Abstract

This paper aims at presenting the results of the topographic fieldwork of a team of professional archaeologists invited by the Ministry of Heritage and Culture of the Sultanate of Oman to excavate and survey three graveyards in the area of Sohar (Falaji as Souq, Wadi al Arad and Liwa) in 2014 and 2015. The construction of the Batinah Express Highway would have led to the destruction of hundreds of burial mounds, therefore the team developed a quick and accurate surveying strategy to document them properly: after a first “test” campaign using monoscopic photogrammetry, the team opted for 3D SfM photogrammetry using a completely open source workflow. This workflow required two surveyors on the field and in the IT lab to ensure the archaeologists updated orthophotos and to update the 2D and 3D vector plans. To manage the huge mass of data coming from the field the team opted for QGIS and the plugin PyArchInit. The mix of surveying methodology and managing system developed on site allowed the team to document the numerous Stratigraphic Units produced during the excavation of hundreds of graves, and also proved to be very helpful as hermeneutic tool as shown in the case of the excavation of Grave 21.

Abstract

Oxidative stress is a key mediator of autoimmune/neurodegenerative disorders. The antioxidant/anti-inflammatory effect of a synthetic conjugated linoleic acid (CLA) mixture in MRL/MpJ-Faslpr mice (MRL/lpr), an animal model of neuropsychiatric lupus, was previously associated with the improvement of Nuclear factor-E2-related factor 2 (Nrf2) defences in the spleen and liver. However, little is known about the neuroprotective ability of CLA mixture. This study investigates the age-dependent progression of oxidative stress and the hyperactivation of redox-sensitive compensatory pathways (macroautophagy, Nrf2) in old/diseased MRL/lpr mice brains, and examines the effect produced by dietary CLA supplementation. Disrupted redox homeostasis was evidenced in the blood, liver and brain of 21- to 22-week old MRL/lpr mice (Old) compared to young animals (Y; 8- to 10-week old). This alteration was associated with significant hyperactivation of compensatory mechanisms (macroautophagy, Nrf2 and astrocyte activation) in the brains of Old mice. Five-week daily supplementation with CLA (650 mg/kg-1 body weight) of 16-week old mice (CLA+Old) diminished all the pathological hallmarks at a level comparable to Y mice or healthy controls (BALB/c). Such data demonstrated that MRL/lpr mice can serve as a valuable model for the evaluation of the effectiveness of neuroprotective drugs. Notably, the preventive effect provided by CLA supplementation against age-associated neuronal damage and hyperactivation of compensatory mechanisms suggests that the activation of an adaptive response is, at least in part, accountable for its neuroprotective ability.

Abstract

The CLUE UV Cerenkov telescope array has started to take data with 8 telescopes in January 1998. The UV Cerenkov images obtained by the CLUE experiment are very different with respect to the visible case, and a new method for reconstructing the shower direction has been worked out. The shower reconstruction is shown and an application to recent data is given.

Abstract

All eukaryotic genomes contain mobile elements and their role in the shaping of eukaryotic genomes is becoming more and more recognized, although the proportion and activity of the different classes of these elements widely varies between genomes. We used an in silico approach to analyze the presence and distribution of one of this family, the V-SINE sequences, in Danio rerio chromosomes. V-SINEs is a superfamily of vertebrate short interspersed repetitive elements, whose central domain is strongly conserved and is even found in SINEs in the lamprey genome, suggesting that V-SINEs might be ~550 Myr old. The central conserved domain might have been subject to some form of positive selection. A total of 108625 V-SINEs fragments have been identified on the 25 assembled Danio rerio chromosomes (danrer6), accounting for about 0.019% of the entire sequence. The average number of V-SINEs fragments per chromosome is about 4372 (±943), ranging from 2628 to 6571. We estimated that about 20% (±3%) of V-SINES per chromosome were found to be located within genic sequences, the main contribution due to their presence in the introns. In particular, we found a striking bias of V-SINES localization in 5? UTRs with respect to the 3? ones. The 5?UTR/3? ratio was estimate to be about 6.73. Our results seem to confirm that V-SINEs preferentially accumulate within relatively gene-rich regions, as previously seen by their in situ mapping on chromosomes. Besides, since, in mice, evidences have already shown that SINEs located in 5?UTR can contribute to the repression of gene expression, we suggest that, in Danio, the biased distribution of V-SINES in the 5?UTR vs 3? UTR regions, could be a scratch of their potential role in gene regulation.

