Abstract The endoplasmic reticulum (ER) is a key organelle in eukaryotic cells, responsible for a wide range of vital functions, including the modification, folding, and trafficking of proteins, as well as the biosynthesis of lipids and the maintenance of intracellular calcium homeostasis. A variety of factors can disrupt the function of the ER, leading to the aggregation of unfolded and misfolded proteins within its confines and the induction of ER stress. A conserved cascade of signaling events known as the unfolded protein response (UPR) has evolved to relieve the burden within the ER and restore ER homeostasis. However, these processes can culminate in cell death while ER stress is sustained over an extended period and at elevated levels. This review summarizes the potential role of ER stress and the UPR in determining cell fate and function in various diseases, including cardiovascular diseases, neurodegenerative diseases, metabolic diseases, autoimmune diseases, fibrotic diseases, viral infections, and cancer. It also puts forward that the manipulation of this intricate signaling pathway may represent a novel target for drug discovery and innovative therapeutic strategies in the context of human diseases.

We conducted a study on the surface compound modification of shot peening and pure carbon DLC coating to simultaneously meet the requirements of wear resistance and fatigue resistance of spline structure. The effects of surface compound modification were investigated on the surface morphology, residual stress profile, microstructure, and nano-indentation hardness of 16Cr3NiWMovNbE gear steel, and conducted a comparative study on fatigue performance. The results show that the surface compound modification inherits the surface morphology and compressive residual stress gradient of shot peening, while the surface residual stress is slightly smaller than that of shot peening. In addition, surface compound modification still reflects the characteristics of high hardness and high fracture resistance of DLC coatings. Under the bending load based on spline tooth root, compared to the original specimen, the fatigue life after shot peening, pure carbon DLC coating, and surface compound modification is increased by 3.68, 2.35, and 3.36 respectively. Although the compound modified surface still maintains the shot peening morphology with a increasing surface roughness and stress concentration coefficient, the 100 μm-depth compressive residual stress profile and the subgrain refinement layer introduced, as well as the hard surface layer with good load-bearing capacity, have played the role of fatigue strengthening.

In the process of infecting plants, plant parasitic nematodes release a series of proteins that play an essential role in the successful infection and pathogenesis of plant cells and tissues through stylets or body walls. In this study, based on transcriptome data, a chorismate mutase gene of Radopholus similis (RsCM) was identified and cloned, which is a single copy gene specifically expressed in the oesophageal gland and highly expressed in juveniles and females. Transient expression of RsCM in tobacco leaves showed that it was localised in the cytoplasm and nucleus of tobacco leaf cells, which inhibited the pattern-triggered immunity (PTI) induced by flg22, including callose deposition and defence gene expression, and cell death induced by immune elicitors BAX, but could not inhibit cell death induced by immune elicitors Gpa2/RBP-1. The RNA interference (RNAi) transgenic tomato of RsCM obviously inhibited the infection, pathogenicity, and reproduction of R. similis. However, the resistance of the overexpression transgenic tomato of RsCM to R. similis infection was significantly reduced, and the expression levels of two salicylic acid (SA) pathway genes (PR1 and PR5) in roots infected by the nematode were significantly down-regulated, which indicated that RsCM might be involved in the inhibition of SA pathway. The results of this study demonstrate that RsCM suppresses the host immune system and might be a new target for the control of R. similis, which also provides new data for the function and mechanism of CM genes of migratory parasitic plant nematodes.

