Pyometra is frequently diagnosed in the female dogs, and it is characterized by endometrial inflammation, accumulation of purulent exudate within the lumen, and bacterial infection. In the dog, pyometra affects more often aged nulliparous bitches during the luteal phase. Pathogenesis of pyometra is multifactorial and progesterone seems to be a key factor. Cystic endometrial hyperplasia has been described as a predisposing condition for canine pyometra. However, a recent study demonstrated that cystic endometrial hyperplasia is not significantly associated with naturally occurring pyometra, whereas there is a significant association of this condition with pseudoplacentational endometrial hyperplasia. The aim of this review is to provide an update on canine pyometra, with focus on its association with uterine hyperplasic lesions, which supports a proposal for adoption of more adequate diagnostic terminology.

Teratoma is a rare neoplasia with differentiation in two or three germ cell lines. Intracranial teratoma in birds has rarely been reported, especially affecting the brain. This is the first report of a brain teratoma in a mallard with neurological clinical signs. The neoplasm was characterized as a mature brain teratoma, extending from the cerebellum to the brainstem, and with one nodule in the cortex.

Background Toxoplasmosis is a zoonotic disease that affects humans and warm-blooded animals. This study describes an outbreak of toxoplasmosis in howler monkeys (Alouatta sp.) and survival of capuchins (Sapajus apella), under the same environmental conditions. Methods Howler monkeys were submitted to post-mortem examination. Tissue samples were processed to histopathology and immunohistochemistry to detect lesions and tachyzoites of Toxoplasma gondii. Tissue samples were also frozen and submitted to PCR and genotyping of T. gondii. Results Typical lesions were observed in several organs including the liver, lymph node, and brain, with intralesional cysts and tachyzoites of T. gondii demonstrated by immunohistochemistry. T. gondii genomic sequences were amplified by PCR, and genotyping characterized the same T. gondii clone in all howler monkeys. Conclusions Our results support the notion that some species of neotropical primates are highly susceptible to toxoplasmosis and the hypothesis that capuchins (S. apella) may be resistant.

Uterine inflammation is the most common and clinically relevant genital change in bitches. The aim of this study was to evaluate the association between uterine hyperplasic lesions and uterine inflammation. Uteri and ovaries from 200 bitches were sampled after elective or pathological ovariohysterectomy and histologically evaluated. Endometrial hyperplastic lesions were classified as cystic endometrial hyperplasia (CEH) or pseudoplacentational endometrial hyperplasia (PEH), whereas cases of uterine inflammation were subdivided into endometritis or pyometra. The frequency of PEH (79.4%) was significantly higher (P 0.05) than CEH (20.6%), whereas 17.5% of the animals did not have any endometrial hyperplasic lesions. There was a statistically significant association between the frequency of PEH and uterine inflammation (P 0.0001) and PEH was also significantly associated with pyometra (P 0.05). There was no significant association between CEH and uterine inflammation. There was a significant association between dioestrus and the frequencies of PEH, CEH, uterine inflammation and pyometra. However, there was no significant association between endometritis and dioestrus. These results support the recommendation that the terms 'cystic endometrial hyperplasia', 'pseudoplacentational endometrial hyperplasia' and 'pyometra' should be used separately, instead of the classical term 'cystic endometrial hyperplasia-pyometra complex'.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A patologia digital tem se tornado um campo próspero nos últimos tempos em razão da introdução no mercado de scanners capazes de gerar imagens digitais de lâminas de vidro com ótima resolução. A patologia digital vem sendo utilizada em laboratórios de diagnósticos clínicos e também na área acadêmica. Métodos de validação já estão disponíveis para avaliar o impacto da patologia digital quando usada como método de diagnóstico primário. Estudos comparando diagnósticos dados em lâminas físicas e digitais apresentam concordância acima de 85%, porém os resultados são bastante variados entre os trabalhos. O trabalho teve como objetivo comparar a habilidade diagnóstica de estudantes e patologistas veterinários na leitura de lâminas histopatológicas digitais e físicas e, com isso, demonstrar aspectos positivos e negativos da patologia digital na formação e atuação de estudantes e profissionais no diagnóstico histopatológico. Os dados encontrados nesse trabalho mostram uma concordância intraexaminador média de 84,87%, além de boa precisão diagnóstica. Uma autoavaliação da experiência vivenciada com a patologia digital foi feita com os examinadores e, apesar da maior familiaridade com a lâmina física, todos foram a favor do uso da patologia digital no ensino. Espera-se que as aplicações e o uso de lâminas digitais no ensino, pesquisa e principalmente na área de diagnóstico aumentem acentuadamente na próxima década. Cabe as instituições acadêmicas se atentarem para importância da implementação da patologia digital no ensino e no treinamento dos profissionais que irão atuar em um mercado de patologia veterinária cada vez mais digital.

