verified that the fertility rate and viability of chicks hatched were higher in the treatment 10/100 (73.6% vs 83.9% and 3.51 chick/hen vs 3.70 chick/hen). The ratio 10/100 allowed a yield increase of 0.48€per present hen in 8 weeks of production when compared with the ratio 8/100.

Dissertação de mestrado em Ciências da Saúde, Early life stress (ELS) or elevated levels of glucocorticoids (GCs) may result in persistent effects in the central nervous system that can lead to maladaptive behavior in adult life and increase the vulnerability to develop psychiatric disorders, such as anxiety, depression or drug addiction. In clinics, synthetic GCs are often prescribed in pregnancies in risk of pre-term labor, to ensure fetal lung maturation. Regardless of its beneficial effect, elevated levels of GCs during this period can lead to deleterious and permanent effects on brain function and development. The mesocorticolimbic dopaminergic circuit, also known as the ‘reward’ system, seems to be a key target of stress/GCs, since it has been shown that animals exposed to GC in utero (iuGC) have structural and molecular alterations in several brain regions of this pathway. Importantly, such changes may underlie the observed addictive-like behavior of iuGC animals. Vulnerability for addictive behavior may be modulated by individual emotional condition and/or specific personality traits. iuGC animals present anxious and depressive-like behavior, but less is known about their novelty-seeking and impulsivity traits, two behavioral dimensions that contribute substantially to drug-seeking behavior. In this perspective, we evaluated iuGC animals in novelty-dependent behavioral tests. iuGC animals did not present major differences in noveltyinduced locomotor activity nor in general exploratory behavior (Novelty Place Preference and Novel Object Recognition). However, iuGC rats explore familiar and novel objects similarly in the 24h retention time test, suggesting long-term memory impairment. In addition, we assessed impulsive action, using the 5-Choice Serial Reaction Time Task (5-CSRTT) and impulsive choice, using the Delay Discounting (DD) test. Whereas no significant differences were found in 5-CSRTT performance, iuGC animals present alterations in the DD task. Since DD task is strongly dependent on the prefrontal cortex (PFC) and also the amygdala, we analyzed the neuronal activation pattern of these brain regions upon task performance. iuGC treatment induced significant changes in c-fos positive cell density in the orbitofrontal cortex, medial PFC and less so in the amygdala, which could potentially explain the observed behavioral differences. Altogether, our results suggest that iuGC treatment does not seem to affect novelty-seeking behavior, but it has an effect in impulsive choice, which may contribute for the observed enhanced drug-seeking behavior., Exposição a stress pré-natal ou a níveis elevados de glucocorticóides (GC) podem resultar em alterações persistentes no sistema nervoso central, aumentando a vulnerabilidade para o desenvolvimento de doenças psiquiátricas tais como a ansiedade, depressão ou dependência de drogas. Na clínica, os GCs sintéticos são vulgarmente prescritos em casos de risco de parto prematuro, para assegurar a maturação pulmonar fetal. Independentemente do seu efeito benéfico, níveis elevados de GCs durante este período podem levar a efeitos nefastos e permanentes na função e desenvolvimento cerebral. O circuito mesocorticolímbico, conhecido como o sistema de recompensa, parece ser um alvo preferencial do stress /GCs, uma vez que foi demonstrado que animais expostos in útero a GCs (iuGC) têm alterações estruturais e moleculares neste circuito. De salientar que tais mudanças podem ser a base do comportamento aditivo observado nestes animais. A vulnerabilidade para o comportamento aditivo pode ser modulada pela condição emocional e/ou características da personalidade do indivíduo. Os animais iuGC são ansiosos e apresentam características do tipo depressivo, mas pouco se sabe sobre o seu efeito em características como a procura de sensação/novidade e impulsividade, duas dimensões comportamentais que contribuem substancialmente para o comportamento aditivo. Nesta perspectiva, estes animais foram avaliados em testes comportamentais baseados na resposta à novidade. Os animais iuGC não apresentam diferenças substanciais na atividade locomotora induzida pela novidade nem no comportamento exploratório geral (Novelty Place Preference e Novel Object Recognition). Contudo, os indivíduos iuGC exploram objectos familiares e novos de forma semelhante no teste com tempo de retenção de 24h. Adicionalmente, avaliámos a acção impulsiva, através do 5-Choice Serial Reaction Time Task (5-CSRTT) e a escolha impulsiva, utilizando o teste Delay Discounting (DD). O grupo iuGC apresenta alterações na tarefa DD, mas não na performance no 5-CSRTT. Como o teste DD é dependente do córtex pré-frontal (PFC) e da amígdala, foi analisado o padrão de activação neuronal destas regiões usando imunohistoquímica para c-fos. O tratamento iuGC induziu alterações na densidade de células cfos positivas no córtex orbito frontal e PFC medial, com menor efeito na amígdala, o que pode ajudar a explicar as diferenças observadas. Em suma, os nossos resultados sugerem que o tratamento iuGC não parece afetar o comportamento exploratório, mas tem um efeito deletério na memória de longo prazo. Em simultâneo, os animais iuGC apresentam maior escolha impulsiva, o que pode contribuir para a maior vulnerabilidade para comportamentos aditivos.