Abstract Due to the highly conserved structure, animal mitochondrial genome (mtDNA) is widely used in classification, evolution, phylogeny, population genetic structure and other fields. We reported on the five circle multipartite mtDNAs of a newly described species of Globodera, Globodera vulgaris (Gv) from potatoes in China. The results showed that the mtDNA of Gv was obtained through second- and third-generation sequencing, with a total length of 42,995 bp. It contained 12 protein-coding genes, two rRNA genes and 17 tRNA genes, which were distributed in different subgenomic circles. Comparison of the differences in mtDNA among Gv, G. rostochiensis, G. pallida and G. ellingtonae showed that the size and arrangement of the genes in the mtDNA of the genus Globodera were variable and not conserved. The codon usage bias of the mitochondrial protein-coding gene of Gv showed that Gv might have originated from locally and more primitive group of existing Globodera. Based on the cytochrome c oxidase subunits I genes (COX1) and the nicotinamide adenine dinucleotide dehydrogenase subunits I genes (ND1), and the results showed that Gv was clustered with Globodera spp. according to the COX1 and ND1 in scmtDNA-V, while Gv was clustered with Meloidogyne spp. according to ND1 in scmtDNA-III. The results of this study provided a new basis for understanding the multipartite structure of mtDNA as a phylogenetic and taxonomic feature of the genus Globodera. The number of subgenomic circles is a diagnostic feature of species and the arrangement order and size of mitochondrial protein-coding genes also have important application value in species identification within the genus.

Tripartite interactions play a fundamental role in the quantum information processing and quantum technology. However, it is generally difficult to realize strong tripartite coupling. We investigate the estimation of a tripartite coupling strength in a hybrid setup composed of a single nitrogen-vacancy (NV) center and a micromagnet. A time-independent parametric drive can be utilized to increase the estimation precision of the tripartite coupling strength. By calculating the quantum Fisher information (QFI), we can obtain the optimal estimation precision by measuring the eigenstate of the tripartite system. At the critical position, the QFI is divergent due to that the preparation time of the eigenstate is divergent. When the system is subjected to a dissipation, the QFI near the critical point of the driven-dissipation phase transition is analytically obtained. The direct intensity measurement is the optimal measurement near the dissipation phase transition point. In addition, we quantify the robustness of an imperfect measurement operator by the measurement noise susceptibility based on the error propagation formula. We find that the direct intensity measurement is robust enough against small measurement disturbance from a coherent drive. But it can be disturbed by the nonlinear anti-harmonic measurement noise, especially near the critical point.

Abstract Objective The percentage of adolescents with test anxiety is increasing rapidly. Working memory (WM) training has been demonstrated to reduce anxiety levels and enhance attentional control in individuals. Therefore, we investigated whether adaptive dual n-back WM training could lower test anxiety level and improve attentional control in adolescents. Methods Forty adolescents were allocated to either adaptive dual n-back WM training (n = 21) or non-adaptive dual 1-back WM training (n = 19) for 10 days. The Test Anxiety Scale was applied to measure individuals’ test anxiety symptoms. The Attentional Control Scale (ACS), the flanker task, and the Go/Nogo task were used to measure attentional control. Results Compared with the control group, the training group reported significantly relief of test anxiety symptoms; however, there were no significant differences between the two groups in pre-to-post changes in ACS scores or performance on the flanker task and Go/Nogo task. Conclusion In sum, adaptive dual n-back WM training effectively reduced adolescents’ level of test anxiety but did not improve their attentional control.

Abstract Helicotylenchus comprises a significant group of plant-parasitic nematodes that primarily feed on plant roots. This study focuses on the description of a newly discovered species, Helicotylenchus zenchengensis n. sp., from China. The distinguishing characteristic of this species, as observed under a microscope, is the absence of head annulation. The stylet, measuring between 23.8 and 29.3 μm, exhibits anteriorly flattened or indented stylet knobs. The elongated tail has a c value ranging from 15.8 to 22.6 and a c’ value between 1.9 and 2.4. It bears 15 to 18 tail annuli, presenting a conical shape with projections. The phasmids are located at the 4th to 10th body annulations posterior to the anus, while no males were observed. Three phylogenetic trees were constructed to determine the evolutionary relationships using the 18S rRNA, ITS, and 28S D2-D3 expansion segments of the newly discovered species and related Helicotylenchus sequences obtained from the database. Based on the ITS sequences, a phylogenetic tree revealed that H. zenchengensis n. sp. clustered with H. cuspicaudatus. However, it was distinguished from H. cuspicaudatus when examining the 18S and 28S rRNA sequences and morphological characteristics. Additionally, comprehensive and diagnostic tables for all known species of Helicotylenchus in China are provided for reference.

