6. Double-crosslinkable poly(urethane)-based hydrogels relying on supramolecular interactions and light-initiated polymerization: promising tools for advanced applications in drug delivery.

Abstract

Physical and chemical hydrogels are promising platforms for tissue engineering/regenerative medicine (TERM). In particular, physical hydrogels are suitable for use in the design of drug delivery systems owing to their reversibility and responsiveness to applied stimuli and external environment. Alternatively, the use of chemical hydrogels represents a better strategy to produce stable 3D constructs in the TERM field. In this work, these two strategies were combined to develop multi-functional formulations integrating both drug delivery potential and TERM approaches in a single device. Specifically, a novel photo-sensitive poly(ether urethane) (PEU) was developed to form supramolecular networks with a-cyclodextrins (a-CDs). The PEU was successfully synthesized using Poloxamers® 407, 1,6-diisocyanatohexane and 2-hydroxyethyl methacrylate, as assessed by infrared spectroscopy, size exclusion chromatography and proton nuclear magnetic resonance (1 H NMR) spectroscopy. Subsequently, PEU thermo-responsiveness was characterized through critical micelle temperature evaluation and dynamic light scattering analyses, which suggested the achievement of a good balance between molecular mass and overall hydrophobicity. Consequently, the formation of supramolecular domains with a-CDs was demonstrated through X-ray diffraction and 1 H NMR spectroscopy. Supramolecular hydrogels with remarkably fast gelation kinetics (i.e., few minutes) were designed using a low PEU concentration (r5% w/v). All formulations were found to be cytocompatible according to the ISO 10993-5 regulation. Notably, the hydrogels were observed to possess mechanical properties and self-healing ability, according to rheological tests, and their fast photo-crosslinking was evidenced (o60 s) by photorheology. A high curcumin payload (570 mg mL-1 ) was encapsulated in the hydrogels, which was released with highly tunable and progressive kinetics in a physiological-simulated environment for up to 5 weeks. Finally, a preliminary evaluation of hydrogel extrudability was performed using an extrusionbased bioprinter, obtaining 3D-printed structures showing good morphological fidelity to the original design. Overall, the developed hydrogel platform showed promising properties for application in the emerging field of regenerative pharmacology as (i) easily injectable drug-loaded formulations suitable for post-application stabilization through light irradiation, and (ii) biomaterial inks for the fabrication of patient-specific drug-loaded patches. Abstract

Catheter-associated urinary tract infections are the most common hospital-acquired infections and cause patient discomfort, increased morbidity, and prolonged stays, altogether posing a huge burden on healthcare services. Colonization occurs upon insertion, or later by ascending microbes from the rich periurethral flora, and is therefore virtually unavoidable by medical procedures. Importantly, the dwell time is a significant risk factor for bacteriuria because it gives biofilms time to develop and mature. This is why we engineer antibacterial and antibiofilm coating through ultrasound- and nanoparticle-assisted self-assembly on silicone surfaces and validate it thoroughly in vitro and in vivo. To this end, we combine bimetallic silver/gold nanoparticles, which exercise both biocidal and structural roles, with dopamine-modified gelatin in a facile and substrate-independent sonochemical coating process. The latter mussel-inspired bioadhesive potentiates the activity and durability of the coating while attenuating the intrinsic toxicity of silver. As a result, our approach effectively reduces biofilm formation in a hydrodynamic model of the human bladder and prevents bacteriuria in catheterized rabbits during a week of placement, outperforming conventional silicone catheters. Abstract

The use of engineered biomimetic substrates combined with neural stem cells (NSCs) constitutes a promising approach to develop reliable in vitro models of the nervous tissue. The fabrication of scaffolds with suitable compositional, topographical, and electrical properties is crucial for directing neural cell adhesion, differentiation, and arrangement. Herein, we propose the development of electroconductive polycaprolactone/polyaniline (PCL/PANI) electrospun mats as functional substrates for NSC culture. A rotating drum collector was employed to obtain fibers with aligned geometry. According to the results, the increase in alignment contributed to the reduction of fiber diameter and the increase of scaffold mechanical properties in terms of elastic modulus and tensile strength. In vitro experiments demonstrated the ability of PCL/PANI membranes to support NSC attachment and growth, as well as significantly foster neuronal differentiation. Furthermore, the presence of an aligned pattern was shown to effectively influence the arrangement of NSCderived neurons, confirming the potential of this substrate for the design of a physiologically relevant culture platform for in vitro investigation of the nervous tissue. Abstract