Abstract

To analyze risk factors for type 2 diabetes among women attending menopause clinics in Italy for counselling about the menopause.Women attending a network of first-level outpatient menopause clinics in Italy for general counselling about menopause or treatment of menopausal symptoms.Cross-sectional study with no exclusion criteria. Type 2 diabetes was defined according to National Diabetes Data Groups Indications and the fasting blood glucose at an oral glucose tolerance test within the previous year.Out of the 44 694 considered in this analysis, 808 had a diagnosis of diabetes type 2 (1.8%). In comparison with women aged50 years, the multivariate odds ratios (OR) of type 2 diabetes were 1.31 (95% confidence interval (CI), 0.99-1.74) for women aged 50-52 years, 1.66 (95% CI, 1.27-2.17) at 53-56 years and 2.84 (95% CI, 2.20-3.67) in women agedor = 57 years. Type 2 diabetes was less frequently reported in more educated women (OR high school/university vs. primary school = 0.44 (95% CI, 0.36-0.55)). Being overweight was associated with an increased risk of type 2 diabetes. In comparison with women reporting a low level of physical activity, the multivariate OR of type 2 diabetes was 0.67 (95% CI, 0.54-0.84) for women reporting regular physical activity. In comparison with premenopausal women, the multivariate OR of type 2 diabetes was 1.38 (95% CI, 1.03-1.84) in women with natural menopause. This finding was present also after allowing for the potential confounding effect of age. The multivariate OR of diabetes for users of hormonal replacement therapy was 0.58 (95% CI, 0.46-0.73).This large cross-sectional study suggests that postmenopausal women are at higher risk of type 2 diabetes after allowance for the effect of age. Other main determinants of risk of type 2 diabetes in women around menopause were low socioeconomic status and being overweight. Diabetes was found less frequently in those taking hormone replacement therapy.

Abstract

OBJECTIVE To obtain data on correlates of climacteric symptoms in women around menopause attending menopause clinics in Italy. METHODS Since 1997 a large cross sectional study has been conducted on the characteristics of women around menopause attending a network of first level menopause outpatient's clinics in Italy. A total of 66,501 (mean age 54.4 years) women are considered in the present paper. RESULTS The odds ratios of moderate and severe hot flashes/night sweats were lower in more educated women and (for severe symptoms only) in women reporting regular physical activity. Depression, difficulty to sleep, forgetfulness and irritability tended to be less frequent in more educated women and (depression only) in women reporting regular physical activity. Parous women reported more frequently these symptoms. CONCLUSIONS This large study confirms in Southern European population that low education, body mass index and low physical activity are associated with climacteric symptoms. Parous women are at greater risk of psychological symptoms.