Improving the measurement precision of low temperature is significant in fundamental science and advanced quantum technology application. However, the measurement precision of temperature T usually diverges as T tends to zero. Here, by utilizing a heavy impurity to measure the temperature of a Bose gas, we obtain the Landau bound to precision δ^{2}T∝T^{2} to avoid the divergence. When the initial momentum of the heavy impurity is close to be fixed and nonzero, the measurement precision can be δ^{2}T∝T^{3} to break the Landau bound. We derive the momentum distribution of the heavy impurity at any moment and obtain the optimal measurement precision of the temperature by calculating the Fisher information. As a result, we find that increasing the expectation value P_{0} and reducing the variance Δ/2 of the initial momentum can help to improve the measurement precision. Moreover, under certain conditions, sqrt[Δ]/P_{0} is a relevant parameter, the smallness of which helps improve the thermometric precision of the probe. In addition, the momentum measurement is the optimal measurement of the temperature in the case that the initial momentum is close to be fixed and not equal to zero. The kinetic energy measurement is the optimal measurement in the case that the expectation value of the initial momentum is close to zero. Finally, we obtain that the temperatures of two Bose gases can be measured simultaneously. The simultaneous measurement precision is proportional to T^{2} when two temperatures are close to T.

PURPOSERadiomics can be used to determine the prognosis of gastric cancer (GC). The objective of this study was to predict the disease-free survival (DFS) after GC surgery based on computed tomography-enhanced images combined with clinical features.METHODSClinical, imaging, and pathological data of patients who underwent gastric adenocarcinoma resection from June 2015 to May 2019 were retrospectively analyzed. The primary outcome was DFS. Radiomics features were selected using Least Absolute Shrinkage and Selection Operator algorithm and converted into the Rad-score. A nomogram was constructed based on the Radscore and other clinical factors. The Rad-score and nomogram were validated in the training and validation groups.RESULTSTotally, 179 patients were randomly divided into the training (n=124) and validation (n=55) groups. In the training group, validation group, and overall population, the Rad-score could be divided into categories indicating low, moderate, and high risk of recurrence, metastasis, or death; all risk categories showed a significant difference between the training, validation, and overall population groups (all P < .001). Positive lymph nodes (hazard ratio (HR)=3.07, 95% CI: 1.52-6.23, P=.002), cancer antigen-125 (HR=3.24, 95% CI: 1.54-6.80, P=.002), and the Radscore (HR=0.73, 95% CI: 0.61-0.87, P < .001) were independently associated with DFS. These 3 variables were used to construct a nomogram. In the training group, the areas under the curve at 3 years were 0.758 and 0.776 for the Rad-score and the nomogram, respectively, while they were both 1.000 in the validation group. The net benefit rate was analyzed using a decision curve in the training and validation groups, and the nomogram was superior to the single Rad-score.CONCLUSIONRad-score is an independent factor for DFS after gastrectomy for GC. The nomogram established in this study could be an effective tool for the clinical prediction of DFS after gastrectomy.

The quantum version of a special classical Wheatstone bridge built with a boundary-driven spin system has recently been proposed. We propose a quantum Wheatstone bridge consisting of Bose systems, which can simulate the general classical Wheatstone bridge. Unknown coupling can be obtained when the quantum Wheatstone bridge is balanced, which can be determined simply by homodyne detection. When the expectation value of the homodyne detection is 0, the quantum Wheatstone bridge is unbalanced. When the expectation value of the homodyne detection is proportional to the square root of the initial number of bosons by regulating a known coupling strength, the quantum Wheatstone bridge is balanced. By calculating the quantum Fisher information, we show that the measurement precision is optimal when the quantum Wheatstone bridge is balanced. And the homodyne detection is close to the optimal measurement in the case of low-temperature baths.

