Abstract Background Non-obstructive azoospermia (NOA) is the most severe form of male infertility and affects approximately 1% of men worldwide. Fanconi anemia (FA) genes were known for their essential role in DNA repair and growing evidence showed the crucial role of FA pathway in NOA. However, the underlying mechanisms for Fance deficiency lead to a serious deficit and delayed maturation of male germ cells remain unclear. Methods We used Fance deficiency mouse model for experiments, and collected testes or epididymides from mice at 8 weeks (8W), 17.5 days post coitum (dpc), and postnatal 11 (P11) to P23. The mice referred to three genotypes: wildtype (Fance +/+), heterozygous (Fance +/-), and homozygous (Fance −/− ). Hematoxylin and eosin staining, immunofluorescence staining, and surface spread of spermatocytes were performed to explore the mechanisms for NOA of Fance −/− mice. Each experiment was conducted with a minimum of three biological replicates and Kruskal-Wallis with Dunn’s correction was used for statistical analysis. Results In the present study, we found that the adult male Fance −/− mice exhibited massive germ cell loss in seminiferous tubules and dramatically decreased sperms in epididymides. During the embryonic period, the number of Fance −/− prospermatogonia decreased significantly, without impacts on the proliferation (Ki-67, PCNA) and apoptosis (cleaved PARP, cleaved Caspase 3) status. The DNA double-strand breaks (γH2AX) increased at the cellular level of Fance −/− prospermatogonia, potentially associated with the increased nonhomologous end joining (53BP1) and decreased homologous recombination (RAD51) activity. Besides, Fance deficiency impeded the progression of meiotic prophase I of spermatocytes. The mechanisms entailed the reduced recruitment of the DNA end resection protein RPA2 at leptotene and recombinases RAD51 and DMC1 at zygotene. It also involved impaired removal of RPA2 at zygotene and FANCD2 foci at pachytene. And the accelerated initial formation of crossover at early pachytene, which is indicated by MLH1. Conclusions Fance deficiency caused massive male germ cell loss involved in the imbalance of DNA damage repair in prospermatogonia and altered dynamics of proteins in homologous recombination, DNA end resection, and crossover, providing new insights into the etiology and molecular basis of NOA.

Abstract To reveal the molecular function of elongation family of very long chain fatty acids(ELO) protein in Cyrtotrachelus buqueti, we have identified 15 ELO proteins from C.buqueti genome. 15 CbuELO proteins were located on four chromosomes. Their isoelectric points ranged from 9.22 to 9.68, and they were alkaline. These CbuELO proteins were stable and hydrophobic. CbuELO proteins had transmembrane movement, and had multiple phosphorylation sites. The secondary structure of CbuELO proteins was mainly α-helix. A total of 10 conserved motifs were identified in CbuELO protein family. Phylogenetic analysis showed that molecular evolutionary relationships of ELO protein family between C. buqueti and Tribolium castaneum was the closest. Developmental transcriptome analysis indicated that CbuELO10, CbuELO13 and CbuELO02 genes were key enzyme genes that determine the synthesis of very long chain fatty acids in pupae and eggs, CbuELO6 and CbuELO7 were that in the male, and CbuELO8 and CbuELO11 were that in the larva. Transcriptome analysis under different temperature conditions indicated that CbuELO1, CbuELO5, CbuELO12 and CbuELO14 participated in regulating temperature stress responses. Transcriptome analysis at different feeding times showed CbuELO12 gene expression level in all feeding time periods was significant downregulation. The qRT-PCR experiment verified expression level changes of CbuELO gene family under different temperature and feeding time conditions. Protein-protein interaction analysis showed that 9 CbuELO proteins were related to each other, CbuELO1, CbuELO4 and CbuELO12 had more than one interaction relationship. These results lay a theoretical foundation for further studying its molecular function during growth and development of C. buqueti.

Abstract DNA binding with one finger(Dof) gene family is a class of transcription factors which play an important role on plant growth and development. Genome-wide identification results indicated that there were 45 Dof genes(ColDof) in C.oleifera genome. All 45 ColDof proteins were non-transmembrane and non-secretory proteins. Phosphorylation site analysis showed that biological function of ColDof proteins were mainly realized by phosphorylation at serine (Ser) site. The secondary structure of 44 ColDof proteins was dominated by random coil, and only one ColDof protein was dominated by α-helix. ColDof genes’ promoter region contained a variety of cis-acting elements, including light responsive regulators, gibberellin responsive regulators, abscisic acid responsive regulators, auxin responsive regulators and drought induction responsive regulators. The SSR sites analysis showed that the proportion of single nucleotide repeats and the frequency of A/T in ColDof genes were the largest. Non-coding RNA analysis showed that 45 ColDof genes contained 232 miRNAs. Transcription factor binding sites of ColDof genes showed that ColDof genes had 5793 ERF binding sites, 4381 Dof binding sites, 2206 MYB binding sites, 3702 BCR-BPC binding sites. ColDof9, ColDof39 and ColDof44 were expected to have the most TFBSs. The collinearity analysis showed that there were 40 colinear locis between ColDof proteins and AtDof proteins. Phylogenetic analysis showed that ColDof gene family was most closely related to that of Camellia sinensis var. sinensis cv.Biyun and Camellia lanceoleosa. Protein-protein interaction analysis showed that ColDof34, ColDof20, ColDof28, ColDof35, ColDof42 and ColDof26 had the most protein interactions. The transcriptome analysis of C. oleifera seeds showed that 21 ColDof genes were involved in the growth and development process of C. oleifera seeds, and were expressed in 221 C. oleifera varieties. The results of qRT-PCR experiments treated with different concentrations NaCl and PEG6000 solutions indicated that ColDof1, ColDof2, ColDof14 and ColDof36 not only had significant molecular mechanisms for salt stress tolerance, but also significant molecular functions for drought stress tolerance in C. oleifera. The results of this study provide a reference for further understanding of the function of ColDof genes in C.oleifera.

Abstract Dof transcription factor family in Cyperus esculentus genome was identified and analyzed using bioinformatics. The analysis results revealed that C.esculentus genome contains 29 Dof genes (CesDof), all of which are located in the nucleus according to subcellular localization prediction. CesDof proteinrs have a range of 124 to 512 amino acids, with most being basic proteins. Their secondary structure was mainly irregular curl. The promoter sequence of CesDof genes contains cis-acting elements that respond to light, drought, hormones, low temperature, and circadian rhythm. Codon preference analysis showed that CesDof genes' codon preference ends in T/A. Collinearity analysis revealed that C.esculentus had three pairs of collinear CesDof genes. Additionally, there were 15 pairs of collinear genes between C.esculentus and Arabidopsis thaliana. The genetic relationship between C.esculentus and Rhynchospora pubera was found to be the closest. Phylogenetic tree analysis revealed that 29 CesDof genes of C.esculentus can be classified into 4 subgroups. Additionally, 144 miRNAs were predicted to target these CesDof genes. Furthermore, protein interaction analysis indicated that 15 Dof proteins in C.esculentus had interactions. The qRT-PCR verification results of drought stress and salt stress treatment experiments showed that most CesDof genes were involved in drought stress and salt stress responses, and the gene expression trends under drought stress and salt stress conditions were consistent. These results lay a theoretical foundation for further studying the molecular functions of Dof gene family in C.esculentus and its molecular mechanisms in regulating the life activities of C.esculentus.

