Emerging viruses are defined as viruses gaining new territories or as known viruses that have mutated or recombined and become more pathogenic and contagious in the population. These viruses represent major public health problems as they can sometimes cause unknown and potentially serious disorders, particularly neuroinvasive diseases. This is the case, for example, with the recent emergence of two neurotropic viruses: Zika virus (ZIKV) and Usutu virus (USUV). ZIKV is responsible for epidemics mainly in French Polynesia in 2013 and Latin America in 2015-2016, where it was also responsible for neurological disorders (encephalitis, meningoencephalitis, Guillain-Barré syndrome, microcephaly, and others) in some patients. USUV has recently emerged in Europe, where it has been implicated in significant avian mortality, mainly in blackbirds, but also in neuroinvasive infections in humans (encephalitis, meningoencephalitis, and others). Despite this, the pathogenesis of these two emerging viruses remained poorly explored. The objective of this thesis was to characterize the effects of ZIKV infection on the blood-brain barrier (BBB) structure and homeostasis and to evaluate and compare the neuropathogenicity of different USUV lineages by combining in vivo and in vitro approaches. Our results first suggest a complex interaction between ZIKV and the BBB that could trigger local inflammation, leukocyte recruitment, and possible long-term cerebrovascular impairment. Secondly, regarding USUV, our data suggest that the virus can reach the brain and cause massive inflammation leading to the recruitment of immune cells, which correlates with human brain inflammatory pathologies such as encephalitis. Also, following the analysis of circulating USUV lineages, our results suggest that some USUV lineages may have a higher degree of neurovirulence, notably the Europe 2 lineage. The emergence and rapid spread of new viruses constitute a complex and constant challenge for human health. Today, many factors such as ecological changes, increased animal movements, world population density, and improved means of transport are contributing to emerging virus appearance and facilitating their transmission. For these reasons, it is necessary to study in greater detail mechanisms of viral infection (entry of the virus into cells, transmission, multiplication, and etc.) to develop appropriate antiviral treatments or specific vaccines.Key Words: Zika Virus, Usutu Virus, flavivirus, central nervous system, neurovirulence and neurotropism; Les virus émergents sont définis comme étant des virus gagnant de nouveaux territoires ou comme des virus connus mais ayant muté ou recombiné au point de devenir beaucoup plus transmissibles et contagieux dans la population. USUV, quant à lui, a émergé récemment en Europe, où il a été impliqué dans une importante mortalité aviaire (principalement chez le merle noir) mais aussi dans des infections neuroinvasives chez l’Homme (encéphalite, méningo-encéphalite, etc.). Malgré cela, la pathogenèse de ces deux virus reste peu explorée. L'objectif de cette thèse était de caractériser l’effet de l’infection du ZIKV sur la structure et l’homéostasie de la barrière hémato-encéphalique (BHE) ainsi que d'évaluer et de comparer la neuropathogénicité de différentes lignées d’USUV en combinant des approches in vivo et in vitro. Nos résultats suggèrent premièrement une interaction complexe entre le ZIKV et la BHE qui pourrait déclencher une inflammation locale, le recrutement de leucocytes et une éventuelle perturbation vasculaire cérébrale sur du long terme. Deuxièmement, pour USUV, nos résultats corrèlent avec les pathologies inflammatoires cérébrales engendrées chez l’Homme comme l’encéphalite. De plus, suite à l’analyse des lignées circulantes d’USUV, nos résultats suggèrent que certaines lignées d’USUV peuvent avoir une neurovirulence plus importante, notamment la lignée Europe 2. L'émergence et la propagation rapide des nouveaux virus illustrent le défi complexe et constant pour la santé humaine. Aujourd’hui, de nombreux facteurs tels que les modifications écologiques, l’augmentation des déplacements animaliers et de la densité de la population mondiale ainsi que l’amélioration des moyens de transport favorisent l’apparition des virus émergents et facilitent leur transmission. Pour ces raisons, il est nécessaire d’étudier plus en détails les mécanismes impliqués dans l’infection virale (entrée du virus dans les cellules, sa transmission, sa multiplication et etc.) pour développer des traitements antiviraux adaptés ou des vaccins spécifiques.Mots clés : Virus Zika, Virus Usutu, flavivirus, système nerveux central, neurovirulence et neurotropisme

- Abstract
Usutu virus (USUV) is an emerging neurotropic arthropod-borne virus recently involved in massive die offs of wild birds predominantly reported in Europe. Although primarily asymptomatic or presenting mild clinical signs, humans infected by USUV can develop neuroinvasive pathologies (including encephalitis and meningoencephalitis). Similar to other flaviviruses, such as West Nile virus, USUV is capable of reaching the central nervous system. However, the neuropathogenesis of USUV is still poorly understood, and the virulence of the specific USUV lineages is currently unknown. One of the major complexities of the study of USUV pathogenesis is the presence of a great diversity of lineages circulating at the same time and in the same location.

