5-HT and noradrenaline are important neurotransmitters that control increase in body mass and are involved in the pathophysiology of obesity and depression. Sibutramine, an established anti-obesity agent, and duloxetine, an anti-depressant agent, are serotonin noradrenaline reuptake inhibitors (SNRIs). The objective of the present study was to compare the anti-obesity effect of duloxetine with sibutramine along with its effect on blood pressure and depression in obese rats. The secondary objective of the study was to determine if a relationship exists between obesity and depression. Obesity was induced by high-fat diet (HFD) in healthy male Sprague-Dawley rats. After 5 weeks of feeding HFD, animals were overweight (17.57%) with high food intake (57.15%) in comparison with normal animals. These obese animals were treated with duloxetine (30 mg·kg-1, p.o.) and sibutramine (5 mg·kg-1, p.o.) for 4 weeks. Control animals were treated with duloxetine alone (30 mg·kg-1, p.o.). Our results depict that duloxetine was as effective as sibutramine in reducing food intake, body mass, and relative adiposity, and increasing rectal temperature with an added advantage of decreasing blood pressure, which sibutramine failed to do. Besides reduction in body mass, unlike sibutramine, duloxetine improved depressive state as evaluated by despair swimming test, tail suspension test, and open field test, speculating its use as an anti-obesity agent in obese-depressive animals. Since obese control animals reflected decreased locomotor activity, a positive relationship can be speculated to exist between obesity and depression. Participatory modelling is an emerging approach in the decision-making process through which stakeholders contribute to the representation of the perceived causal linkages of a complex system. The use of fuzzy cognitive maps (FCMs) for participatory modelling helps policy-makers develop dynamic quantitative models for strategising development interventions. The aggregation of knowledge from multiple stakeholders provides consolidated and more reliable results. Average aggregation is the most common aggregation method used in FCMs-based modelling for weighted interconnections between concepts. This paper proposes a new aggregation method using learning OWA (ordered weighted averaging) operators for aggregating FCM weights assigned by various stakeholders. Besides, we report a comparative analysis of 'OWA learning aggregation' with the conventional average aggregation method, while evaluating the theory of change for the world's most extensive poverty alleviation programme in India. The results of the FCMWizard web-based tool show that the proposed method provides an opportunity to policy-makers for evaluating outcomes of proposed policies while addressing social resilience and economic mobility., Competing Interests: The authors have declared that no competing interests exist.

A slew of participatory and community-demand-driven approaches have emerged in order to address the multi-dimensional nature of poverty in developing nations. The present study identifies critical factors responsible for poverty alleviation in India with the aid of fuzzy cognitive maps (FCMs) deployed for showcasing causal reasoning. It is through FCM-based simulations that the study evaluates the efficacy of existing poverty alleviation approaches, including community organisation based micro-financing, capability and social security, market-based and good governance. Our findings confirm, to some degree, the complementarity of various approaches to poverty alleviation that need to be implemented simultaneously for a comprehensive poverty alleviation drive. FCM-based simulations underscore the need for applying an integrated and multi-dimensional approach incorporating elements of various approaches for eradicating poverty, which happens to be a multi-dimensional phenomenon. Besides, the study offers policy implications for the design, management, and implementation of poverty eradication programmes. Background: The present study was designed as an open label, multiple-dose, randomized, parallel trial to evaluate the pharmacodynamic drug-drug interaction of lisinopril and concomitantly administered diclofenac sodium in non-diabetic and diabetic, mild to moderate hypertensive, osteoarthritic patients., Methods: Post-screening and on inclusion, patients were put on a 2-week washout period and then randomly assigned to either only lisinopril 10 mg or combination of lisinopril 10 mg and diclofenac sodium 100 mg treatments for 8-12 weeks in diseased states of hypertension and osteoarthritis with or without type 2 diabetes mellitus., Results: The blood pressure (BP) control with lisinopril was reduced by concomitantly administered diclofenac sodium in non-diabetic (SBP: p=0.00002; DBP: p=0.000008) and diabetic (SBP: p=0.002; DBP: p=0.001) patients when compared with the patients receiving lisinopril alone. Insulin sensitivity was improved (p=0.00002) and urinary albumin excretion rate was better controlled (p=0.0096) in lisinopril-treated patients when compared with the combination treatment in diabetic pool. Serum creatinine levels increased significantly in non-diabetic patients (p=0.00004) receiving combination treatment. In addition, creatinine clearance (CLCR) and blood urea nitrogen (BUN) were significantly higher in diabetic (CLCR: p<0.00001; BUN: p=0.0098) as well as in non-diabetic (CLCR: p<0.00001; BUN: p=0.03) patients treated with combination treatment. The alterations in serum electrolytes, reduction in % platelet aggregation activity and improvement in lipid profile was more profound with combination treatment in comparison to lisinopril alone., Conclusions: The antihypertensive efficacy and insulin sensitivity improving property of lisinopril along with the renal function might get worse in hypertensive osteoarthritic patients receiving concomitant treatment of oral diclofenac sodium with lisinopril. In addition to this, close monitoring of serum electrolytes is also suggested to rule out any long-term detrimental effect.