Abstract Ceracris kiangsu Tsai (C. kiangs) is a kind of forest pest, which can harm nearly 100 kinds of weeds and crops. In this study, based on 314 species distribution points of C. kiangsu which were obtained from Chinese herbaria, literatures and investigation, and data of three future climate scenarios presented by CMIP6, two niche models (Garp, Maxent) were used to predict the suitable area of C. kiangsu in China. The result shows that the main environmental factors affecting the distribution of C. kiangsu are precipitation of driest month (bio14) and min temperature of coldest month (bio6). No matter now and future, the potential distribution areas of C. kiangsu in China are mainly in the south of Qinling–Huaihe River. Under current scenarios, the areas of the total, highly, moderately and poorly suitable of C. kiangsu in China are 160.65 × 104 km2, 31.70 × 104 km2, 60.36 × 104 km2 and 68.59 × 104 km2 respectively. The southern Hubei, western Jiangxi and eastern Hunan are highly-suitable areas. Under SSP1-2.6 and SSP2-4.5 scenarios, both the total suitable area and the highly suitable show a decreasing tread in 2050s. Compared to the 2050s, the total suitable area will coninue to decease in 2090s under SSP1-2.6, while it will increase under SSP2-4.5. The highly suitable area will increase in both scenarios, and the increased percentage under SSP2-4.5 is greater than that under SSP1-2.6. Under SSP5-8.5 scenarios, the total suitable area will increase by 1.83% in the 2050s, and decrease by 1.17% in the 2090s. The highly suitable area in the 2050s and 2090s under this scenarios is larger than under current scenarios. No matter what the scenario, the southern part of Yunnan, the southeast of Sichuan and the southwest of Chongqing will become highly-suitable areas as the climate continues to warm and should be monitored more cosely.

ABSTRACT Ceracris nigricornis Walker is an insect of the Acrididae, which can harm bamboo, rice, corn, sorghum and other crops, and can cause serious economic losses. In this study, based on 234 occurrence sites of C. nigricornis obtained from the Global Biodiversity Information Facility and literature, and data of three future climate scenarios presented by CMIP6, two niche models (GARP, MaxEnt) were used to predict the suitable area of C. nigricornis in China. The result shows that the main environmental factors affecting the distribution of C. nigricornis are min temperature of coldest month (bio6), mean temperature of coldest quarter (bio11), precipitation of driest month (bio14) and precipitation of wettest quarter (bio16). From the result of MaxEnt model, it can be seen that the suitable area of C. nigricornis in China is 128.91 × 104 km2 under current scenario. It will decrease by 3.19% in the 2050s and then increase by 12.04% in the 2090s under the SSP1‐2.6 scenario, increase by 5.79% in the 2050s and then decrease by 7.53% in the 2090s under the SSP2‐4.5 scenario, and increase by 33.03% in 2050s and then decrease by 23.31% in the 2090s under SSP5‐8.5 scenario. From the result of GARP model, it can be seen that the suitable area of C. nigricornis in China is 166.09 × 104 km2 under current scenario. It will increase by 8.41% in 2050s and then continue to increase by 6.11% in 2090s under SSP1‐2.6 scenario, increase by 23.84% in the 2050s and then decrease by 0.88% in the 2090s under the SSP2‐4.5 scenario, and increase by 34.37% in 2050s and then decrease by 1.75% in 2090s under SSP5‐8.5 scenario. The boundaries of suitable areas will expand to the north and southwest of China under future climate change scenarios, specially in Sichuan, Chongqing and Yunnan. Local forestry authorities should strengthen the monitoring of bamboo forests to prevent the damage caused by the introduction of C. nigricornis.

Global climate warming has led to changes in the suitable habitats for fungi. Colletotrichum acutatum, a common fungus causing anthracnose disease, is widely distributed in southern China. Currently, research on the relationship between C. acutatum and environmental warming was limited. In this study, MaxEnt and ArcGIS software were used to predict the suitable habitats of C. acutatum under current and future climate conditions based on its occurrence records and environmental factors. The optimal MaxEnt model parameters were set as feature combination (FC) = lp and regularization multiplier (RM) = 2.6. Bio15, Bio12, Bio09, and Bio19 were identified as the main environmental factors influencing the distribution of C. acutatum. Under current climate conditions, C. acutatum was distributed across all continents globally, except Antarctica. In China, C. acutatum was primarily distributed south of the Qinling-Huaihe Line, with a total suitable area of 259.52 × 104 km2. Under future climate conditions, the potential suitable habitat area for C. acutatum was expected to increase and spread towards inland China. The results of this study provided timely risk assessment for the distribution and spread of C. acutatum in China and offer scientific guidance for monitoring and timely controlled of its distribution areas.

Abstract Background Insect Cytochrome P450 monooxygenase (CYPs or P450s) plays an important role in detoxifying insecticides, causing insect populations to develop resistance. However, the molecular functions of P450 gene family in Cyrtotrachelus buqueti genome are still lacking. Results In this study, 71 CbuP450 genes have been identified. The amino acids length of CbuP450 proteins was between 183 aa ~ 1041 aa. They are proteins with transmembrane domains. The main component of their secondary structure is α-helix and random coils. Phylogenetic analysis showed that C. buqueti and Rhynchophorus ferrugineus were the most closely related. This gene family has 29 high-frequency codons, which tend to use A/T bases and A/T ending codons. Gene expression analysis showed that CbuP450_23 in the female adult may play an important role on high temperature resistance, and CbuP450_17 in the larval may play an important role on low temperature tolerance. CbuP450_10, CbuP450_17, CbuP450_23, CbuP450_10, CbuP450_16, CbuP450_20, CbuP450_23 and CbuP450_ 29 may be related to the regulation of bamboo fiber degradation genes in C. buqueti. Protein interaction analysis indicates that most CbuP450 proteins are mainly divided into three aspects: encoding the biosynthesis of ecdysteroids, participating in the decomposition of synthetic insecticides, metabolizing insect hormones, and participating in the detoxification of compounds. Conclusions We systematically analyzed the gene and protein characteristics, gene expression, and protein interactions of CbuP450 gene family, revealing the key genes involved in the stress response of CbuP450 gene family in the resistance of C. buqueti to high or low temperature stress, and identified the key CbuP450 proteins involved in important life activity metabolism. These results provided a reference for further research on the function of P450 gene family in C. buqueti.

Levulinic acid, a hydrolysis product of lignocellulose, can be metabolized into important compounds in the field of medicine and pesticides by engineered strains of Saccharomyces cerevisiae. Levulinic acid, as an intermediate product widely found in the conversion process of lignocellulosic biomass, has multiple applications. However, its toxicity to Saccharomyces cerevisiae reduces its conversion efficiency, so screening Saccharomyces cerevisiae genes that can tolerate levulinic acid becomes the key. By creating a whole-genome knockout library and bioinformatics analysis, this study used the phenotypic characteristics of cells as the basis for screening and found the HMX1 gene that is highly sensitive to levulinic acid in the oxidative stress pathway. After knocking out HMX1 and treating with levulinic acid, the omics data of the strain revealed that multiple affected pathways, especially the expression of 14 genes related to the cell wall and membrane system, were significantly downregulated. The levels of acetyl-CoA and riboflavin decreased by 1.02-fold and 1.44-fold, respectively, while the content of pantothenic acid increased. These findings indicate that the cell wall-membrane system, as well as the metabolism of acetyl-CoA and riboflavin, are important in improving the resistance of Saccharomyces cerevisiae to levulinic acid. They provide theoretical support for enhancing the tolerance of microorganisms to levulinic acid, which is significant for optimizing the conversion process of lignocellulosic biomass to levulinic acid.

