Benchmark datasets play an important role in evaluating Natural Language Understanding (NLU) models. However, shortcuts -- unwanted biases in the benchmark datasets -- can damage the effectiveness of benchmark datasets in revealing models' real capabilities. Since shortcuts vary in coverage, productivity, and semantic meaning, it is challenging for NLU experts to systematically understand and avoid them when creating benchmark datasets. In this paper, we develop a visual analytics system, ShortcutLens, to help NLU experts explore shortcuts in NLU benchmark datasets. The system allows users to conduct multi-level exploration of shortcuts. Specifically, Statistics View helps users grasp the statistics such as coverage and productivity of shortcuts in the benchmark dataset. Template View employs hierarchical and interpretable templates to summarize different types of shortcuts. Instance View allows users to check the corresponding instances covered by the shortcuts. We conduct case studies and expert interviews to evaluate the effectiveness and usability of the system. The results demonstrate that ShortcutLens supports users in gaining a better understanding of benchmark dataset issues through shortcuts, inspiring them to create challenging and pertinent benchmark datasets., 15 pages, 6 figures

Mesenchymal stem cells (MSCs) are multi-potent cells that are able to differentiate and mature into various types of cells under a certain microenvironment for cell therapy and tissue regeneration. Scandium (Sc), an important rare earth element, recently has been intensively investigated in biomedical fields as well as industrial engineering, and chloride channels have been proven to be able to affect osteogenic differentiation. Thus, it is significant to investigate effects of ScCl3 on cell activities of MSCs. In this paper, rat bone MSCs (rBMSCs) were co-cultured with various concentrations of ScCl3 (1 × 10−8, 1 × 10−6, and 1 × 10−4 mol/L) to evaluate their influence on cell proliferation as well as osteogenic and adipogenic differentiation in vitro. The results indicate that ScCl3 promotes the proliferation of rBMSCs initially, which is yet reduced upon ion accumulation. We used immunofluorescence staining, quantitative real time polymerase chain reactions, and assays measuring alkaline phosphatase activity, mineralized deposits, and intracytoplasmic lipids to reveal that rBMSCs treated with ScCl3 at concentrations of 1 × 10−8 –1 × 10−6 mol/L can enhance levels of osteogenic differentiation in a dose-dependent manner and reduce adipogenic differentiation to a certain degree through Wnt/β-catenin signaling pathway. These results indicate that appropriate concentrations of ScCl3 can improve osteogenic differentiation in the lineage commitment of rBMSCs, and thus, promote bone remodeling. This study implies that ScCl3 possesses great potentials in the treatment of bone diseases and would provide new strategy of designing composites by SiCl3 doping for biomedical applications in the future.

Microbial induced calcium carbonate precipitation (MICP) is widely common in nature, which belongs to biomineralization, and has been explored carefully in recent decades. The research studied the main factors including temperature, initial pH value and Ca2+ concentration on bacterial growth and carbonic anhydrase activity by a kind of bacteria producing extracellular carbonic anhydrase, and revealed the biomineralization process of MICP by Ca2+ concentration and calcification rate in alkali environment. Meanwhile, microbial healing agent containing spores and calcium nitrate was prepared for the early age concrete cracks. The self-healing efficiency was assessed by crack closure rate and water permeability repair rate. After 28 d healing time of dry-wet cycles, compared with reference, the crack closure rate with the crack width up to 0.339 mm could reach 95.62%, the water permeability repair rate was 87.54%. XRD analysis showed that the precipitations at the crack mouth were calcite CaCO3. Meanwhile, the self-healing mechanism of mortar cracks was discussed in detail. In particular, there is no other pollution in the whole mineralization process, and the self-healing system is environmentally friendly, which provides a novel idea and method for the application of microbial self-healing concrete.