Dissertação de mestrado em Ciências da Saúde, Diariamente, os indivíduos filtram a informação sensorial do ambiente que os rodeia. A atribuição de valência permite aos mamíferos concentrarem-se em estímulos salientes ao determinar se estes são recompensadores e devem ser abordados ou se são aversivos, e devem ser evitados, de forma a prosperarem e sobrevirem. O processo ativo de codificação de valência é gerado pelo sistema límbico que o traduz numa reação fisiológica e/ou comportamental. Uma pequena estrutura do tronco cerebral, o núcleo tegmental dorso-lateral (LDT), tem ganho atenção devido ao seu papel em diversos comportamentos relacionados com recompensa e aversão. O LDT é um núcleo heterogéneo sendo composto maioritariamente por células glutamatérgicas, colinérgicas e GABAérgicas. O seu papel no circuito de recompensa/aversão foi inicialmente associado ao controlo de células dopaminérgicas da área tegmental ventral (VTA). Recentemente, o nosso grupo descobriu projeções para outro elemento importante deste circuito, o núcleo accumbens (NAc). Há algumas evidências que o LDT responde a vários estímulos de valência positiva e negativa. Contudo, não se sabe ao certo que populações codificam valência e se estas populações apresentam um baixo ou alto grau de sobreposição/segregação. Nesta dissertação, avaliámos a ativação neuronal no LDT, através de marcação de c-Fos, associados a estímulos de valência negativa (choque na pata) ou positiva (cocaína ou sacarose). Depois, usando o modelo de murganho TRAP2 (Fos 2A-iCreERT2), comparamos os grupos neuronais recrutados por dois eventos com valência temporalmente distintos no LDT. Utilizando o modelo, também realizamos ativação optogenética de neurónios do LDT responsivos a estímulos positivos ou negativos. Adicionalmente, analisamos anatomicamente a inervação do NAc pelas populações neuronais responsivas a estímulos provenientes do LDT. Os nossos dados mostram que o LDT contém populações neuronais que respondem ao choque, cocaína ou sacarose, medido por uma maior densidade de células c-Fos+. Comparando os grupos neuronais, verificamos que o choque e a cocaína recrutaram maioritariamente populações celulares distintas no LDT. Adicionalmente, a ativação optogenética dos neurónios do LDT ativados por choque ou cocaína nos testes de real time place preference (RTPP) ou conditioned place preference (CPP) foi inconclusiva. Por último, as células do LDT responsivas a cocaína não mostraram inervação preferencial para nenhuma das sub-regiões do NAc. No entanto, as células do LDT responsivas a choque projetaram preferencialmente para a sub-região posterior do NAc shell. Estes resultados demonstram que o LDT é um participante ativo na atribuição de sinais de valência que podem revelar princípios fundamentais sobre o processamento emocional., Individuals must filter the overwhelming environmental sensory information that every day surrounds them. Assigning affective valence allows mammalians to focus on salient stimuli by determining whether they are rewarding and should be approached or are aversive and should be avoided, in order to thrive and survive. The active process of valence encoding is generated by the brain’s limbic system that translates it, ultimately, in a physiological and/or behavioral reaction. A small brainstem structure, the laterodorsal tegmental nucleus (LDT), has gained much attention in recent years due to its prominent role in a myriad of reward- and aversion-related behaviors. The LDT is a heterogeneous nucleus, being mainly composed by glutamatergic, cholinergic and GABAergic cells. Its role in the reward/aversion circuitry was initially attributed to the influence of this region on midbrain dopaminergic cells of the ventral tegmental area (VTA). However, recently, our group found direct projections from the LDT to another important player of the circuit, the nucleus accumbens (NAc). Moreover, there is sparse evidence showing that the LDT responds to several positive- and negative-valence stimuli. However, it is not known which neuronal populations encode valence and if these populations are segregated or overlap. In this thesis we studied cell activation following negative- (footshock) and positive-valence stimuli (cocaine and sucrose) in the LDT using c-Fos immunolabelling. Then, using the TRAP2 (Fos 2A-iCreERT2) mouse model, we evaluated in the same subject the neuronal ensembles activated by temporally distinct valence containing events. Using this model, we also performed optogenetic activation of positive or negative stimulus-responsive neurons in the LDT. Furthermore, we analyzed the pattern of NAc innervation of these LDT responsive ensembles. Our data shows that the LDT contains neuronal populations that respond to shock, cocaine or sucrose, measured by a higher c-Fos+ cell density in comparison to controls. Analyzing the responsive ensembles, we verified that shock and cocaine recruited mainly distinct cell populations in the LDT. The assessment of behavioral phenotype in the real time place preference (RTPP) or conditioned place preference (CPP) tasks to optogenetic activation of footshock- or cocaine-responsive neurons in the LDT was inconclusive. The NAc innervation analysis revealed that cocaine-responsive cells do not appear preferentially innervate different subregions of the NAc. Conversely, shock-responsive LDT cells projected preferentially to the posterior shell subregion of the NAc.