In horses, there is an increasing interest in developing long-lasting drug formulations, with bi-opolymers as viable carrier alternatives in addition to their use as scaffolds, suture threads, screws, pins, and plates for orthopedic surgeries. This communication focuses on the prolonged biocompatibility and biodegradation of PLA, prepared by hot pressing at 180 ºC. Six samples were implanted subcutaneously on the lateral surface of the neck of one horse. The polymers remained implanted for 24 to 57 weeks. Physical examination, plasma fibrinogen, and the mechanical noci-ceptive threshold (MNT) were performed. After 24, 28, 34, 38, and 57 weeks, the materials were removed for histochemical analysis using hematoxylin-eosin and scanning electron microscopy (SEM). There were no essential clinical changes. MNT decreased after the implantation procedure, returning to normal after 48h. A foreign body response was observed by histopathologic evalua-tion up to 38 weeks. At 57 weeks, no polymer or fibrotic capsules were identified. SEM showed surface roughness suggesting a biodegradation process, with an increase in the average pore di-ameter. As in the histopathological evaluation, it was not possible to detect the polymer 57 weeks after implantation. PLA showed biocompatible degradation and these findings may contribute to future research in the biomedical area.

Cardiac disease is of importance in captive chimpanzee (Pan troglodytes) health. Here we report an eosinophilic and necrotizing myocarditis in a 17-y-old chimpanzee with no previous history of cardiac disease that progressed to death within 48 h. Toxic and infectious causes were ruled out. The chimpanzee had eosinophilia at different occasions in previous years. The animal had a severe, diffuse, and acute monophasic necrotizing myocarditis, with a moderate lymphoplasmacytic infiltrate that was rich in eosinophils. Ante- and postmortem investigations are compatible with an unusual eosinophilic myocarditis with clinical evolution and morphology comparable with human eosinophilic myocarditis secondary to hypereosinophilic syndrome.

Squamous metaplasia of the rete ovarii is an ovarian pathologic change characterized by replacement of the normal single layered cuboidal epithelium of the rete ovarii by a stratified squamous keratinized epithelium. Uterus and ovaries from a local slaughterhouse pregnant crossbreed cow were evaluated through ultrasound, macroscopically and histologically. Grossly, there were multiple cysts in both ovaries, which were histologically characterized as rete ovarii cysts with squamous metaplasia and intraluminal accumulation of keratinized material. Squamous metaplasia of the rete ovarii has been previously reported in cows, however this is the first report of this condition in a pregnant animal, demonstrating that this ovarian change is compatible with pregnancy.

