Identification of the cellular mechanisms that mediate cancer cell chemosensitivity is important for developing new cancer treatment strategies. Several chemotherapeutic drugs increase levels of the posttranslational modifier ISG15, which suggests that ISGylation could suppress oncogenesis. However, how ISGylation of specific target proteins controls tumorigenesis is unknown. Here, we identified proteins that are ISGylated in response to chemotherapy. Treatment of a human mammary epithelial cell line with doxorubicin resulted in ISGylation of the p53 family protein p63. An alternative splice variant of p63, ΔNp63α, suppressed the transactivity of other p53 family members, and its expression was abnormally elevated in various human epithelial tumors, suggestive of an oncogenic role for this variant. We showed that ISGylation played an essential role in the downregulation of ΔNp63α. Anticancer drugs, including doxorubicin, induced ΔNp63α ISGylation and caspase-2 activation, leading to cleavage of ISGylated ΔNp63α in the nucleus and subsequent release of its inhibitory domain to the cytoplasm. ISGylation ablated the ability of ΔNp63α to promote anchorage-independent cell growth and tumor formation in vivo as well to suppress the transactivities of proapoptotic p53 family members. These findings establish ISG15 as a tumor suppressor via its conjugation to ΔNp63α and provide a molecular rationale for therapeutic use of doxorubicin against ΔNp63α-mediated cancers., Introduction ISG15, the product of IFN-stimulated gene 15, is the first reported ubiquitin-like protein (1). A general method for the assay of deubiquitinating enzymes was described in detail using 125I-labeled ubiquitin-fused &agr;NH-MHISPPEPESEEEEEHYC (referred to as Ub-PESTc) as a substrate. Since the tyrosine residue in the PESTc portion of the fusion protein was almost exclusively radioiodinated under a mild labeling condition, such as using IODO-BEADS, the enzymes could be assayed directly by simple measurement of the radioactivity released into acid soluble products. Using this assay protocol, we could purify six deubiquitinating enzymes from chick skeletal muscle and yeast and compare their specific activities. Since the extracts of E. coli showed little or no activity against the substrate, the assay protocol should be useful for identification and purification of eukaryotic deubiquitinating enzymes cloned and expressed in the cells.

Ubiquitin-fold modifier 1 (UFM1) is a newly identified ubiquitin-like protein. Like ubiquitin, UFM1 is conjugated to its target proteins through a three-step enzyme system: UBA5 (E1), UFC1 (E2), and UFL1 (E3), but with an additional essential component, UFBP1. This protein modification by UFM1 (ufmylation) can be reversed by UFM1-specific proteases (UFSPs). So far only a handful of target proteins for ufmylation have been identified, and they are mostly associated with either promotion or suppression of tumorigenesis. Here, we summarize the recent progress in the knowledge of tumor-suppressive and tumorigenic functions of ufmylation as well as in the development of therapeutic drugs against ufmylation-associated cancer. Northern blot analysis and patch-clamp techniques were utilized to determine the developmental regulation and functional role of inwardly rectifying K+ channels during chick myoblast differentiation. Northern blot analysis of replicating myoblasts indicated the presence of increasing levels of IRK activity that were initially absent in the first stages of myogenesis. Furthermore, IRK currents mediated K+ permeability during the fusion process of myoblasts.

Calcium influx is known to be prerequisite for membrane fusion of myoblasts. However, little is known about the channels that are responsible for the entry of calcium into the cells. Here we show that [K.sub.Ca] channels and stretch-activated channels are involved in the calcium influx. Upon analysis of single-channel recordings, calcium sensitivity of [K.sub.Ca] channels in myoblasts was found to be about sixfold higher than that in myotubes. Their density in myoblasts (1.68 [[[micro]meter].sup.-2]) was also about sixfold higher than that in myotubes (0.27 [[[micro]meter].sup.-2]). In addition, the opening of the calcium-permeable cationic channels in myoblasts was found to increase with membrane stretching and could be blocked by gadolinium. The density of stretch-activated channels was 0.22 [[[micro]meter].sup.-2] for myoblasts, and the relative permeability of calcium to potassium was [P.sub.Ca]/[P.sub.K] [approximately equals] 3.6. The channels could generate inward calcium currents to open [K.sub.Ca] channels in physiological solution. Furthermore, the activation of [K.sub.Ca] channels by phloretin dramatically hyperpolarized the resting membrane potential of myoblasts and this effect could be reversed upon treatment of tetraethylammonium. While phloretin induced precocious fusion, tetraethylammonium or gadolinium blocked not only the phloretin-induced precocious fusion but also the spontaneous fusion of myoblasts. These results suggest that hyperpolarization generated by reciprocal activation of stretch-activated channels and [K.sub.Ca] channels is involved in the calcium influx that triggers myoblast fusion.

Phosphorylation assays and cell cultures of chick embryonic myoblasts reveal that cyclic AMP (cAMP) inhibits Ca2+/calmodulin based phosphorylation of a 100-kDa protein and the cultured myoblast membrane fusion. However, cAMP does not affect the accumulation of the 100-kDa protein, CaM kinase III phosphorylation and muscle-specific protein levels. Addition of theophylline or forskolin enhances the negative effects of cAMP and suggests a correlation between the cAMP-inhibition of myoblast fusion and 100-kDa protein phosphorylation.

KMnO4 and N-ethylmaleimide at low concentrations (i.e., below 0.2 and 1.5 mM, respectively) are known to interact specifically with four to five sulfhydryl residues per Ca2+/Mg2+-ATPase molecule in sarcoplasmic reticulum. Purified calpain preferentially hydrolyzes the ATPase that was treated with either agent but not the native form of the enzyme. Exposure to each agent with increasing concentrations results in a greater loss of the ATPase activity and renders the enzyme more susceptible to calpain. In addition, β,r-methylene-ATP, when added during the treatment of KMnO4 or N-ethylmaleimide, can partially protect the ATPase against the degration. These results suggest that the covalent modification at the specific sulfhydryl residues in sarcoplasmic reticulum ATPase may mark the enzyme for degradation by intracellular proteinases, such as calpain.
