Ants are an important arthropod group due to their involvement in ecological processes amongst others as ecosystem engineers or predators, but some invasive alien species are also implicated in detrimental environmental, economic and human health effects. Despite recent advancements, the ant biodiversity of Cyprus is still in need of further research with previous online species inventories synthesising a checklist of just 65 native and 10 alien species. The “Ants of Cyprus” website (https://sites.google.com/view/ants-of-cyprus) aims to: (1) raise public awareness and increase local knowledge on the biodiversity and ecological significance of ants, (2) provide ecological data and (3) enhance conservation efforts and management practices for native and alien ant species, respectively. This dynamic, user-friendly website has been designed for use by the scientific community, conservation officers and the wider public and it will be regularly updated as new information and data are generated. Additionally, this information will be critical for reporting on progress towards biodiversity targets and goals.

Biodiversity data platforms including databases, websites and data repositories underpin conservation efforts by collecting spatiotemporal data of discovered native and alien species and maps of their distributions. Chalcid wasps (Hymenoptera, Chalcidoidea) are one of the most diverse insect groups estimated to include half a million species. Being mostly parasitoids of other arthropods, they have been successfully used as biological control agents against serious agricultural pests worldwide. In Cyprus, only 124 species of chalcid wasps have been recorded, with 53 species being alien to the island. Their true biodiversity is predicted to be much larger because the island is both under-sampled and under-researched. A number of biodiversity data platforms focusing on the biodiversity of Cyprus are currently online; however, an online platform dedicated on the chalcid wasps of Cyprus is lacking. In the framework of the Darwin Plus Fellowship (DPLUS202) “Species richness and biological invasions of Chalcid wasps in Akrotiri Peninsula”, the “Chalcid wasps of Cyprus” website (https://sites.google.com/view/chalcidwaspscyprus) is presented. This online, dynamic database aims to: (1) raise public awareness regarding a rather neglected and yet ecologically important insect group, (2) provide data on the morphology, ecology and biodiversity of Chalcidoidea on Cyprus, as well as (3) promote conservation needs by setting a baseline for the future assessment of both native and alien chalcid wasp species. This online platform will be regularly revised in order to provide an up-to-date, user-friendly digital environment to the scientific community, policy-makers and citizens.

Abstract For high rate water electrolysers, minimising Ohmic losses through efficient gas bubble evacuation away from the active electrode is as important as minimising activation losses by improving the electrode’s electrocatalytic properties. In this work, by a combined experimental and computational fluid dynamics (CFD) approach, we identify the topological parameters of flow-engineered 3-D electrodes that direct their performance towards enhanced bubble evacuation. In particular, we show that integrating Ni-based foam electrodes into a laterally-graded bi-layer zero-gap cell configuration allows for alkaline water electrolysis to become Proton Exchange Membrane (PEM)-like, even when keeping a state-of-the-art Zirfon diaphragm. Detailed CFD simulations, explicitly taking into account the entire 3-D electrode and cell topology, show that under a forced uniform upstream electrolyte flow, such a graded structure induces a high lateral velocity component in the direction normal to and away from the diaphragm. This work is therefore an invitation to start considering PEM-like cell designs for alkaline water electrolysis as well, in particular the use of square or rectangular electrodes in flow-through type electrochemical cells.

Umbilical cord blood (UCB) T cells exhibit distinct naive ontogenetic profiles and may be an attractive source of starting cells for the production of chimeric antigen receptor (CAR) T cells. Pre-selection of UCB-T cells on the basis of CD62L expression was investigated as part of a machine-based manufacturing process, incorporating lentiviral transduction, CRISPR-Cas9 editing, T-cell expansion and depletion of residual TCReeeT cells. This provided stringent mitigation against the risk of graft versus host disease (GVHD), and was combined with simultaneous knockout of CD52 to enable persistence of edited T cells in combination with preparative lymphodepletion using Alemtuzumab. Under compliant manufacturing conditions, two cell banks were generated with high levels of CAR19 expression and minimal carriage of TCReeeT cells. Sufficient cells were cryopreserved in dose-banded aliquots at the end of each campaign to treat dozens of potential recipients. Molecular characterisation captured vector integration sites and CRISPR editing signatures and functional studies, including in vivo potency studies in humanised mice, confirmed antileukaemic activity comparable to peripheral blood-derived universal CAR19 T cells. Machine manufactured UCB derived T cells banks offer an alternative to autologous cell therapies and could help widen access to CAR T cells.

