Eph receptors and ligands are two families of proteins that control axonal guidance during development. Their expression was originally thought to be developmentally regulated but recent work has shown that several EphA receptors are expressed postnatally. The EphB3 receptors are expressed during embryonic development in multiple regions of the central nervous system but their potential expression and functional role in the adult brain is unknown. We used in situ hybridization, immunohistochemistry, and receptor affinity probe in situ staining to investigate EphB3 receptors mRNA, protein, and ligand (ephrin-B) expression, respectively, in the adult rat brain. Our results indicate that EphB3 receptor mRNA and protein are constitutively expressed in discrete regions of the adult rat brain including the cerebellum, raphe pallidus, hippocampus, entorhinal cortex, and both motor and sensory cortices. The spatial profile of EphB3 receptors was co-localized to regions of the brain that had a high level of EphB3 receptor binding ligands. Its expression pattern suggests that EphB3 may play a role in the maintenance of mature neuronal connections or re-arrangement of synaptic connections during late stages of development.

In this study, we examined the effects of nerve growth factor (NGF) administration on cholinergic enzyme activity in both normal and ethylcholine mustard aziridinium (AF64A)-treated rats. Choline acetyltransferase (ChAT) and acetylcholinesterase activity were measured in the hippocampus and septum of rats chronically administered NGF (0.36-2.85 micrograms/day) into the lateral ventricle for 14 days. In both normal and AF64A-treated rats, NGF increased cholinergic enzyme activity in a dose-dependent manner. Furthermore, although NGF increased ChAT activity in normal rats by 147%, it had a greater effect in AF64A-treated rats, increasing ChAT activity as much as 273%. NGF increased acetylcholinesterase activity in normal rats by only 125% but produced a 221% increase in this activity in AF64A-treated rats. These data indicate that AF64A produces an increased sensitivity to NGF in cholinergic neurons.

Neuronal survival during the developmental period of naturally occurring cell death is mediated through a successful competition for limiting concentrations of neurotrophic factors, and the deprived neurons will die. New results show that induced death through the p75 neurotrophin receptor (p75 NTR ), a member of the p55TNF/Fas family of cell death receptors, may also influence survival during development. We find that eliminating p75 NTR or neurotrophin 4 (NT4) in mice leads to a marked attenuation of apoptosis during the programmed cell death period of the trigeminal ganglion neurons, suggesting that NT4 can induce the death of these neurons through the p75 NTR . These in vivo findings were reproduced in primary cell cultures, where NT4 was found to induce death in a p75 NTR -dependent pathway. Analysis of p75 deficient and wild-type cells revealed two separate cell death pathways, a p75 NTR - and caspase-3-independent pathway activated by trophic factor deprivation, and a p75 NTR - and caspase-3-dependent pathway initiated by NT4. Crossing in the NT4 null alleles in brain-derived neurotrophic factor (BDNF) null mutant mice led to a rescue of a large proportion of BDNF-dependent neurons from excessive cell death, indicating that trophic factor deprivation is not sufficient for the death of many neurons and that additional death inducing signals might be required. Our results suggest that NT4 competitively signals survival and death of sensory neurons through trkB and p75 NTR , respectively.

There are two populations of neurons which are continually renewed in the adult, the dentate gyrus granule neurons and the olfactory bulb granule and periglomerular neurons. In the dentate gyrus, a secondary proliferative zone termed the subgranular zone is established along the interface between the dentate gyrus and the hilus where granule cells are born throughout life. Olfactory bulb neurons are generated in the anterior subventricular zone of the lateral ventricle and migrate via the rostral migratory stream to the olfactory bulb. We examined animals lacking brain-derived neurotrophic factor (BDNF) in order to establish whether this neurotrophin could be involved in the generation and/or survival of these neurons in vivo. We find that cells in nestin-positive regions of both the subgranular layer of the dentate gyrus and the subventricular zone of the olfactory bulb undergo apoptosis starting 2 weeks after birth in the absence of BDNF. However, increased apoptosis was not limited to precursors, as apoptotic cells were also found in the granule cell layer of the dentate gyrus and in the granule and periglomerular layers of the olfactory bulb. The excessive cell death was limited to these populations of neurons as no excessive cell death was detected in other forebrain areas. We conclude that BDNF is essential for the survival of neurons specifically in populations which are continuously being regenerated in the brain.