Herein, nano-sized α-Al2O3 powders were successfully synthesized at low temperatures via unidirectional ball milling of Al(OH)3 and addition of an AlF3 additive. The effects of the rotational direction in planetary ball milling on the α-phase transformation and morphology of α-Al2O3 were studied. Unidirectional rotation was observed to be significantly more helpful than bidirectional rotation with regard to improving the α-phase transformation and microstructure of α-Al2O3. Further, α-Al2O3 was obtained at a lower temperature (800 °C) by employing unidirectional ball milling; this was because gibbsite agglomerates (20–100 μm) could be refined into micro-sized curled flocs with a high specific surface area (124.95 m2 ∙ g−1) by performing unidirectional ball milling for 9 h. The addition of 1 wt% AlF3 accelerated the gas-phase mass transfer, which could effectively promote α-phase transformation and decrease the temperature at which α-Al2O3 was obtained. In comparison with the pyramidal α-Al2O3 polyhedron that exhibited a size range of 0.2–2 μm, homogeneous α-Al2O3 polyhedron with the size of approximately 100 nm was prepared at the temperature range of 910–940 °C via unidirectional ball milling.

Mesenchymal stem cells (MSCs) have been recognized as one of the most promising pharmaceutical multipotent cells, and a key step for their wide application is to safely and efficiently regulate their activities. Various methods have been proposed to regulate the directional differentiation of MSCs during tissue regeneration, such as nanoparticles and metal ions. Herein, nanoscale zeolitic imidazolate framework-8 (ZIF-8), a Zn-based metal-organic framework, is modified to direct MSCs toward an osteoblast lineage. Specifically, ZIF-8 nanoparticles are encapsulated using stem cell membranes (SCMs) to mimic natural molecules and improve the biocompatibility and targeted ability toward MSCs. Neural stem cell (NSC)-based therapy holds great promise for the treatment of neurodegenerative diseases. Presently, however, it is hindered by poor functional neuronal differentiation. Electrical stimulation is considered one of the most effective ways to promote neuronal differentiation of NSCs. In addition to surgically implanted electrodes, traditional electrical stimulation includes wires connected to the external power supply, and an additional surgery is required to remove the electrodes or wires following stimulation, which may cause secondary injuries and infections. Herein, a novel method is reported for generation of wireless electrical signals on an Au nanostrip array by leveraging the effect of electromagnetic induction under a rotating magnetic field. The intensity of the generated electrical signals depends on the rotation speed and magnetic field strength. The Au nanostrip array-mediated electric stimulation promotes NSC differentiation into mature neurons within 5 days, at the mRNA, protein, and function levels. The rate of differentiation is faster by at least 5 days than that in cells without treatment. The Au nanostrip array-based wireless device also accelerates neuronal differentiation of NSCs in vivo. The novel method to accelerate the neuronal differentiation of NSCs has the advantages of wireless, timely, localized and precise controllability, and noninvasive power supplementation.

Objective: We aimed to study the types and characteristics of TUBB1 mutations in a large Chinese cohort with congenital hypothyroidism (CH) and thyroid dysgenesis (TD). Methods: Mutation of the entire coding region of TUBB1 was analysed by Sanger sequencing in 289 children with CH and TD from China. Functional studies were further used to identify the effect of novel mutations on thyroid cells. Results: Among the 289 children with CH and TD, 4 (1.4%) had a c.952C>T(p.R318W) heterozygous mutation in TUBB1, resulting in a change from tryptophan to arginine at codon 318 of the TUBB1 protein. Functional studies indicated that the p.R318W mutant decreased TUBB1 expression and inhibited cell proliferation in a human thyroid cell line. Conclusions: A novel heterozygous missense mutation of TUBB1 in children with CH and TD was identified first in China, laying the foundations to expand the genotype-phenotype spectrum of TD.

Although engineered nanoparticles loaded with specific growth factors are used to regulate differentiation of stem cells, the low loading efficiency and biocompatibility are still great challenges in tissue repair. A nature-inspired biomimetic delivery system with targeted functions is attractive for enhancing cell activity and controlling cell fate. Herein, a stem cell membrane (SCM)-wrapped dexamethasone (DEX)-loaded zeolitic imidazolate framework-8 (ZIF-8) is constructed, which integrates the synthetic nanomaterials with native plasma membrane, to achieve efficient DEX delivery and DEX-mediated bone repair. The DEX@ZIF-8-SCM enables high DEX loading capacity, modulates the sustained release, and facilitates the specific uptake of mesenchymal stem cells (MSCs), owing to the porous property of ZIF-8 and the innate targeting capability of SCM. The endocytosed DEX@ZIF-8-SCM shows high cytocompatibility and greatly enhances the osteogenic differentiation of MSCs. Furthermore, RNA-sequencing data reveal that the phosphoinositide 3-kinase (PI3K)-Akt signaling pathways are activated and dominantly involved in the accelerated osteogenesis. In the bone defect model, the administrated DEX@ZIF-8-SCM exerts excellent biocompatibility and effectively promotes bone regeneration. Overall, the SCM-derived biomimetic nanoplatform achieves targeted delivery, excellent biosafety, and enhanced osteogenic differentiation and bone repair, which provides a new and valid strategy for treating various tissue injuries.

