1. Improving Teachers' Professional Vision through a Video-Based Reflection Program: A Case Study in Mexican Primary Schools

Abstract

Teachers' professional vision (TPV) is an essential contributor to enhancing the quality of teaching-learning processes thanks to its potential effect on teachers' self-reflective skills. This is of particular importance in Physical Education (PE), due to the fact that this subject presents different challenges and more dynamic situations compared to others. One of the latest approaches for enhancing TPV is video-based reflection, allowing teachers to observe and analyze recorded lessons. This research aimed to test the effect of a video-based reflection program on TPV in PE. Two teachers participated in a 3-step video-analysis program consisting of self-reflection, peer reflection, and expert feedback. Notable changes were found in TPV at the end of the 6-month intervention, both in terms of teachers' selective attention to classroom events and their knowledge-based reasoning. Our findings suggest that video-based reflection interventions could represent an important component of any teacher training program aiming to increase PE teachers' ability to evaluate and respond to a variety of in-class situations.

Abstract

Abstract Background Surfactant is a well-established therapy for preterm neonates affected by respiratory distress syndrome (RDS). The goals of different methods of surfactant administration are to reduce the duration of mechanical ventilation and the severity of bronchopulmonary dysplasia (BPD); however, the optimal administration method remains unknown. This study compares the effectiveness of the INtubate-RECruit-SURfactant-Extubate (IN-REC-SUR-E) technique with the less-invasive surfactant administration (LISA) technique, in increasing BPD-free survival of preterm infants. This is an international unblinded multicenter randomized controlled study in which preterm infants will be randomized into two groups to receive IN-REC-SUR-E or LISA surfactant administration. Methods In this study, 382 infants born at 24+0–27+6 weeks’ gestation, not intubated in the delivery room and failing nasal continuous positive airway pressure (nCPAP) or nasal intermittent positive pressure ventilation (NIPPV) during the first 24 h of life, will be randomized 1:1 to receive IN-REC-SUR-E or LISA surfactant administration. The primary outcome is a composite outcome of death or BPD at 36 weeks’ postmenstrual age. The secondary outcomes are BPD at 36 weeks’ postmenstrual age; death; pulse oximetry/fraction of inspired oxygen; severe intraventricular hemorrhage; pneumothorax; duration of respiratory support and oxygen therapy; pulmonary hemorrhage; patent ductus arteriosus undergoing treatment; percentage of infants receiving more doses of surfactant; periventricular leukomalacia, severe retinopathy of prematurity, necrotizing enterocolitis, sepsis; total in-hospital stay; systemic postnatal steroids; neurodevelopmental outcomes; and respiratory function testing at 24 months of age. Randomization will be centrally provided using both stratification and permuted blocks with random block sizes and block order. Stratification factors will include center and gestational age (24+0 to 25+6 weeks or 26+0 to 27+6 weeks). Analyses will be conducted in both intention-to-treat and per-protocol populations, utilizing a log-binomial regression model that corrects for stratification factors to estimate the adjusted relative risk (RR). Discussion This trial is designed to provide robust data on the best method of surfactant administration in spontaneously breathing preterm infants born at 24+0–27+6 weeks’ gestation affected by RDS and failing nCPAP or NIPPV during the first 24 h of life, comparing IN-REC-SUR-E to LISA technique, in increasing BPD-free survival at 36 weeks’ postmenstrual age of life. Abstract

Resting heart rate is associated with cardiovascular diseases and mortality in observational and Mendelian randomization studies. The aims of this study are to extend the number of resting heart rate associated genetic variants and to obtain further insights in resting heart rate biology and its clinical consequences. A genome-wide meta-analysis of 100 studies in up to 835,465 individuals reveals 493 independent genetic variants in 352 loci, including 68 genetic variants outside previously identified resting heart rate associated loci. We prioritize 670 genes and in silico annotations point to their enrichment in cardiomyocytes and provide insights in their ECG signature. Two-sample Mendelian randomization analyses indicate that higher genetically predicted resting heart rate increases risk of dilated cardiomyopathy, but decreases risk of developing atrial fibrillation, ischemic stroke, and cardio-embolic stroke. We do not find evidence for a linear or non-linear genetic association between resting heart rate and all-cause mortality in contrast to our previous Mendelian randomization study. Systematic alteration of key differences between the current and previous Mendelian randomization study indicates that the most likely cause of the discrepancy between these studies arises from false positive findings in previous one-sample MR analyses caused by weak-instrument bias at lower P-value thresholds. Abstract