In man, acute ethanol administration decreases lower esophageal sphincter pressure (LESP), prolongs the duration, and lowers the amplitude of esophageal contractions. These changes may contribute to gastroesophageal reflux and esophagitis. To evaluate the underlying mechanisms of these changes an animal model is needed. Hence, we studied the effect of various doses of ethanol on esophageal motility in cats, an animal with an esophagus similar to man's. Similar to man, intravenous administration of ethanol significantly decreased LESP and amplitude of lower (smooth muscle portion) esophageal contractions. It also prolonged the duration of lower esophageal contractions, even through it had no effect on contraction velocity. The effect of ethanol on upper (striated muscle portion) esophagus was different. Ethanol had no effect on the amplitude of contractions, whereas it prolonged their duration and decreased their velocity. The effect of acute ethanol on LESP in four withdrawing alcoholic cats was similar to controls. However, the acute effect of ethanol on the esophageal contractions was less marked in alcoholics. Bilateral cervical vagotomy and intravenous injection of the neurotoxin tetrodotoxin before the administration of ethanol did not prevent the effect of ethanol on LESP. This data suggests that cat esophagus is a good model for studying the underlying mechanisms of the effects of acute ethanol because its response is similar to the esophagus of man, and the acute effect of ethanol on LESP is not neurally mediated but is the result of its direct effect on muscle.

There continues to be disagreement regarding the effects of ethanol on gastric emptying. This study was designed to assess different degrees of ethanol exposure on gastric emptying. Scintigraphic gastric emptying studies were performed in healthy adult cats and the half time (t 1/2) of gastric emptying was obtained from each study. Repeat measures of gastric emptying were again obtained in these cats under the conditions of both acute and chronic ethanol exposure and 24 hr following ethanol withdrawal. Gastric emptying half times were prolonged significantly during acute and chronic exposure to ethanol. The mean value for t 1/2 under control conditions was 46.46 min, while for acute and chronic exposure to ethanol the mean t 1/2 values were 69.34 and 91.33 min, respectively. In addition, we demonstrated a similar delay in gastric emptying during active withdrawal from alcohol (t 1/2=70). We conclude that both acute and chronic alcohol exposure significantly delay gastric emptying.

Spinal cord injury (SCI) releases a cascade of events that leads to the onset of an inhibitory milieu for axonal regeneration. Some of these changes result from the presence of repulsive factors that may restrict axonal outgrowth after trauma. The Eph receptor tyrosine kinase (RTK) family has emerged as a key repellent cue known to be involved in neurite outgrowth, synapse formation, and axonal pathfinding during development. Given the nonpermissive environment for axonal regeneration after SCI, we questioned whether re-expression of one of these molecules occurs during regenerative failure. We examined the expression profile of EphA3 at the mRNA and protein levels after SCI, using the NYU contusion model. There is a differential distribution of this molecule in the adult spinal cord and EphA3 showed an increase in expression after several injury models like optic nerve and brain injury. Standardized semi-quantitative RT-PCR analysis demonstrated a time-dependent change in EphA3 mRNA levels, without alterations in beta-actin levels. The basal level of EphA3 mRNA in the adult spinal cord is low and its expression was induced 2 days after trauma (the earliest time point analyzed) and this upregulation persisted for 28 days post-injury (the latest time point examined). These results were corroborated at the protein level by immunohistochemical analysis and the cell phenotype identified by double labeling studies. In control animals, EphA3 immunoreactivity was observed in motor neurons of the ventral horn but not in lesioned animals. In addition, GFAP-positive cells were visualized in the ventral region of injured white matter. These results suggest that upregulation of EphA3 in reactive astrocytes may contribute to the repulsive environment for neurite outgrowth and may be involved in the pathophysiology generated after SCI.

Eph receptors and ligands represent two families of proteins that control axonal guidance during development. Recent work has shown that several Eph receptors are expressed postnatally. Because the Eph molecules represent a class of axon guidance molecules that are mainly inhibitory to axonal growth, we investigated whether EphB3 expression was upregulated in both spinal cord and four supraspinal nuclei (locus coeruleus, vestibular, raphe pallidus, and red) 1 week after a complete spinal cord thoracic transection. Injured rats had a significant increase in EphB3 mRNA and protein expression in the spinal cord. The increased EphB3 expression was colocalized with GFAP staining and indicated that astrocytes play a role in EphB3 expression after spinal cord injury. No change in EphB3 expression was seen in supraspinal brain nuclei, which further demonstrated that changes in expression were due to changes in the local microenvironment at the injury site. The expression of EphB3 was colocalized to regions of the CNS that had a high level of EphB3 binding ligands. These data indicate upregulation of EphB3 expression after injury may also contribute to an environment in the spinal cord that is inhibitory to axonal regeneration.