Traditional bone defect treatments are limited by an insufficient supply of autologous bone, the immune rejection of allogeneic bone grafts, and high medical costs. To address this medical need, bone tissue engineering has emerged as a promising option. Among the existing tissue engineering materials, the use of electroactive scaffolds has become a common strategy in bone repair. However, single-function electroactive scaffolds are not sufficient for scientific research or clinical application. On the other hand, multifunctional electroactive scaffolds are often complicated and expensive to prepare. Therefore, we propose a new tissue engineering strategy that optimizes the electrical properties and biocompatibility of carbon-based materials. Here, a hydroxyapatite/carbon nanofiber (HAp/CNF) scaffold with optimal electrical activity was prepared by electrospinning HAp nanoparticle-incorporated polyvinylidene fluoride (PVDF) and then carbonizing the fibers. Biochemical assessments of the markers of osteogenesis in human adipose-derived stem cells (h-ADSCs) cultured on HAp/CNF scaffolds demonstrate that the material promoted the osteogenic differentiation of h-ADSCs in the absence of an osteogenic factor. The results of this study show that electroactive carbon materials with a fibrous structure can promote the osteogenic differentiation of h-ADSCs, providing a new strategy for the preparation and application of carbon-based materials in bone tissue engineering.

Polyetheretherketone (PEEK) is widely used in orthopedic and craniomaxillofacial surgeries as it exhibits excellent biocompatibility, mechanical property, and chemical stability. However, its clinical application is limited by the biological inertness of PEEK. Numerous efforts have been made to improve the bioactivity of this polymer over the years. However, modification methods that can not only promote osteogenesis but also maintain excellent properties are still limited. Hence, a facile hot die formation technique is developed for establishing patterned nanorod arrays on the PEEK surface in situ. This method can maintain the excellent properties of PEEK and can be used in implantation as it can facilitate osteogenic activity in the absence of any organic/inorganic differentiation-inducing factors. PEEK with 200-nm patterned nanorod arrays on the surface exhibits excellent osteogenic properties. This result is obtained by assessing the osteogenic differentiation properties of rat adipose-derived stem cells at the gene and protein levels in vitro. In vivo experimental results reveal that the surface-modified cylindrical PEEK 200 implants present with excellent osseointegration properties. Moreover, they can tightly bind with the surrounding bone tissue. A practical method for manufacturing single-component PEEK implants with excellent osseointegration properties is reported, and the materials can be possibly used as orthopedic implants.

One of the major issues in tissue engineering is regulation of stem cell differentiation toward specific lineages. Unlike biological and chemical signals, physical signals with adjustable properties can be applied to stem cells in a timely and localized manner, thus making them a hot topic for research in the fields of biomaterials, tissue engineering, and cell biology. According to the signals sensed by cells, physical signals used for regulating stem cell fate can be classified into six categories: mechanical, light, thermal, electrical, acoustic, and magnetic. In most cases, external macroscopic physical fields cannot be used to modulate stem cell fate, as only the localized physical signals accepted by the surface receptors can regulate stem cell differentiation via nanoscale fibrin polysaccharide fibers. However, surface receptors related to certain kinds of physical signals are still unknown. Recently, significant progress has been made in the development of functional materials for energy conversion. Consequently, localized physical fields can be produced by absorbing energy from an external physical field and subsequently releasing another type of localized energy through functional nanostructures. Based on the above concepts, we propose a methodology that can be utilized for stem cell engineering and for the regulation of stem cell fate via nanostructure-mediated physical signals. In this review, the combined effect of various approaches and mechanisms of physical signals provides a perspective on stem cell fate promotion by nanostructure-mediated physical signals. We expect that this review will aid the development of remote-controlled and wireless platforms to physically guide stem cell differentiation both in vitro and in vivo, using optimized stimulation parameters and mechanistic investigations while driving the progress of research in the fields of materials science, cell biology, and clinical research.