No. of bitstreams: 0 Previous issue date: 2017-11-15 The use of silvopastoral systems (SPS) can be a good alternative to reduce the environmental impacts of livestock breeding in Brazil. One of the reasons for its scarce adoption is the lack of information on health and productivity of cattle raised under these conditions. The experiment reported here was designed to compare the infestation by external parasites - the cattle tick (Rhipicephalus microplus), horn fly (Haematobia irritans), and larvae of the botfly (Dermatobia hominis) - in beef cattle raised in a SPS and a conventional pasture system (CPS), evaluated for 24 months. Data on air and soil temperature, solar radiation, wind incidence and water balance were used to characterize the SPS and CPS. R. microplus adult females and D. hominis larvae were counted on the body of each animal to determine the parasites burdens, but we did not find significant differences between the two systems. Horn flies counts on animals' body, and analysis of the horn fly and its pupal parasitoids associated with the dung pats were obtained in the two systems. Horn fly infestation was significantly lower (p = 0.01) in the SPS (13.17 +/- 3.46) in comparison with the CPS (24.02 +/- 4.43). In SPS and CPS, respectively, the mean densities of pupae of H. irritansin dung pats were 9.8 and 10.7; the mean density of adults of H. irritans, 3.7 and 3.5; and the density of its pupal parasitoids were 20.5 and 5.4. The effect of production system was significant (p < 0.05) only for the occurrence of pupal parasitoids of the horn fly, where the greatest occurrences of these natural enemies were in the SPS. These data indicate that natural enemies were able to control, at least partially, the horn fly populations in the cattle. Samples with the anticoagulant EDTA were used for DNA extraction and without anticoagulant for separation of the blood serum. The quantification of the NC mt-cyB of B. bovis was carried out by the quantitative PCR technique (qPCR), while the anti-B. bovis IgG antibody titers (S/P) were quantified by the ELISA method. The NC and S/P data were log10-transformed to improve the approximation to the normal distribution and were analyzed using mixed models. The correlations between NC mt-cyB and S/P were estimated, as well as the repeatability values for each trait. The results obtained showed the high sensitivity of the techniques, with 100% of the animals being positive for B. bovis, detected by both the serological and molecular tests. The correlations estimated between NC and S/P were low, 0.10 and 0.12, in the first and second collection, respectively. The repeatability estimated for NC was 0.06, whereas for the SIP it was 0.42. The low correlations between S/P and NC in the two collections demonstrated that the variation in the NC value is independent of the level of antibodies. This results indicated that animals with a higher levels of antibodies against B. bovis in the first collection continued to have a higher levels in the second one. However, the very low values for the repeatability value of NC, and for the correlations between S/P and NC, demonstrates that neither NC or S/P could be used to discriminate animals for resistance/susceptibility to B. bovis. Despite the advantages offered by public policies, many producers hesitate to use this system. One of the reasons is the lack of information on health and productivity of cattle raised under these conditions. The experiment reported here was designed to compare the behavior of infection by gastrointestinal nematodes and weight gain of beef cattle raised in a SPS and a conventional pasture system. We monitored the number of eggs per gram of feces, the prevalent nematode genus, data on climate, forage availability, weight gain and packed cell volume (PCV) of the animals bred in the two systems. The infection by nematodes was significantly higher in the cattle raised in the SPS (p 0.05). Despite the higher prevalence of nematodes in the SPS, no negative impact was detected on the animals’ weight gain and health. The results of this experiment indicate that under the conditions studied, there is no need to alter the parasite management to assure good productive performance of cattle. Six buffalo calves constituted the control group (CNT) and six were orally inoculate with 108 CFU of S. Dublin (INF). Clinical evaluation was performed, rectal swabs to detect S. Dublin strains were collected and venous blood was sampled before and throughout seven days after inoculation. The APP fractions β-haptoglobin, α-haptoglobin, ceruloplasmin and transferrin were analyzed by 1-D and 2-D electrophoresis. Proteins were identified using LC/ESI-MS/MS and NCBI database. Plasma fibrinogen, serum iron and serum haptoglobin concentrations were measured. The inoculation of 108 CFU of S. Dublin was effective in inducing clinical signs of Salmonellosis, such as hyperthermia and diarrhea. 1-DE showed that β and α-haptoglobin increased 204% (p = 0.008) and 184% (p = 0.022) 48 h after inoculation (HAI), respectively, with highest concentrations 120 HAI (498% increased, p = 0.012; 431% increased, p = 0.011) and 168 HAI (492% increased, p = 0.019; 523% increased, p = 0.028). 2-DE showed that the expression of two spots, identified as β-haptoglobin, were increased 693% (p = 0.0006) and 580% (p = 0.0003) 168 HAI, respectively, while one spot, identified as α-haptoglobin, increased 714% (p = 0.040). Haptoglobin concentrations increased 1339% (p < 0.0001) 168 HAI. 1-DE showed that ceruloplasmin increased 42% (p = 0.034) 48 HAI, with highest concentration 120 HAI (133% increased, p = 0.022). 