Lignocellulosic biomass (LCB) comprising of wheat bran, coconut husk, rice husk, cereals straw, and other hardwood and softwoods is a good source for the production of xylooligosaccharides (XOS) (prebiotic). XOS produced are nondigestible carbohydrates being stable under stomach pH and digestive enzymes so they can be easily delivered to the intestine in native form, thus stimulating the growth of probiotics. Here we review about the raw material, production, purification, and application of XOS with health benefits. Importance of XOS being valuable food ingredient is increasing as they perform a variety of functions, including reduction in cholesterol levels, gastrointestinal health maintenance, anticancer and antioxidant properties, and modulation of immune system. We also discuss the different characterization methods which are necessary to determine the degree of polymerization (DP) of XOS. Low DP (xylobiose and xylotriose) is usually preferred for the application of XOS in various sectors. Waterlogging represents a substantial agricultural concern, inducing harmful impacts on crop development and productivity. In the present study, 142 diverse sesame genotypes were examined during the early vegetative phase to assess their response under waterlogging conditions. Based on the severity of symptoms observed, 2 genotypes were classified as highly tolerant, 66 as moderately tolerant, 69 as susceptible, and 5 as highly susceptible. Subsequent investigation focused on four genotypes, i.e., two highly tolerant (JLT-8 and GP-70) and two highly susceptible (R-III-F6 and EC-335003). These genotypes were subjected to incremental stress periods (0 h, 24 h, 48 h, 72 h, and 96 h) to elucidate the biochemical basis of tolerance mechanisms. Each experiment was conducted as a randomized split-plot design with three replications, and the statistical significance of the treatment differences was determined using the one-way analysis of variance (ANOVA) followed by the Fisher least significant difference (LSD) test at p ≤ 0.05. The influence of waterlogging stress on morphological growth was detrimental for both tolerant and susceptible genotypes, with more severe consequences observed in the latter. Although adventitious roots were observed in both sets of genotypes above flooding levels, the tolerant genotypes exhibited a more rapid and vigorous development of these roots after 48 h of stress exposure. Tolerant genotypes displayed higher tolerance coefficients compared to susceptible genotypes. Furthermore, tolerant genotypes maintained elevated antioxidant potential, thereby minimizing oxidative stress. Conversely, susceptible genotypes exhibited higher accumulation of hydrogen peroxide (H2O2) and malondialdehyde content. Photosynthetic efficiency was reduced in all genotypes after 24 h of stress treatment, with a particularly drastic reduction in susceptible genotypes compared to their tolerant counterparts. Tolerant genotypes exhibited significantly higher activities of anaerobic metabolism enzymes, enabling prolonged survival under waterlogging conditions. Increase in proline content was observed in all the genotypes indicating the cellular osmotic balance adjustments in response to stress exposure. Consequently, the robust antioxidant potential and efficient anaerobic metabolism observed in the tolerant genotypes served as key mechanisms enabling their resilience to short-term waterlogging exposure. These findings underscore the promising potential of specific sesame genotypes in enhancing crop resilience against waterlogging stress, offering valuable insights for agricultural practices and breeding programs. Amaranth is an annual or short-perennial, dicotyledonous plant, globally cultivated for its grains and leaves. The plant is recognized as one of the earliest domesticated vegetables with exceptional nutraceutical and therapeutic properties. The present investigation aims to evaluate the genetic diversity and breeding potential of 22 vegetable amaranthus genotypes. The experiment was conducted in randomized block design with three replications during summer 2023, with observations recorded on 14 agro-morphological traits. The analysis of variance revealed significant differences for all the traits, depicting a wide range of variation for these traits. Further, the genotype VRAM-45 was found most promising for leaf yield, while VRAM-44 excelled for seed yield. A significant and positive correlation exists between leaf yield/plant, leaf length and leaf width, implying a potential link between leaf area and the efficiency of photosynthesis. Principal component analysis highlighted the significance of the first two components, explaining a substantial 53.71% of the total variation. Traits such as plant height at the vegetative stage, leaf length, and days to 50% flowering emerged as significant contributors to the variance. Clustering analysis categorized the 22 genotypes into four clusters. Cluster IV, though was found monogenotypic, but found superior for most of the economic traits, followed by cluster III. Thus, focusing on these clusters in breeding or selection programs could lead to more favorable outcomes in terms of economic productivity and other desirable characteristics. [ABSTRACT FROM AUTHOR]