This study examines the osteogenic differentiation promotion effect of micro/nanostructures of raffia on human adipose-derived stem cells to confirm the potential application of plant-derived micro/nanotopographies in tissue regeneration. The results confirm that the nanorod array on the front surface and the honeycomb-like microstructure on the back surface of raffia can not only regulate the adhesion, spreading, and migration of stem cells but also promote the osteogenic differentiation of the stem cells at a subsequent stage of cell culture. The osteocalcin expressions by the 21-day cultured cells on the front and back surfaces of raffia were 55-fold and 36-fold higher compared to the expression on a tissue culture plate. This indicates that plant-derived micro/nanotopographies can significantly promote stem cell differentiation. Furthermore, a general strategy for the application of plant-derived materials to stem-cell differentiation and bone-tissue engineering is suggested.

Objectives: Maternal cigarette smoke exposure (SE) causes intrauterine undernutrition, resulting in increased risk for metabolic disorders and type 2 diabetes in the offspring without sex differences. L-leucine supplementation has been shown to reduce body weight and improve glucose metabolism in both obese animals and humans. In this study, we aimed to determine whether postnatal L-leucine supplementation in female offspring can ameliorate the detrimental impact of maternal SE.Methods: Female Balb/c mice (6-week) were exposed to cigarette smoke (SE, 2 cigarettes/day) prior to mating for 5 weeks until the pups weaned. Sham dams were exposed to air during the same period. Half of the female offspring from the SE and SHAM dams were supplied with L-leucine via drinking water (1.5% w/w) after weaning (21-day) for 10 weeks and sacrificed at 13 weeks (adulthood).Results: Maternal SE during pregnancy resulted in smaller body weight and glucose intolerance in the offspring. L-leucine supplement in Sham offspring reduced body weight, fat mass, and fasting blood glucose levels compared with their untreated littermates; however somatic growth was not changed. L-leucine supplement in SE offspring improved glucose tolerance and reduced fat mass compared with untreated littermates.Conclusions: Postnatal L-leucine supplement could reduce fat accumulation and ameliorate glucose metabolic disorder caused by maternal SE. The application of leucine may provide a potential strategy for reducing metabolic disorders in offspring from mothers who continued to smoke during pregnancy.

In our previous study, we found that safrole oxide (SFO) could induce bone marrow mesenchymal stem cell differentiation into neuron-like cells. However, which kind of neuron cells was induced by SFO was unknown. Here, we found that SFO could induce BMSC differentiation into 5-hydroxytryptamine (5-HT) neuron-like cells. Microarray analysis of BMSCs treated with SFO for 6 h revealed a total of 35 genes changed more than twice. We selected G9a, a histone methyltransferase for further study. The upregulation of G9a was confirmed by RT-PCR and Western blot analysis. Small interfering RNA knockdown of G9a blocked SFO-induced BMSC differentiation. These results demonstrated that G9a was the pivotal factor in SFO-medicated 5-HT neuronal differentiation of BMSCs. Our findings provide a new clue for further investigating the gene control of BMSC differentiation into 5-HT neuron-like cells and provide a putative strategy for depression diseases therapies.

Background. The abnormal expression and prognosis prediction of epithelial cell transforming sequence 2 (ECT2) in gastric cancer (GC) has been reported. However, the effect of ECT2 on 5-fluorouracil (5-Fu) resistance in GC is unclear. This research aims to solve the abovementioned problems. Methods. Gene expression was detected by RT-qPCR and Western blot analysis. Cell viability was evaluated by the colony formation assay, MTT assay, and flow cytometric analysis. Transwell and wound healing assays were used to detect cell metastasis. Results. Upregulation of ECT2 was found in stomach adenocarcinoma (STAD) and GC tissues. In addition, high ECT2 expression can predict adverse clinical outcomes in GC patients. More importantly, ECT2 knockdown weakened the resistance of 5-FU in GC cells. ECT2 silencing reduced the cell migratory and invasive abilities of GC cells treated with 5-FU. We also found that downregulation of ECT2 increased 5-FU sensitivity in GC cells by downregulating P-gp, MRP1, and Bcl-2. Conclusion. Upregulation of ECT2 can predict adverse clinical outcomes and increase 5-FU resistance in GC patients.