Climate change is a major threat to the Middle East and North Africa (MENA) region, which can cause significant harm to its plant and animal species. We predicted the habitat distribution of Cataglyphis nodus (Brullé, 1833) in MENA using MaxEnt models under current and future climate conditions. Our analysis indicates that the cooler regions of the MENA are projected to experience temperature increases of 1–2 °C by 2040 and 2–4 °C by the 2070s. Similarly, the warmer regions may anticipate rises of 0.5–2 °C by 2040 and 2–4 °C by the 2070s. MaxEnt model results for the current climate show good agreement with observations (mean area under the curve value of 0.975 and mean true statistical skill value of 0.8), indicating good potential habitat suitability for C. nodus. Significant factors affecting habitat suitability are elevation, mean monthly precipitation of the coldest quarter, temperature seasonality, and precipitation amount of the driest month. The research predicts that under Shared Socioeconomic Pathway (SSP) 1.2.6, the habitat suitability area may increase by 6% in 2040, while SSP 3.7.0 (0.3%) and SSP 5.8.5 (2.6%) predict a decrease. For 2070, SSP 5.8.5 predicts a 2.2% reduction in habitat suitability, while SSP 1.2.6 (0.4%) and SSP 3.7.0 (1.3%) predict slight increases. The results provide insight into the potential impacts of climate change on the species and regional biodiversity changes associated with the projected species distribution.

Messor is a diverse genus of Myrmicinae with 168 extant species and subspecies. In the Mediterranean, some of its taxa historically were classified as members of the Messor instabilis group (sensu Santschi), of which 19 are known from the eastern Mediterranean. Here, the Messor semirufus complex of the Balkan Peninsula that assembles a distinct subsection of members of the instabilis group is defined and treated. In total, five species are recorded, including three that are new. Messor atanassovii Atanassov, 1982 is redescribed and confirmed for Bulgaria (Thracian Plain, Struma, and Mesta Valley, Pirin Mt., and Eastern Rhodopi) and Greece (Epirus, Ionian Islands, Central and Eastern Macedonia, and Thraki). Three species are described as new to science: Messor danaes Salata, Georgiadis & Borowiec, sp. nov. (Cyclades: Serifos), Messor kardamenae Salata & Borowiec, sp. nov. (Dodecanese: Kos, Nisyros, Rhodes, and Tilos), and Messor veneris Salata, Georgiadis & Borowiec, sp. nov. (Cyclades: Milos). The fifth member of the complex, Messor creticus Borowiec & Salata, 2019, maintains its status of Cretan endemic.