IntroductionAnthracnose is a significant fungal disease that affects tree growth and development, with Colletotrichum spp. exhibiting host non-specificity and targeting various organs, making disease control challenging.MethodsThis study aimed to identify the pathogenic species causing anthracnose in Ilex macrocarpa in Nanchong, Sichuan Province, and screen effective fungicides, particularly biological ones. The pathogen was identified as Colletotrichum fioriniae through morphological observation, pathogenicity assays, and molecular biological methods. Three biological and five chemical fungicides were evaluated for their effects on the mycelial growth and spore germination rate of the pathogen.ResultsThe results indicated that prochloraz was the most effective chemical fungicide, while the cell-free supernatant (CFS) of Bacillus velezensis had the most significant inhibitory effect among the biological fungicides. Transcriptome analysis revealed that the CFS of B. velezensis significantly reduced the expression of genes associated with ribosomes, genetic information processing, membrane lipid metabolism, and sphingolipid biosynthesis in C. fioriniae. Additionally, the glutathione pathway’s expression of various genes, including key genes such as GST, GFA, Grx, TRR, and POD, was induced. Furthermore, the expression of 17 MFS transporters and 9 ABC transporters was increased. Autophagy-related ATGs were also affected by the B. velezensis CFS.DiscussionThese findings suggest that the B. velezensis CFS may inhibit C. fioriniae through interference with ribosomes, genetic information processing, cell membrane metabolism, and energy metabolism. These results provide potential target genes for the B. velezensis CFS and insights into the antifungal mechanism by which B. velezensis inhibits C. fioriniae.

Abstract In bioethanol production, the main by-product, 5-hydroxymethylfurfural (HMF), significantly hinders microbial fermentation. Therefore, it is crucial to explore genes related to HMF tolerance in Saccharomyces cerevisiae for enhancing the tolerance of ethanol fermentation strains. A comprehensive analysis was conducted using genome-wide deletion library scanning and SGAtools, resulting in the identification of 294 genes associated with HMF tolerance in S. cerevisiae. Further KEGG and GO enrichment analysis revealed the involvement of genes OCA1 and SIW14 in the protein phosphorylation pathway, underscoring their role in HMF tolerance. Spot test validation and subcellular structure observation demonstrated that, following a 3-h treatment with 60 mM HMF, the SIW14 gene knockout strain exhibited a 12.68% increase in cells with abnormal endoplasmic reticulum (ER) and a 22.41% increase in the accumulation of reactive oxygen species compared to the BY4741 strain. These findings indicate that the SIW14 gene contributes to the protection of the ER structure within the cell and facilitates the clearance of reactive oxygen species, thereby confirming its significance as a key gene for HMF tolerance in S. cerevisiae.

Chicory (Cichorium intybus L., 2n = 18), belonging to the Asteraceae family, exhibits significant edible, medicinal, and pasture values. Moderate research has been performed on identifying Chicory species’ chromosomes using fluorescence in situ hybridization (FISH) and C-banding. Detailed karyotype comparisons with chromosome nomenclature have not yet been performed for Chicory and similar species. In this study, the tandem repeats (TRs) were predicted and mapped to chromosomal regions based on released C. intybus L. ASM2352571 genome assembly v1, and then compared to the genome of Lettuce (Lactuca sativa L.). Nine new oligo probes were then developed and employed for karyotypic investigation of endive, Lettuce, and Chicory mitotic metaphase using non-denaturing FISH (ND-FISH). By combining the conserved oligo probes for 5S rDNA and 18S rDNA with the unique ND-FISH signals of new TR-oligo probes, we can develop a high-resolution standard karyotype for the cultivars of Lettuce and Chicory. The occurrence of chromosome structure variations from the natural population of Chicory and Lettuce was also revealed by ND-FISH with multiple oligo probes. The current observation of the karyotype differences and divergences of Lactuca and Cichorium and the genomic research offers crucial information about the Asteraceae family’s genetic diversity, chromosomal dynamics, and evolutionary routes.

The Asiatic rhinoceros beetle, Oryctes rhinoceros (Linnaeus, 1758) (Coleoptera: Scarabaeidae), is a destructive invasive species that poses a serious threat to palms, oil palms, and other plants. Defining a suitable area for the distribution of O. rhinoceros is essential for the development of appropriate policies and preventive measures. In this work, the MaxEnt niche model and ArcGIS software were used to predict the potential geographic distribution of O. rhinoceros in the world based on occurrence data and related environmental variables and to speculate on the influence of environmental variables on the distribution of O. rhinoceros. The results showed that the suitable areas of O. rhinoceros beetle were mainly distributed in 30° N–30° S, and the highly suitable areas were concentrated in South Asia, East Asia, Southeast Asia, and northern Oceania. The key environmental variables that determine the distribution location of O. rhinoceros are Precipitation of Wettest Month (bio13), Temperature of July (tmin7), Minimum Temperature of November (tmin11), and Precipitation of September (prec9). The prediction results of the MaxEnt model can reflect the global distribution of O. rhinoceros. This study can provide a theoretical basis for the prevention and control of O. rhinoceros and the development of relevant quarantine measures.

Spodoptera frugiperda (J.E. Smith) is a significant economic pest that has recently invaded Africa and Asia. However, much of the information regarding its ecological capabilities in these newly invaded environments remains largely unknown. In this study, the life history traits of the fall armyworm under conditions of increased temperature, different photoperiods, and varying humidity levels were systematically evaluated. Among 43 studies, a total of 20 studies were included in the analysis by passing the screening criteria, and random-effects meta-analysis, fixed-effects meta-analysis, and meta-regression were conducted. It has been found that with the increase in temperature above 20 °C, various physiological indicators of the fall armyworm are significantly enhanced. When the temperature reaches 32 °C, the physiological activities of S. frugiperda are at their highest point. As the temperature increases, the duration of each developmental stage of the fall armyworm decreases significantly, accompanied by an increase in oviposition quantity and period in females. Additionally, the pupal development time is shortened, which leads to an increase in the lifespan of the adult moth. Using temperature and relative humidity as environmental variables, the optimal survival conditions for each insect state of the fall armyworm were calculated. These findings can assist in predicting the population dynamics of the fall armyworm and in formulating appropriate and practical management strategies.

Peanut pod shape is a heritable trait which affects the market acceptance of in-shell peanut products. In order to determine the genetic control of pod shape, six component traits of pod shape (pod length, pod width, pod length/width ratio, pod roundness, beak degree and constriction degree) were measured using an image-based phenotyping method. A recombinant inbred line (RIL) population consisting of 181 lines was phenotyped across three environments. Continuous distributions and transgressive segregations were demonstrated in all measured traits and environments. Significant correlations were found among most component traits with broad-sense heritability ranging from 0.87 to 0.95. Quantitative trait locus (QTL) analysis yielded 26 additive QTLs explaining 3.79 to 52.37% phenotypic variations. A novel, stable and major QTL region conditioning multiple shape features was detected on chromosome 2, which spans a 10.81-Mb genomic region with 543 putative genes. Bioinformatics analysis revealed several candidate genes in this region. In addition, 73 pairs of epistatic interactions involving 92 loci were identified for six component traits explaining 0.94–6.45% phenotypic variations. These results provide new genetic loci to facilitate genomics-assisted breeding of peanut pod shape.

Abstract Fanconi anemia (FA) gene mutations are critical components in the genetic etiology of premature ovarian insufficiency (POI). Fance −/− mice detected meiotic arrest of primordial germ cells (PGCs) as early as embryonic day (E) 13.5 and exhibited decreased ovarian reserve after birth. However, the mechanism of Fance defect leading to dysgenesis of PGCs is unclear. We aimed to explore the effect of Fance defects on mitotic proliferation of PGCs. Combined with transcriptomic sequencing and validation, we examined the effect of Fance defects on cell cycle, transcription–replication conflicts (TRCs), and multiple DNA repair pathways in PGCs during active DNA replication at E11.5 and E12.5. Results showed Fance defects cause decreased numbers of PGCs during rapid mitosis at E11.5 and E12.5. Mitotic cell cycle progression of Fance −/− PGCs was blocked at E11.5 and E12.5, shown by decreased cell proportions in S and G2 phases and increased cell proportions in M phase. RNA-seq suggested the mechanisms involved in DNA replication and repair. We found Fance −/− PGCs accumulate TRCs during active DNA replication at E11.5 and E12.5. Fance −/− PGCs down-regulate multiple DNA repair pathways at E11.5 and E12.5 including the FA pathway, homologous recombination (HR) pathway, and base excision repair (BER) pathway. In conclusion, Fance defect impaired the mitotic proliferation of PGCs leading to rapidly decreased numbers and abnormal cell cycle distribution. Proliferation inhibition of Fance −/− PGCs was associated with accumulated TRCs and down-regulation of FA, HR, BER pathways. These provided a theoretical basis for identifying the inherited etiology and guiding potential fertility management for POI.