Abstract

SCOPE: The involvement of oxidative stress in gluten-induced toxicity has been evidenced in vitro and in clinical studies but has never been examined in vivo. We recently demonstrated the protective activity of conjugated linoleic acid (CLA), which functions by the activation of nuclear factor erythroid 2-related factor2 (Nrf2), a key transcription factor for the synthesis of antioxidant and detoxifying enzymes (phase 2). Here, we evaluate the involvement of nuclear factor erythroid 2-related factor2 in gliadin-mediated toxicity in human Caco-2 intestinal cells and in gliadin-sensitive human leukocyte antigen-DQ8 transgenic mice (DQ8) and the protective activity of CLA. METHODS AND RESULTS: Gliadin effects in differentiated Caco-2 cells and in DQ8 mice, fed with a gliadin-containing diet with or without CLA supplementation, were evaluated by combining enzymatic, immunochemical, immunohistochemical, and quantitative real-time PCR (qRT-PCR) assays. Gliadin toxicity was accompanied by downregulation of phase 2 and elevates proteasome-acylpeptide hydrolase activities in vitro and in vivo. Notably, gliadin was unable to generate severe oxidative stress extent or pathological consequences in DQ8 mice intestine comparable to those found in celiac patients and the alterations produced were hampered by CLA. CONCLUSION: The beneficial effects of CLA against the depletion of crucial intestinal cytoprotective defenses indicates a novel nutritional approach for the treatment of intestinal disease associated with altered redox homeostasis.

Abstract

We describe a trigger architecture developed for shower selection in the CLUE Cherenkov telescopes. The selection is made first in each telescope of the array looking at the low resolution images produced by fast electronics, than requiring temporal coincidence between units. Big effort has been put to simplify the programming of trigger configurations to help the user to switch easily between different observational and calibration requests. Abstract

BackgroundApproximately two hundred human burials were discovered on the edge of a paleolake in Niger that provide a uniquely preserved record of human occupation in the Sahara during the Holocene ( approximately 8000 B.C.E. to the present). Called Gobero, this suite of closely spaced sites chronicles the rapid pace of biosocial change in the southern Sahara in response to severe climatic fluctuation.Methodology/principal findingsTwo main occupational phases are identified that correspond with humid intervals in the early and mid-Holocene, based on 78 direct AMS radiocarbon dates on human remains, fauna and artifacts, as well as 9 OSL dates on paleodune sand. The older occupants have craniofacial dimensions that demonstrate similarities with mid-Holocene occupants of the southern Sahara and Late Pleistocene to early Holocene inhabitants of the Maghreb. Their hyperflexed burials compose the earliest cemetery in the Sahara dating to approximately 7500 B.C.E. These early occupants abandon the area under arid conditions and, when humid conditions return approximately 4600 B.C.E., are replaced by a more gracile people with elaborated grave goods including animal bone and ivory ornaments.Conclusions/significanceThe principal significance of Gobero lies in its extraordinary human, faunal, and archaeological record, from which we conclude the following: The early Holocene occupants at Gobero (7700-6200 B.C.E.) were largely sedentary hunter-fisher-gatherers with lakeside funerary sites that include the earliest recorded cemetery in the Sahara.Principal components analysis of craniometric variables closely allies the early Holocene occupants at Gobero with a skeletally robust, trans-Saharan assemblage of Late Pleistocene to mid-Holocene human populations from the Maghreb and southern Sahara.Gobero was abandoned during a period of severe aridification possibly as long as one millennium (6200-5200 B.C.E).More gracile humans arrived in the mid-Holocene (5200-2500 B.C.E.) employing a diversified subsistence economy based on clams, fish, and savanna vertebrates as well as some cattle husbandry.Population replacement after a harsh arid hiatus is the most likely explanation for the occupational sequence at Gobero.We are just beginning to understand the anatomical and cultural diversity that existed within the Sahara during the Holocene.

Abstract

The CLUE experiment uses a new cosmic ray detector array planned to operate for the next decade. It utilises a MWPC chambers sensitive to UV, to image Cherenkov radiation produced in cosmic ray showers. This approach is unique in that the instrument is insensitive to skylight backgrounds, has a threshold similar to that of visible Cherenkov experiments but a longer duty cycle. These features make possible a class of interesting cosmic ray physics experiments. The CLUE experiment has started operation with two of the ten telescopes that are foreseen for its beginning. These telescopes are at present at Roque de Los Muchachos in the Canary Islands, in the same site as the HEGRA experiment. A description of the telescopes is given and some data on the tests performed are presented.