Globally, abiotic stresses, such as temperature (heat or cold), water (drought and flooding), and salinity, cause significant losses in crop production and have adverse effects on plant growth and development. A variety of DNA-based molecular markers, such as SSRs, RFLPs, AFLPs, SNPs, etc., have been used to screen germplasms for stress tolerance and the QTL mapping of stress-related genes. Such molecular-marker-assisted selection strategies can quicken the development of tolerant/resistant cultivars to withstand abiotic stresses. Oilseeds such as rapeseed, mustard, peanuts, soybeans, sunflower, safflower, sesame, flaxseed, and castor are the most important source of edible oil worldwide. Although oilseed crops are known for their capacity to withstand abiotic challenges, there is a significant difference between actual and potential yields due to the adaptation and tolerance to severe abiotic pressures. This review summarizes the applications of molecular markers to date to achieve abiotic stress tolerance in major oilseed crops. The molecular markers that have been reported for genetic diversity studies and the mapping and tagging of genes/QTLs for drought, heavy metal stress, salinity, flooding, cold and heat stress, and their application in the MAS are presented. Iron (Fe) and zinc (Zn) contents in hexaploid wheat are very low and are further reduced because of the removal of micronutrient-rich bran of wheat grains during milling and processing. Therefore, hexaploid wheat, its wild species and wheat–Aegilops kotschyi substitution lines were evaluated to identify the genome(s) carrying gene(s) for high Fe and Zn concentrations in bran and endosperm fractions of grains. It is reflected from the results that Triticum monococcum (acc. W463) may serve as a promising donor for biofortification of Fe, and Aegilops speltoides (acc. 3804) may serve as a promising donor for biofortification of Zn in the endosperm of cultivated wheat. Further, among the three wheat–Ae. kotschyi substitution lines, the higher concentration of Fe and Zn in endosperm fraction was observed in BC2F4 63-2-13-1. PurposeTo transfer the 2S chromosomal fragment(s) ofAegilops kotschyi(2Sk) into the bread wheat genome which could lead to the biofortification of wheat with high grain iron and zinc content. Materials and methodsWheat-Ae. kotschyi2A/2Sksubstitution lines with high grain iron and zinc content were used to transfer the gene/loci for high grain Fe and Zn content into wheat using seed irradiation approach. ResultsBread wheat plants derived from 40 krad-irradiated seeds showed the presence of univalents and multivalents during meiotic metaphase-I. Genomic in situ hybridization analysis of seed irradiation hybrid F2seedlings showed several terminal and interstitial signals indicated the introgression ofAe. kotschyichromosome segments. This proves the efficacy of seed radiation hybrid approach in gene transfer experiments. All the radiation-treated hybrid plants with high grain Fe and Zn content were analyzed with wheat group 2 chromosome-specific polymorphic simple sequence repeat markers to identify the introgression of small alien chromosome fragment(s). ConclusionRadiation-induced hybrids showed more than 65% increase in grain iron and 54% increase in Zn contents with better harvest index than the elite wheat cultivar WL711 indicating effective and compensating translocations of 2Skfragments into wheat genome. Thirty different wheat genotypes, including landraces, cultivars, and related wild species, were analyzed for puroindoline genes and proteins. Full-length Pina and Pinb genes were amplified using gene-specific markers where 13 genotypes amplified both genes, while 17 genotypes had either one or none of the puroindoline genes. None of the puroindoline genes were detected in any of the Triticum durum cultivars. Genotypes with both puroindoline genes had a soft grain texture, while other genotypes had hard grains. Related nonprogenitor species with A, C, M, U, and S genomes also showed the presence of either one or both the puroindoline genes, indicating that these species can also be utilized for development of extra-soft wheat. SDS-PAGE analysis of selected wheat genotypes revealed that puroindoline proteins are approximately 13-14 kDa and the protein results were in agreement with the PCR results. Puroindoline proteins of selected 3 soft wheat genotypes HPW 89, Naphal, and IITR-9 also exhibited antimicrobial activity against gram-positive and gram-negative bacteria. Both genes of a soft wheat cultivar, HPW 89, were successfully cloned in the Escherichia coli DH-5α strain. ABSTRACT Legumes represent a significant source of protein, roughage, minerals and carbohydrates