2-DE showed that the expression of two spots, identified as ceruloplasmin, were increased 218% (p = 0.0153) and 85% (p = 0.0143) 168 HAI, respectively. Fibrinogen increased 78% (p = 0.012) 96 HAI, with highest concentration 120 HAI (increased 114%, p = 0.002). Iron decreased 33% 24 HAI (p = 0.015) and 37% 72 HAI (p = 0.029), and began to be restored 96 HAI. 1-DE showed that transferrin decreased 23% 120 HAI (p = 0.047), and that values were restored 168 HAI. 2-DE showed that expression patterns of transferrin comparing 0 h and 168 HAI were similar, evidencing that values were restored 168 HAI. In conclusion, the inoculation of 108 CFU was effective in inducing hyperthermia and diahrrea. β and α-haptoglobin, ceruloplasmin and fibrinogen worked as positive APP during the APR to S. Dublin infection and are potential biomarker candidates. Concentrations of iron and transferrin decreased during the infection, highlighting the fact that mechanisms for restricting iron availability are part of the APR triggered against S. Dublin infection in buffalo calves. No. of bitstreams: 0 Previous issue date: 2017-07-01 University of Glasgow Establishing of reference intervals (RI) for hematologic variables and blood serum acute phase proteins (APP) of healthy newborn buffaloes is an important tool for monitoring alterations during infection and inflammation. Considering the scarcity of published data on newborns, the aim of the study was to establish RI for hematologic variables and APP from healthy newborn buffaloes. Blood samples from 28 healthy Murrah buffalo calves, 10–30 days old, were selected to determine RI. Fourteen hematologic and four blood APP variables were analyzed. Before collection of blood samples, calves were subjected to physical examination (rectal temperature, degree of dehydration, and fecal consistency) and only calves that were considered healthy were included in the study. The Anderson-Darling test was used to assess normal distribution of values. The Dixon test and Tukey test were used to identify outliers. RI and 90% CI were determined using standard/robust methods and Box-Cox transformation. RI for variables analyzed were the following: (1) hematologic variables: RBC 7.5–12.9 × 106/μL, HGB 10.6–19.0 g/dL, packed cell volume 33.1–54.8%, mean corpuscular volume 36.2–50.6 fL, mean corpuscular hemoglobin 12.1–17.3 pg, mean corpuscular hemoglobin concentration 28.1–42.9 g/dL, platelets 361–1081 × 103/μL, WBC 6.56–18.2 × 103/μL, lymphocytes 4.15–12.8 × 103/μL, segmented neutrophils 0.950–10.6 × 103/μL, band neutrophils 0–0.160 × 103/μL, monocytes 0–0.754 × 103/μL, eosinophils 0–0.326 × 103/μL, and basophils 0–0.149 × 103/μL and (2) APP variables: fibrinogen 2.49-9.50 g/L, haptoglobin 0.02-0.56 g/L, serum amyloid A (SAA) 3.70-97.51 μg/mL, and C-reactive protein (CRP) 0.02-2.78 μg/mL. In conclusion, hematologic and acute phase protein RI have been documented and can be used as a physiologic database to help the interpretation of laboratory results of newborn buffaloes during infection and inflammation conditions. This study aimed to evaluate the profile of predominant inflammatory cells by immunohistochemistry in cutaneus squamous cell carcinoma (SCC) of dogs, comparing it with the degree of differentiation of the tumor. Twenty samples of SCC were analyzed. The tumors were histologically classified into two groups, differentiated SCC (SCCd=12) and undifferentiated SCC (SCCu=08). The tumor inflammatory infiltrate was determined by immunohistochemistry, in order to identify macrophages, lymphocytes and plasma cells. The comparison between groups, SCCd and SCCu, was not significant concerning the density of macrophages (P=1.0), T lymphocytes (P=0.335) and plasma cells (P=0.075). Fifty-one contemporaneous Angus cattle were evaluated on two occasions. The number of standard female Rhipicephalus microplus ticks present on the left side of the body was counted and blood samples were drawn from the tail vein into tubes containing the anticoagulant EDTA. The blood samples were submitted to DNA extraction and used to quantify the number of copies (NC) of DNA from B. bovis and B. bigemina by qPCR. The data on tick count and number of DNA copies were transformed for normalization and analyzed by a mixed model method. A multivariate model with repeated measures of the same animal, including the effects of collection, parasite species and their interaction, was used. The repeatability values were obtained from the matrix of (co)variances and were expressed for each species. The correlations between the counts of different species on the same animal, in the same collection or different collections, were also estimated. The results showed the qPCR could distinguish the two between infection by the two Babesia species. Infection levels by B. bovis and B. bigemina were detected in 100% and 98% of the animals, respectively. Significant differences were found (P < 0.05) between the NC of the two Babesia species, B. bovis 1.49 +/- 0.07 vs. B. bigemina 0.82 +/- 0.06. Low repeatabilities were found for the counts of R. microplus and NC of B. bovis and B. bigemina: 0.05, 0.10 and 0.02, respectively. The correlations between R. microplus count and NC Of B. bovis and B. bigemina were both very near zero. However, an association was observed between the NC of the two species, with a correlation coefficient of 0.30 for measures from the same collection. The absence of associations between the quantity of DNA from B. bovis and B. bigemina and the tick counts suggests that the variation of parasitemia by the hemoparasites did not depend on the tick infestation levels at the moment of each collection. The repeatability values estimated indicate that under the study conditions, the variations in the tick infestation levels and of parasitemia by B. bovis and B. bigemina depend more on factors related to each collection than on intrinsic factors of the animal.
