Standardized patient encounters are being utilized more often in occupational therapy education as a replacement for traditional fieldwork. While there is a growing body of research to support the use of this model in developing student skillsets and confidence, there remains limited information on the topic, and no studies which look at its use with occupational therapy assistant students. Twenty-four occupational therapy assistant students participated in standardized patient encounters which served as a replacement for traditional Level I fieldwork. All students completed an anonymous supplementary course evaluation regarding their experiences. A secondary analysis of the data looked at their perceptions of a standardized patient encounter model for fieldwork and how it did or did not prepare them for future Level II clinicals. Quantitative and qualitative data exposed aspects of these experiences which students found to be most and least effective. Overall, data showed moderate support for use of standardized patient encounters to support student confidence and skills in anticipation of Level II fieldwork. The student perception of these types of experiences is influenced by environmental (e.g., structure) and personal (e.g., student personality) factors. Success requires adequate preparation of all involved parties. Use of standardized patient encounters should be researched further to determine its impact on fieldwork performance evaluation scores and future employment.

Background: Abdominoplasty is a common surgical procedure in which excess abdominal skin and fat are reduced to improve body contouring. Fibrin sealant has been proposed to reduce postsurgical bleeding and exudation. In this study, we evaluated whether there was a significant statistical difference in surgical output between the use of fibrin glue and its nonuse in abdominoplasty surgery, specifically in reducing bleeding and exudation. Material and methods: A retrospective chart review of 68 postbariatric abdominoplasty patients (58 females, 10 males) was performed. We divided the patients into Group A (30 cases, 44%), in which we used fibrin sealant, and Group B (38 cases, 56%), in which we did not use fibrin glue. We calculated the total amount of liquid in suction drainages until the day of their removal. Statistical analysis included the independent t-test with a significance level of 0.05. Results: The average drainage output in Group A was 620.0 ± 375.0 mL, whereas in Group B, it was 500.0 ± 290.0 mL. Results indicate an insignificant correlation between the use of fibrin glue and the amount of liquid in the surgical drains (t = 1.52, p = 0.13). The result is not significant at p

Small interfering RNA (siRNA) is essential for the process of gene silencing, especially for cancer. Despite its considerable promise, siRNA faces challenges due to stability issues of formulation and undesirable off-target side effects. In order to address these difficulties, it is essential to carefully monitor the levels of siRNA. The existing point-of-care (POC) systems cannot precisely and efficiently detect or monitor siRNA levels. In light of these challenges, this review gives the prospects of siRNA detection by proposing a novel hypothesis of existing electrical and optical-based detection of DNA/RNA with the POC platform. This hypothesis offers an interesting novel perspective to potentially fill the existing gaps, in detecting siRNA. By utilising these technologies, there is high potential to develop a proof-of-concept system that will not only overcome the existing challenges, but it will also allow effective and precise monitoring of siRNA, in real-world healthcare environments. In summary, the prospects for siRNA in the realm of POC platforms are quite encouraging, since it allows precise and effective monitoring.

SARS-CoV-2 viral-derived particles have been proposed to have a causal role in tissue inflammation. Macrophage is the culprit cell in the pathogenesis of destructive inflammatory response to the SARS-CoV-2 virus. We investigated whether the spike protein might play a role in perturbing the physiological process of resolution of inflammation. Using an in vitro model of M2 polarized macrophages, we found that recombinant spike protein produced typical M1 morphological features in these alternative differentiated cells. In the presence of spike, M2-macrophages lose their elongated morphology, become rounded and acquire a strong capability to stimulate lymphocyte activation and proliferation. Moreover, in M2 macrophages, spike activated the signal transducer and activator-1 (STAT1) the pivotal mediator of pro-inflammatory macrophages. We observed STAT1 activation also in endothelial cells cultured with recombinant spike, accompanied by Bax upregulation and cell death. Blockade of beta3 integrin with the RGD mimetic tirofiban reverted the spike-induced costimulatory effects on M2 macrophages. Also, tirofiban counteracted STAT1 and Bax activation in endothelial cells cultured with spike and reduced endothelial cell death. In conclusion, we found that some proinflammatory effects of the spike protein can involve the integrin pathway and provide elements supporting use of RGD mimetics against SARS-Cov-2.