in global diets. They are cultivated primarily for human consumption, forage and as a green manure. However, the growth and productivity of legumes are often hindered by a range of biological and environmental factors. The most prevalent diseases afflicting legumes include downy mildew, fusarium root rot, southern blight and common leaf spot. Additionally, environmental stresses such as salinity and drought represents significant challenges. Over time, the development of cultivars with enhanced tolerance has emerged as a sustainable approach to combat the impact of these stresses. Although environmentally friendly, traditional breeding methods entail lengthy screening and cross‐breeding protocols, which constrain their efficacy in addressing climate challenges and ensuring global food security. Furthermore, these approaches rely on the natural availability of genetic variation, which may not always be exist for specific traits, particularly in the context of rapidly changing environmental conditions. Additionally, the traditional breeding method is not sufficiently precise, which can result in the unintended introduction of undesirable traits. Furthermore, the genetic intricacy of legumes presents an additional challenge making it difficult to isolate and enhance desired traits. These constrain impede the capacity to expeditiously develop cultivars that can withstand emerging stresses, such as climate change. Genome editing has emerged as a powerful tool to overcome the limitations of traditional breeding methods. By enabling precise and targeted genetic manipulation, genome editing can enhance desired traits or introduce new ones, thereby facilitating the development of climate‐resilient agronomic varieties. In particular, clustered regularly interspaced short palindromic repeat/CRISPR associated protein (CRISPR/Cas9)‐mediated editing has demonstrated considerable potential in enhancing legume adaptability to diverse stress conditions and increasing crop yield. This study highlights the advancements in genome editing, with a specific focus on CRISPR/Cas9 technology, as a means to improve legume crops' resilience and productivity in response to environmental pressures. An adaptive catalytic system that provides control over the nitroarene hydrogenation network to prepare a wide range of aniline and hydroxylamine derivatives is presented. This system takes advantage of a delicate interplay between a rhodium(III) center and a Lewis acidic borane introduced in the secondary coordination sphere of the metal. The high chemoselectivity of the catalyst in the presence of various potentially vulnerable functional groups and its readiness to be deployed at a preparative scale illustrate its practicality. Mechanistic studies and density functional theory (DFT) methods were used to shed light on the mode of functioning of the catalyst and elucidate the origin of adaptivity. The competition for interaction with boron between a solvent molecule and a substrate was found crucial for adaptivity. The role of oxidative stress management was evaluated in two maize (Zea mays L.) genotypes - Parkash (drought-resistant) and Paras (drought-sensitive), subjected to drought stress during reproductive stage. Alterations in their antioxidant pools - glutathione (GSH) and ascorbic acid (AsA) combined with activities of enzymes glutathione reductase (GR), ascorbate peroxidase (APX), peroxidase (POX) and catalase (CAT) involved in defense against oxidative stress and stress parameters, namely chlorophyll (Chl), hydrogen peroxide (H2O2) and malondialdehyde (MDA) were investigated in flag leaves from silk emergence till maturity. The drought caused transient increase in GR, APX, POX and CAT activities in drought-tolerant genotype (Parkash) which decreased at later stages with the extended period of drought stress. However, in Paras, drought stress caused decrease in activities of GR and CAT from initial period of stress till the end of experiment, except for POX which showed slight increase in activity. A significant increase in GSH content was observed in Parkash till 35 days after silking (DAS), whereas in Paras, GSH content remained lower than irrigated till maturity. Parkash which had higher AsA and Chl contents, also showed lower H2O2 and MDA levels than Paras under drought stress conditions. However, at the later stages, decline in antioxidant enzyme activities in Parkash due to severe drought stress led to enhanced membrane damage, as revealed by the accumulation of MDA. Our data indicated that significant activation of antioxidant system in Parkash might be responsible for its drought-tolerant behavior under drought stress and helped it to cope with the stress up to a definite period. Thus, the results indicate that antioxidant status and lipid peroxidation in flag leaves can be used as indices of drought tolerance in maize plants and also as potential biochemical targets for the crop improvement programmes to develop drought-tolerant cultivars.