Abstract Hypoxic pulmonary hypertension (PH) is a progressive disease characterized by hyper-proliferation of pulmonary vascular cells including pulmonary artery smooth muscle cells (PASMCs) and can lead to right heart failure and early death. Selective degradation of mitochondria by mitophagy during hypoxia regulates mitochondrial functions in many cells, however, it is not clear if mitophagy is involved in the pathogenesis of hypoxic PH. By employing the hypoxic mitophagy receptor Fundc1 knockout (KO) and transgenic (TG) mouse models, combined hypoxic PH models, the current study found that mitophagy is actively involved in hypoxic PH through regulating PASMC proliferation. In the pulmonary artery medium from hypoxic PH mice, mitophagy was upregulated, accompanied with the increased active form of FUNDC1 protein and the enhanced binding affinity of FUNDC1 with LC3B. In PASMCs, overexpression of FUNDC1 increased mitophagy and cell proliferation while knockdown of FUNDC1 inhibited hypoxia-induced mitophagy and PASMC proliferation. Stimulation of mitophagy by FUNDC1 in PASMCs elevated ROS production and inhibited ubiquitination of hypoxia inducible factor 1α (HIF1α), and inhibition of mitophagy by FUNDC1 knockdown or knockout abolished hypoxia-induced ROS-HIF1α upregulation. Moreover, Fundc1 TG mice developed severe hemodynamics changes and pulmonary vascular remodeling, and Fundc1 KO mice were much resistant to hypoxic PH. In addition, intraperitoneal injection of a specific FUNDC1 peptide inhibitor to block mitophagy ameliorated hypoxic PH. Our results reveal that during hypoxic PH, FUNDC1-mediated mitophagy is upregulated which activates ROS-HIF1α pathway and promotes PASMC proliferation, ultimately leads to pulmonary vascular remodeling and PH.

We mainly investigate the quantum measurement of Kerr nonlinearity in the driven-dissipative system. Without the dissipation, the measurement precision of the nonlinearity parameter χ scales as “super-Heisenberg scaling” 1/N2 with N being the total average number of particles (photons) due to the nonlinear generator. Here, we find that “super-Heisenberg scaling” 1/N3/2 can also be obtained by choosing a proper interrogation time. In the steady state, the “super-Heisenberg scaling” 1/N3/2 can only be achieved when the nonlinearity parameter is close to 0 in the case of the single-photon loss and the one-photon driving or the two-photon driving. The “super-Heisenberg scaling” disappears with the increase of the strength of the nonlinearity. When the system suffers from the two-photon loss in addition to the single-photon loss, the optimal measurement precision will not appear at the nonlinearity χ=0 in the case of the one-photon driving. Counterintuitively, in the case of the two-photon driving we find that it is not the case that the higher the two-photon loss, the lower the measurement precision. It means that the measurement precision of χ can be improved to some extent by increasing the two-photon loss.

Background: As there is a growing concern about the cerebral embolism events secondary to non-valvular atrial fibrillation (NVAF), novel oral anticoagulant (NOAC) has been more and more widely used as an anticoagulation treatment for the prevention of stroke. However, in the face of life-threatening bleeding or emergency surgery/treatment, NOAC-related antagonists such as idarucizumab need to be urgently used to reverse the NOAC. Using recombinant tissue plasminogen activator (rt-PA) intravenous thrombolysis for acute ischemic stroke requires a time window of 4.5 h. This case reports rt-PA intravenous thrombolysis after reversal of dabigatran anticoagulation with idarucizumab in patients with acute ischemic stroke.Case Presentation: We report the case of 62-year-old Chinese female with NVAF treated with dabigatran 110 mg twice daily, and missed a dose on the eve of the stroke. The patient presented with acute ischemic stroke causing the angle of mouth deviated to right side and left limb weakness in the early morning of the next day. However, the last dosing time of dabigatran was between 24 and 48 h, the patients were given rt-PA intravenous thrombolysis after reversal of dabigatran anticoagulation with idarucizumab, while any potential relative contraindication had been excluded by means of laboratory test and CT scan in the hospitalization services. National Institute of Health stroke scale (NIHSS) score was reduced from 4 to 1, and the patient was discharged after 2 weeks.Conclusion: Our case report adds to the evidence that idarucizumab administration is safe and effective in the setting of patients with atrial fibrillation treated with dabigatran who develop acute ischemic stroke requiring rt-PA intravenous thrombolysis.