Engineering Saccharomyces cerevisiae for biodegradation and transformation of industrial toxic substances such as catechol (CA) has received widespread attention, but the low tolerance of S. cerevisiae to CA has limited its development. The exploration and modification of genes or pathways related to CA tolerance in S. cerevisiae is an effective way to further improve the utilization efficiency of CA. This study identified 36 genes associated with CA tolerance in S. cerevisiae through genome-wide identification and bioinformatics analysis and the ERG6 knockout strain (ERG6Δ) is the most sensitive to CA. Based on the omics analysis of ERG6Δ under CA stress, it was found that ERG6 knockout affects pathways such as intrinsic component of membrane and pentose phosphate pathway. In addition, the study revealed that 29 genes related to the cell wall-membrane system were up-regulated by more than twice, NADPH and NADP+ were increased by 2.48 and 4.41 times respectively, and spermidine and spermine were increased by 2.85 and 2.14 times, respectively, in ERG6Δ. Overall, the response of cell wall-membrane system, the accumulation of spermidine and NADPH, as well as the increased levels of metabolites in pentose phosphate pathway are important findings in improving the CA resistance. This study provides a theoretical basis for improving the tolerance of strains to CA and reducing the damage caused by CA to the ecological environment and human health.

Water bodies provide humans with important hydrological ecosystem services (HESs), directly or indirectly. Water yield, water conservation, and soil conservation are essential to HESs. Since China's reform and opening up, and with its rapid socio-economic development, land use in Jiangxi Province has undergone drastic change, resulting in threats to the ecological environment. This paper evaluates three HESs, water yield, water conservation, and soil conservation, in Jiangxi Province based on land use and rainfall data, quantifies the impacts of different land classes on each ecosystem, predicts future land use using the patch-generating land use simulation (PLUS) model, and finally, discusses the ecological risks in the study area. The following results were obtained: (1) The HESs in the basin increased and then decreased from 2000 to 2020, and the spatial distribution of water yield and water conservation was greatly influenced by rainfall. Soil conservation was mostly consistent with the elevation distribution. (2) Over time, the overall aggregation of HESs in the study area increased. There were small differences in the effects of various land uses on water yield and water conservation, and large differences in the effects on soil conservation. (3) The distribution of ecological risks was not affected by different land use strategies, with the lower ecological risk level 1 dominating. Most risk areas were present in Ganzhou, Ji'an, Shangrao, and Jiujiang. The ecological risk from urban sprawl (US) accounted for the most significant proportion, and that from the ecological protection (EP) strategy accounted for the lowest proportion. This study provides reference for sustainable land use development and ecological risk prevention in the study area.

With the wave of the digital economy and industrial ecological construction, it is more essential for developing countries to focus on improving the structural quality of the service industry rather than just the quantitative aspect of the service industry. This study utilizes panel data from 30 provinces in China and spatial Dubin models to estimate the impact of service industry structure upgrading on industrial ecologicalization efficiency and its spatial effect. Our results reveal that productive and high-end service industries play a vital role in promoting industrial ecological efficiency. At the level of spatial effect, the productive service industry has a negative spatial correlation with the adjacent area, while the effect of the high-end service industry on the adjacent area is not highlighted. This study focuses on clarifying the spatial role of service industry structure upgrading on the improvement of industrial ecological efficiency, further expanding the theory of industrial structure adjustment, and providing insights for developing countries on how to optimize the structure of the service industry and achieve high-quality development of industrial ecologicalization.

Background Bactrocera minax (Enderlein, 1920) (Diptera: Tephritidae) is a destructive citrus pest. It is mainly distributed throughout Shaanxi, Sichuan, Chongqing, Guizhou, Yunnan, Hubei, Hunan, and Guangxi in China and is considered to be a second-class pest that is prohibited from entering that country. Climate change, new farming techniques, and increased international trade has caused the habitable area of this pest to gradually expand. Understanding the suitable habitats of B. minax under future climate scenarios may be crucial to reveal the expansion pattern of the insect and develop corresponding prevention strategies in China. Methods Using on the current 199 distribution points and 11 environmental variables for B. minax, we chose the optimal MaxEnt model to screen the dominant factors that affect the distribution of B. minax and to predict the potential future distribution of B. minax in China under two shared socio-economic pathways (SSP1-2.6, SSP5-8.5). Results The current habitat of B. minax is located at 24.1–34.6°N and 101.1–122.9°E, which encompasses the provinces of Guizhou, Sichuan, Hubei, Hunan, Chongqing, and Yunnan (21.64 × 104 km2). Under future climate scenarios, the potential suitable habitat for B. minax may expand significantly toward the lower-middle reaches of the Yangtze River. The land coverage of highly suitable habitats may increase from 21.64 × 104 km2 to 26.35 × 104 × 104 km2 (2050s, SSP5-8.5) ~ 33.51 × 104 km2 (2090s, SSP5-8.5). This expansion area accounts for 29% (2050s, SSP1-2.6) to 34.83% (2090s, SSP1-2.6) of the current habitat. The center of the suitable habitat was predicted to expand towards the northeast, and the scenario with a stronger radiative force corresponded to a more marked movement of the center toward higher latitudes. A jackknife test showed that the dominant variables affecting the distribution of B. minax were the mean temperature of the driest quarter (bio9), the annual precipitation (bio12), the mean diurnal range (bio2), the temperature annual range (bio7), and the altitude (alt). Discussion Currently, it is possible for B. minax to expand its damaging presence. Regions with appropriate climate conditions and distribution of host plants may become potential habitats for the insects, and local authorities should strengthen their detection and prevention strategies. Climate changes in the future may promote the survival and expansion of B. minax species in China, which is represented by the significant increase of suitable habitats toward regions of high altitudes and latitudes across all directions but with some shrinkage in the east and west sides.

Background Gibberellins (GAs) play important roles in regulating peanut growth and development. GA20ox and GA3ox are key enzymes involved in GA biosynthesis. These enzymes encoded by a multigene family belong to the 2OG-Fe (II) oxygenase superfamily. To date, no genome-wide comparative analysis of peanut AhGA20ox and AhGA3ox-encoding genes has been performed, and the roles of these genes in peanut pod development are not clear. Methods A whole-genome analysis of AhGA20ox and AhGA3ox gene families in peanut was carried out using bioinformatic tools. The expression of these genes at different stage of pod development was analyzed using qRT-PCR. Results In this study, a total of 15 AhGA20ox and five AhGA3ox genes were identified in peanut genome, which were distributed on 14 chromosomes. Phylogenetic analysis divided the GA20oxs and GA3oxs into three groups, but AhGA20oxs and AhGA3oxs in two groups. The conserved pattern of gene structure, cis-elements, and protein motifs further confirmed their evolutionary relationship in peanut. AhGA20ox and AhGA3ox genes were differential expressed at different stages of pod development. The strong expression of AhGA20ox1/AhGA20ox4, AhGA20ox12/AhGA20ox15, AhGA3ox1 and AhGA3ox4/AhGA3ox5 in S1-stage indicated that these genes could have a key role in controlling peg elongation. Furthermore, AhGA20ox and AhGA3ox also showed diverse expression patterns in different peanut tissues including leaves, main stems, flowers and inflorescences. Noticeably, AhGA20ox9/AhGA20ox11 and AhGA3o4/AhGA3ox5 were highly expressed in the main stem, whereas the AhGA3ox1 and AhGA20ox10 were strongly expressed in the inflorescence. The expression levels of AhGA20ox2/AhGA20ox3, AhGA20ox5/AhGA20ox6, AhGA20ox7/AhGA20ox8, AhGA20ox13/AhGA20ox14 and AhGA3ox2/AhGA3ox3 were high in the flowers, suggesting their involvement in flower development. These results provide a basis for deciphering the roles of AhGA20ox and AhGA3ox in peanut growth and development, especially in pod development.