Nanoplastics (NPs) are plastic particles, typically less than 100 nm in size, that result from daily life products as well as the degradation of larger plastic debris. Due to their small size and chemical composition, they can interact with biological systems in ways that larger plastic particles cannot. Humans are continuously exposed to NPs and several studies showed the potentially toxic effects of these latter on health. Polystyrene nanoplastics (PS-NPs) are the prevalent form of nanoparticles found in the environment and their cellular uptake can cause cytotoxicity and structural alteration of biomolecules. Thus, there is an urgent need for evaluation of the genotoxic effects of PS-NPs on human cell models. Through different and complementary experimental approaches, we investigated the potential genotoxic and cytotoxic effects of PS-NPs exposure on HeLa cell lines. We highlighted the genotoxic effects of polystyrene nanoplastics by showing the formation of multinuclei and micronuclei in all the studied concentrations and time points, also at short incubation time (6 h) and low concentration. At higher concentrations, we demonstrate the presence of apoptotic and necrotic cells outlining the acute cytotoxic effects of nanoplastics. The genotoxic potential is further highlighted by the presence of low molecular weight DNA fragments in PS-NPs treated cells, and by the relationship between polystyrene nanoplastics and γ-H2AX. Thus, our data provide important insights at a cellular level into the possible risks produced by these nanoparticles and recommend further deeper research studies to address the impacts of nanoplastics on human health.

Abstract

This paper describes the spatial and temporal variability of the hydrogeochemistry of shallow aquifer in the western Po Plains, located in Piedmont (northwestern Italy), using groundwater monitoring network data for the main chemical–physical parameters. Hydrochemical maps for the 2015–2020 period were created to identify the main natural and anthropogenic factors responsible for the remarkable spatial variability. Temporal variations in the 2000–2020 period were defined to show the existence of variabilities that were previously not investigated. The spatial and temporal elaborations show significant and various variabilities. The hydrochemical maps confirm the existence of several natural and anthropogenic factors, including the lithological compositions of aquifers, agricultural practices and pressure from urban centres. The temporal variations suggest different resilience capacities of the aquifers to the several impacting factors. The availability of this knowledge is crucial to create the basis for groundwater protection.

Abstract

The United Nations Educational, Scientific and Cultural Organization (UNESCO) states that quality physical education (QPE) is essential for promoting health in youth, and that teachers have a prominent role towards achieving it (UNESCO, 2015). Previous research linked PE teachers’ hard and soft skills with their students’ in-class motivation. Teachers can foster students’ willingness to learning (Johnson, 2017) by using professional competences – hard skills - (Simón-Chico et al., 2023), and by promoting positive relationships – soft skills - (Opdenakker, 2023). However, the mechanism by which these skills affect students’ motivation is unclear because other variables, such as, students’ enjoyment, may intervene. In fact, based on the most recognized international motivation-focused theories, enjoyment is considered as one of the most prominent determinants of motivation (Hagger & Chatzisarantis, 2015). The aim of the present work was to analyse the mediation role of enjoyment towards PE in the relationship between teachers’ skills and students’ intrinsic motivation for PE. A total of 1,319 adolescents (girls = 51.8%, 14.28 ± 2.18 years) filled in the Intrinsic Motivation Inventory for PE (4 dimensions: interest in PE, perceived competence, importance attributed to PE, and tension felt during PE), the Teachers Personal and Professional Skills Questionnaire (2 dimensions: soft skills, and hard skills), and the Questionnaire for the Measurement of Enjoyment in PE in Adolescents (3 dimensions: pleasure, flow, and recreation). Regarding hard skills, results of the path analysis showed signs of mediation for flow (p = .035) and pleasure (p = .023) towards interest in PE, whereas the paths to perceived competence and importance attributed to PE were both fully mediated by pleasure (p = .032). Hard skills also directly increased students’ perceived tension during PE (p = .017). Soft skills positively, directly, and indirectly (through all dimensions of enjoyment) affected students’ interest in PE, importance attributed to PE, and students’ tension during PE (p < .001), whereas their impact on perceived competence was fully mediated by all enjoyment dimensions (p < .05). Our outcomes confirm the key role of teachers’ hard and soft skills, additionally supporting the idea that triggering enjoyment in students may lead to higher motivation to actively adhere to PE. In conclusion, the mediating role of enjoyment is confirmed in most of the paths leading to students’ motivation for PE; hence, it is recommended that teachers focus on using both their sets of skills to propose fun and engaging activities, which may increase participation both on the short and long term. Abstract