The preparation of superhydrophobic (SH) surface on stainless steel by chemical etching method is challenging due to the good corrosion resistance of the material. In this work, SH surface with water contact angle (WCA) as high as 163.21° was accomplished on 304 stainless steel surface by a rapid ultrasonic-assisted chemical etching method within 7 min and a low-cost fluorine-free modification treatment. The mechanism of ultrasonic field on the etching process was explored by detecting the cavitation and oscillation energy in the reactor. It is the first time to found that the ultrasonic cavitation effect enhanced the etching process by both chemical and physical facilitation resulting in hierarchical lamellar micro-structures, “mountain-like” micro-structure clusters and “coral-reef-like” nano-scale structures on the surface. With the ultrasonic power increasing, the ultrasonic cavitation effect not only enhanced the superhydrophobicity of sample surface, but also improved the uniformity of surface wettability. The samples also showed excellent performance of oil/water separation for various organics (all separation efficiencies up to 96%) and remarkable mechanical stability.

Peptide transporter 2 (PepT2) in mammals plays essential roles in the reabsorption and conservation of peptide-bound amino acids in the kidney and in maintaining neuropeptide homeostasis in the brain. It is also of significant medical and pharmacological significance in the absorption and disposing of peptide-like drugs, including angiotensin-converting enzyme inhibitors, β-lactam antibiotics and antiviral prodrugs. Understanding the structure, function and regulation of PepT2 is of emerging interest in nutrition, medical and pharmacological research. In this review, we provide a comprehensive overview of the structure, substrate preferences and localization of PepT2 in mammals. As PepT2 is expressed in various organs, its function in the liver, kidney, brain, heart, lung and mammary gland has also been addressed. Finally, the regulatory factors that affect the expression and function of PepT2, such as transcriptional activation and posttranslational modification, are also discussed.

Transmission engineering components need to fulfill requirements for adequate wear resistance and fatigue resistance, which are related to their surface properties. In this paper, we combined shot peening and nitrogen ion implantation to improve the surface properties of 16Cr3NiWMoVNbE gear steel. The average surface roughness decreased slightly after the ion implantation because of the high-speed impact of implanted ions having the surface etching role. The maximum compressive residual stress of the near-surface layer after the surface treatment increased by more than 11.8–15.9% compared with shot peening. The nitrogen ions diffused through the peening deformation channel, and the deformation degree and the implantation temperature were positively correlated with the diffusion process. The surface nano-hardness obtained by ion implantation after shot peening was increased by 124.4% compared to the AR state.

We investigate the parameter estimation in a magnon–cavity–magnon coupled system. PT symmetrical two magnons system can be formed in the gain magnetic materials by the adiabatic elimination of the cavity field mode. We show that the optimal estimation will not appear at the exceptional point due to that the quantum fluctuations are the strongest at the exceptional point. Moreover, we demonstrate that the measurements at the exceptional point tend to be optimal with the increase of prepared time. And the direct photon detection is the optimal measurement for the initial state in the vacuum input state. For the open PT symmetrical two magnons system, the quantum fluctuations will greatly reduce the degree of entanglement. Finally, we show that a higher estimated magnetic sensitivity can be obtained by measuring the frequency of one magnon in the PT symmetrical two magnons system.