After spinal cord injury (SCI), the inability of supraspinal neurons to regenerate or reform functional connections is likely due to proteins in the surrounding microenvironment restricting regeneration. EphAs are a family of receptor tyrosine kinases that are involved in axonal guidance during development. These receptors and their ligands, the Ephrins, act via repulsive mechanisms to guide growing axons towards their appropriate targets and allow for the correct developmental connections to be made. In the present study, we investigated whether EphA receptor expression changed after a thoracic contusion SCI. Our results indicate that several EphA molecules are upregulated after SCI. Using semiquantitative RT-PCR to investigate mRNA expression after SCI, we found that EphA3, A4, and A7 mRNAs were upregulated. EphA3, A4, A6, and A8 receptor immunoreactivity increased in the ventrolateral white matter (VWM) at the injury epicenter. EphA7 had the highest level of immunoreactivity in both control and injured rat spinal cord. EphA receptor expression in the white matter originated from glial cells as coexpression in both astrocytes and oligodendrocytes was observed. In contrast, gray matter expression was localized to neurons of the ventral gray matter (motor neurons) and dorsal horn. After SCI, specific EphA receptor subtypes are upregulated and these increases may create an environment that is unfavorable for neurite outgrowth and functional regeneration.

Spinal cord injury (SCI) in adult rats initiates a cascade of events producing a nonpermissive environment for axonal regeneration. This nonfavorable environment could be due to the expression of repulsive factors. The Eph receptor protein tyrosine kinases and their respective ligands (ephrins) are families of molecules that play a major role in axonal pathfinding and target recognition during central nervous system (CNS) development. Their mechanism of action is mediated by repellent forces between receptor and ligand. The possible role that these molecules play after CNS trauma is unknown. We hypothesized that an increase in the expression of Eph proteins and/or ephrins may be one of the molecular cues that restrict axonal regeneration after SCI. Rats received a contusive SCI at T10 and in situ hybridization studies 7 days posttrauma demonstrated: (i) a marked up-regulation of Eph B3 mRNA in cells located in the white matter at the lesion epicenter, but not rostral or caudal to the injury site, and (ii) an increase in Eph B3 mRNA in neurons in the ventral horn and intermediate zone of the gray matter, rostral and caudal to the lesion. Immunohistochemical analyses localizing Eph B3 protein were consistent with the mRNA results. Colocalization studies performed in injured animals demonstrated increased Eph B3 expression in white matter astrocytes and motor neurons of the gray matter. These results suggest that Eph B3 may contribute to the unfavorable environment for axonal regeneration after SCI.

In neurodegenerative diseases, such as Alzheimer’s and Parkinson’s disease, neurons in specific brain regions atrophy, become dysfunctional (e.g. reduced ability to produce neurotransmitters), and die. This cell death results in a loss of selective neuronal populations and the neurotransmitters that they synthesize and release. In Alzheimer’s disease, the loss of basal forebrain cholinergic neurons results in a reduction of acetylcholine and its synthesizing enzyme, choline acetyltransferase (ChAT) (Nagei et al., 1983). The loss of cholinergic neurons in the basal forebrain is thought to play a role in the memory and cognitive deficits in patients with Alzheimer’s disease (Hefti, 1994). Neurotrophic factors, which are known to interact with these neurons, have been proposed as a treatment for Alzheimer’s disease (Hefti, 1994).

We evaluated whether brain-derived neurotrophic factor (BDNF) can stimulate choline acetyltransferase (ChAT) activity in the septo-hippocampal pathway of ethylcholine mustard aziridinium (AF64A) and non-AF64A-treated rats. Rats received either AF64A (1.5 nmol/ ventricle) or a sham (non-AF64A-treated) injection into the lateral ventricles. BDNF infusion (at a dose range of 0.1-171 microg) into either the lateral ventricle or hippocampus of AF64A-treated rats for 14 days increased septal ChAT activity (26% and 41%, respectively). BDNF did not reverse the decrease in hippocampal ChAT activity (20-60%) produced by AF64A. BDNF infusion did not change ChAT activity in the septo-hippocampal pathway of non-AF64A-treated rats. Thus, septo-hippocampal cholinergic neurons in AF64A-treated rats are sensitive to BDNF while those of non-AF64A-treated rats (normal) are not responsive to this neurotrophin.

Abnormal esophageal motility has been reported in alcoholics. This includes abnormally high esophageal contraction amplitudes (ECA) and elevated lower esophageal sphincter pressures (LESP). However, an animal model is lacking for evaluating the underlying mechanisms. To develop such a model of ethanol-induced esophageal motor dysfunction, esophageal manometry was done in control, chronically intoxicated, and withdrawing cats. Ethanol was administered for at least 30 days via gastrostomy tube and blood alcohol levels, typically 300 mg/dl, were adjusted to maintain inebriation. LESP and upper (striated muscle) and lower (smooth muscle) ECA were elevated in male withdrawing cats. In intoxicated cats only upper ECA was elevated. Surprisingly, LESP and ECA were not significantly affected in females. Duration of ECA was prolonged in both male and female intoxicated cats. These findings indicate that: (1) similar to withdrawing men, ECA and LESP are elevated in withdrawing male cats; (2) ethanol-treated cats are an appropriate model for studying the mechanism of ethanol effects on esophageal motor function; (3) gender has a significant role in influencing the effects of alcohol.