Chronic wounds (CWs) are a growing issue for the health care system. Their treatment requires a synergic approach to reduce both inflammation and the bacterial burden. In this work, a promising system for treating CWs was developed, comprising cobalt-lignin nanoparticles (NPs) embedded in a supramolecular (SM) hydrogel. First, NPs were obtained through cobalt reduction with phenolated lignin, and their antibacterial properties were tested against both Gram-negative and Gram-positive strains. The anti-inflammatory capacity of the NPs was proven through their ability to inhibit myeloperoxidase (MPO) and matrix metalloproteases (MMPs), which are enzymes involved in the inflammatory process and wound chronicity. Then, the NPs were loaded in an SM hydrogel based on a blend of a-cyclodextrin and custom-made poly(ether urethane)s. The nano-enabled hydrogel showed injectability, self-healing properties, and linear release of the loaded cargo. Moreover, the SM hydrogel’s characteristics were optimized to absorb proteins when in contact with liquid, suggesting its capacity to uptake harmful enzymes from the wound exudate. Abstract

Nowadays, human skin constructs (HSCs) are required for biomaterials, pharmaceuticals and cosmetics in vitro testing and for the development of complex skin wound therapeutics. In vitro three-dimensional (3D) dermal-epidermal based interfollicular, full-thickness, human skin equivalent (HSE) was here developed, recapitulating skin morphogenesis, epidermal differentiation, ultra-structure, tissue architecture, and barrier function properties of human skin. Different 3D cell culture conditions were tested to optimize HSE maturation, using various commercially available serum/animal component-free and/or fully defined media, and air-liquid interface (ALI) culture. Optimized culture conditions allowed the production of HSE by culturing normal human dermal fibroblasts (NHDFs) for 5–7 days in CELLnTEC-Prime Fibroblast (CnT-PR-F) medium and then culturing normal human epidermal keratinocytes (NHEKs) for 3 days in CELLnTEC-Prime Epithelial culture (CnT-PR) medium on them. Co-culture was then submerged overnight in CELLnTEC-Prime-3D barrier (CnT-PR-3D) medium to stimulate cell-cell contact formation and finally placed at ALI for 15–20 days using CnT-PR-3D medium. Histological analysis revealed uniform distribution of NHDFs in the dermal layer and their typical elongated morphology with filopodia. Epidermal compartment showed a multi-layered structure, consisting of stratum basale, spinosum, granulosum, and corneum. NHDFs and keratinocytes of basal layer were positive for the proliferation marker Kiel 67 (Ki-67) demonstrating their active state of proliferation. The presence of typical epidermal tissue proteins (keratins, laminins, filaggrin, loricin, involucrin, and β-tubulin) at their correct anatomical position was verified by immunohistochemistry (IHC). Moreover, transmission electron microscopy (TEM) analyses revealed basement membrane with lamina lucida, lamina densa, hemidesmosomes and anchoring fibers. The epidermal layers showed abundant intracellular keratin filaments, desmosomes, and tight junction between keratinocytes. Scanning electron microscopy (SEM) analyses showed the interwoven network of collagen fibers with embedded NHDFs and adjacent stratified epidermis up to the stratum corneum similar to native human skin. HSE physiological static contact angle confirmed the barrier function. The developed HSE represents a fundamental in vitro tool to assess biocompatibility of biomaterials, pharmacotoxicity, safety and effectiveness of cosmetics, as well as to investigate skin biology, skin disease pathogenesis, wound healing, and skin infection.