Listeria monocytogenes is a Gram-positive facultative intracellular bacterium that causes central nervous system infection. We report a case of rhombencephalitis caused by L. monocytogenes infection, which mimicked Bickerstaff’s brainstem encephalitis, and GQ1b antibody positivity and multiple intracranial foci were observed. A 68-year-old male patient presented with a nonspecific prodrome of faintness, forehead tightness, and walking instability. This was followed by progressive cranial nerve palsies, limb weakness, cerebellar signs, hyperpyrexia, and impaired consciousness. Brain imaging showed multiple abnormal brainstem and cerebellar signals that were accompanied by blood infiltration without any lesion enhancement. Serum GQ1b antibody positivity led to an initial diagnosis of Bickerstaff’s brainstem encephalitis, which was treated with immunosuppressive therapy with limited efficacy. A pathogen examination helped confirm L. monocytogenes infection. A combination of meropenem and trimethoprim-sulfamethoxazole therapy was applied and the patient recovered without sequelae. The symptoms and imaging of Listeria rhombencephalitis are nonspecific. Accurate diagnosis and prompt treatment of this condition are essential. Whether Listeria infection triggers an autoimmune response remains unclear.

Abstract Background Great auricular nerve schwannoma is extremely rare. Herein, we reported the first case of schwannoma arising from great auricular nerve trunk. Case presentation A 29 year-old female complained of a slowly-growing superfacial neck mass for 6 months. MRI revealed a high possibility of schwannoma. Although the patient underwent successfully surgical removal of the tumor, ipsilateral numbness of both auricle and peripheral skin developed due to traction of the nerve. Immunohistochemistry staining confirmed the diagnosis of schwannoma. And the patient has been followed regularly. Conclusion For superficial cervical tumors, the cervical plexus cutaneous nerve should be considered if MRI and other imaging findings suggest neurogenic tumors.

We investigate simultaneous estimation of multi-parameter with time-dependent Hamiltonians. We analytically obtain the maximum quantum Fisher information matrix for two-parameter encoded in time-dependent three-level systems. The optimal coherent control scheme is proposed to increase the simultaneous multi-parameter estimation precisions. In an example of a spin-1 particle in a uniformly rotating magnetic field, the optimal coherent Hamiltonians for different parameters can be chosen to be completely same. However, in general, the optimal coherent Hamiltonians for different parameters are incompatible. In this situation, we suggest a variance method to obtain the optimal coherent Hamiltonian for simultaneously estimating multiple parameters, and obtain the optimal simultaneous estimation precision of two-parameter in a three-level Landau-Zener Hamiltonian.

In recent years, a series of research experiments have been conducted on WiFi-based gesture recognition. However, current recognition systems are still facing the challenge of small samples and environmental dependence. To deal with the problem of performance degradation caused by these factors, we propose a WiFi-based gesture recognition system, WiGAN, which uses Generative Adversarial Network (GAN) to extract and generate gesture features. With GAN, WiGAN expands the data capacity to reduce time cost and increase sample diversity. The proposed system extracts and fuses multiple convolutional layer feature maps as gesture features before gesture recognition. After fusing features, Support Vector Machine (SVM) is exploited for human activity classification because of its accuracy and convenience. The key insight of WiGAN is to generate samples and merge multi-grained feature maps in our designed GAN, which not only enhances the data but also allows the neural network to select different grained features for gesture recognition. According to the result of experiments conducted on two existing datasets, the average recognition accuracy of WiGAN reaches 98% and 95.6%, respectively, outperforming the existing system. Moreover, the recognition accuracy under different experimental environments and different users shows the robustness of WiGAN.