Chronic hepatitis B is the leading cause of hepatocellular carcinoma and a significant global health issue, affecting over 296 million people worldwide, with 15 million people coinfected with hepatitis delta virus (HDV) suffering accelerated disease progression. Recent advances in single-cell sequencing and spatial transcriptomics offer promising insights to improve the understanding of the liver’s immune responses and hepatitis B virus (HBV)-infected cell distribution, with the final goal being the achievement of an HBV ‘functional cure’. In this issue of eGastroenterology, Cross et al used the GeoMx nanostring digital spatial profiling (DSP) technology to study gene expression in the liver tissues of three patients (one HBV-monoinfected, one HBV/HDV coinfected and one HBV/human immunodeficiency virus (HIV) coinfected). Unlike other spatial transcriptomics techniques, GeoMx DSP allows targeted selection of specific tissue regions (regions of interest) for analysis, enabling precise gene expression mapping. The study revealed spatially distinct transcriptomic signatures related to immune features and viral burden, identifying a component of underinvestigated immune cells. Despite the small sample size, this proof-of-concept study demonstrates the feasibility of spatial transcriptomics in analysing HBV infections. Future advances, such as integrating viral proteins and nucleic acids, will enhance the understanding of spatial viral replication. Challenges in tissue processing, data analysis and costs remain before spatial transcriptomics can be applied as a diagnostic tool, but ongoing multiomics approaches offer promise for improved diagnosis and therapy.

Abstract

Abstract Multiple oncogenic alterations contribute to breast cancer development. Metabolic reprogramming, deeply contributing to tumor microenvironment (TME) education, is now widely recognized as a hallmark of cancer. The reverse Warburg effect induces cancer-associated fibroblasts (CAFs) to produce and secrete L-lactate, enhancing malignant characteristics such as neoangiogenesis, metastatic dissemination, and treatment resistance. Monocarboxylate transporter (MCT) 4 is involved in lactate efflux from CAFs into stromal and epithelial cells. Here, we first assess the expression of miR-425-5p and its target MCT4 in breast cancer CAFs and normal fibroblasts. We analyzed the metabolic changes induced by miR-425-5p in CAFs and its role in the education of breast cancer epithelial cells. We show that miR-425-5p-induced MCT4 knockdown decreased lactate extrusion from CAFs and its availability in the TME. miR-425-5p overexpression induced profound metabolic transformation in CAFs, ultimately influencing breast cancer metabolism. Furthermore, miR-425-5p impaired the capacity of CAFs to sustain vessel formation and breast cancer cell migration, viability, and proliferation. These findings emphasize the key role of miR-425-5p in breast cancer metabolism and aggressiveness, and its possible importance for breast cancer therapy and monitoring.

27. Ketamine-Medetomidine General Anesthesia Occurs With Alternation of Cortical Electrophysiological Activity Between High and Low Complex States

Abstract

Anesthetic agents are known to induce a range of alterations in cortical electrophysiological activity, such as the rise of signature patterns, changes in statistical properties, and altered dynamic behavior of neural records. Plenty of methods can be used to monitor these changes, among them complexity metrics demonstrated to have the power to discriminate states involving distinct levels of awareness. There is a consensus that anesthetic drugs can interfere with neural activities at different levels and time scales, being able to induce alterations both locally and in the spatiotemporal patterns established throughout the whole cortex. However, it is still unclear how such changes in the complexity of cortical activity are supposed to occur, and experimental evidence is still needed. For this purpose, we have analyzed an ECoG records database of a Ketamine-Medetomidine anesthetic induction in a non-human primate subject. The MDR-ECoG technique provided records of cortical activity with both high temporal and spatial resolution allied with extensive coverage of the cortical surface. The Permutation Entropy and the Fractal Dimension were employed to evaluate the complexity of the neural time series. It was found that the complexity of cortical activity was relatively constant during awakened conditions. The transition to unconsciousness occurred relatively fast; it required about 30 to 40 seconds for the first remarkable changes to take place. During anesthesia, the complexity assumed considerable variation at local levels and fluctuated apparently without a defined period. The cortex dynamically alternated between high and low complex states on a global scale. This study provides novel evidence of the effects of anesthetics on neural activity and cortical dynamics, contributing to the elucidation of anesthetic mechanisms in terms of circuits, pathways, and global brain functioning., Comment: 22 pages, 11 figures