43. White Paper on MPS

Abstract

Microphysiological systems (MPS) are technological platforms that recapitulate functional units of human organs in vitro. There is no general unanimity in the definition of MPS and several platforms have been described as MPS technologies. Among these, are spheroids, static co-cultures, static micro patterned technologies, organ on chips (OOC), multi-organs on chips and human on chips (HOC). In this white paper we define MPS as advanced systems over the classic bi-dimensional (2D) cultures by including some of the following aspects: a three-dimensional (3D) framework (commonly referred to as scaffold) based on biomaterials of natural or synthetic origin; a 3D structure; a microfluidic counterpart; sensors to monitor MPS behaviour; an appropriate apparatus for MPS stimulation (e.g., electrical, mechanical cues); primary or stem cell-derived cells. MPS permit the study of human physiology in an organ-specific context, and they can find application in several fields. For instance, MPS can 1) help to unravel phenomena involved in the regulation of disease onset and progression, 2) support the drug development pipeline, and 3) contribute to chemical risk assessment. MPS may overcome the limits associated with traditional animal models, that are often not enough predictive of the human reality. Moreover, MPS present clear advantages in terms of cost, time, and ethical issues if compared with in vivo models. MPS demonstrated to be superior also to traditional 2D static cell culture models, that are not able to accurately reconstitute the in vivo cellular micro environment, tissue micro-architecture, and physiological functionality. The final goal is to develop systems, which combine 3D tissue models and flow-mimetic conditions, to recapitulate in vitro the complexity of human patho-physiology, thus effectively reproducing human clinical responses to drugs, and other exogenous stimuli.

44. Collagen-derived biomaterials in tissue engineering and organ modelling

Abstract

Tissue Engineering (TE) reconstructs damaged tissues/organs exploiting biocompatible and biodegradable scaffolds, taking often advantage of natural materials' bioactive/biomimetic cues together with advanced 3D processing techniques. In this scenario, collagen-derived materials (e.g. gelatin) have played a pivotal role. This paper will review past and recent outcomes of my research in the field, focusing on how collagen-based materials have been transferred from TE to the realization of 3D tissue/organ models supporting disease monitoring and drug design in an increasingly sustainable and ethical approach. Pioneering experiments have shown the potential of enzymatic/genipin crosslinking to realize new biomaterials. Engineered fillers of porous hollow tubes have also been designed to guide peripheral nerve regeneration. A procedure for preparing bone-derived biomimetic soluble collagen was also developed “in-house”. More recently, collagen coatings with custom polyurethanes supported the modelling of healthy and osteoporotic bone. Collagen is, moreover, a key component to design 3D skin equivalents for assessing biological safety. Gelatin is also a key ingredient as bioink for 3D-bioprinting. For instance, gelatin/nano-hydroxyapatite bioinks have permitted the realization of bone models. Cellularized gelatin bioinks are also used to colonize polyurethane scaffolds mimicking the cardiac tissue. Electrospinning of gelatin and collagen membranes has been implemented, successfully, in the design of lung and pancreatic tissue replicas in disease modelling.

45. Design of 3D bioengineered cardiac tissue models for the evaluation of chemical cardiotoxicity

Abstract

Humans are continuously exposed to a huge amount and a variety of chemicals. Animal tests are the gold standard for toxicity testing. However, they often fail in finely replicating the real physio-pathological scenario and their use is associated with ethical issues. 3D in vitro tissue models more efficiently mimic the native human environment and bring clear ethical advantages.1 In order to design a 3D bioengineered tissue model, the 3D matrix used to guide cell behavior, extracellular matrix (ECM) production and new tissue formation should replicate the architecture and composition of the native tissue. In this context, the proper selection of the biomaterial, the fabrication method and the functionalization protocol plays a pivotal role. In cardiac tissue engineering (TE), elastomeric polymers are required as constituents of porous structs replicating in vitro the myocardium architecture and mechanical properties. Moreover, surface functionalization with cardiac ECM proteins replicates in vitro the biochemical cues present in the native tissue. In this work the versatility of poly(urethane) (PU) chemistry was exploited to design a plethora of polymers with a wide range of physico-chemical properties. The most promising material for cardiac TE was then microfabricated through melt extrusion additive manufacturing (AM). Scaffolds were surface functionalized with cardiac ECM proteins (e.g., laminin, LN) and seeded with cardiac progenitor cells (CPCs) to establish cardiac tissue models.
