This is an edited transcript of a panel discussion at a Pacific preconference of the World Journalism Education Congress (WJEC) congress in Auckland in July 2016 that relates to fundamentally crucial issues about development in the region. As the world becomes more intensely interested in what is going on in the Pacific, numerous international treaties have been signed with interest in the Pacific from the European Union, the World Bank, the Asian Development Bank in partnership with the South Pacific Forum as well as massive interest from foreign donors. How these resources are being deployed is actually crucial to successful development and many news media are trying to trace where the money goes. This is probably one of the biggest challenges, aside from global climate change and depleting fishery resources, facing the Pacific and is a threat to cultural identity. Panel convenor was Fuimaono Tuiasau of Transparency International New Zealand. Panellists were: Dr Shailendra Singh, coordinator of the University of the South Pacific journalism programme, Alexander Rheeney, editor-in-chief of the PNG Post-Courier, and Kalafi Moala, owner, publisher and editor of Taimi 'o Tonga. Associate Professor Camille Nakhid, chair of the Pacific Media Centre, summed up. Disruption of processes involved in tissue development and homeostatic self-renewal is increasingly implicated in cancer initiation, progression, and recurrence. The adrenal cortex is a dynamic tissue that undergoes life-long turnover. Here, using genetic fate mapping and murine adrenocortical carcinoma (ACC) models, we have identified a population of adrenocortical stem cells that express delta-like non-canonical Notch ligand 1 (DLK1). These cells are active during development, near dormant postnatally but are re-expressed in ACC. In a study of over 200 human ACC samples, we have shown DLK1 expression is ubiquitous and is an independent prognostic marker of recurrence-free survival. Paradoxically, despite its progenitor role, spatial transcriptomic analysis has identified DLK1 expressing cell populations to have increased steroidogenic potential in human ACC, a finding also observed in four human and one murine ACC cell lines. Finally, the cleavable DLK1 ectodomain is measurable in patients' serum and can discriminate between ACC and other adrenal pathologies with high sensitivity and specificity to aid in diagnosis and follow-up of ACC patients. These data demonstrate a prognostic role for DLK1 in ACC, detail its hierarchical expression in homeostasis and oncogenic transformation and propose a role for its use as a biomarker in this malignancy., Competing Interests: Declaration of interests All authors declare no conflicts of interest.

Benchtop 99 Mo/ 99m Tc and 188 W/ 188 Re generators enable economical production of molecular theranostic 99m Tc and 188 Re radiopharmaceuticals, provided that simple, kit-based chemistry exists to radiolabel targeting vectors with these radionuclides. We have previously described a diphosphine platform that efficiently incorporates 99m Tc into receptor-targeted peptides. Here, we report its application to label a prostate-specific membrane antigen (PSMA)-targeted peptide with 99m Tc and 188 Re for diagnostic imaging and systemic radiotherapy of prostate cancer. Methods: Two diphosphine-dipeptide bioconjugates, DP1-PSMAt and DP2-PSMAt, were formulated into kits for radiolabeling with 99m Tc and 188 Re. The resulting radiotracers were studied in vitro, in prostate cancer cells, and in vivo in mouse xenograft models, to assess similarity of uptake and biodistribution for each 99m Tc/ 188 Re pair of agents. Results: Both DP1-PSMAt and DP2-PSMAt could be efficiently radiolabeled with 99m Tc and 188 Re using kit-based methods to furnish the isostructural compounds M-DP1-PSMAt and M-DP2-PSMAt (M = [ 99m Tc]Tc, [ 188 Re]Re). All 99m Tc/ 188 Re radiotracers demonstrated specific uptake in PSMA-expressing prostate cancer cells, with negligible uptake in prostate cancer cells that did not express PSMA or in which PSMA uptake was blocked. M-DP1-PSMAt and M-DP2-PSMAt also exhibited high tumor uptake (18-30 percentage injected dose per gram at 2 h after injection), low retention in nontarget organs, fast blood clearance, and excretion predominantly via a renal pathway. Importantly, each pair of 99m Tc/ 188 Re radiotracers showed near-identical biologic behavior in these experiments. Conclusion: We have prepared and developed novel pairs of isostructural PSMA-targeting 99m Tc/ 188 Re theranostic agents. These generator-based theranostic agents have potential to provide access to the benefits of PSMA-targeted diagnostic imaging and systemic radiotherapy in health care settings that do not routinely have access to either reactor-produced 177 Lu radiopharmaceuticals or PET/CT infrastructure., (© 2024 by the Society of Nuclear Medicine and Molecular Imaging.)