Abstract

Common single-nucleotide polymorphisms (SNPs) are predicted to collectively explain 40-50% of phenotypic variation in human height, but identifying the specific variants and associated regions requires huge sample sizes1. Here, using data from a genome-wide association study of 5.4 million individuals of diverse ancestries, we show that 12,111 independent SNPs that are significantly associated with height account for nearly all of the common SNP-based heritability. These SNPs are clustered within 7,209 non-overlapping genomic segments with a mean size of around 90 kb, covering about 21% of the genome. The density of independent associations varies across the genome and the regions of increased density are enriched for biologically relevant genes. In out-of-sample estimation and prediction, the 12,111 SNPs (or all SNPs in the HapMap 3 panel2) account for 40% (45%) of phenotypic variance in populations of European ancestry but only around 10-20% (14-24%) in populations of other ancestries. Effect sizes, associated regions and gene prioritization are similar across ancestries, indicating that reduced prediction accuracy is likely to be explained by linkage disequilibrium and differences in allele frequency within associated regions. Finally, we show that the relevant biological pathways are detectable with smaller sample sizes than are needed to implicate causal genes and variants. Overall, this study provides a comprehensive map of specific genomic regions that contain the vast majority of common height-associated variants. Although this map is saturated for populations of European ancestry, further research is needed to achieve equivalent saturation in other ancestries.

ObjectiveSince there is no scientifically validated German version of the Behavioral Regulation in Exercise Questionnaire (BREQ-3), the aim of this study was to assess its psychometric parameters and invariance across sex in a sample of German-speaking young adults. The BREQ-3 is an instrument measuring the social and internal influences of motivation toward exercising. This tool is widespread within the scientific community and has been validated in several languages.MethodsA total of 271 participants (45% women; mean age = 20.67 ± 2.17 years; effect size ≥ 0.5) filled in the BREQ-3 at one time point, with a small sub-sample (n = 37) responding it a second time after 15 days. Confirmatory Factorial Analysis, Structural Modeling, and Intraclass Correlation Coefficient were used to examine the German version of the questionnaire.ResultsResults highlighted a good fit of the six-dimensional model after the removal of two items (CFI = 0.912; SRMR = 0.0594; RMSEA = 0.064), as well as full invariance across sex (pχ2 = 0.218; ΔCFI < 0.01). Internal consistency and reliability were moderate to good.ConclusionsThe 22-item German BREQ-3 is a scientifically valid instrument that can be used in cross-national studies dealing with social aspects of exercise behaviors.

Abstract

Abstract X-chromosomal genetic variants are understudied but can yield valuable insights into sexually dimorphic human traits and diseases. We performed a sex-stratified cross-ancestry X-chromosome-wide association meta-analysis of seven kidney-related traits (n = 908,697), identifying 23 loci genome-wide significantly associated with two of the traits: 7 for uric acid and 16 for estimated glomerular filtration rate (eGFR), including four novel eGFR loci containing the functionally plausible prioritized genes ACSL4, CLDN2, TSPAN6 and the female-specific DRP2. Further, we identified five novel sex-interactions, comprising male-specific effects at FAM9B and AR/EDA2R, and three sex-differential findings with larger genetic effect sizes in males at DCAF12L1 and MST4 and larger effect sizes in females at HPRT1. All prioritized genes in loci showing significant sex-interactions were located next to androgen response elements (ARE). Five ARE genes showed sex-differential expressions. This study contributes new insights into sex-dimorphisms of kidney traits along with new prioritized gene targets for further molecular research.

