Disturbances are important for renewal of North American forests. Here we summarize more than 180 site years of eddy covariance measurements of carbon dioxide flux made at forest chronosequences in North America. The disturbances included stand-replacing fire (Alaska, Arizona, Manitoba, and Saskatchewan) and harvest (British Columbia, Florida, New Brunswick, Oregon, Quebec, Saskatchewan, and Wisconsin) events, insect infestations (gypsy moth, forest tent caterpillar, and mountain pine beetle), Hurricane Wilma, and silvicultural thinning (Arizona, California, and New Brunswick). Net ecosystem production (NEP) showed a carbon loss from all ecosystems following a stand-replacing disturbance, becoming a carbon sink by 20 years for all ecosystems and by 10 years for most. Maximum carbon losses following disturbance (g C m−2y−1) ranged from 1270 in Florida to 200 in boreal ecosystems. Similarly, for forests less than 100 years old, maximum uptake (g C m−2y−1) was 1180 in Florida mangroves and 210 in boreal ecosystems. More temperate forests had intermediate fluxes. Boreal ecosystems were relatively time invariant after 20 years, whereas western ecosystems tended to increase in carbon gain over time. This was driven mostly by gross photosynthetic production (GPP) because total ecosystem respiration (ER) and heterotrophic respiration were relatively invariant with age. GPP/ER was as low as 0.2 immediately following stand-replacing disturbance reaching a constant value of 1.2 after 20 years. NEP following insect defoliations and silvicultural thinning showed lesser changes than stand-replacing events, with decreases in the year of disturbance followed by rapid recovery. NEP decreased in a mangrove ecosystem following Hurricane Wilma because of a decrease in GPP and an increase in ER.

Background and Purpose: Prostanoid EP2 receptor agonists exhibit several activities including ocular hypotension, tocolysis and anti-inflammatory activity. This report describes the affinity and selectivity of a structurally novel, non-prostanoid EP2 receptor agonist, PGN-9856, and its therapeutic potential.Experimental Approach: The pharmacology of a series of non-prostanoid EP2 receptor agonists was determined according to functional and radioligand binding studies, mostly using human recombinant prostanoid receptor transfectants. The selectivity of PGN-9856, as the preferred compound, was subsequently determined by using a diverse variety of non-prostanoid target proteins. The therapeutic potential of PGN-9856 was addressed by determining its activity in relevant primate cell, tissue and disease models.Key Results: PGN-9856 was a selective and high affinity (pKi ≥ 8.3) ligand at human recombinant EP2 receptors. In addition to high affinity binding, it was a potent and full EP2 receptor agonist with a high level of selectivity at EP1 , EP3 , EP4 , DP, FP, IP and TP receptors. In cells overexpressing human recombinant EP2 receptors, PGN-9856 displayed a potency (pEC50 ≥ 8.5) and a maximal response (increase in cAMP) comparable to that of the endogenous agonist PGE2 . PGN-9856 exhibited no appreciable affinity (up 10 μM) for a range of 53 other receptors, ion channels and enzymes. Finally, PGN-9856 exhibited tocolytic, anti-inflammatory and long-acting ocular hypotensive properties consistent with its potent EP2 receptor agonist properties.Conclusions and Implications: PGN-9856 is a potent, selective and efficacious prostanoid EP2 receptor agonist with diverse potential therapeutic applications: tocolytic, anti-inflammatory and notably anti-glaucoma. Objective Caspase-1 is required for nephritis and robust autoantibody development in the pristane model of murine lupus. The objective of this study was to evaluate the immune response and to study the splenic B and T cell populations in wild-type (WT) and caspase-1 -/- mice following pristane injection in order to develop an understanding of why absence of caspase-1 is protective in pristane-induced lupus. Methods Immunization responses to NP-Ficoll and NP-ovalbumin were assessed in WT and caspase-1 -/- mice. In vitro IgM and IgG responses to R848 were measured by ELISA. Serum IgM anti-dsDNA and IL-1β were also measured by ELISA. B and T cell populations 2 weeks and 6 months following pristane injection were measured by flow cytometry in WT and caspase-1 -/- mice. Results Caspase-1 -/- mice generate equivalent IgG responses to NP-Ficoll and NP-ova antigens when compared to wild-type mice. Additionally, they secrete IgM and IgG in response to TLR7 activation. Pristane injected WT and caspase-1 -/- mice generate robust IgM anti-dsDNA responses. Caspase-1 -/- mice have a significant reduction in marginal zone B cell populations compared to WT 6 months after pristane exposure whereas T cell responses are intact in these mice. Conclusions Caspase-1 -/- mice have intact immune responses but do not develop an expanded marginal zone B cell population in response to pristane-induced lupus. Hydrologic variability during 2005-2011 was observed and analyzed at an upland oak/pine forest in the New Jersey Pinelands. The forest experienced defoliation by Gypsy moth ( Lymantria dispar L.) in 2007, drought conditions in 2006 and a more severe drought in 2010. By using sap flux and eddy covariance measurements, stream discharge data from USGS, soil water changes, precipitation ( P) and precipitation throughfall, a local water balance was derived. Average annual canopy transpiration ( EC) during 2005-2011 was 201 mm a−1 ± 47 mm a−1. A defoliation event reduced EC by 20% in 2007 compared with the 2005-2011 mean. During drought years in 2006 and 2010, stand transpiration was reduced by 8% in July 2006 and by 18% in 2010, respectively, compared with the overall July average. During July 2007, after the defoliation and subsequent reflushing of half of the leaves, EC was reduced by 25%. This stand may experience higher sensitivity to drought when recovering from a defoliation event as evidenced by the higher reduction of EC in 2010 (post-defoliation) compared with 2006 (pre-defoliation). Stream water discharge was normalized to the watershed area by dividing outflow with the watershed area. It showed the greatest correlation with transpiration for time lags of 24 days and 219 days, suggesting hydrological connectivity on the watershed scale; stream water discharge increases when transpiration decreases, coinciding with leaf-on and leaf-off conditions. Thus, any changes in transpiration or precipitation will also alter stream water discharge and therefore water availability. Under future climate change, frequency and intensity of precipitation and episodic defoliation events may alter local water balance components in this upland oak/pine forest.