Twenty populations of Radopholus similis from three countries and different hosts (19 populations from ornamental plants and one population from ginger) were compared using morphological characters, morphometrics and karyotype between progeny from both single females and 30 females of each population. Morphological diversity existed in and among the populations, even within the progeny nematodes from single nematodes compared to that of 30 females. The labial disc shape, the number of head annuli, the terminated position of lateral lips, the number of genital papillae before cloacal apertures and female and male tail terminal shape showed variation. In addition, genital papillae arranged in a double row before cloacal apertures was first found in two ornamental populations. The karyotype of all the 20 populations was n = 5. Combining our results and previous studies, we support that R. citrophilus is a synonym of R. similis, and that it is not possible to distinguish physiological races or pathotypes of R. similis according to morphological characters or karyotype.

High infection rates of roots of wild kiwifruit (Actinidia chinensis Planch) and soil infestation by a root-knot nematode were found in Anshun, GuiZhou Province, China. Morphology, esterase phenotype and molecular analyses confirmed that this nematode was different from previously described root-knot nematodes. In this report, the species is described, illustrated and named Meloidogyne aberrans sp. nov. The new species has a unique combination of characters. A prominent posterior protuberance, round and faint perineal pattern and a medium-length stylet (13.6-15.5 μm) characterized the females. Second-stage juveniles (J2) were characterized by a smooth lip region with distinctly protruded medial lips and a depression in outline at the oral aperture, a relatively long stylet (15.9-16.8 μm), four incisures in the lateral field and a very short, even poorly defined, hyaline tail terminus (2.2-5.5 μm). More incisures (11-15) existed in the lateral field of males, and the stylet and spicules of males were 18.2-19.6 μm and 22.7-36.8 μm long respectively. Egg masses were typically produced within the roots of kiwifruit. The new species had a rare Est phenotype, S2. Phylogenetic trees inferred from SSU, LSU D2D3, ITS, and partial coxII-16S rRNA revealed that M. aberrans sp. nov. was within the Meloidogyne clade and was distinguished from all described root-knot nematodes. Moreover, from histopathological observations, M. aberrans sp. nov. induced the formation of multinucleate giant cells.

Mitochondrial dysfunction underlies many prevalent diseases including heart disease arising from acute ischemia/reperfusion (I/R) injury. Here, we demonstrate that mitophagy, which selectively removes damaged or unwanted mitochondria, regulated mitochondrial quality and quantity in vivo. Hypoxia induced extensive mitochondrial degradation in a FUNDC1-dependent manner in platelets, and this was blocked by in vivo administration of a cell-penetrating peptide encompassing the LIR motif of FUNDC1 only in wild-type mice. Genetic ablation of Fundc1 impaired mitochondrial quality and increased mitochondrial mass in platelets and rendered the platelets insensitive to hypoxia and the peptide. Moreover, hypoxic mitophagy in platelets protected the heart from worsening of I/R injury. This represents a new mechanism of the hypoxic preconditioning effect which reduces I/R injury. Our results demonstrate a critical role of mitophagy in mitochondrial quality control and platelet activation, and suggest that manipulation of mitophagy by hypoxia or pharmacological approaches may be a novel strategy for cardioprotection.

Abstract Objective This retrospective study aimed to analyze the clinical characteristics and toxicological spectrum of acute poisoning in children treated at our pediatric intensive care unit. The findings provide a basis for the treatment and prevention of acute poisoning in children. Methods Clinical data of hospitalized children aged 0 to14 years with acute poisoning, treated between January 2015 and December 2023, were collected and analyzed. The analysis included the types of poisoning, sex and age distribution of children, routes and causes of poisoning, time from poisoning to medical consultation, length of hospital stays, and treatment outcomes. Results Among the 493 cases of acute poisoning in children, pesticide poisoning (n = 252, 51.1%) was the most common type, followed by medication poisoning (n = 182, 36.9%). Chemical poisoning accounted for 7.7% (n = 38), while food poisoning and inhalation of harmful gases accounted for 3.0% (n = 15) and 1.2% (n = 6), respectively. The main route of poisoning was oral ingestion (n = 477, 96.8%), while accidental ingestion accounted for a large proportion (n = 379, 79.4%), and adolescents aged 11 to 14 years accounted for 20.6% (n = 98).Apart from the predominance of girls attempting suicide by self-administering antipsychotic medications (n = 88, 75.9%) due to depression, there was no significant difference in sex distribution among other types of poisoning children. The highest incidence of poisoning except children who self-administer antipsychotic drugs occurred in children aged 1–3 years (n = 267, 54.2%) and 8–14 years (n = 135, 27.4%), with the highest proportion in the preschool age group. The average time from poisoning to medical consultation was 33.4 ± 54.9 h, and majority of children received treatment within 48 h after poisoning (n = 423, 85.8%). The overall effective treatment rate was 96.8%. Conclusion Acute poisoning in children has complex and diverse causes, with accidental ingestion being the main route. Prevention should be emphasized, including strengthening child safety education and improving parental supervision. Moreover, psychological health education is crucial for adolescents during their puberty. In the event of acute poisoning, prompt medical consultation and appropriate treatment are necessary to mitigate adverse outcomes.