Biological invasions are considered a major driver of biodiversity loss, particularly on islands. Invasive alien ants can often have severe consequences on native biodiversity. Here, we review published and new information on alien ant species found on the Mediterranean island of Cyprus, a biodiversity hotspot. Our checklist of alien ants of Cyprus includes a total of 17 species, of which nine are reported from Cyprus for the first time (*): Camponotus cf. vitiosus Smith, Cardiocondyla mauritanica Forel, 1890, Cardiocondyla obscurior Wheeler, W.M., 1929*, Hypoponera punctatissima (Roger, 1859)*, Monomorium bicolor Emery, 1877, Nylanderia jaegerskioeldi (Mayr, 1904), Paratrechina longicornis (Latreille, 1802), Pheidole fadli Sharaf, 2007*, Pheidole indica Mayr, 1879, Solenopsis sp. (thief ant)*, Tetramorium bicarinatum (Nylander, 1846)*, Tetramorium caldarium (Roger, 1857)*, Tetramorium immigrans Santschi, 1927*, Tetramorium lanuginosum Mayr, 1870*, Trichomyrmex destructor (Jerdon, 1851), Trichomyrmex mayri (Forel, 1902)*, and Wasmannia auropunctata (Roger, 1863). We did not include three previously reported alien species for which we could not find supporting specimens [Monomorium pharaonis (Linnaeus, 1758), Nylanderia vividula (Nylander, 1846), Solenopsis geminata (Fabricius, 1804)], one based on a previous misidentification [Cardiocondyla nuda (Mayr, 1866)], and two species now considered native to Cyprus [Hypoponera eduardi (Forel, 1894), Monomorium subopacum (F. Smith, 1858)]. Literature records, specimens from field surveys and museum collections, the geographic origin of species, occupied habitats in Cyprus, and notes on invasiveness (spread and impact) are presented for each species. An identification key to distinguish alien from native ant species in Cyprus is provided, including widespread alien ants not yet known from Cyprus in order to support early detection, monitoring, and management efforts.

Biological invasions represent one of the main drivers of biodiversity loss with adverse impacts on human societies, economies and public health. More than 500 ant species have been transported outside their native range with the help of humans, while the majority of them have managed to establish viable populations in the wild. Nevertheless, data from the Mediterranean region suggest that most alien ants occupy anthropogenic habitats with little spread in semi-natural and natural habitats. Research on biological invasions of ants in Greece had previously identified a total of 15 alien ant species. In this article, an extensive literature investigation and material examination provide a revised checklist of the alien myrmecofauna of Greece. Although the number of alien ant species remains the same, the checklist’s composition is largely altered to provide an up-to-date overview of the country’s alien myrmecofauna in order to enhance management decisions and future research. The presence and distribution of alien ants within Greek administrative divisions, NATURA 2000 sites and Corine Land Cover types are analysed and presented. In particular, the species richness of alien ants seems to be highest in the Aegean Archipelago (Crete and Southern Aegean Islands) probably due to uneven collecting efforts and increased climatic suitability. Alien ant species are mostly associated with anthropogenic habitats including urban and agricultural areas, although a significant percentage has managed to spread into forest and semi-natural areas, including protected NATURA 2000 sites. Future research directions enhancing the monitoring of alien ants and their impacts are indicated to safeguard native ant biodiversity and conservation efforts of rare and endemic taxa.

Lyciasalamandra species, like most amphibians, secrete a wide array of compounds from their granular and mucous skin glands, including the internally synthesized samandarine alkaloids, making their skin a complex organ performing a variety of functions. Lyciasalamandra helverseni and L. luschani basoglui are insular endemics of the Dodecanese islands of SE Greece, bearing distinct isolated populations, with well-documented phylogenetic profiles. Here, we employ a metabolomics approach, utilizing UPLC-ESI-HRMS/MS data of the skin secretions sampled from a number of specimens found in the islands of Karpathos, Kasos and Kastellorizo, in an effort to reveal aspects of their chemistry and diversity across populations. The results indicated statistically significant variation between all taxa examined, based on various secreted compounds. The underlying factors of variation highlighted by the multivariate analysis were differences in samandarine and other alkaloid content as well as in animal size. Metabolite annotation, based on dereplication tools and most importantly HRMS and HRMS/MS spectra, yielded a number of known samandarine alkaloids, reported for the first time in the currently studied Lyciasalamandra species. We also present documentation for novel members of the samandarine alkaloid family, as well as preliminary evidence for a possible dietary alkaloid sequestration. This work can set the basis for further research of this often-neglected endemic species of the Salamandridae, as well as the structural investigation of the samandarine alkaloid group.

