4. Effects of a Dietary Multi-Mineral Bolus on Udder Health in Dairy Cows: A Clinical Assessment.

Abstract

Simple Summary: Nutritional status has a pivotal role in animal health and production. Unfortunately, the animal's trace mineral status is often overlooked until an animal's performance falls below expectation or illness is detected; therefore, data exploring the effects of supplementation on udder health are still incomplete. Hence, this study aimed to assess the impact of trace elements—copper, iodine, cobalt, and selenium—delivered via a slow-release bolus on dairy cow udder health. The administered product might have contributed to promoting a reduction in the overall number of mastitis events, limiting the duration of the disease process, leading to a decrease in overall severity, and an increase in total milk production. Considering the observed effects, its use in an integrated approach that includes correct clinical strategies for mastitis prevention management might be supposed. Further studies involving a larger sample size, as well as considering the metabolic and trace element status of enrolled animals, should be performed to confirm the hypotheses. The clinical effects on the udder health of several trace elements—copper, iodine, cobalt, and selenium—contained in an intraruminal slow-release bolus were explored for the first time. Fifty-four dairy cows received the bolus (treated group, TG), while fifty-three were left untreated (control group, CG). Monthly composite milk samples were collected from 30 to 300 days in milk to measure somatic cell count (SCC); milk production was also recorded on the same days. Cows with SCC > 200 × 103 cells/mL were considered as affected by mastitis (with or without clinical signs). The effects on udder health were evaluated using several clinical indices employed for mastitis monitoring. The TG cows had a higher average daily milk yield than CG (p < 0.001), as well as a lower overall daily average of SCC (p < 0.0001). Fewer overall mastitis cases were detected in TG than in CG (p < 0.0001), although no significant differences were observed in new or cured mastitis cases. Finally, fewer failures of existing mastitis to cure cases were detected in TG as compared to CG (p < 0.0001), as well as fewer chronic mastitis cases (p < 0.0001). By looking at the clinical findings, some potential benefits on udder health might be hypothesized; nevertheless, additional studies are necessary to confirm these encouraging results. Abstract

The expression of several key molecules is altered in parathyroid tumors due to gene mutations, the loss of heterozygosity, and aberrant gene promoter methylation. A set of genes involved in parathyroid tumorigenesis has been investigated in sporadic parathyroid adenomas (PAds). Thirty-two fresh PAd tissue samples surgically removed from patients with primary hyperparathyroidism (PHPT) were collected and profiled for gene, microRNA, and lncRNA expression (n = 27). Based on a gene set including MEN1, CDC73, GCM2, CASR, VDR, CCND1, and CDKN1B, the transcriptomic profiles were analyzed using a cluster analysis. The expression levels of CDC73 and CDKN1B were the main drivers for clusterization. The samples were separated into two main clusters, C1 and C2, with the latter including two subgroups of five PAds (C2A) and nineteen PAds (C2B), both differing from C1 in terms of their lower expression of CDC73 and CDKN1B. The C2A PAd profile was also associated with the loss of TP73, an increased expression of HAR1B, HOXA-AS2, and HOXA-AS3 lncRNAs, and a trend towards more severe PHPT compared to C1 and C2B PAds. C2B PAds were characterized by a general downregulated gene expression. Moreover, CCND1 levels were also reduced as well as the expression of the lncRNAs NEAT1 and VLDLR-AS1. Of note, the deregulated lncRNAs are predicted to interact with the histones H3K4 and H3K27. Patients harboring C2B PAds had lower ionized and total serum calcium levels, lower PTH levels, and smaller tumor sizes than patients harboring C2A PAds. In conclusion, PAds display heterogeneous transcriptomic profiles which may contribute to the modulation of clinical and biochemical features. The general downregulated gene expression, characterizing a subgroup of PAds, suggests the tumor cells behave as quiescent resting cells, while the severity of PHPT may be associated with the loss of p73 and the lncRNA-mediated deregulation of histones. Abstract

The study aims to verify the usefulness of new intervals-based algorithms for clinical interpretation of animal behavior in dairy cows around calving period. Thirteen activities associated with feeding-ruminating-locomotion-behaviors of 42 adult Holstein-Friesian cows were continuously monitored for the week (wk) -2, wk -1 and wk +1 relative to calving (overall 30'340 min/animal). Soon after, animals were retrospectively assigned to group-S (at least one spontaneous diseases; n = 24) and group-H (healthy; n = 18). The average activities performed by the groups, recorded by RumiWatch�� halter and pedometer, were compared at the different weekly intervals. The average activities on the day of clinical diagnosis (dd0), as well as one (dd-1) and two days before (dd-2) were also assessed. Differences of dd0 vs. dd-1 (��D1), dd0 vs. wk -1 (��D2), and wk +1 vs. wk -1 (��weeks) were calculated. Variables showing significant differences between the groups were used for a univariate logistic regression, a receiver operating characteristic analysis, and a multivariate logistic regression model. At wk +1 and dd0, eating- and ruminating-time, eating- and ruminate-chews and ruminating boluses were significantly lower in group-S as compared to group-H, while other activity time was higher. For ��D2 and ��weeks, the differences of eating- and ruminating-time, as well as of eating-and ruminate-chews were significantly lower in group-S as compared to group-H. Concerning the locomotion behaviors, the lying time was significantly higher in group-S vs. group-H at wk +1 and dd-2. The number of strides was significantly lower in group-S compared to group-H at wk +1. The model including eating-chews, ruminate-chews and other activity time reached the highest accuracy in detecting sick cows in wk +1 (area under the curve: 81%; sensitivity: 73.7%; specificity: 82.4%). Some of the new algorithms for the clinical interpretation of cow behaviour as described in this study may contribute to monitoring animals' health around calving.

Abstract

The aims of the investigation were to establish for the first time (i) clinical efficacy and (ii) pharmacokinetic profile of meloxicam intravenously (IV) administered in male Mediterranean buffalo calves after surgical orchiectomy. The study was performed on 10 healthy buffalo calves, between 4 and 5 months old and between 127 and 135 kg of body weight (b.w.). An IV injection of 0.5 mg/kg b.w. of meloxicam was administered in six calves (treated group, TG) immediately after surgery; the other four animals were used as untreated control group (CG). The clinical efficacy of meloxicam was evaluated pre- and post-surgery by monitoring respiratory rate (RR), heart rate (HR), rectal temperature (T°C), serum cortisol levels (SCL) and pain score (PS).