To promote the development of the renewable energy (RE) industry, China officially implemented renewable portfolio standard (RPS) in 2020, the policy effect of which is closely related to the amount of renewable power offered to users by power-selling enterprises. We use evolutionary game theory to analyze the behavioral strategies of regional governments, regulatory authorities, and power-selling enterprises under RPS, and build a system dynamics (SD) model to determine the influence of the relevant parameters on stakeholders’ strategy making considering quota heterogeneity. The results show that: (1) enterprises evolve to being stable earlier in high-quota area than in low-quota area, which infers that RE development has a certain bottleneck in the initial stage and that RPS can play an effective role; (2) a high certificate price can not only help power selling companies evolve to being stable, but also promote the withdrawal of governments subsidies; (3) to increase the proportion of renewable electricity, the net profit of RE power should not be lower than that of conventional energy; and (4) the incentive effect of subsidy income is not stronger than that of resale income, while when compared with penalty, the incentive effect is stronger, and penalty is not more severe and actually better. Importantly, the results provide policy suggestions for the development of RPS.

Abstract Background DNA methylation is an essential factor in the progression of cervical intraepithelial neoplasia (CIN) to cervical cancer. The aim was to investigate the diagnostic value provided by methylation biomarkers of six tumor suppressor genes (ASTN1, DLX1, ITGA4, RXFP3, SOX17 and ZNF671) for cervical precancerous lesions and cervical cancer. Methods The histological cervical specimens of 396 cases including 93 CIN1, 99 CIN2, 93 CIN3 and 111 cervical cancers were tested for methylation-specific PCR assay (GynTect®) of score and positive rate. Among them, 66 CIN1, 93 CIN2, 87 CIN3 and 72 cervical cancers were further used for paired analysis. A chi-square test was used to analyze the difference of methylation score and positive rate in cervical specimens. The paired t-test and paired chi-square test were for analyzing the methylation score and positive rate in paired CIN and cervical cancer cases. The specificity, sensitivity, odds ratio (OR) and 95% confidence interval (95% CI) of the GynTect® assay for CIN2 or worse (CIN2 +) and CIN3 or worse (CIN3 +) were evaluated. Results According to the chi-square test trend, hypermethylation increased with severity of the lesions as defined by histological grading (P = 0.000). The methylation score above 1.1 was more common in CIN2 + than in CIN1. The DNA methylation scores in the paired groups of CIN1, CIN3 and cervical cancer were significant differences (P = 0.033, 0.000 and 0.000, respectively), except for CIN2 (P = 0.171). Abstract Background Carbon nanoparticles (CNPs) are a new tracer for lymph node mapping, which can quickly reach and develop lymph nodes through a lymphatic network. This research investigated the characteristics of systematic lymph node dissection and sentinel lymph node biopsy mapped with CNPs in endometrial carcinoma. Methods We first applied CNPs to systematic lymph node dissection in 18 endometrial carcinoma patients as the study group and another 18 endometrial carcinoma patients who were not injected with anything served as the control group. Then, we applied CNPs to sentinel lymph nodes biopsy in 54 endometrial carcinoma patients. All 54 patients received systematic lymph node dissection after sentinel lymph node biopsy. The detection rate, sensitivity, specificity, and accuracy of systematic lymph node dissection and sentinel lymph node biopsy by CNPs were respectively analyzed. NAC transcription factors are an important genes that regulate plant growth and development, and can regulate functions such as fruit ripening in plants. Based on genome data of Dendrobium nobile, the NAC gene family was identified and analyzed by bioinformatics methods. In this study, we identified 85 NAC genes in Dendrobium nobile genome, and systematically analyzed the NAC gene family. We found that they were distributed unevenly in the nineteen chromosomes. The amino acid length of D. nobile NAC gene family (DnoNACs) ranged from 80 to 1065, molecular weight ranged from 22.17 to 119.02 kD, and isoelectric point ranged from 4.61~9.26. Its promoter region contains multiple stress responsive elements, including light responsive, gibberellin-responsive, abscisic acid responsiveness, MeJA-responsiveness and drought-inducibility elements. Phylogenetic analysis indicates that the D. nobile NAC gene family is most closely related to Dendrobium catenatum and Dendrobium chrysotoxum. Analysis of SSR loci indicates that the fraction of mononucleotide repeats was the largest, as was the frequency of A/T. Non-coding RNA analysis showed that these 85 NAC genes contain 397 miRNAs. The collinearity analysis shows that 9 collinear locis were found on the chromosomes of D. nobile with Arabidopsis thaliana, and 75 collinear locis with D.chrysotoxum. QRT-PCR experiment under different salt concentration and temperature conditions verified the response mechanism of DnoNAC gene family under stress conditions. Most DnoNAC genes are sensitive to salt stress and temperature stress. The results of this study provide a reference for further understanding the function of NAC gene in D. nobile.

With the depletion of natural resources and the requirement of higher strength-weight ratio, lightweight aggregate concrete has attracted more and more attention because of its good thermal properties, fire resistance and seismic performance. However, exposure to low temperature environments accelerates deterioration of concrete, thereby, reduce the service life of lightweight aggregate concrete. Even worse, in cold and arid regions, lightweight aggregate concrete often experiences accidental impacts, wind erosion, earthquakes, and other disasters during service, these damage significantly impact its frost-resistance. Therefore, accurately and quantitatively describing and predicting the frost-resistance of lightweight aggregate concrete under specific disaster conditions is crucial. In this study, take the initial damage degree and freeze-thaw cycles as input variables, while the relative dynamic elastic modulus (RDEM) as an out variable, a frost resistance prediction models for stress-damaged lightweight aggregate concrete was established based on back propagation neural network (BPNN). The results show that the predicted values of BPNN model are in good agreement with the experimental values, and the results are also compared with the revised Loland model which is proposed by another author. Results demonstrate that the average relative error between predicted values of BPNN and experimental values is only 1.69%, whereas the one of revised Loland model is 9.13%, which indicating that the proposed BPNN prediction model can achieve a relatively accurate quantitative assessment of frost-resistance throughout the entire post-disaster lifecycle of lightweight aggregate concrete, it also broadened the idea and provided a reference for the frost resistance prediction of stress-damaged lightweight aggregate concrete.

Abstract To achieve the goal of energy conservation and emission reduction, China has launched its national carbon-emission trading scheme (ETS). Therefore, it is of great significance to evaluate the implementation of ETS from the perspective of regional ecologicalization efficiency of industry. Based on the panel data of 30 provinces and cities from 2007 to 2019, this study takes the SBM-DEA model to measure the regional ecologicalization efficiency of industry, uses the Difference-in-Difference (DID) model to evaluate the influence of ETS on the regional ecologicalization efficiency of industry, and verifies the outcome by propensity score matching method and counterfactual test. The mediating effect model is used to test the acting pattern of ETS on the regional ecologicalization efficiency of industry. The results show that: (1) ETS can significantly improve the ecologicalization efficiency of industry and shows regional heterogeneity. (2) ETS promotes regional energy conservation, emission reduction, and economic development because of the upgrading of structural optimization, agglomeration of resource synergy, and optimization of ecology. This study offers penetrating insights into the positive operation of the carbon market and provides some useful hints for the development of China’s regional economy.