A dead individual of Metamasius hemipterus (Linnaeus, 1758) was collected by a citizen scientist in Athens, Greece from a hand of imported bananas. The equatorial origin of the fruit as well as the extensive growth of a fungus on the specimen, both suggest an unintentional introduction as a contaminant. Nevertheless, this record highlights the valu-able contribution of citizen scientists in the study of alien species. Although the species is not presently considered as established in the country, citizen-science initiatives in order to raise awareness and monitor its presence are ongoing.

The Nearctic Sceliphron caementarium (Drury, 1773) is widely distributed in the Western Palearctic and is spreading to new territories. Despite the large quantities of data on citizen-science platforms, these records have been mostly overlooked. In this publication, the first records of S. caementarium from six Eastern European and Balkan countries (Albania, Greece, Montenegro, Poland, Serbia, and Slovakia) are presented. Data derived from online citizen-science observations and museum specimens supplement our knowledge of this species’ range. The distribution of this species in Europe and possible ecological implications are discussed.

Recessive dystrophic epidermolysis bullosa is a debilitating blistering skin disorder caused by loss-of-function mutations in COL7A1, which encodes type VII collagen, the main component of anchoring fibrils at the dermal−epidermal junction. Although conventional gene therapy approaches through viral vectors have been tested in preclinical and clinical trials, they are limited by transgene size constraints and only support unregulated gene expression. Genome editing could potentially overcome some of these limitations, and CRISPR/Cas9 has already been applied in research studies to restore COL7A1 expression. The delivery of suitable repair templates for the repair of DNA cleaved by Cas9 is still a major challenge, and alternative base editing strategies may offer corrective solutions for certain mutations. We show highly targeted and efficient cytidine deamination and molecular correction of a defined recessive dystrophic epidermolysis bullosa mutation (c.425A>G), leading to restoration of full-length type VII collagen protein expression in primary human fibroblasts and induced pluripotent stem cells. Type VII collagen basement membrane expression and skin architecture were restored with de novo anchoring fibrils identified by electron microscopy in base-edited human recessive dystrophic epidermolysis bullosa grafts recovered from immunodeficient mice. The results show the potential and promise of emerging base editing technologies in tackling inherited disorders with well-defined single nucleotide mutations.

Native to the Neotropics, Wasmannia auropunctata (Roger, 1863) has been unintentionally introduced around the world, heavily impacting native ant biodiversity, societies, and economies as well as human and animal health due to its potentially dangerous stings. Herein we report on the first record of W. auropunctata in Cyprus. Specimens were collected from plant nurseries and tourist facilities in Paphos and Limassol district. Wasmannia auropunctata is believed to spread via the horticultural pathway to locations with sufficient humidity. Further research is necessary to determine the distribution and assess possible negative impacts of W. auropunctata to native biodiversity, society, the economy and human health in Cyprus.

Alien invasive species not only have adverse environmental impacts, they may also pose socioeconomic and human health threats. The increase in detected non-native (alien) and cryptogenic species, followed by the necessity for up-to-date scientific information on biological invasions, prompted a thorough literature investigation on the non-native entomofauna of Greece. Supplementing previous studies concerning Europe and Greece, a checklist for the non-native insect species of the country is provided, while their number is elevated from 266 to 469. Current trends regarding species richness and taxonomy, deliberate or accidental means of introduction, as well as their native range and trophic preferences are analysed and discussed. This publication illustrates the current state of non-native insects of Greece, highlighting the need for an effective early warning system integrated in national phytosanitary legislation, survey protocols and strategies towards the eradication and mitigation of their detrimental impacts.