In comparison with the typically worm-like α-Al2O3 powders formed from an unground Al(OH)3 precursor calcined at 1450°C, spherical α-Al2O3 powders with ~1 μm in size were prepared by the calcination of a ground Al(OH)3 precursor with 5 wt.% [NH4]+BF4 − under the same calcination conditions. The influence of a high-energy ball milling pretreatment as well as of the additives on the morphological evolution of α-Al2O3 powders was studied using the commercial precursor Al(OH)3 as raw material. The results indicate that the morphology of α-Al2O3 powders is closely related to the morphology of the Al(OH)3 precursor and to the introduction of different additives. The refinement of the Al(OH)3 precursor in aggregate size and of the primary crystal size by high-energy ball milling has effectively suppressed the neck growth of α-Al2O3 grains. In contrast to the findings made previously with the introduction of 5 wt.% [NH4]+Cl−, the morphology of the α-Al2O3 particles could be significantly improved from a ground Al(OH)3 precursor with the addition of 5 wt.% [NH4]+BF4 −, which resulted in the formation of spherical α-Al2O3 powders with 1 μm size at 1450°C.

Neural stem cells (NSCs) therapy is promising for treating neurodegenerative disorders and neural injuries. However, the limited in vitro expansion, spontaneous differentiation, and decrease in stemness obstruct the acquisition of high quantities of NSCs, restricting the clinical application of cell-based therapies and tissue engineering. This article reports a facile method of promoting NSCs expansion and maintaining stemness using wireless electrical stimulation triggered by piezoelectric nanomaterials. A nanofibrous membrane of poly L-lactic acid (PLLA) is prepared by electrostatic spinning, and the favorable piezoelectric property of PLLA facilitates the freeing of electrons after transformation. These self-powered electric signals generated by PLLA significantly enhance NSCs proliferation. Further, an undifferentiated cellular state is maintained in the NSCs cultured on the surfaces of PLLA nanofibers exposed to ultrasonic vibration. In addition, the neural differentiation potencies and functions of NSCs expanded by piezoelectric-driven localized electricity are not attenuated. Moreover, cell stemness can be maintained by wireless electric stimulation. Taken together, the electronic signals mediated by PLLA nanofibers facilitate NSCs proliferation. This efficient and simple strategy can maintain the stemness of NSCs during proliferation, which is essential for their clinical application, and opens up opportunities for the mass production of NSCs for use in cell therapy.

Electrical stimulation is an efficient approach to inducing neural differentiation of stem cells. However, most demonstrations of conventional electrical stimulation for the regulation of stem cell differentiation generally involve three components—an electrical signal generator, conductive culture substrate, and pair of lines, which limits its use in clinical applications for neural degeneration treatments. Herein, we proposed a facile method to generate localized electrical signals on the surface of a piezoelectric poly (vinylidene fluoride) (PVDF) film with a well-designed nanopillar array driven by ultrasound irradiation based on a piezotronic effect, which proved to induce neuronal differentiation of rat bone marrow mesenchymal stem cells (rBMSCs) without any biological or chemical neural inducing factors. The assessment of rBMSCs on the surface of the PVDF nanopillar array at the gene and protein levels confirmed that rBMSCs could differentiate into neuron-like cells. This demonstration provides a practical approach for the regulation of adult stem cells to differentiate into neurons, which will be a great achievement to overcome the shortage of neural stem cells in the adult human body and realize the autologous stem cell treatment of neurodegeneration. This work creates a new therapeutic avenue for contactless, controlled neuroregenerative therapies.

Neural stem cells (NSCs) are thought to be the best seed cells for neurodegenerative diseases therapy due to their potential to differentiate into either neurons or glial cells. However, the lack of resources for autologous NSCs prohibits their clinical applications. Although mesenchymal stem cells can be conveniently obtained from patients, neural differentiation is difficult to be realized, which becomes the great challenge for stem cell-based neurodegenerative diseases therapy. Herein, we report a novel method to induce neural differentiation of rat bone-marrow-derived mesenchymal stem cells (rBMSCs) based on piezotronics without any neural inducing factors (NIFs). In this work, a hybrid nanofibrous membrane was prepared by assembling a layer of FeOOH nanorods on the surface of polyvinylidene fluoride (PVDF) electrospun nanofibers. Under ultrasonic irradiation, piezoelectric-driven localized electric potential and electrical-driven iron ion (Fe3+) release based on piezotronics induced neural differentiation of rBMSCs cultured on the surface of these membranes. Genetic and molecular assays on the differentiated cells demonstrated that the synergistic effects of electrical signals and iron ion release induced the differentiation of rBMSCs into neurons (γ-aminobutyric acid (GABA)ergic, cholinergic and aminergic neurons) without any NIFs. Intracellular calcium imaging experiments showed that the differentiated cells generated fast peaking spontaneous [Ca2+]i-transients under the effects of neurotransmitters, especially GABA, indicating that rBMSCs-derived neurons had neuronal function. This study provides a novel strategy for inducing neural differentiation of rBMSCs via wireless stimulation with piezotronic effect, which is of great significance in clinical and neural tissue engineering.