Biomedical applications relying on optical radiation, particularly with the advent of lasers, have experienced exponential growth in the last 20 years. Powerful optical sources are now found not only in universities, hospitals, and industries but also in beauty centers, used for tasks such as tattoo removal, and even in our homes. Despite their widespread use, managing the risks associated with lasers, particularly in non-research contexts, has not kept pace with their proliferation. While the risks associated with direct exposure to radiation to the eye and skin are relatively well understood, the hazards posed by reflected and diffuse radiation remain less characterized and monitored. Therefore, there is a critical need to assess potential eye and skin hazards in spaces where lasers and non-coherent light sources are used. This necessitates a detailed analysis of reflective surfaces, with particular emphasis on evaluating their reflectance characteristics at relevant wavelength ranges. This study investigates the reflectance and transmittance (where relevant) properties of commonly used materials in biomedical settings, including fabrics, plastics, and metals, across a broad spectrum from 250 nm (UVA) to visible light and into the infrared (IR) region up to 25 μm. Both specular (at 45° incidence) and diffuse reflectance spectra were measured using spectrophotometric techniques and used to provide a straightforward parameter to classify the specular/diffusive behavior of the different surfaces. Besides, small-angle reflectance measurements in the IR range were performed by Fourier transform infrared spectrometry. The knowledge of the material optical properties used in environments where optical radiation is employed allows for accurate assessment of associated risks. This facilitates the determination of appropriate preventive measures and the establishment of safer protocols, for both operators and, where applicable, patients and the general public. For this scope, the creation of a database of material reflective properties has been initiated.

Abstract The growth of liquid biopsy, i. e., the possibility of obtaining health information by analysing circulating species (nucleic acids, cells, proteins, and vesicles) in peripheric biofluids, is pushing the field of sensors and biosensors beyond the limit to provide decentralised solutions for nonspecialists. In particular, among all the circulating species that can be adopted in managing cancer evolution, both for diagnostic and prognostic applications, microRNAs have been highly studied and detected. The development of electrochemical devices is particularly relevant for liquid biopsy purposes, and the screen‐printed electrodes (SPEs) represent one of the building blocks for producing novel portable devices. In this work, we have taken miR‐2115‐3p as model target (it is related to lung cancer), and we have developed a biosensor by exploiting the use of a complementary DNA probe modified with methylene blue as redox mediator. In particular, the chosen sensing architecture was applied to serum measurements of the selected miRNA, obtaining a detection limit within the low nanomolar range; in addition, various platforms were interrogated, namely commercial and hand‐made SPEs, with the aim of providing the reader with some insights about the optimal platform to be used by considering both the cost and the analytical performance.

Cellular movement is essential for many vital biological functions where it plays a pivotal role both at the single cell level, such as during division or differentiation, and at the macroscopic level within tissues, where coordinated migration is crucial for proper morphogenesis. It also has an impact on various pathological processes, one for all, cancer spreading. Cell migration is a complex phenomenon and diverse experimental methods have been developed aimed at dissecting and analysing its distinct facets independently. In parallel, corresponding analytical procedures and tools have been devised to gain deep insight and interpret experimental results. Here we review established experimental techniques designed to investigate specific aspects of cell migration and present a broad collection of historical as well as cutting-edge computational tools used in quantitative analysis of cell motion.

In the framework aimed at evaluating the close link between the chemical composition of poorly explored organs of local edible medicinal plants in the Cilento region (Southern Italy) and their recovery for new health products, Cichorium intybus L. was of interest. Ultra-High Performance Liquid Chromatography Quadrupole-Time-Of-Flight tandem Mass Spectrometry (UHPLC-QqTOF-MS/MS) investigation revealed that sesquiterpene lactones characterized roots and stems, while hydroxycinnamic derivatives accumulated in florets, bracts, and leaves. Florets diverged for anthocyanin derivatives, while roots differed by low flavonoid content. Despite shared phytochemical constituents, their ratio was responsible for different antioxidant responses. Notably, extracts, particularly rich in hydroxycinnamoyl derivatives, showed protective effects against oxidative stress induced by hydrogen peroxide in SH-SY5Y and HaCaT cell lines. The wound healing assay revealed marked inhibition of SH-SY5Y cell migration, avoiding wound closure. Overall, the research encourages the use of different organs from Cilentan wild chicory to obtain antioxidant safe and consumer-guaranteed products.

Active learning methods are needed to promote student motivation and facilitate the development of technical and soft skills. Previous research in software engineering education shows that LEGO® Serious Play (LSP) fully aligns with these needs. However, prior works are usually based on insufficiently robust research methods that do not include a large sample, a variety of evaluation instruments, and/or rigorous comparative methods such as randomized controlled trials, which makes it difficult to obtain reliable and solid conclusions. This article presents an original LSP activity to teach software development life cycle models and core software engineering activities, tackling learning objectives different from those addressed by the LSP activities reported in prior works. The LSP activity was validated through a cluster-randomized controlled trial involving 217 computer science students. These students were divided into a control group that received a traditional lecture and an experimental group that performed the LSP activity. The research was supported by pre and post-tests that allowed the study of the knowledge attained by the students, as well as a questionnaire to gather students’ perceptions. The results indicate that the students in the experimental group learned significantly more and were more motivated than their counterparts in the control group. This leads to the conclusion that LSP-based activities such as the one reported in this article are highly effective in terms of knowledge acquisition and motivation to teach some software engineering topics compared to traditional lectures.