ObjectivesTo evaluate the efficacy of Optical Coherence Tomography (OCT) for detecting cervical lesions in women with minor abnormal cytology results (atypical squamous cells of undetermined significance (ASC-US) and low-grade squamous intraepithelial lesion (LSIL)).MethodsA prospective study was conducted at gynecologic clinic from Mar 2021 to Sep 2021. The recruited women with cervical cytological findings of ASC-US or LSIL were inspected with OCT before colposcopy-directed cervical biopsy. The diagnostic performance of OCT, alone and in combination with high-risk human papillomavirus (hrHPV) testing were evaluated to detect cervical intraepithelial neoplasia of grade 2 or worse (CIN2+)/CIN3 or worse (CIN3+). The rate of colposcopy referral and the immediate risk of CIN3+ of OCT were calculated.ResultsA total of 349 women with minor abnormal cervical cytology results were enrolled. For detection of CIN2+/CIN3+, the sensitivity and NPV of OCT were lower than those of hrHPV testing (CIN2+: 71.3% vs. 95.4%, 89.0% vs. 91.1%, P < 0.001; CIN3+: 75% vs. 93.8%, 96.5% vs. 95.6%, P < 0.001), but the specificity, accuracy and PPV were higher than those of hrHPV testing (CIN2+: 77.5% vs. 15.6%, 75.9% vs. 35.5%, 51.2% vs. 27.3%, P < 0.001; CIN3+: 69.4% vs. 13.6%, 69.9% vs. 20.9%, 19.8% vs. 9.9%, P < 0.001). OCT combined with hrHPV testing (CIN2+: 80.9%; CIN3+: 72.6%) showed higher specificity than that of OCT alone (P < 0.001). The colposcopy referral rate base on OCT classification was lower than that based on hrHPV testing (34.7% vs. 87.1%, P < 0.001). Patients with hrHPV-positive ASC-US and hrHPV-negative LSIL cytology, the immediate CIN3+ risk in OCT negative cases was less than 4%.ConclusionsOCT alone or combination with hrHPV testing shows good performance for detecting CIN2+/CIN3+ in patients with ASC-US/LSIL cytology. OCT is an effective method for colposcopy triage in women with hrHPV-positive ASC-US and hrHPV-negative LSIL cytology.

Fanconi anemia (FA) genes contribute to tumorigenesis by regulating DNA repair. Despite its importance for assembly and functionality of the FA core complex, no pan-cancer analysis of FANCE was performed. We aimed to provide a comprehensive understanding of the role of FANCE in cancers. Based on The Cancer Genome Atlas (TCGA), Cancer Cell Line Encyclopedia (CCLE), Genotype Tissue-Expression (GTEx), Human Protein Atlas (HPA), Gene Expression Omnibus (GEO), and Cancer Single-cell Atlas (CancerSEA) databases, we investigated the carcinogenicity of FANCE using various bioinformatics methods, including FANCE expression and prognosis, immune invasion, tumor mutation burden, microsatellite instability, and neoantigens. We monitored Fance mutations in mice that caused tumorigenesis. FANCE expression and activity scores were upregulated in 15 and 21 cancers. High expression of FANCE affected shorter overall survival (OS) in seven cancers and longer overall survival in three cancers. It was correlated with shorter overall survival and progression-free interval (PFI) in endometrial cancer and longer overall survival and PFI in cervical cancer. FANCE expression negatively correlated with stromal/immune scores in 21 cancers including cervical cancer, endometrial cancer, and ovarian cancer. FANCE expression negatively correlated with CD8 T cells in endometrial cancer and positively correlated with M1 macrophages in cervical cancer, possibly related to cancer prognosis. FANCE positively correlated with immune checkpoint inhibitors PD-1, PD-L1, and CTLA4 in endometrial cancer and ovarian cancer. FANCE expression positively correlated with microsatellite instability, tumor mutational burden, and neoantigens in 7, 22, and five cancers, especially in endometrial cancer, potentially increasing the effectiveness of immunotherapy. Single-cell sequencing data showed FANCE was primarily expressed in cancer cells in cervical and ovarian cancer, and in fibroblasts in endometrial cancer. Fance heterozygous mutant mice had increased tumor incidences and shorter overall survival and tumor-free survival (TFS) than Fance homozygous mutant mice and wild-type mice. Conclusively, FANCE potential to serve as a biomarker for cancer prognosis and may predict cancer immunotherapy responses. Fance heterozygous mutant resulted in increased tumorigenesis and poor prognosis in mice.

In order to improve the identification accuracy of damage detection and evaluation based on the vibration response, this paper presents a structural damage identification method based on the fractal dimension, data fusion and a revised counter-propagation network (RCPN). Firstly, the fractal dimensions of the original signal response are extracted through data preprocessing. Secondly, the first-time fusion of data (i.e., the feature-level fusion) is carried out, after which these data are used as the input for the RCPN, to identify and decide the initial damage. Finally, the second-time data fusion (i.e., based on the decision results of the feature-level fusion) is carried out, leading to decision-level fusion. In order to verify the validity of the proposed method, a four-storey benchmark structure of ASCE is used for damage identification and comparison, using a single RCPN decision and the data fusion damage identification method, respectively. The results show that the proposed method is more accurate and reliable than the results of single RCPN decision and feature-level fusion decision, and has good noise resistance and robustness.

Abstract Background We assessed the image quality of endometrial cancer lesions by readout segmentation of long variable echo-trains (RESOLVE) diffusion-weighted imaging (DWI) compared with that by single-shot echo-planar imaging (SS-EPI) DWI, aimed to explore the value of RESOLVE DWI for determining myometrial invasion and clinical stage in endometrial cancer. Materials and methods From April 2017 to March 2018, a total of 30 endometrial cancer patients (mean age 52.8 ± 9.0 years), who had undergone RESOLVE DWI and SS-EPI DWI, were included in the study. The image quality of endometrial carcinoma by two kinds of DWI scanning methods was compared qualitatively and quantitatively. The Spearman rank correlation test was used to assess the correlation of qualitative image quality scores between two readers. The accuracy of two DWI methods in detecting myometrial invasion and staging of endometrial carcinoma was calculated according to postoperative pathological results. The indexes were analyzed including sensitivity, specificity, accuracy, positive predictive value (PPV), and negative predictive value (NPV). Abstract Background With the ageing of China’s population, the incidence and mortality of coronary atherosclerotic heart disease (CAD) is increasing year by year, which brings a heavy burden to the family and society [1]. We aimed to analyse the strategy of coronary artery bypass grafting (CABG) in the right coronary artery and to compare the haemodynamic characteristics of the sequential grafts with those of single grafts and to observe the patency rate of those grafts for 1 week after the operation. Methods A total of 242 patients (178 men, mean age 62.6 ± 8.8 years) underwent right coronary artery bypass grafting in our hospital from October 2016 to January 2019. The blood flow (Q, ml/min), pulsatility index (PI) and related parameters of the grafts were measured and recorded by TTFM during the CABG. The patency of the grafts was evaluated by coronary computed tomography (CT) for 1 week after the operation. Results The most common material used for the graft in the right coronary system of CABG is the greater saphenous vein (92.3%), followed by the radial artery (5.5%) and the internal mammary artery (1.9%). The highest frequency target of the right coronary artery is the posterior descending artery (PDA) (47.6%), followed by the right main coronary artery (RCA) (29.1%) and the posterior branch of the left ventricle (PL) (23.3%). The proportion of single grafts was the highest for the right coronary artery in CABG (178 cases, 67.9%), followed by a graft of the PDA-PL (42 cases, 16.0%) and other sequential grafts among the different coronary artery systems (including the system of the left anterior descending artery (LAD) and the left circumflex (LCX)). Whether there were sequential grafts of the PDA-PL or other sequential grafts among the different systems of the coronary artery, the instantaneous flow of a group of sequential grafts was higher than that of a single graft, and the difference had statistical significance (P

