The soil nematode Caenorhabditis elegans was used in 24-h acute exposures to arsenic (As), copper (Cu) and glyphosate (GPS) and to mixtures of As/Cu and As/GPS to investigate the effects of mixture exposures in the worms. A synergistic type of interaction was observed for acute toxicity with the As/Cu and As/GPS mixtures. Sublethal 24-h exposures of 1/1000, 1/100 and 1/10 of the LC50 concentrations for As, Cu and GPS individually and for As/Cu and As/GPS mixtures were conducted to observe responses in locomotory behavior (head thrashing), reproduction, and heat shock protein expression. Head thrash frequency and reproduction exhibited concentration dependent decreases in both individual and combined exposures to the tested chemical stressors, and showed synergistic interactions even at micromolar concentrations. Furthermore, the HSP70 protein level was significantly increased following exposure to individual and combined chemical stressors in wild-type C. elegans. Our findings establish for the first time the effects of exposure to As/GPS and As/Cu mixtures in C. elegans.

The effects of di-(2-ethylhexyl) phthalate (DEHP) at five different doses from 10 to 1000 mg kg−1 soil on biological properties were investigated over a period of 56 days. Meanwhile, the dissipation of DEHP was also monitored. The results indicated that the microbial biomass C (Cmic) fluctuated at around 70 mg kg−1 soil for the control, whereas the Cmic varied significantly for the soil samples contaminated by DEHP. The catalase activities in all five treatments were stimulated at most time, and the activities of phosphatase in the soils treated by DEHP with 500 mg kg−1 or 1000 mg kg−1 were significantly higher than the other treatments from the 20th day. Urease was more sensitive and inhibited significantly during the initial period of incubation. Additionally, the dose–response relationship of invertase was presented in the later phase of incubation. The activities of urease and invertase might indicate soil perturbations caused by the introduction of DEHP. The dissipation of DEHP was found to follow the pseudo first-order kinetics behavior.

The key of tissue section technique is the selection and application of the embedding agent. Based on the specific characteristics of the tissue and outer skeleton of insects, a new kind of embedding agent, colophony and paraffin (CP), was used without destroying the integrity of insect organs. The tissue can be sliced continuously by the use of CP embedding technique. It solves the 3 difficult problems in the serial section of the organ and tissue of insect, namely stiff materials, thin section and continuity of sections. These difficult problems are not solved by any unitary tissue section technique that is used now, including the paraffin section technique. By using the CP embedding technique, any parts or organs (like antenna, leg and wing) and even the whole body of insects (mini body) can be sliced up continuously. It will open up wide possibilities for the study of inner structure, the interrelations among organs, the micro-characteristics and the physiological functions of insects systematically.

Immediately following the discovery of tryptophan hydroxylase in Drosophila, we demonstrated the presence of tryptophan hydroxylase in the brain of the beetle Harmonia axyridis (Coleoptera: Coccinellidae). However, whether tryptophan hydroxylase is present in the brains of other insects is still a matter of discussion. In the current study, sheep anti-tryptophan hydroxylase polyclonal antibody has been applied to test for tryptophan hydroxylase immunoreactivity in a broader taxonomic range of insect brains, including holometabolous and hemimetabolous insects: one species each of Coleoptera, Hymenoptera, Diptera, and Blattaria, and two species of Lepidoptera. All species show consistent tryptophan hydroxylase immunoreactivity with distribution patterns matching that of serotonin. The immuno-positive results of such an antibody in brains from diverse orders of insects suggest that specific tryptophan hydroxylase responsible for central serotonin synthesis is probably present in the brains of all insects.

In this work we report a novel scalable strategy to prepare a lithium-air battery electrode from 3D N-doped pierced graphene microparticles (N-PGM) with highly active performance. This approach has combined the merits of spray drying technology and the hard template method. The pierced structured graphene microparticles were characterized physically and electrochemically. An x-ray photoelectron spectrometer and Raman spectra have revealed that the novel structure possesses a higher N-doping level than conventional graphene without the pierced structure. A much higher BET surface area was also achieved for the N-PGM than the conventional N-doped graphene microparticles (N-GM). Cyclic voltammetry indicated that the lithium-air battery with the N-PGM electrode has a better utilization for the graphene mass and a higher void volume for Li2O2 formation than that of the N-GM electrode. N-PGM also exhibits improved decomposition kinetics for Li oxide species yielded in the cathodic reaction. Charge and discharge measurements showed that the N-PGM lithium-air battery achieved an improved specific capacity and an enhanced cycle performance than when an N-GM electrode is used.

Rabbit anti-serotonin and mouse monoclonal anti-tryptophan hydroxylase antisera were applied on the brain sections of the beetle Harmonia axyridis, butterfly Childrena zenobia, moth Antheraea pernyi and ant Camponotus japonicus, using the Streptavidin-Peroxidase immunohistochemical method and Colophony-Paraffin embedded section technique. Results revealed that all the experimental insects showed notable serotonin-like immunoreactivity in the brain. However, only the brain sections of the beetle H. axyridis were strongly labeled by mouse monoclonal anti-tryptophan hydroxylase antiserum, with the distribution pattern matching that of serotonin. These results demonstrate that specific tryptophan hydroxylase may exist in the brain of the beetle and likely reflect the diversity of serotonin synthetic mechanisms as well as the evolution of aromatic amino acid hydroxylase genes.