Abstract

Fluorene-benzothiadiazole (PFBT), commercially known as F8BT, and fluorene-thiophene (PFT) copolymers were synthesized by the Suzuki-Miyaura coupling polycondensation technique in the presence of a phase transfer catalyst (Aliquat 336). These novel copolymers showed higher molar masses, lower dispersivity, and thermal stability above 400 °C when synthesized under the modified (M) action of Aliquat 336 (F8BT-M). Polymer Light-emitting diodes (PLEDs) were assembled with the novel synthesized copolymers as the emitter layer. For F8BT-M, a maximum luminance of 327 cd/m2 was achieved, while a diode containing just the commercial F8BT resulted in only 210 cd/m2. As for the PFT copolymers, it was observed that for the Aliquat 336 sample synthesized in a 60h extended (E) (PFT-ME), the luminance of the PLED was much higher than that where the reaction time was 48 h (PFT-M), with a luminance of 168 cd/m2 and 54 cd/m2, respectively. Without Aliquat 336, the PFT copolymer did not show an electroluminescent signal. The improved electroluminescence in the copolymers with Aliquat 336 was attributed to the improved synthesis conditions, i.e., reaction time and catalytic system employed. The results suggest that copolymers with luminescent and morphological properties, more suitable for light-emitting devices, might be obtained with simple modifications during synthetic polycondensation reactions.

Abstract

The health status (HS) of children is influenced by a variety of factors, including physical fitness (PF) or social and environmental characteristics. We present a 4-year longitudinal study carried out with 263 primary school children. PF was assessed yearly using the German Motor Performance Test 6–18. Demographic data, leisure time behavior and socioeconomic factors were collected using questionnaires for children and parents. Based on parents' ratings in year 4, children were categorized as either “very good health status” (VGHS) or “good health status or below” (GHSB). Children with VGHS (73%) showed a larger improvement of global PF (p < 0.001), a significantly higher proportion of being/playing outside (p < 0.001), significantly lower proportions of overweight (p < 0.001), of media availability in the bedroom (p = 0.011) and of daily media consumption > 2 h (p = 0.033) compared to children with GHSB. Regarding socio-economic factors, children with VGHS revealed significantly fewer parents with lower education (p = 0.002), lower physical activity levels (p = 0.030) and lower migration background (p < 0.001). Physical fitness (p = 0.019) and outdoors exercising (p = 0.050) were the only variables to provide significantly higher chances of perceiving one's own health as very good when tested within a complex model including all the variables studied in this work. Considering the little focus on PF in the current Austrian physical education curriculum and the favorable environmental features of the Tyrolean region, more emphasis should be given to promoting didactical and pedagogical approaches that allow schoolers to be active in the nature.

Abstract

This work presents a sustainability assessment approach to evaluate technological innovations in secondary steel production making use of Electric Arc Furnace (EAF) technology. The assessment covers the environmental, social, and economic dimensions of sustainability by combining different tools (Life Cycle Assessment and Analytic Hierarchy Process), and also provides an integrated assessment of the overall sustainability. The approach, which can also be used to support decision-making, has been applied to a real case study of a steel plant located in Northern Italy. In the case study, environmental sustainability is positively impacted mainly by increased metal yield and reduced furnace energy consumption. The greatest social sustainability benefits are mainly related to improved ergonomic and safety conditions for workers (reduced demand for physical effort, manual handling and repetitiveness, and lower risk of accidents), as a consequence of the introduction of Industry 4.0 technologies. Regarding economic sustainability, a positive impact related to reduced cycle time, increased metal yield and quality yield, reduced maintenance and quality control costs was observed. The integrated assessment of the overall sustainability has proven to be a viable approach to manage trade-offs between the different dimensions of sustainability.

Abstract

Despite the wide distribution of textile microfibers in the marine environment, there is still limited knowledge on microfiber ingestion in fish species intended for human consumption, mostly due to analytical issues. The present study aims to assess the occurrence of microfibers in red mullet (Mullus barbatus) samples collected from the Italian waters of the central Adriatic Sea. M. barbatus is a bottom fish that lives in contact with sediment and therefore was proposed as a sentinel species for the monitoring plastic pollution. A visual approach based on the evaluation of specific microfiber surface morphology was applied for the identification of particles of different origins. The preliminary findings showed the presence of microfibers in 80% of red mullet samples with a mean of 5.95 microfibers/individual. The majority (>80%) of the isolated microfibers were of natural/artificial origin, while the dominant colors were blue and black. The obtained results confirmed that benthic fish species are susceptible to microfiber ingestion and indicate the high availability of these particles in the Adriatic basin. Considering the spectroscopic drawbacks in microfiber analyses and the need to improve the current knowledge on the rate of contamination of fishery products, the visual approach could be a feasible, easy, and accessible method in the study of microfiber pollution, and the assessment of consumer health risks.