Background: Cultivated peanut (Arachis hypogaea. L) represents one of the most important oil crops in the world. Although much effort has been expended to characterize microsatellites or Simple Sequence Repeats (SSRs) in peanut, the quantity and quality of the markers in breeding applications remain limited. Here, genome-wide SSR characterization and marker development were performed using the recently assembled genome of the cultivar Tifrunner. Results: In total, 512,900 microsatellites were identified from 2556.9-Mb genomic sequences. Based on the flanking sequences of the identified microsatellites, 7757 primer pairs (markers) were designed, and further evaluated in the assembled genomic sequences of the tetraploid Arachis cultivars, Tifrunner and Shitouqi, and the diploid ancestral species, A. duranensis and A. ipaensis. In silico PCR analysis showed that the SSR markers had high amplification efficiency and polymorphism in four Arachis genotypes. Notably, nearly 60% of these markers were single-locus SSRs in tetraploid Arachis species, indicating they are more specific in distinguishing the alleles of the A and B sub-genomes of peanut. In addition, two markers closely related with purple testa color and 27 markers near to FAD2 genes were identified, which could be used for breeding varieties with purple testa and high-oleic acid content, respectively. Moreover, the potential application of these SSR markers in tracking introgressions from Arachis wild relatives was discussed. Conclusions: This study reported the development of genomic SSRs from assembled genomic sequences of the tetraploid Arachis Tifrunner, which will be useful for diversity analysis, genetic mapping and functional genomics studies in peanut. It is urgent to improve the comprehensive utilization rate of tailings, especially to solve the problem of fine iron ore tailings treatment and the shortage of natural sand, so iron ore tailings concrete with different replace rates was prepared in this paper. At the same time, in order to further popularize the wide application of iron tailings concrete, it is of great significance to study the frost resistance and life prediction of it in cold regions. Thereby, a series of experimental research on iron ore tailings concrete under freezing-thawing cycles was carried out to analyze the attenuation law of quality, compressive strength, and relative dynamic elastic modulus after different freezing-thawing cycles. In this paper, the relative dynamic elastic modulus was used to define the damage factor, and the freezing-thawing damage model of iron ore tailings concrete was established based on Weibull distribution. Here, the specific function form of two-parameter Weibull distribution of iron ore tailings concrete under freezing-thawing cycle was proposed, and the model was used to predict life of it. The results showed that, compared with the replacement rates of 20%, 60%, and 80%, the iron ore tailings with 40% replacement rate had the best improvement effect on the frost resistance of concrete and can delay the attenuation of the compressive strength of concrete. This model can not only predict the durability life of iron ore tailings concrete under freezing-thawing conditions, but also provide theoretical reference for the application of iron ore tailings concrete in cold areas.

Objective. Cervical cancer (CC) is a prevalent cancer in women. Hypoxia plays a critical role in CC cell ferroptosis resistance. This study explored the mechanism of hypoxia in CC cell ferroptosis resistance by regulating HIF1α/KDM4A/H3K9me3. Methods. Cultured SiHa and Hela cells were exposed to CoCl2 and treated with Erastin. Cell viability was detected by MTT assay, and concentrations of iron ion, MDA and GSH were determined using corresponding kits. Expressions of KDM4A, HIF1α, TfR1, DMT1, and H3k9me3 were detected by RT-qPCR, Western blot, and ChIP assay. The correlation of KDM4A and HIF1α was analyzed on Oncomine, UALCAN, and Starbase. CC cells were co-transfected with shKDM4A or/and pcDNA3.1-HIF1α. Iron uptake and release were assessed using the isotopic tracer method. The binding relationship between HIF1α and HRE sequence was verified by dual-luciferase assay. Results. Cell viability and GSH were decreased while iron concentration, MDA, KDM4A, and HIF1α levels were increased in hypoxia conditions. The 2-h hypoxia induced ferroptosis resistance. KDM4A and HIF1α were highly-expressed in CC tissues and positively correlated with each other. KDM4A knockdown attenuated cell resistance to Erastin, increased H3K9me3 level in the HIF1α promoter region, and downregulated HIF1α transcription and translation. H3K9me3 level was increased in the HIF1α promoter after hypoxia. HIF1α overexpression abrogated the function of KDM4A knockdown on ferroptosis in hypoxia conditions. Iron uptake/release and TfR1/DMT1 levels were increased after hypoxia. Hypoxia activated HRE sequence in TfR1 and DMT1 promoters. Conclusion. Hypoxia upregulated KDM4A, enhanced HIF1α transcription, and activated HRE sequence in TfR1 and DMT1 promoters via H3K9me3, thus inducing ferroptosis resistance in CC cells.

Green innovation has become the goal for promoting the transformation and upgrading heavy pollution industries in the context of high-quality development, and the key factor for the success of green innovation is increasing the green innovation efficiency of heavy pollution industries. To understand the current situation of China's industrial innovation and get out of the dilemma, we use non-expected Slacks-based model (SBM) to measure green innovation efficiency in Chinese industry, Lasso regression to screen the influencing factors of heavy pollution industries, tobit regression to study the influence degree and direction of different influencing factors on green innovation efficiency of heavy pollution industry. The results show that: (1) The green innovation efficiency of the 16 heavily polluting industries studied in this paper is generally low; (2) Coordination, green and openness all have a positive impact on the green innovation efficiency of the industry. (3) A certain degree of government scientific research support is conducive to improving the efficiency of industrial green innovation and exceeding the limit will have a restraining effect on enterprise innovation. According to the results, we put forward the corresponding policy implications.

The core problem of low energy consumption anaerobic fermentation reactor is that the reactor can make reasonable and effective use of energy from two aspects of quantity and quality, so as to ensure the anaerobic fermentation performance of the reactor and make it run efficiently and energy-saving. However, serious corrosion of metal for reactors was found during operation, and the selection of reactor materials became the key to restrict biogas production. In this paper, the corrosion characteristics of the four metals including Q235A steel, Q345A steel, 45# steel and 3Cr13 steel were determined by corrosion morphology, mechanical and electrochemical experiments. The results showed that the corrosion product particles of Q235A steel were polygonal, showing a good cross-linking feature, which was better than that of Q345A and 45# steels. However, there was no obvious boundary between corrosion product particles of 3Cr13 steel. The presence of Fe _3 C in the corrosion products of 3Cr13 steel and the observation of micro-cracks on the surface at nanometer scale indicated that the intergranular corrosion of 3Cr13 steel was dominated. For another aspect, with the increase of CO _2 and CH _4 content, the corrosion rate of the four metals was generally accelerated, in which the Q235A steel showed the best performance under different conditions. In addition, Q345A and 45# steels showed relatively good corrosion resistance, under which the total mole of mixed gas (CO _2 /H _2 O/CH _4 ) was lowest.