Aujourd’hui, de nombreux facteurs tels que les modifications écologiques, l’augmentation des déplacements animaliers et de la densité de la population mondiale ainsi que l’amélioration des moyens de transport favorisent l’apparition des virus émergents et facilitent leur transmission. Usutu virus (USUV) is an emerging arbovirus that was first isolated in South Africa in 1959. This Flavivirus is maintained in the environment through a typical enzootic cycle involving mosquitoes and birds. USUV has spread to a large part of the European continent over the two decades mainly leading to substantial avian mortalities with a significant recrudescence of bird infections recorded throughout Europe within the few last years. USUV infection in humans is considered to be most often asymptomatic or to cause mild clinical signs. Nonetheless, a few cases of neurological complications such as encephalitis or meningoencephalitis have been reported. USUV and West Nile virus (WNV) share many features, like a close phylogenetic relatedness and a similar ecology, with co-circulation frequently observed in nature. However, USUV has been much less studied and in-depth comparisons of the biology of these viruses are yet rare. In this review, we discuss the main body of knowledge regarding USUV and compare it with the literature on WNV, addressing in particular virological and clinical aspects, and pointing data gaps.

National audience; Usutu virus (USUV) is an emerging arbovirus, close to the West Nile virus (WNV), which was first isolated in South Africa in 1959. This flavivirus has spread to a large part of the European continent, causing bird deaths, particularly in 2018. Although human infection seems to be mostly asymptomatic, several cases of neurological complications (encephalitis, meningoencephalitis) have been described. The description in Montpellier of an atypical case of Bell's palsy suggests that the clinical spectrum of USUV-related infections may be more extensive than expected and highlights our limited knowledge of the pathophysiology of this emerging virus.; Le virus Usutu (USUV) est un arbovirus émergent, proche du virus West Nile (WNV), isolé pour la première fois en Afrique du Sud en 1959. Ce flavivirus s’est propagé sur une grande partie du continent européen, provoquant des mortalités aviaires importantes, notamment en 2018. Usutu virus, an arbovirus discovered in Africa in 1959, has spread over a large part of Europe over the last twenty years causing significant bird mortality as reported in France since 2015. The zoonotic risk, associated with this succession of avian epizootics in Europe, deserves to be taken into account even if human cases remain rare to date. Human infections are most often asymptomatic or present a benign clinical expression. However, neurological complications such as encephalitis or meningoencephalitis have been described. In addition, the recent description of an atypical case of facial paralysis in France suggests that the clinical spectrum of infections caused by Usutu virus is not fully characterized. Finally, the recent history of other arboviral outbreaks invites the scientific community to be extremely vigilant., Le virus Usutu, un arbovirus découvert en 1959 en Afrique, s’est propagé au cours des vingt dernières années sur une grande partie du continent européen provoquant notamment des mortalités aviaires importantes, comme cela est rapporté en France depuis 2015. Le risque zoonotique associé à cette succession d’épizooties aviaires en Europe mérite d’être considéré, même si à ce jour les cas humains restent exceptionnels. L’infection humaine est le plus souvent asymptomatique, ou d’une expression clinique bénigne. Toutefois, des complications neurologiques telles qu’encéphalites ou méningoencéphalites ont été décrites. L’observation récente en France d’un cas atypique de paralysie faciale a frigore1 suggère que le spectre clinique des infections dues au virus Usutu n’est pas complétement connu. L’histoire récente de flambées épidémiques d’autres arboviroses invite ainsi la communauté scientifique à la plus grande vigilance. Les connaissances concernant la physiopathologie de ce virus émergent sont, pour l’heure, très sommaires. Les travaux en cours visent donc à mieux appréhender sa biologie et les mécanismes associés aux atteintes neurologiques.
