A synthetic strategy based on reactions of cyclic imine oxides, namely 2H-imidazole 1-oxides, with thiophenols mediated by acetyl chloride was successfully applied as a convenient tool to obtain a series of novel azaheterocyclic molecules, including water-soluble hydrochloride forms. Optimized reaction conditions found herein for the nucleophilic substitution of hydrogen (SNH) in non-aromatic azaheterocyclic substrates via the “addition-elimination” (SNH AE) scheme enabled 15 arylthiolated 2H-imidazoles to be prepared in yields of up to 90%. The developed methodology discloses an original synthetic way to obtain numerous azaheterocyclic molecules, which are of interest in the field of medicinal chemistry and materials science.

We investigated the inhibitory activities of novel 9-phosphoryl-9,10-dihydroacridines and 9-phosphorylacridines against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and carboxylesterase (CES). We also studied the abilities of the new compounds to interfere with the self-aggregation of β-amyloid (Aβ42) in the thioflavin test as well as their antioxidant activities in the ABTS and FRAP assays. We used molecular docking, molecular dynamics simulations, and quantum-chemical calculations to explain experimental results. All new compounds weakly inhibited AChE and off-target CES. Dihydroacridines with aryl substituents in the phosphoryl moiety inhibited BChE; the most active were the dibenzyloxy derivative 1d and its diphenethyl bioisostere 1e (IC50 = 2.90 ± 0.23 µM and 3.22 ± 0.25 µM, respectively). Only one acridine, 2d, an analog of dihydroacridine, 1d, was an effective BChE inhibitor (IC50 = 6.90 ± 0.55 μM), consistent with docking results. Dihydroacridines inhibited Aβ42 self-aggregation; 1d and 1e were the most active (58.9% ± 4.7% and 46.9% ± 4.2%, respectively). All dihydroacridines 1 demonstrated high ABTS•+-scavenging and iron-reducing activities comparable to Trolox, but acridines 2 were almost inactive. Observed features were well explained by quantum-chemical calculations. ADMET parameters calculated for all compounds predicted favorable intestinal absorption, good blood–brain barrier permeability, and low cardiac toxicity. Overall, the best results were obtained for two dihydroacridine derivatives 1d and 1e with dibenzyloxy and diphenethyl substituents in the phosphoryl moiety. These compounds displayed high inhibition of BChE activity and Aβ42 self-aggregation, high antioxidant activity, and favorable predicted ADMET profiles. Therefore, we consider 1d and 1e as lead compounds for further in-depth studies as potential anti-AD preparations.

This study explores the problem of describing viscous fluid motion for Navier–Stokes equations in curved channels, which is important in applications like hemodynamics and pipeline transport. Channel curvature leads to vortex flows and closed vortex zones. Asymptotic models of the flux problem are useful for describing viscous fluid motion in long pipes, thus considering geometric parameters like pipe diameter and characteristic length. This study provides a representation for the vorticity vector and energy dissipation in the flow problem for a curved channel, thereby determining the magnitude of vorticity and energy dissipation depending on the channel’s central line curvature and torsion. The accuracy of the asymptotic formulas are estimated in terms of small parameter powers. Numerical calculations for helical tubes demonstrate the effectiveness of the asymptotic formulas.

Forests being mostly non-renewable, miscanthus seems to be a promising alternative energy resource. This study investigates the enzymatic hydrolysis of pretreated miscanthus biomass. The research featured Miscanthus sinensis Strictus biomass. The methods included raw material pretreatment, enzymatic hydrolysis, matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF) of monosaccharide residues, and nuclear magnetic resonance (1H NMR) spectrometry of non-hydrolyzed lignin. A hammer mill with a particle size of 0.2–0.3 mm proved to be effective for preliminary mechanical processing of miscanthus. This type of treatment provided the maximal yield of carbohydrate-containing biomass (62.33 ± 1.87%). The optimal multi-enzyme composition included Cellulase Ultra obtained from Trichoderma reeseii, xylanase obtained from Thermomyces lanuginosus, and β-gluconase obtained from Myceliophtorafergusii. The fermentation temperature was 50 ± 1°C (72 ± 1 h). At these parameters, the conversion of holocellulose of miscanthus biomass was 96.0 ± 4.8%, and the yield of reducing substances was 97.00 ± 4.85%. The article introduces the optimal enzymatic hydrolysis parameters for pretreating Miscanthus sinensis Strictus biomass as a source of carbohydrate-containing substrates and describes their subsequent use for bacterial cellulose biosynthesis. The carbohydrate composition of hydrolysates included residual lignin, such polysaccharides as glucan and xylan, and various monosaccharides, namely arabinose, xylose, galactose, glucose, and uronic acids. Hydrolyzed miscanthus proved to be an effective renewable and environmentally friendly biodiesel.

Herein, we report a convenient one-pot synthesis of 1,2,4-triazinyl derivatives of benzocoumarins. The proposed approach consists in the nucleophilic addition of tetrahydrobenzo annulated dimethoxycoumarin to 1,2,4-triazines followed by double oxidation of both dihydrotriazine and tetrahydrobenzo groups with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ). The nucleophilic addition of the dimethoxycoumarin to 1,2,4-triazines was carried out in the presence of three-fold excess of methanesulfonic acid in DCM at room temperature and take place between positions 4 and 5 of coumarin and 1,2,4-triazine, respectively. The double oxidation step was achieved with 3.6 equivalent of DDQ. Selective oxidation of dihydrotriazine moiety, without affecting the tetrahydrobenzo fragment, was achieved using 1.2 equivalent of tetrachlorobenzoquinone (TCQ). The differences in the oxidation with TCQ and DDQ appear to be related to the higher oxidative potential of DDQ in contrast to TCQ. The advantages of the method are the elimination of the use of transition metals, the availability of starting materials, and the simplicity of the procedure. The proposed approach provides a two-step one-pot protocol for the synthesis of triazinyl benzocoumarins, precursors for the preparation of push-pull pyridinyl chromophore.