Background: The most severe insect damage to bamboo shoots is the bamboo-snout beetle (Cyrtotrachelus buqueti). Bamboo is a perennial plant that has significant economic value. C. buqueti also plays a vital role in the degradation of bamboo lignocellulose and causing damage. The genome sequencing and functional gene annotation of C. buqueti are of great significance to reveal the molecular mechanism of its efficient degradation of bamboo fiber and the development of the bamboo industry.Results: The size of C. buqueti genome was close to 600.92 Mb by building a one paired-end (PE) library and k-mer analysis. Then, we developed nine 20-kb SMRTbell libraries for genome sequencing and got a total of 51.12 Gb of the original PacBio sequel reads. Furthermore, after filtering with a coverage depth of 85.06×, clean reads with 48.71 Gb were obtained. The final size of C. buqueti genome is 633.85 Mb after being assembled and measured, and the contig N50 of C. buqueti genome is 27.93 Mb. The value of contig N50 shows that the assembly quality of C. buqueti genome exceeds that of most published insect genomes. The size of the gene sequence located on chromosomes reaches 630.86 Mb, accounting for 99.53% of the genome sequence. A 1,063 conserved genes were collected at this assembled genome, comprising 99.72% of the overall genes with 1,066 using the Benchmark Uniform Single-Copy Orthology (BUSCO). Moreover, 63.78% of the C. buqueti genome is repetitive, and 57.15% is redundant with long-term elements. A 12,569 protein-coding genes distributed on 12 chromosomes were acquired after function annotation, of which 96.18% were functional genes. The comparative genomic analysis results revealed that C. buqueti was similar to D. ponderosae. Moreover, the comparative analysis of specific genes in C. buqueti genome showed that it had 244 unique lignocellulose degradation genes and 240 genes related to energy production and conversion. At the same time, 73 P450 genes and 30 GST genes were identified, respectively, in the C. buqueti genome.Conclusion: The high-quality C. buqueti genome has been obtained in the present study. The assembly level of this insect’s genome is higher than that of other most reported insects’ genomes. The phylogenetic analysis of P450 and GST gene family showed that C. buqueti had a vital detoxification function to plant chemical components.

Abstract Background As an important biomass raw material, the lignocellulose in bamboo is of significant value in energy conversion. The conversion of bamboo lignocellulose into fermentable reducing sugar, i.e. the degradation of bamboo lignocellulose, is an important step in lignocellulose conversion. However, little research has focussed on excavating the enzymes and microbes that are related to the degradation of bamboo lignocellulose, which is important for its utilisation. This study used Cyrtotrachelus buqueti (bamboo snout beetle) to evaluate the efficiency of bamboo lignocellulose degradation. Results RNA sequencing was conducted to sequence the transcriptome of the insect before and after feeding on bamboo shoots. The expression levels of genes encoding several carbohydrate-active enzymes, such as endoglucanase (evgtrinloc27093t1 and evgtrinloc16407t0) and laccase (evgtrinloc15173t0 and evgtrinloc11252t0), were found to be upregulated after feeding. Faecal component analysis showed that the degradation efficiencies of cellulose, hemicellulose and lignin were 61.82%, 87.65% and 69.05%, respectively. After 6 days of co-culture with crude enzymes in vitro, the degradation efficiencies of cellulose, hemicellulose and lignin in bamboo shoot particles (BSPs) were 24.98%, 37.52% and 26.67%, respectively. These results indicated that lignocellulosic enzymes and related enzymes within the insect itself co-degraded bamboo lignocellulose. These finding can potentially be used for the pre-treatment and enzymatic hydrolysis of bamboo lignocellulose. Conclusion Our results showed that intestinal digestive enzymes from C. buqueti degraded bamboo shoot lignocellulose both in vivo and in vitro. In addition, the expression levels of many carbohydrate-active enzyme (CAZyme) genes were upregulated in the transcriptome, including those for cellulase, xylanase and ligninase genes. Therefore, we proposed a scheme for applying the lignocellulolytic enzymes from C. buqueti to degrade bamboo lignocellulose using genetic, enzymatic and fermentation engineering techniques to overexpress the lignocellulolytic enzymes genes in vitro and obtain large quantities of enzymes that could efficiently degrade bamboo lignocellulose and be used for lignocellulose bioconversion.

Abstract Background Gut symbiotic microbiota plays a critical role in nutrient supply, digestion, and absorption. The bamboo snout beetle, Cyrtotrachelus buqueti, a common pest of several bamboo species, exhibits high lignocellulolytic enzyme activity and contains various CAZyme genes. However, to date, no studies have evaluated the role of gut symbiotic microbiota of the snout beetle on bamboo lignocellulose degradation. Therefore, the present study investigated the role of gut symbiotic microbiota of C. buqueti on bamboo lignocellulose degradation. Results Gut symbiotic microbiota of female (CCJ), male (XCJ), and larvae (YCJ) beetles was used to treat bamboo shoot particles (BSPs) in vitro for 6 days. Scanning electron microscopy (SEM) revealed significant destruction of the lignocellulose structure after treatment, which was consistent with the degradation efficiencies of CCJ, XCJ, and YCJ for cellulose (21.11%, 17.58% and 18.74%, respectively); hemicellulose (22.22%, 27.18% and 34.20%, respectively); and lignin (19.83%, 24.30% and 32.97%, respectively). Gut symbiotic microbiota of adult and larvae beetles was then identified using 16sRNA sequencing, which revealed that four microbes: Lactococcus, Serratia, Dysgonomonas and Enterococcus, comprise approximately 84% to 94% of the microbiota. Moreover, the genomes of 45 Lactococcus, 72 Serratia, 86 Enterococcus and 4 Dysgonomonas microbes were used to analyse resident CAZyme genes. These results indicated that gut symbiotic microbiota of adult and larvae C. buqueti is involved in the lignocellulose degradation traits shown by the host. Conclusions This study shows that the gut symbiotic microbiota of C. buqueti participates in bamboo lignocellulose degradation, providing innovative findings for bamboo lignocellulose bioconversion. Furthermore, the results of this study will allow us to further isolate lignocellulose-degrading microbiota for use in bamboo lignocellulose bioconversion.

Pseudoregma bambucicola is an economically significant bamboo pest which is highly reproductive found in Eastern Asia that influences the normal growth of bamboo, induces the formation of sooty molds, and even causes substantial injury or death among young bamboos. This study provides a theoretical and practical basis for the comprehensive prevention and control of Pseudoregma bambucicola. The results showed that 10 geographical populations of Pseudoregma bambucicola showed high genetic diversity (Hd = 0.879 PI + 0.075, =0.08295 + 0.03901) and the average genetic distance was 0.018. In the haplotype phylogenetic tree constructed, the branches of MP tree and BI tree were basically the same, the Sichuan Basin was clustered into one and the populations from other provinces were clustered into a large branch and had a high posterior probability (the posterior probability of MP tree was 99% and the posterior probability of BI tree was 1). The NJ tree showed the differences among the 10 geographies. It was found that the experimental results were consistent with the actual geographical conditions because different geographical Pseudoregma bambusicola experienced natural isolation due to the possible impact of the natural environment, resulting in no genetic exchange between them, thus having a great genetic variability and high genetic diversity.

This article presents a hybrid method of structural modal parameter identification, based on improved empirical mode decomposition (EMD) and autoregressive and moving average (ARMA). Special attention is given to some implementation issues, such as the modal mixing, false modes, the judgment of the real intrinsic mode function (IMF) of classical EMD, and the difficulty of fixing the order of ARMA. To resolve the existing defects of EMD, an improved EMD (IEMD) that combines frequency band filtering and cluster analysis is proposed in this paper, where frequency band filtering divides the signal into several narrowband signals before the EMD process, and cluster analysis is used to determine the real IMFs. Euclidean distance is used to cluster the decomposition results, with no need to adjust any indexes or thresholds, and only by means of using the nearest distance to efficiently determine the real IMF. Moreover, IEMD is used as a pre-processing tool for ARMA, to resolve the difficulty of fixing its order. The capabilities of the proposed method were compared and assessed using a numerical simulation and an experimental model. The numerical simulation and experimental results showed that the improved method could resolve the modal mixing and false modal problems in the classical EMD process and could automatically identified the real IMFs, while the proposed IEMD was combined with ARMA to successfully identify the frequency and mode shape of the structure. Additionally, since each IMF is a single component signal, it is easy to determine the order of the ARMA model.