Emerging base editing technology exploits CRISPR RNA-guided DNA modification effects for highly specific C > T conversion, which has been used to efficiently disrupt gene expression. These tools can enhance synthetic T cell immunity by restricting specificity, addressing histocompatibility leukocyte antigen (HLA) barriers, and promoting persistence. We report lentiviral delivery of a hepatitis B-virus (HBV)-specific recombinant T cell receptor (rTCR) and a linked CRISPR single-guide RNA for simultaneous disruption of endogenous TCRs (eTCRs) when combined with transient cytosine deamination. Discriminatory depletion of eTCR and coupled expression of rTCR resulted in enrichment of HBV-specific populations from 55% (SEM, ±2.4%) to 95% (SEM, ±0.5%). Intensity of rTCR expression increased 1.8- to 2.9-fold compared to that in cells retaining their competing eTCR, and increased cytokine production and killing of HBV antigen-expressing hepatoma cells in a 3D microfluidic model were exhibited. Molecular signatures confirmed that seamless conversion of C > T (G > A) had created a premature stop codon in TCR beta constant 1/2 loci, with no notable activity at predicted off-target sites. Thus, targeted disruption of eTCR by cytosine deamination and discriminatory enrichment of antigen-specific T cells offers the prospect of enhanced, more specific T cell therapies against HBV-associated hepatocellular carcinoma (HCC) as well as other viral and tumor antigens.

Considering the emerging challenges posed by the spread of COVID-19, this study was designed to evaluate citizens’ perceptions of the role of urban green areas in the era of COVID-19 in Greece. The evaluation was based on the implementation of an electronic questionnaire survey through the Google Forms platform, which was conducted nationwide. The survey was conducted in 2020 and 735 responses were collected in total based on 14 structured questions. Among the key findings of the study, of notable importance is that citizens considered urban green areas as an important means of improving public health, while citizens were willing to accept an increase of EUR1 to EUR20 in their municipal taxes for improving the services offered by the urban green areas. Results indicate that in a period of both climatic and public health crises, healthy and green urban environments can play a seminal role for alleviating and mitigating different challenges and impacts, while at the same time ensuring sustainability of urban ecosystems. A certain necessity arises for investigating the socioeconomic importance of urban green areas both from an ecosystemic and public health perspective considering the novel challenges of COVID-19 to public policy and decision making.

Key Clinical Message With few cases of PUNLMPs in young adults being reported in the literature, we hope to raise clinical awareness of prompt and effective diagnosis, while maintaining a high index of suspicion among health professionals. Even in the absence of red blood cells in the urine and subsequent negative imaging workup, clinicians should not delay performance of diagnostic cystoscopy.

Abstract Background Zinner syndrome represents a rare congenital malformation of the urinary tract. It comprises a constellation of Wolffian duct anomalies and is almost exclusively encountered as a classic triad of seminal vesicle cysts, ejaculatory duct obstruction and renal agenesis. Patients can be either asymptomatic or symptomatic. Recently, minimally invasive surgical techniques have emerged, superseding traditional surgery for select symptomatic cases. Our case highlights the finding of a rare clinical syndrome that was incidentally detected during a routine mass screening of military recruits in the Greek Armed Forces. Case presentation Herein, we present a case of a 19-year-old male who reported having a solitary right kidney when examined in a military training center of Northern Greece. No additional clinical information was available; thus, referral to a tertiary urology department for further investigation ensued. Imaging studies, namely, computed tomography and magnetic resonance imaging, revealed left renal aplasia, multiple left seminal vesicle cysts, and ejaculatory duct obstruction. Laboratory values and urinalysis were within normal range. Semen analysis was significant for cryptozoospermia. Our patient remained asymptomatic during the entire hospitalization. Long-term follow-up was recommended. Nevertheless, he declined further investigation and sought treatment in a private practice setting. Conclusions This article aims to present the incidental diagnosis of a rare syndrome in a military setting. Population screening conducted in the armed forces permits the identification of undiagnosed diseases that warrant further investigation. To the best of our knowledge, this was the first report of Zinner syndrome in a military recruit and the second case cited of a Greek patient in the published literature. Regular follow-up is the key to timely intervention in conservatively managed cases.