Decreased bone formation is often associated with increased bone marrow adiposity. The molecular mechanisms that are accountable for the negative correlation between bone mass and bone marrow adiposity are incompletely understood. Focal adhesion kinase (FAK) has critical functions in proliferation and differentiation of many cell types; however, its roles in osteoblast lineage cells are largely unknown. We show herein that mice lacking FAK in Osterix-expressing cells exhibited decreased osteoblast number and low bone mass as well as increased bone marrow adiposity. The decreased bone mass in FAK-deficient mice was accounted for by decreased proliferation, compromised osteogenic differentiation, and increased adipogenic differentiation of bone marrow Osterix-expressing cells resulting from downregulation of Wnt/β-catenin signaling due to the reduced expression of canonical Wnt ligands. In contrast, FAK loss in calvarial preosteoblasts had no adverse effect on their proliferation and osteogenic differentiation and these cells had intact Wnt/β-catenin signaling. © 2016 American Society for Bone and Mineral Research.

These investigations necessitate the development of sensitive and reliable methods of tracking stem cells. As the unique physicochemical properties of β-NaGdF4:Yb/Er upconversion nanoparticles make them highly efficient fluorescent probes, they could be utilized to track stem cells through bio-imaging. However, their biocompatibility constitutes a major challenge to their use in biomedical applications. In this paper, we prepared ligand-free spherical β- NaGdF4:Yb/Er nanoparticles of two different sizes (~15 and ~30 nm in diameter) and investigated their internalization into rat bone marrow-derived MSCs (rBMSCs), as well as their effects on cell proliferation, osteogenic and adipogenic differentiation. Even though particles of both sizes were efficiently taken up by the cells, the larger particles had a stronger fluorescence intensity but their proliferation was not significantly affected; this makes them superior for cell imaging. Analysis of multiple markers revealed that the nanoparticles, especially the larger ones, promoted the process of osteogenic differentiation. In contrast, adipogenesis was slightly hindered by the larger particles, whereas the smaller ones did not affect the process. As a whole, this study suggests that ligand-free spherical β-NaGdF4:Yb/Er particles of appropriate size are compatible with stem cell proliferation and differentiation, which makes them promising agents for biomedical applications.

MicroRNAs (miRNAs) have been proven to have important effects on the proliferation and metastasis of multiple cancers, including hepatocellular carcinoma (HCC). In the present study, our aim was to explore the biological function of miR-106b in HCC cell proliferation and metastasis. qPCR analysis showed that miR-106b was expressed at higher levels, while disabled homolog 2 (DAB2) was expressed at lower levels in HCC tissues and cells. Moreover, the aberrant miR-106b expression in HCC affected the cell proliferative and migratory ability by MTT and Transwell assay. DAB2 was identified as a specific target of miR-106b in HCC by luciferase reporter assay and regression analysis showed a negative correlation between DAB2 and miR-106b expression. In addition, DAB2 may attenuate the miR-106b promotion effect on HCC cell proliferation and migration. In short, miR-106b may promote HCC cell proliferation and migration by targeting DAB2.

Along with the continuous progress of the information technology, Internet of Things is the inevitable way for realizing the fusion of communication and traditional network technology. Network coding, an important breakthrough in the field of communication, has many applied advantages in information network. This article analyses the eavesdropping problem of Internet of Things and presents an information secure network coding scheme against the eavesdropping adversaries. We show that, if the number of links the adversaries can eavesdrop on is less than the max-flow of a network, the proposed coding scheme not only `achieves the prefect information secure condition but also the max-flow of the network.