ABSTRACT: In the buffalo dairy sector, a huge effort is still needed to improve mastitis prevention, detection, and management. Electrical conductivity (EC) and total somatic cell count (SCC) are well-known indirect indicators of mastitis. Differential somatic cell count (DSCC), which represents the proportion of neutrophils and lymphocytes on the total SCC, is instead a novel phenotype collected in the dairy cattle sector in the last lustrum. As little is known about this novel trait in dairy buffalo, in the present study we explored the nongenetic factors affecting DSCC, as well as EC and total somatic cell score (SCS), in the Italian Mediterranean buffalo. The data set used for the analysis included 14,571 test-day (TD) records of 1,501 animals from 6 herds, and climatic information of the sampling locations. The original data were filtered to exclude animals with less than 3 TD per lactation and, for the investigated traits, outliers beyond 4 standard deviations. In the statistical model we included the fixed effects of herd (6 classes), days in milk (DIM; 10 classes of 30 d, with the last being an open class until 360 d), parity (6 classes, from 1 to 6+), year-season of calving (11 classes, from summer 2019 to winter 2021/2022), year-season of sampling (9 classes, from spring 2020 to spring 2022), production level (4 classes based on quartiles of average milk yield by herd), and temperature-humidity index (THI; 4 classes based on quartiles, calculated using the average temperature and relative humidity of the 5 d before sampling). Average EC, SCS, and DSCC vary across herds. Considering DIM, greater EC values were observed at the beginning and the end of lactation; SCS was slightly lower, but DSCC was greater around the lactation peak. Increased EC, SCS, and DSCC levels with increasing parity were reported. Year-season calving and year-season sampling only slightly affected the variation of the investigated traits. Milk of high-producing buffaloes was characterized by lower EC and SCS mean values, nevertheless it had slightly greater DSCC percentages. Buffaloes grouped in the highest THI classes (classes 3 and 4) showed, on average, greater EC, SCS, and DSCC in comparison to the lower classes, especially to class 2. Results of the present study represent a preliminary as well as necessary step for the possible future inclusion of EC, SCS, or DSCC in breeding programs aimed to improve mastitis resistance in dairy buffaloes.

ABSTRACT: Until now, the genetic evaluation of the Italian Mediterranean Buffalo has been mainly focused on production traits. However, female fertility affects the efficiency of the dairy industry as it is essential to maintain the profitability of dairy farms. Indeed, the estimation of its genetic component is crucial for its improvement. In this study, 3 measures of buffalo's fertility were analyzed: the age at first calving (AFC), the interval between first and second calving (CIV1), and the interval between second and successive calvings (CIV2_12). Milk yield at 270 d (MY270) was used as a correlated trait. First, genetic parameters were estimated using 7,915 buffalo cows with first calving from 1991 to 2018, then breeding values were calculated from 236,087 buffalo cows. Genetic parameters were estimated by Bayesian inference fitting a multiple-trait animal model using the GIBBS1F90 program, and BLUPF90 was used for estimation of breeding value. The heritability and repeatability estimates of fertility traits were low. The genetic correlations among fertility traits ranged from 0.10 (AFC-CIV1) to 0.92 (CIV1-CIV2_12). Genetic correlation between MY270 and fertility traits was unfavorable, ranging from 0.23 to 0.48. The results from this study can be used as a basis for the future genetic improvement of fertility traits in the Italian Mediterranean Buffaloes.

Abstract Clonal hematopoiesis (CH) is defined as a single hematopoietic stem/progenitor cell (HSPC) gaining selective advantage over a broader range of HSPCs. When linked to somatic mutations in myeloid malignancy-associated genes, such as TET2-mediated clonal hematopoiesis of indeterminate potential or CHIP, it represents increased risk for hematological malignancies and cardiovascular disease. IL1β is elevated in patients with CHIP, however, its effect is not well understood. Here we show that IL1β promotes expansion of pro-inflammatory monocytes/macrophages, coinciding with a failure in the demethylation of lymphoid and erythroid lineage associated enhancers and transcription factor binding sites, in a mouse model of CHIP with hematopoietic-cell-specific deletion of Tet2. DNA-methylation is significantly lost in wild type HSPCs upon IL1β administration, which is resisted by Tet2-deficient HSPCs, and thus IL1β enhances the self-renewing ability of Tet2-deficient HSPCs by upregulating genes associated with self-renewal and by resisting demethylation of transcription factor binding sites related to terminal differentiation. Using aged mouse models and human progenitors, we demonstrate that targeting IL1 signaling could represent an early intervention strategy in preleukemic disorders. In summary, our results show that Tet2 is an important mediator of an IL1β-promoted epigenetic program to maintain the fine balance between self-renewal and lineage differentiation during hematopoiesis.