Abstract Background Growth hormone-secreting pituitary neuroendocrine tumors can be pathologically classified into densely granulated (DGGH) and sparsely granulated types (SGGH). SGGH is more aggressive and associated with a poorer prognosis. While epigenetic regulation is vital in tumorigenesis and progression, the role of N6-methyladenosine (m6A) in aggressive behavior has yet to be elucidated. Methods We performed m6A-sequencing on tumor samples from 8 DGGH and 8 SGGH patients, complemented by a suite of assays including ELISA, immuno-histochemistry, -blotting and -fluorescence, qPCR, MeRIP, RIP, and RNA stability experiments, aiming to delineate the influence of m6A on tumor behavior. We further assessed the therapeutic potential of targeted drugs using cell cultures, organoid models, and animal studies. Results We discovered a significant reduction of m6A levels in SGGH compared to DGGH, with an elevated expression of fat mass and obesity-associated protein (FTO), an m6A demethylase, in SGGH subtype. Series of in vivo and in vitro experiments demonstrated that FTO inhibition in tumor cells robustly diminishes hypoxia resistance, attenuates growth hormone secretion, and augments responsiveness to octreotide. Mechanically, FTO-mediated m6A demethylation destabilizes desmoplakin (DSP) mRNA, mediated by the m6A reader FMR1, leading to prohibited desmosome integrity and enhanced tumor hypoxia tolerance. Targeting the FTO-DSP-SSTR2 axis curtailed growth hormone secretion, therefor sensitizing tumors to octreotide therapy. Conclusion Our study reveals the critical role of FTO in the aggressive growth hormone-secreting pituitary neuroendocrine tumors subtype and suggests FTO may represent a new therapeutic target for refractory/persistent SGGH.

Through the integration and analysis platform of particulate matter components, a preliminary comparison was conducted on the pollution contribution of the three winter transit air masses in December to environmental quality, and the changes and related relationships of water-soluble ions and particulate matter elements were analyzed. The results showed that winter transit air masses had a significant impact on the air quality in Hengyang. In the quantitative comparison of the primary pollutant contribution during the transit period of air masses, local sources and transit sources each accounted for half, and the impact of transit source on ambient air quality was much greater than that of seasonal base increase. Fine particulate matter was closely related to secondary ions, and particulate matter was closely related to primary ions. The transit of air masses promoted the improvement of secondary ion conversion rate, and the unit increment of fine particulate matter was greater than that of particulate matter. During the transit period, the mass concentrations of most water-soluble ions and most particulate matter elements maintained synchronous growth, with a superimposed effect. The composition ratio of organic carbon and nitrate ions increased, while the composition ratio of ammonia and sulfate ions decreased. Both disposable ions and secondary conversions in the composition of PM2 s had increases and decreases. The high conversion rate of nitrate and the high composition ratio of ammonia radical in Hengyang indicated that transportation source factors had a significant impact on the local environmental quality of Hengyang.