Hydrocele is the most common benign cause of painless scrotal enlargement and only very rarely can be reactive to an underlying testicular tumor. We present the case of a healthy young man, complaining of mild left scrotal discomfort and swelling. Physical examination revealed a non-tender fluctuant left scrotum and serum tumor markers were normal. Scrotal ultrasonography (US) showed a normal right hemiscrotum and testicle and a fluid collection among thickened irregular septations in the left hemiscrotum, a finding which was considered as a complex hydrocele. Intraoperatively the presumed “complex hydrocele” was in fact a multicystic testicular tumor. We proceeded with orchiectomy through the scrotal incision and pathology revealed a mixed germ cell tumor of the testis consisting of cystic teratoma, in situ germ cell neoplasia unclassified (IGCNU) and Sertoli cell tumor. This is the first reported case of this type of testis tumor presenting as complex hydrocele. The aim of this case presentation is to underline the need for an accurate preoperative diagnosis in cases of suspected scrotal pathology in young males.

Invasive alien species have been increasingly acknowledged as a major threat to native biodiversity and ecosystem services, while their adverse impacts expand to human health, society and the economy on a global scale. Insects represent one of the most numerous alien organismic groups, accounting for about one fifth of their total number. In Greece, a large number of alien insects have been identified, currently reaching 469 species. In recent decades, the contribution of citizen science towards detecting and mapping the distribution of alien insects has been steeply increasing. Addressing the need for up-to-date information on alien species as well as encouraging public participation in scientific research, the Alientoma website—derived from “alien” and the Greek word “entoma”, meaning insects, is presented. The website aims towards providing updated information on alien species of insects to the public as well as the scientific community, raising awareness about biological invasions and addressing their distribution and impacts inter alia. By maintaining a dynamic online database alongside a strong social media presence since its launch, Alientoma has attracted individuals mainly from Greece and Cyprus, interacting with the website through a total of 1512 sessions. Alientoma intends to establish a constantly increasing network of citizen scientists and to supplement early detection, monitoring and management efforts to mitigate the adverse impacts of alien insects in Greece.

Vesicouterine fistula (VUF) represents a rare urogenital complication. It is considered to be the least common type of urogenital fistulas. Iatrogenic reasons have been shown to be the most prominent cause, with lower segment caesarean section accounting for approximately two-thirds of the cases. The highest incidence concerns young females of reproductive age. VUF can present with clinical symptoms varying from cyclic hematuria, amenorrhea, and vaginal leakage of urine to secondary infertility and first-trimester abortion. Quality of life (QoL) for patients having this pathology is strongly affected due to the psychological burden. Surgical excision of the fistula remains the mainstay of treatment, as less than 5% of patients respond to conservative therapy. Recently laparoscopic and robotic-assisted VUF repair started gaining ground with comparable results to open surgery. Herein, we presented the successful delayed surgical repair of VUF in a 32-year-old female patient. A review of the published literature was also performed, summarizing all the available evidence regarding this rare clinical entity.

Transverse testicular ectopia (TTE) with fused vas deferens is an extremely rare clinical entity. Herein, we present a case of a 19-year-old patient with persistent left testicular pain lasting for a week. Clinical examination revealed an empty right hemiscrotum, a normal left-sided descended testis, and in close proximity a mass-like structure resembling testicular parenchyma. Laboratory tests were significant for elevated follicle-stimulating hormone (FSH), while sperm count revealed azoospermia. Ultrasound imaging (US) of the scrotum demonstrated the presence of both testes in the same left hemiscrotum with varicocele and no signs of inguinal hernia. Magnetic resonance imaging (MRI) of the penis and scrotum revealed TTE with a single, fused vas deferens, and hypoplastic seminal vesicles. Surgical intervention by means of microsurgical sperm retrieval and transseptal orchidopexy were considered but not performed, primarily owing to the patient’s unwillingness and to a lesser extent due to the restriction that the short and fused vas would pose in an attempt to transpose the ectopic testis. Therefore, an annual follow-up was recommended.
