Abstract

Brachypodium distachyon was proposed as new model plant for grasses because of its small genome and its phylogenetic position between rice and Triticeae crops. Since, a range of genetic and genomic resources has been developed and the whole genome sequence is now available. We use Brachypodium as a model system for studying how to modify grass crops for increased biofuel production made from lignocellulosic cell walls. We are involved in developing many resources for Brachypodium: TILLING collection, natural accessions, different methods of phenotyping, and tools and method for gene transfer via Agrobacterium tumefaciens. Some lignin mutants from the monolignol biosynthesis pathway were already identified by phenotyping and TILLING, and their functional analysis is underway notably through complementation by overexpression of concerned gene in mutant background.

Abstract

Procedures are now available for the exploitation of protoplast technology for the genetic manipu- lation of lettuce and for studies of lettuce/virus interactions. These techniques have made it possible to overcome interspecific barriers by protoplast fusion. Following the procedures we developed, we were able to obtain hybrids between Lactuca sativa and two wild Lactuca species (L. tatarica and L. perennis). Moreover, protoplast technology can overcome some of the limitations in the application of mutation techniques in crops. In this paper, we demonstrated the feasibility and the power of our in vitro techni- ques, in combination with induced mutations, for creating new variability in lettuce. Furthermore, we adapted protocols that can provide the opportunity to start studies on mechanisms associated with the different LMV-resistance genes at the cellular level. It is presently possible to obtain protoplasts from different Lactuca species containing different resistance genes. A protocol for specific staining by immu- nofluorescence of infected lettuce protoplasts is described and preliminary electroporation experiments in some Lactuca protoplasts were performed.

Abstract

Lettuce mosaic potyvirus (LMV) has been a destructive virus on lettuce crops and the emergence of new strains requires to find out new resistance genes. The coat protein mediated protection approach was shown previously to confer some protection to LMV. Additional transformed lettuce lines were analysed for resistance against LMV-0. The R2 homozygous progenies of four R1 resistant Jessy plants showed a protection against LMV similar to the one obtained in the R2 Cocarde line (Cocarde 9a-91) initially described. Only few plants showed a complete resistance and a large majority of R2 plants of each progeny showed a recovery phenotype. This LMV protection phenotype was transmitted after cross with untransformed lettuce. A new screening of transgenic lettuce plants was performed on R1 plants and did not allow the characterization of highly resistant plants. Therefore, the protection conferred by the LMV-0 coat protein seems to be triggered only after a period 5 to 7 weeks after inoculation, period during which plants can not develop any resistance to virus infection.

Abstract

Protoplasts were isolated from callus, cotyledons, hypocotyls and leaves of cotton (Gossypium hirsutum L. var. Coker 310) using different digestion media. Protoplast yield per gram of fresh mass of leaves, callus cotyledons and hypocotyls was 22.2, 5.5, 1.7 and 1. 1 million respectively. Protoplasts were most viable in basic medium in presence of thidiazuro 0.663 gm/1 and 2,4,5-trichlorophenoxy acetic acid 7 mg/1. Hypocotyl and leaf-derived protoplasts formed microcolonies. Addition of CaC12 and glutamine to the basic medium improved protoplast survival, and division of hypocotyl and leaf-derived protoplasts. The growth conditions, such as the light and carbon source of donor plant were the important factors for protoplast isolation and culture.

Abstract

The effects of Mepiquat chloride (Pix) on the performances of 58 cotton plots were studied during 1994 and 1995 at Chaibadan district, Lop Buri Province. Pix application at early blooming reduced significantly vegetative growth (node production, inter-node length) during the reproductive period and shortened the crop duration. Over all plots the average seed-cotton yield was not improved by Pix despite significant changes in boll distribution pattern and boll-size. Large variation in yield response existed between plots. The response depended on the rate of increase in internode length prior to Pix application together with fruit retention of the first fruiting branches.

In a search for model systems in plant cell genetics studies mesophyll protoplasts from eleven species of Nicotiana with low chromosome number (N. acuminata, N. alata, N. glauca, N. glutinosa, N. Iangsdorffii, N. longiflora, N. otophora, N. paniculata, N. plumbaginifolia, N. suaveolens, N. sylvestris) were shown to divide in a liquid culture medium. Plants were recovered from calli originating from protoplasts of all these species except N. glutinosa. [ABSTRACT FROM AUTHOR]