Abstract

The study of globin genes has greatly contributed to the understanding of universal mechanisms of gene evolution and of gene expression regulation. In Notothenioidei, the dominant suborder of Antarctic fish, the investigation of the organization of globin genes is particularly interesting in order to characterize the evolutionary mechanisms which led to the hemoglobin loss of the family Channichthyidae. The Antarctic fish Trematomus newnesi has an oxygen transport system with unique characteristics among notothenioids. The cloning and sequence analysis of the ?- and ?-globin cDNAs of the major hemoglobin of T. newnesi, with the determination of the linkage of the relative genes, is herewith reported. The orientation of T. newnesi Hb 1 ?- and ?-globin genes was head-to-head, corresponding to the classical organization of fish globin genes. Seven of the eight notothenioid families are red-blooded but have reduced erythrocyte number and hemoglobin multiplicity; all 16 species of the eighth family (Channichthyidae) are completely devoid of hemoglobin (white-blooded icefish) and represent a novel and unique phenotype in the evolutionary history of fish and vertebrates. Therefore, studies have aimed at evaluating the pathway of genomic change and the mechanisms that have led to the loss of hemoglobin expression by the white-blooded icefish. Adult and embryonic/juvenile gene complexes from a closely related, red-blooded notothenioid species were isolated and characterized. The results indicate that in channichthyids the adult ?-globin gene is absent and that the inability to express hemoglobin in these fishes might have arisen from a large-scale deletional event.

42. Tail-to-head linkage of alpha- and beta-globin genes in the Antarctic skate Bathyraja eatonii

Abstract

Introduction: Extensive investigations of the organisation of globin genes has greatly contributed to the understanding of universal mechanisms of gene evolution and of regulation of gene expression. A single primordial globin gene is believed to have duplicated to produce an ?-like and a ?-like globin gene approximately 450 million year ago. The duplication event that led to the evolution of the vertebrate ?- and ?-globin genes would initially have left the two genes adjacent. The ?- and ?-globin genes are tightly linked in teleosts, where they are organised as consecutive ?/? sets with the genes linked in head-to-head (5' to 5') orientation, and in amphibians, where they form ?- and ?-type clusters with the genes having the same transcriptional direction. In mammals and birds the ?- and ?-gene clusters are separated and located on different chromosomes, and usually arranged along the order of ontogenic expression. The duplication and differentiation of the ancestral globin gene into the ? and ? genes, including the evolution of the hemoglobin tetramer, the formation of the subunit interfaces, and the mechanism of the quaternary structure change, took place within a comparatively short period between the branching points of hagfish and lampreys from cartilaginous fish. Up to now, there was complete lack of data about the organisation of globin genes in chondrichthyans. Results: In Antarctica the chondrichthyan genera are represented in the modern fauna with only three species of sharks and eight species of skates, that account for about 4% of the total fauna. Bathyraja (six species) is the dominant rajid south of 60°S. In the framework of the investigations on globin genes in Antarctic red-blooded and hemoglobinless fish, by RT-PCR, we obtained the cloning of the ?- and ?-globin cDNAs of the main hemoglobin of the skate Bathyraja eatonii. Then, PCR-based gene linkage analysis was used to assess the relative orientation of the globin gene pair. A genomic fragment of about 6700 bp was obtained where the ? and ? genes are linked in a tail-to-head (3' to 5') orientation. To confirm this finding and to contribute to our Antarctic/Arctic comparison studies, work is currently in progress with the aim to elucidate the organisation of globin genes in another chondrichthyan: the Arctic skate Raja hyperborea. Conclusions: The finding of a tail-to-head linkage of ?- and ?-globin genes in the Antarctic skate B. eatonii is the first characterisation of the organisation of globin genes in chondrichthyes; such finding will offer a substantial contribution to the understanding of the general evolution of this class of genes.
